Objective:To observe the effect and possible mechanism of action of Bushen Bitong recipe(BSBT)containing serum on IL-1β-induced chondrocyte apoptosis.Methods:Generation 3 rat chondrocytes were randomized into Control,IL-1β,IL-1β+BSBT(L),IL-1β+BSBT(M),and IL-1β+BSBT(H)groups(5%,10%and 15%BSBT-containing serum),and then 24h after intervention respectively,the cell proliferation and Apoptosis rate;Western blot detected the expression levels of Bcl-2,BAX,Caspase-3,SOX9,NF-κB p65,MMP-13 proteins in chondrocytes.ELISA detected the levels of TNF-α,IL-6,and bFGF in the supernatants of chondrocyte culture.Results:Compared with Control group,cell proliferation activity decreased,apoptosis rate increased,NF-κB p65,MMP-13 protein level and TNF-α,IL-6 level increased,and SOX9 protein level and bFGF level decreased in IL-1βgroup;compared with IL-1βgroup,different concentrations of BSBT-containing serum group,cell proliferation activity increased,and apoptosis rate decreased.NF-κB p65,MMP-13 protein level and TNF-α,IL-6 level decreased,SOX9 protein level and bFGF level increased;compared with IL-1β+BSBT(L)group,cell proliferation activity increased,apoptosis rate decreased in IL-1β+BSBT(M)and IL-1β+BSBT(H)groups,and NF-κB p65,MMP-13 protein level and TNF-αlevel decreased.13 protein levels and TNF-αand IL-6 levels decreased,and SOX9 protein levels and bFGF levels increased.Conclusion:BSBT-containing serum may promote IL-1β-induced proliferation of chondrocytes,reduce apoptosis,improve the microenvironment of chondrocytes,and promote cartilage repair through the SOX9/NF-κB/MMP-13 signaling pathway. 展开更多

四海舒郁丸介导miRNA-494对甲状腺乳头状癌细胞的干预作用

【目的】探讨四海舒郁丸含药血清对甲状腺乳头状癌细胞miRNA-494及靶基因PTEN和相关通路PI3K/AKT的干预机制。【方法】将2月龄SPF级雄性SD大鼠随机分为四海舒郁丸低、中、高剂量组与空白组,每组5只,灌胃给药后制备四海舒郁丸含药血清和... 【目的】探讨四海舒郁丸含药血清对甲状腺乳头状癌细胞miRNA-494及靶基因PTEN和相关通路PI3K/AKT的干预机制。【方法】将2月龄SPF级雄性SD大鼠随机分为四海舒郁丸低、中、高剂量组与空白组,每组5只,灌胃给药后制备四海舒郁丸含药血清和空白组血清(等量生理盐水)并作用于体外培养的细胞。试验组甲状腺乳头状癌细胞选用KTC-1细胞株,分为空白组(2 mL/(100 g·d)生理盐水),低、中、高剂量组(分别为0.24、0.48、0.96 g/(100 g·d)),正常组甲状腺组织细胞选用Nthy-ori-3-1。通过CCK-8探究细胞的活力的影响;RTqPCR检测miRNA-494-5p及PTEN的mRNA表达水平,Western Blot法检测PTEN及下游PI3K/AKT通路蛋白表达水平。【结果】四海舒郁丸能抑制KTC-1细胞的活性,且作用随浓度的增加而增强(P<0.01),但无时间依赖性;相比较于KTC-1空白组,四海舒郁丸低、中、高剂量组中miRNA-494-5p的mRNA表达均降低(P<0.01),PTEN的mRNA表达量增加,其中高剂量组差异有统计学意义(P<0.05);高、中、低剂量组中p-PI3K/p-AKT信号通路蛋白表达均明显下降(P<0.05),而PTEN蛋白表达均有所增加(P<0.01)。【结论】四海舒郁丸含药血清通过下调miRNA-494-5p后负向调控PTEN进而阻断PI3K/AKT通路对PTC发挥辅助抑癌作用。 展开更多

