目的 通过观察不同毒力型的幽门螺旋杆菌(helicobacter pylori,Hp)感染对胃液、胃蛋白酶及炎症因子的影响。方法 收集2021年12月至2023年3月收治的110例患者,根据碳13呼气试验结果,分为Hp阳性组(n=79),Hp阴性组(n=31)。其中Hp阳性组根据... 目的 通过观察不同毒力型的幽门螺旋杆菌(helicobacter pylori,Hp)感染对胃液、胃蛋白酶及炎症因子的影响。方法 收集2021年12月至2023年3月收治的110例患者,根据碳13呼气试验结果,分为Hp阳性组(n=79),Hp阴性组(n=31)。其中Hp阳性组根据其Hp抗体分型,分为Ⅰ型组(n=52)、Ⅱ型组(n=11)、未定型组(n=16);Hp阴性组挑选抗体全阴者分为空白对照组(n=12)。所有患者均进行胃液pH值、钠离子(Na+)、钾离子(K+)、氯离子(Cl-)、白介素6 (Interleukin-6,IL-6)、白介素8 (Interleukin-8,IL-8)、胃泌素17 (Gastrin-17,G-17)、胃蛋白酶原Ⅰ(Pepsinogen I,PGⅠ)、胃蛋白酶原Ⅱ(Pepsinogen I,PGⅡ)检测。结果 Hp阳性组与Hp阴性组相比pH值、Na+、Cl-、K+,差异无统计学意义(P> 0.05);Hp阳性组相比Hp阴性组Cl-含量降低(P <0.05)。Hp阳性组较Hp阴性组相比IL-6、IL-8、G-17、PGⅠ、PGⅡ均有上升(P <0.05)。Ⅰ型组与Ⅱ型组、未定型组、空白对照组相比pH值、Na+、K+,差异均无统计学意义(P> 0.05)。Ⅰ型组、未定型组与空白对照组比较,Cl-含量均降低(P <0.05)。Ⅰ型组分别与Ⅱ型组、未定型组、空白对照组比较,IL-6、IL-8、PGⅠ均有上升(P <0.05)。空白对照组分别与其他各组之间PGⅡ存在明显差异(P <0.05)。Ⅰ型组与未定型组的G-17含量差异无统计学意义(P> 0.05)。Ⅰ型组与Ⅱ型组、空白对照组比较G-17均有上升(P <0.05)。结论 Ⅰ型Hp菌感染可能通过增加IL-6、IL-8、G-17的表达造成胃黏膜损伤,进而导致消化功能异常。 展开更多

Argatroban promotes recovery of spinal cord injury by inhibiting the PAR1/JAK2/STAT3 signaling pathway

Argatroban is a synthetic thrombin inhibitor approved by U.S.Food and Drug Administration for the treatment of thrombosis.However,whether it plays a role in the repair of spinal cord injury is unknown.In this study,we established a rat model of T10 moderate spinal cord injury using an NYU Impactor ModerⅢand performed intraperitoneal injection of argatroban for 3 consecutive days.Our results showed that argatroban effectively promoted neurological function recovery after spinal cord injury and decreased thrombin expression and activity in the local injured spinal cord.RNA sequencing transcriptomic analysis revealed that the differentially expressed genes in the argatroban-treated group were enriched in the JAK2/STAT3 pathway,which is involved in astrogliosis and glial scar formation.Western blotting and immunofluorescence results showed that argatroban downregulated the expression of the thrombin receptor PAR1 in the injured spinal cord and the JAK2/STAT3 signal pathway.Argatroban also inhibited the activation and proliferation of astrocytes and reduced glial scar formation in the spinal cord.Taken together,these findings suggest that argatroban may inhibit astrogliosis by inhibiting the thrombin-mediated PAR1/JAK2/STAT3 signal pathway,thereby promoting the recovery of neurological function after spinal cord injury. 展开更多

