文摘Gibberellin(GA)is one of the major plant hormones that promote parthenocarpy,a highly valuable agronomic trait. Here,we demonstrated that exogenous GA3application triggered the formation of parthenocarpic fruits with smaller size but unchanged shape in tomato(Solanum lycopersicum). These fruits exhibited a thicker pericarp, undeveloped ovules, and few jelly tissues, leading to smaller locules with empty cavities.Histological investigation showed that GA treatment produced more cell layers with larger cells in the pericarp, suggesting its promotion in both cell division and expansion.Transcriptomic analyses between GA-3and mock-treated unpollinated ovaries/fruits identified a large number of differentially expressed genes related to hormones, cell division, cell expansion, and transcription factors, implying that they coordinately regulated parthenocarpy conferred by GA.In particular, the downregulation of five reported repressors of tomato parthenocarpy, including two auxin signaling components,AUXIN RESPONSE FACTOR5(SlARF5)and Sl ARF7,and three MADS-box genes,TOMATO APETALA3(TAP3),TOMATO PISTILLATA(TPI),and AGAMOUS-LIKE6(SlAGL6),after GA treatment might play a key role in this process.Furthermore,we found that the knockdown of a GA signaling factor SlMYB33,which was depressed by GA treatment, induced parthenocarpic fruit set in tomato,an effect that might have been achieved by enhancing GA biosynthesis and decreasing the expression of some repressors of tomato parthenocarpy. Thus, our results provide a basis for understanding the regulatory mechanism of GA in tomato parthenocarpy.

文摘Background:As reported,γ-tubulin(TuBG1)is related to the occurrence and development of various types of malignant tumors. However, its role in hepatocellular cancer(HCC)is not clear. The present study was to investigate the relationship between TuBG1 and clinical parameters and survival in HCC patients. Methods:The correlation between TuBG1 and clinical parameters and survival in HCC patients was ex-plored by bioinformatics analysis.Immunohistochemistry was used for the verification. The molecular function of TuBG1 was measured using colony formation, scratch assay,trans-well assay and flow cytometry.Gene set enrichment analysis(GSEA)was used to pick up the enriched pathways, followed by investigating the target pathways using Western blotting. The tumor-immune system interactions and drug bank database(TISIDB)was used to evaluate TuBG1 and immunity.Based on the TuBG1-related immune genes,a prognostic model was constructed and was further validated internally and externally. Results:The bioinformatic analysis found high expressed TuBG1 in HCC tissue, which was confirmed us-ing immunohistochemistry and Western blotting. After silencing the TuBG1 in HCC cell lines, more G1 arrested cells were found, cell proliferation and invasion were inhibited, and apoptosis was promoted.Furthermore, the silence of TuBG1 increased the expressions of Ataxia-Telangiectasia and Rad-3(ATR),phospho-P38 mitogen-activated protein kinase(P-P38MAPK),phospho-P53(P-P53),B-cell lymphoma-2 associated X protein(Bax),cleaved caspase 3 and P21;decreased the expressions of B-cell lymphoma-2(Bcl-2),cyclin D1,cyclin E2,cyclin-dependent kinase 2(CDK2)and CDK4.The correlation analysis of immunohistochemistry and clinical parameters and survival data revealed that TuBG1 was negatively corre-lated with the overall survival. The constructed immune prognosis model could effectively evaluate the prognosis. Conclusions:The increased expression of TuBG1 in HCC is associated with poor prognosis, which might be involved in the occurrence and development of HCC.

文摘Electromagnetic wave(EMW)absorbing materials have an irreplaceable position in the field of military stealth as well as in the field of electromagnetic pollution control. And in order to cope with the complex electromagnetic environment, the design of multifunctional and multiband high efficiency EMW absorbers remains a tremendous challenge. In this work,we designed a three-dimensional porous structure via the salt melt synthesis strategy to optimize the impedance matching of the absorber. Also, through interfacial engineering,a molybdenum carbide transition layer was introduced between the molybdenum selenide nanoparticles and the three-dimensional porous carbon matrix to improve the absorption behavior of the absorber. The analysis indicates that the number and components of the heterogeneous interfaces have a significant impact on the EMW absorption performance of the absorber due to mechanisms such as interfacial polarization and conduction loss introduced by interfacial engineering.Wherein, the prepared MoSe_(2)/MoC/PNC composites showed excellent EMW absorption performance in C,X,and Ku bands, especially exhibiting a reflection loss of−59.09 dB and an effective absorption bandwidth of 6.96 GHz at 1.9 mm.The coordination between structure and components endows the absorber with strong absorption, broad bandwidth, thin thickness, and multi-frequency absorption characteristics.Remarkably,it can effectively reinforce the marine anticorrosion property of the epoxy resin coating on Q235 steel substrate. This study contributes to a deeper understanding of the relationship between interfacial engineering and the performance of EMW absorbers, and provides a reference for the design of multifunctional, multiband EMW absorption materials.