Development and validation of a prediction model for early screening of people at high risk for colorectal cancer 被引量:2

BACKGROUND Colorectal cancer(CRC)is a serious threat worldwide.Although early screening is suggested to be the most effective method to prevent and control CRC,the current situation of early screening for CRC is still not optimistic.In China,the incidence of CRC in the Yangtze River Delta region is increasing dramatically,but few studies have been conducted.Therefore,it is necessary to develop a simple and efficient early screening model for CRC.AIM To develop and validate an early-screening nomogram model to identify individuals at high risk of CRC.METHODS Data of 64448 participants obtained from Ningbo Hospital,China between 2014 and 2017 were retrospectively analyzed.The cohort comprised 64448 individuals,of which,530 were excluded due to missing or incorrect data.Of 63918,7607(11.9%)individuals were considered to be high risk for CRC,and 56311(88.1%)were not.The participants were randomly allocated to a training set(44743)or validation set(19175).The discriminatory ability,predictive accuracy,and clinical utility of the model were evaluated by constructing and analyzing receiver operating characteristic(ROC)curves and calibration curves and by decision curve analysis.Finally,the model was validated internally using a bootstrap resampling technique.RESULTS Seven variables,including demographic,lifestyle,and family history information,were examined.Multifactorial logistic regression analysis revealed that age[odds ratio(OR):1.03,95%confidence interval(CI):1.02-1.03,P<0.001],body mass index(BMI)(OR:1.07,95%CI:1.06-1.08,P<0.001),waist circumference(WC)(OR:1.03,95%CI:1.02-1.03 P<0.001),lifestyle(OR:0.45,95%CI:0.42-0.48,P<0.001),and family history(OR:4.28,95%CI:4.04-4.54,P<0.001)were the most significant predictors of high-risk CRC.Healthy lifestyle was a protective factor,whereas family history was the most significant risk factor.The area under the curve was 0.734(95%CI:0.723-0.745)for the final validation set ROC curve and 0.735(95%CI:0.728-0.742)for the training set ROC curve.The calibration curve demonstrated a high correlation between the CRC high-risk population predicted by the nomogram model and the actual CRC high-risk population.CONCLUSION The early-screening nomogram model for CRC prediction in high-risk populations developed in this study based on age,BMI,WC,lifestyle,and family history exhibited high accuracy. 展开更多

In order to enhance the accuracy of Air Traffic Control(ATC)cybersecurity attack detection,in this paper,a new clustering detection method is designed for air traffic control network security attacks.The feature set for ATC cybersecurity attacks is constructed by setting the feature states,adding recursive features,and determining the feature criticality.The expected information gain and entropy of the feature data are computed to determine the information gain of the feature data and reduce the interference of similar feature data.An autoencoder is introduced into the AI(artificial intelligence)algorithm to encode and decode the characteristics of ATC network security attack behavior to reduce the dimensionality of the ATC network security attack behavior data.Based on the above processing,an unsupervised learning algorithm for clustering detection of ATC network security attacks is designed.First,determine the distance between the clustering clusters of ATC network security attack behavior characteristics,calculate the clustering threshold,and construct the initial clustering center.Then,the new average value of all feature objects in each cluster is recalculated as the new cluster center.Second,it traverses all objects in a cluster of ATC network security attack behavior feature data.Finally,the cluster detection of ATC network security attack behavior is completed by the computation of objective functions.The experiment took three groups of experimental attack behavior data sets as the test object,and took the detection rate,false detection rate and recall rate as the test indicators,and selected three similar methods for comparative test.The experimental results show that the detection rate of this method is about 98%,the false positive rate is below 1%,and the recall rate is above 97%.Research shows that this method can improve the detection performance of security attacks in air traffic control network. 展开更多