Maintaining the integrity of the blood-spinal cord barrier is critical for the recove ry of spinal cord injury.Ferro ptosis contributes to the pathogenesis of spinal cord injury.We hypothesized that ferroptosis is involved in disruption of the blood-s pinal cord barrier.In this study,we administe red the ferroptosis inhibitor liproxstatin-1 intraperitoneally after contusive spinal co rd injury in rats.Liproxstatin-1 improved locomotor recovery and somatosensory evoked potential electrophysiological performance after spinal cord inju ry.Liproxstatin-1 maintained blood-spinal cord barrier integrity by upregulation of the expression of tight junction protein.Liproxstatin-1 inhibited ferroptosis of endothelial cell after spinal cord injury,as shown by the immunofluorescence of an endothelial cell marker(rat endothelium cell antigen-1,RECA-1) and fe rroptosis markers Acyl-CoA synthetase long-chain family member 4 and 15-lipoxygenase.Liproxstatin-1reduced brain endothelial cell ferroptosis in vitro by upregulating glutathione peroxidase 4 and downregulating Acyl-CoA synthetase long-chain family member4 and 15-lipoxygenase.Furthermore,inflammatory cell recruitment and astrogliosis were mitigated after liproxstatin-1 treatment.In summary,liproxstatin-1im proved spinal cord injury recovery by inhibiting ferroptosis in endothelial cells and maintaining blood-s pinal co rd barrier integrity. 展开更多

Compared with the control,yield per mu increases by 5. 08- 18. 86%; N content increases by 4. 27%-11. 28%; P content increases by 2. 13%- 13. 57%; K content increases by 2. 00%- 18. 48%; crude protein increases by 4. 16%-19. 94%; vitamin C increases by 2. 71%- 11. 79%; nitrate content decreases by 5%- 15%. A novel anchorage for long-span suspension bridges,called pile-caisson composite structures,was recently proposed by the authors in an attempt to reduce the construction period and costs.This study aims to investigate the displacement and force behavior of piles in a pile-caisson composite structure under eccentric inclined loading considering different stratum features.To this end,both 1g model tests and three-dimensional numerical simulations were performed.Two groups of 1g model tests were used to validate the finite-element(FE)method.Parametric studies were then performed to investigate the effects of groundwater level,burial depth of the pile-caisson composite structure,and distribution of soil layers on the performance of the pile-caisson composite structure.The numerical analyses indicated that the influence of the groundwater level on the stability of the caisson was much greater than that of the piles.In addition,increasing the burial depth of the pile-caisson composite structure can assist in reducing the displacements and improving the stability of the pile-caisson composite structure.In addition,the distribution of soil layers can significantly affect the stability of the pile-caisson composite structure,especially the soil layer around the caisson. 展开更多

We previously identified that hepatitis B virus(HBV)encodes a microRNA(HBV-miR-3)that restrains HBV replication by targeting the HBV transcript.However,whether HBV-miR-3 affects host innate immunity to modulate HBV replication remains unclear.Here,we examined the vital functions of HBV-miR-3 in the innate immune response after HBV infection.We found that HBV-miR-3 expression gradually increased in a dose-and time-dependent manner in HBV-infected HepG2-NTCP cells.HBV-miR-3 activated the JAK/STAT signaling pathway by downregulating SOCS5 in hepatocytes,thereby enhancing the IFN-induced anti-HBV effect.In addition,HBVmiR-3 in exosomes facilitated the Ml polarization of macrophages.Furthermore,exosomes containing HBV-miR-3 enhanced the secretion of IL-6 via inhibiting the SOCS5-mediated ubiquitination of EGFR.In short,these results demonstrate that HBV-miR-3 activates the innate immune response to restrain HBV replication by multiple pathways,which may suppress HBV-induced acute liver cell injury and affect the progression of persistent HBV infection. 展开更多

To describe the distribution of sandstone reservoirs between wells finely,this paper takes the sandstone reservoir of Layer Nm II-4 in Qinhuangdao 32-6 Oilfield for example,and uses the seismic forward simulation and well loggings to study the channel sandstone reservoir. Under the guidance of the modern rivers and outcrops sandstone distribution pattern,the predicting limitations of the seismic forward simulation about channel sandstone boundaries such as mudstone interlayer between channel sandstone and elevation distance between adjacent channel sandstone are clear. The research shows that the mudstone interlayer can be described by seismic forward simulation seismic when the mudstone interlayer is thicker than 2 m and the channel sandstone is thicker than 10 m because of the appearance of the seismic peak when the main frequency of seismic data is nearly 60 Hz. And the elevation distance between adjacent channel sandstone can be described by the seismic forward simulation when elevation distance is longer than 6 m and the channel sandstone is thicker than 10 m because of the appearance of the seismic peak. And the seismic waveform features of single channel sandstone boundaries such as elevation distance between channels,overbank sandstone and abandoned channels. Under the study mentioned above,the single channel boundaries are described in Layer Nm II-4 of Qinhuangdao 32-6 Oilfield,and the predicting boundaries are confirmed by the chemical tracers. The results show that the channel sandstone reservoir architecture characterization can be improved by the seismic forward simulation. 展开更多