文摘Formal credit is critical in agricultural production, allowing more expenditure and productive input, thereby improving farmers'welfare. In pastoral China, formal financial institutions are gradually increasing. However,a limited understanding remains of how formal credit affects herders'household expenses.Based on a survey of 544 herders from the Qinghai-Xizang Plateau of China, this study adopted the propensity score matching approach to identify the effect of formal credit on herders'total household expenses, daily expenses, and productive expenses. The results found that average age, grassland mortgage, and other variables significantly affected herders'participation in formal credit.Formal credit could significantly improve household expenses, especially productive expenses.A heterogeneity analysis showed that formal credit had a greater impact on the household total expense for those at higher levels of wealth;however,it significantly affected the productive expense of herders at lower wealth levels. Moreover, the mediating effect indicated that formal credit could affect herders'household income, thus influencing their household expenses. Finally, this study suggests that policies should improve herders'accessibility to formal credit.

文摘Lightweight thin-walled structures with lattice infill are widely desired in satellite for their high stiffness-to-weight ratio and superior buckling strength resulting fromthe sandwich effect.Such structures can be fabricated bymetallic additive manufacturing technique, such as selective laser melting(SLM).However, the maximum dimensions of actual structures are usually in a sub-meter scale, which results in restrictions on their appliance in aerospace and other fields. In this work,a meter-scale thin-walled structure with lattice infill is designed for the fuel tank supporting component of the satellite by integrating a self-supporting lattice into the thickness optimization of the thin-wall.The designed structure is fabricated by SLM of AlSi10Mg and cold metal transfer welding technique.Quasi-static mechanical tests and vibration tests are both conducted to verify the mechanical strength of the designed large-scale lattice thin-walled structure. The experimental results indicate that themeter-scale thin-walled structure with lattice infill could meet the dimension and lightweight requirements of most spacecrafts.

文摘Coptis chinensis Franch.,also named Chinese goldthread is a member of Ranunculaceae in the order Ranunculales and represents an important lineage of early eudicots with traditional medicinal value. In our study,by using syntenic analysis combined with phylogenomic analysis of C.chinensis and four other representative genomes from basal and core eudicots,we confirmed that the WGD event in C.chinensis was shared by Aquilegia coerulea and Papaver somniferum L.and quickly occurred after Ranunculales diverged from other eudicots, likely a Ranunculales common tetraploidization(RCT).The synonymous nucleotide substitutions at synonymous sites distribution of syntenic blocks across these genomes showed that the evolutionary rate of the P.somniferum genome is faster than that of the C.chinensis genome by approximately 13.7%,possibly due to Papaveraceaes having an additional special tetraploidization event(PST).After Ks correction, the RCT dated to 115—130 million years ago(MYA),which was close to the divergence of Ranunculaceaes and Papaveraceaes approximately115.45—130.51 MYA.Moreover,we identified homologous genes related to polyploidization and speciation and constructed multiple sequence alignments with different reference genomes.Notably, the event-related subgenomes in the basal genomes all showed genomic fractionation bias, suggesting a likely allopolyploid nature of the RCT,PST and T-Alpha and T-Beta events in Tetracentron sinense. In addition,we detected that the sixteen P450 subfamilies were markedly expanded in the genomes of Ranunculales, and most of them were related to the RCT and PST events. We constructed a new platform for Early Eudicot Comparative Genomic Research(http://www.cgrpoee.top/index.html)to store more information. In summary, our findings support the WGD of C.chinensis shared by Ranunculales, which is likely an allotetraploidization event. This present effort offered new insights into the evolution of key polyploidization events and the genes related to secondary metabolites during the diversification of early eudicots.