With the popularization of the Internet and the development of technology,cyber threats are increasing day by day.Threats such as malware,hacking,and data breaches have had a serious impact on cybersecurity.The network security environment in the era of big data presents the characteristics of large amounts of data,high diversity,and high real-time requirements.Traditional security defense methods and tools have been unable to cope with the complex and changing network security threats.This paper proposes a machine-learning security defense algorithm based on metadata association features.Emphasize control over unauthorized users through privacy,integrity,and availability.The user model is established and the mapping between the user model and the metadata of the data source is generated.By analyzing the user model and its corresponding mapping relationship,the query of the user model can be decomposed into the query of various heterogeneous data sources,and the integration of heterogeneous data sources based on the metadata association characteristics can be realized.Define and classify customer information,automatically identify and perceive sensitive data,build a behavior audit and analysis platform,analyze user behavior trajectories,and complete the construction of a machine learning customer information security defense system.The experimental results show that when the data volume is 5×103 bit,the data storage integrity of the proposed method is 92%.The data accuracy is 98%,and the success rate of data intrusion is only 2.6%.It can be concluded that the data storage method in this paper is safe,the data accuracy is always at a high level,and the data disaster recovery performance is good.This method can effectively resist data intrusion and has high air traffic control security.It can not only detect all viruses in user data storage,but also realize integrated virus processing,and further optimize the security defense effect of user big data. 展开更多

【目的】克隆白来航鸡干扰素基因刺激因子基因(stimulator of interferon genes,STING)CDS区序列并进行生物信息学和组织表达分析,为阐明STING基因在抗病毒免疫应答中的作用奠定基础。【方法】采用PCR扩增并克隆白来航鸡STING基因CDS区... 【目的】克隆白来航鸡干扰素基因刺激因子基因(stimulator of interferon genes,STING)CDS区序列并进行生物信息学和组织表达分析,为阐明STING基因在抗病毒免疫应答中的作用奠定基础。【方法】采用PCR扩增并克隆白来航鸡STING基因CDS区,测序后对其编码氨基酸序列进行相似性比对及系统进化树构建,利用生物信息学预测STING蛋白的理化特性及结构功能,并利用实时荧光定量PCR技术检测STING基因在鸡心脏、肝脏等14个组织中的表达情况。【结果】白来航鸡STING基因CDS区序列全长1140 bp,编码379个氨基酸。相似性比对和系统进化树分析结果表明,白来航鸡STING基因与原鸡的相似性最高(99.7%),亲缘关系最近,与冠小嘴乌鸦亲缘关系最远。STING蛋白为酸性、亲水性蛋白,分子质量为42.625 ku,等电点(pI)为6.67,不稳定系数为69.26,脂肪系数为105.01。该蛋白大部分在线粒体和内质网上合成,含有跨膜结构,不含信号肽。STING蛋白二级结构包括α-螺旋(54.62%)、延伸链(10.29%)、β-转角(3.43%)及无规则卷曲(31.66%)。蛋白互作分析表明,白来航鸡STING蛋白与NFKB1、DDX41、cGAS、TBK1等蛋白存在相互作用。实时荧光定量PCR结果显示,STING基因在白来航鸡组织中广泛表达,其中在肺脏中表达量最高,且显著高于其他组织(P<0.05);在胸肌中表达量最低。【结论】本研究成功克隆了白来航鸡STING基因,其CDS区序列全长1140 bp,编码379个氨基酸。白来航鸡STING蛋白为酸性、亲水性蛋白,含有跨膜结构。STING基因在白来航鸡肺脏中表达量最高。研究结果为深入探究白来航鸡STING基因编码蛋白功能提供了材料。 展开更多

研究多目标约束下的智能路侧设施优化布设方法,构建设备成本、平均可靠性、平均管控效果等多目标约束的优化框架,并引入遗传算法结合AHP层次分析法等评价方法,提出多目标约束的智能路侧设施优化布设模型。其次,选用多品牌、多种类智能... 研究多目标约束下的智能路侧设施优化布设方法,构建设备成本、平均可靠性、平均管控效果等多目标约束的优化框架,并引入遗传算法结合AHP层次分析法等评价方法,提出多目标约束的智能路侧设施优化布设模型。其次,选用多品牌、多种类智能路侧设施开展案例分析,以遗传算法求解最优解。为验证模型灵敏度,本文还通过灵敏度分析验证了所设计的模型在侧重某一指标的应用效果:相较于专家给定的α、β、γ(平均管控效果、设备成本、平均可靠性对应权重值),侧重平均可靠性指标提升6.67%,侧重设备成本指标降低了27.82%,侧重平均管控效果指标有一定提升。实验结果表明本文模型在面向多品牌、多种类的智能路侧设施选型布设和实际动态需求具有可行性。 展开更多

Objective:To evaluate the diagnostic value of fluorescence in situ hybridization(FISH)in bladder cancer.Methods:We enrolled healthy volunteers and patients who were clinically suspected to have bladder cancer and conducted FISH tests and cytology examinations from August 2007 to December 2008.Receiver operating characteristic(ROC)curve analysis was performed and the area under curve(AUC)values were calculated for both the FISH and urine cytology tests.Results:A cohort of 988 healthy volunteers was enrolled to establish a reference range for the normal population.A total of 4807 patients with hematuria were prospectively,randomly enrolled for the simultaneous analysis of urine cytology,FISH testing,and a final diagnosis as determined by the pathologic findings of a biopsy or a surgically-excised specimen.Overall,the sensitivity of FISH in detecting transitional-cell carcinoma was 82.7%,while that of cytology was 33.4%(p<0.001).The sensitivity values of FISH for non-muscle invasive and muscle invasive bladder transitional-cell carcinoma were 81.7%and 89.6%,respectively(p=0.004).The sensitivity values of FISH for low and high grade bladder cancer were 82.6%and 90.1%,respectively(p=0.002).Conclusion:FISH is significantly more sensitive than voided urine cytology for detecting bladder cancer in patients evaluated for gross hematuria at all cancer grades and stages.Higher sensitivity using FISH was obtained in high grade and muscle invasive tumors. 展开更多

The spatial distribution of forest biomass is closely related with carbon cycle, climate change, forest productivity, and biodiversity. Efficient quantification of biomass provides important information about forest quality and health. With the rising awareness of sustainable development, the ecological benefits of forest biomass attract more attention compared to traditional wood supply function. In this study, two nonparametric modeling approaches, random forest(RF) and support vector machine were adopted to estimate above ground biomass(AGB) using widely used Landsat imagery in the region,especially within the ecological forest of Fuyang District in Zhejiang Province, China. Correlation analysis was accomplished and model parameters were optimized during the modeling process. As a result, the best performance modeling method RF was implemented to produce an AGB estimation map. The predicted map of AGB in the study area showed obvious spatial variability and demonstrated that within the current ecological forest zone, as well as the protected areas, the average of AGB were higher than the ordinary forest. The quantification of AGB was proven to have a close relationship with the local forest policy and management pattern, which indicated that combining remote-sensing imagery and forest biophysical property would provide considerable guidance for making beneficial decisions. 展开更多

Collisions of spatial solitons occurring in the nonlinear Schroeinger equation with harmonic potential are studied, using conservation laws and the split-step Fourier method. We find an analytical solution for the separation distance between the spatial solitons in an inhomogeneous nonlinear medium when the light beam is self-trapped in the transverse dimension. In the self-focusing nonlinear media the spatial solitons can be transmitted stably, and the interaction between spatial solitons is enhanced due to the linear focusing effect (and also diminished for the linear defocusing effect). In the self-defocusing nonlinear media, in the absence of self-trapping or in the presence of linear self-defocusing, no transmission of stable spatial solitons is possible. However, in such media the linear focusing effect can be exactly compensated, and the spatial solitons can propagate through. 展开更多

目的:探究鲢鱼、青鱼内源性转谷氨酰胺酶(transglutaminase,TGase)的酶学性质差异。方法:采用80%(NH_(4))_(2)SO_(4)盐析、Q-Sepharose FF和Sephacryl S-200 HR层析法从鲢鱼、青鱼肌肉中分离纯化出鲢鱼TGase(STG)、青鱼TGase(BTG),并对... 目的:探究鲢鱼、青鱼内源性转谷氨酰胺酶(transglutaminase,TGase)的酶学性质差异。方法:采用80%(NH_(4))_(2)SO_(4)盐析、Q-Sepharose FF和Sephacryl S-200 HR层析法从鲢鱼、青鱼肌肉中分离纯化出鲢鱼TGase(STG)、青鱼TGase(BTG),并对酶的相对分子质量、肽段序列、二级结构、适宜反应条件、热失活动力学等指标进行测定。结果:纯化后的STG和BTG的比酶活分别为14.34,12.67 U/mg,二者具有相近的相对分子质量。二者的肽段序列存在一定差异,其二级结构均以β-折叠为主,但STG的β-折叠含量略高于BTG的。STG和BTG的适宜反应温度均为50℃,适宜反应pH分别为8.0,7.5,完全激活两种TGase活性所需的Ca^(2+)浓度均为1 mmol/L,DTT可使两种TGase的酶活性增强,而PMSF、NH_(4)Cl、NEM、EDTA、Cu^(2+)、Ba^(2+)、Zn^(2+)、Mg^(2+)则会抑制其酶活。当温度为37~50℃时,热处理对STG和BTG的钝化均符合一级指数衰减动力学,二者的动力学参数E a值相近。结论:STG和BTG的一级结构及二级结构差异明显,但仍具有相似的适宜反应条件和热失活动力学特征。 展开更多

为确定复合调理狮子头的最优加工工艺,该研究以猪肉/鱼肉复合调理狮子头为研究对象,研究搅打时间、成型温度、成型时间对复合调理狮子头色度、质构特性、持水性、感官品质的影响,并运用响应面分析法优化复合调理狮子头的制作工艺。结果... 为确定复合调理狮子头的最优加工工艺,该研究以猪肉/鱼肉复合调理狮子头为研究对象,研究搅打时间、成型温度、成型时间对复合调理狮子头色度、质构特性、持水性、感官品质的影响,并运用响应面分析法优化复合调理狮子头的制作工艺。结果表明,搅打时间、成型温度、成型时间对复合调理狮子头的品质有显著影响(P<0.05)。随着搅打时间的延长,产品的破断力、破断距离、持水性先升高后降低,并在搅打8 min时达到最大值。随着成型温度的上升,破断距离、凝胶强度及硬度总体上呈先升高后降低的趋势,持水性、感官评分随温度的上升先逐渐增加,在温度超过90℃后基本保持不变。随成型时间的延长,产品的破断距离、凝胶强度及硬度、内聚性、咀嚼性先升高后降低,持水性、感官评分先升高后无显著变化,且在16 min时达到最大值。经响应面优化后,复合调理狮子头的最优加工工艺参数为:搅打时间8 min、成型温度91℃、成型时间17 min,该条件下产品质地优良,适口性好,风味独特。 展开更多

目的运用网络药理学和实验验证探讨补肾痹通方治疗膝骨关节炎(KOA)的作用机制。方法运用中药系统药理学数据库与分析平台获取补肾痹通方的有效成分与作用靶点,运用GeneCards、OMIM、PharmGkb、TTD、DrugBank数据库检索KOA疾病相关靶点,... 目的运用网络药理学和实验验证探讨补肾痹通方治疗膝骨关节炎(KOA)的作用机制。方法运用中药系统药理学数据库与分析平台获取补肾痹通方的有效成分与作用靶点,运用GeneCards、OMIM、PharmGkb、TTD、DrugBank数据库检索KOA疾病相关靶点,将药物和疾病靶点取交集,将获得的有效成分及相关靶点导入Cytoscape软件,绘制“补肾痹通方-有效成分-KOA作用靶点”的网络图,筛选关键有效成分,将潜在靶点导入STRING数据库绘制蛋白质-蛋白质相互作用网络图。筛选补肾痹通方治疗KOA的核心靶点,进行GO富集与KEGG注释分析。选取人软骨细胞设置对照组、损伤组、给药组,对照组正常培养,损伤组和给药组在含白细胞介素(IL)-1β(10 ng/ml)DMEM/F12培养基中培养,给药组给予0.4447μg/ml补肾痹通方药液。培养24 h后,观察三组细胞形态,采用酶联免疫吸附试验检测炎症因子水平,qRT-PCR与免疫印迹法检测三组软骨修复因子的表达。结果共筛选出补肾痹通方有效成分186个,主要为槲皮素、木犀草素、山柰酚等,主要通过TNF、MMP1、CXCL2、IL1B等关键靶点和PI3K/Akt、T细胞受体、Toll样受体、低氧诱导因子-1、TNF、IL-17等信号通路发挥作用,涉及氧化应激反应、细胞凋亡、炎症反应等生物过程。显微镜下可见,损伤组细胞数量减少,给药组细胞数量增加。损伤组肿瘤坏死因子-α(TNF-α)、IL-1β、IL-6水平高于对照组,转化生长因子-β(TGF-β)、胰岛素样生长因子-1(IGF-1)、碱性成纤维细胞生长因子(BFGF)水平及SOX9、COL2A1基因和蛋白的表达水平均低于对照组(P<0.05)。给药组TNF-α、IL-1β、IL-6水平低于损伤组,TGF-β、IGF-1、BFGF及SOX9、COL2A1基因和蛋白的表达水平均高于损伤组(P<0.05)。结论补肾痹通方可能通过缓解炎症细胞浸润并参与SOX9、COL2A1调控来促进损伤软骨细胞修复。 展开更多

目的:通过代谢组学和网络药理学探究豨芍方(Xi Shao Formula,XSF)治疗神经性疼痛(neuropathic pain,NP)的机制。方法:建立NP的坐骨神经慢性压缩损伤(chronic compression injury,CCI)模型。造模后,分别给予生理盐水、XSF或阳性对照药物... 目的:通过代谢组学和网络药理学探究豨芍方(Xi Shao Formula,XSF)治疗神经性疼痛(neuropathic pain,NP)的机制。方法:建立NP的坐骨神经慢性压缩损伤(chronic compression injury,CCI)模型。造模后,分别给予生理盐水、XSF或阳性对照药物普瑞巴林灌胃治疗21天。通过代谢组学分析,明确XSF在体内的主要血清代谢产物和代谢途径。通过网络药理学和液相色谱–质谱仪分析,探究XSF治疗NP的主要活性成分和可能作用靶点。在此基础上,进一步整合代谢组学和网络药理学研究结果,使用Cytoscape 3.9.1绘制XSF治疗NP的“靶点–代谢途径–代谢物”网络关系图,并通过分子对接进行结果验证。结果:通过代谢组学分析,共获得XSF治疗NP中具有显著变化的差异代谢物11个,并通过功能富集分析获得了8条相关代谢通路。进一步通过联合代谢组学和网络药理学研究发现,“单胺氧化酶A(MAOA)/单胺氧化酶B(MAOB)/酪氨酸酶(TYR)–酪氨酸代谢途径–龙胆酸钠”可能是XSF治疗NP的重要网络途径。此外,分子对接结果显示XSF的活性成分与MAOA、MAOB和TYR(结合能<-5 kcal/mol)之间均具有高亲和力且相互作用稳定。结论:我们的发现进一步明确了XSF治疗NP的镇痛药效及作用机制,为XSF的临床应用和机制研究提供了科学基础。 展开更多