文摘AIM:To explore the therapeutic effect and main molecular mechanisms of acteoside in a glaucoma model in DBA/2J mice.METHODS:Proteomics was used to compare the differentially expressed proteins of C57 and DBA/2J mice. After acteoside administration in DBA/2J mice, anterior segment observation, intraocular pressure(IOP)monitoring, electrophysiology examination, and hematoxylin and eosin staining were used to analyze any potential effects.Immunohistochemistry(IHC)assays were used to verify the proteomics results.Furthermore, retinal ganglion cell 5(RGC5)cell proliferation was assessed with cell counting kit-8(CCK-8)assays.Serta domain-containing protein 4(Sertad4)mRNA and protein expression levels were measured by qRT-PCR and Western blot analysis, respectively.RESULTS:Proteomics analysis suggested that Sertad4 was the most significantly differentially expressed protein.Compared with the saline group, the acteoside treatment group showed decreased IOP,improved N1-P1 wave amplitudes, thicker retina, and larger numbers of cells in the ganglion cell layer(GCL).The IHC results showed that Sertad4 expression levels in DBA/2J mice treated with acteoside were significantly lower than in the saline group.Acteoside treatment could improve RGC5 cell survival and reduce the Sertad4 mRNA and protein expression levels after glutamate injury.CONCLUSION:Sertad4 is differentially expressed in DBA/2J mice.Acteoside can protect RGCs from damage, possibly through the downregulation of Sertad4,and has a potential use in glaucoma treatment.

文摘D-Psicose,as a low-calorie rare sugar, has attracted a lot of attention in recent years for alternating to sucrose. The anti-obesity effect of D-psicose has been extensively confirmed in previous studies, however, the impact of D-psicose on colitis remains vague. Here,we firstly evaluated the effect of the D-psicose prophylactic intervention on dextran sulfate sodium-induced colitis in C57BL/6 mice. The pathological symptoms, inflammatory cytokines levels, gut microbiota composition, short chain fatty acids(SCFAs)production and colonic barrier integrity were comprehensively evaluated. The results confirmed that D-psicose intervention aggravated colitis, characterized by the exacerbation of colon shortening, increase of colonic inflammatory infiltration, and marked exaltation of disease activity indices and IL-6,IL-1βand TNF-αlevels.Further,the dysfunction of gut microbiota was identified in the psicose group. The abundance of pro-inflammatory bacteria Lachnospiraceae_NK4A136_group was significantly up-regulated while the abundance of probiotics Akkermansia and Lactobacillus were significantly down-regulated in the psicose group compared to the model group. Moreover, the production of SCFAs was suppressed in the psicose group, accompanied by a decrease in the level of mucin 2(Muc-2).Collectively,the underlying mechanism of the exacerbation of colitis by D-psicose intervention might be attributed to microbiota dysfunction accompanied by the reduction of SCFAs,which leads to the damage of the mucosal barrier and the intensifi cation of inflammatory invasion.

文摘Artificial immune detection can be used to detect network intrusions in an adaptive approach and proper matching methods can improve the accuracy of immune detection methods. This paper proposes an artificial immune detection model for network intrusion data based on a quantitative matching method. The proposed model defines the detection process by using network data and decimal values to express features and artificial immune mechanisms are simulated to define immune elements. Then,to improve the accuracy of similarity calculation,a quantitative matching method is proposed. The model uses mathematical methods to train and evolve immune elements, increasing the diversity of immune recognition and allowing for the successful detection of unknown intrusions. The proposed model’s objective is to accurately identify known intrusions and expand the identification of unknown intrusions through signature detection and immune detection, overcoming the disadvantages of traditional methods. The experiment results show that the proposed model can detect intrusions effectively. It has a detection rate of more than 99.6%on average and a false alarm rate of 0.0264%.It outperforms existing immune intrusion detection methods in terms of comprehensive detection performance.

文摘Diferent from necrosis, apoptosis, autophagy and other forms of cell death, ferroptosis is a mechanism that catalyzes lipid peroxidation of polyunsaturated ftty acids under the action of iron divalent or lipoxygenase, leading to cell death.Apatinib is currently used in the third line standard treatment of advanced gastric cancer, targeting the anti-angiogenesis pathway. However,Apatinib mediated ferroptosis in vascular endothelial cells has not been reported yet.Tumor.secreted exosomes can be taken up into target cells to regulate tumor development, but the mechanism related to vascular endothelial cell ferroptosis has not yet been discovered. Here,we show that exosomes secreted by gastric cancer cells carry miR-214.3p into vascular endothelial cells and directdy target zinc finger protein A20 to negatively regulate ACSL4,a key enzyme of lipid peroxidation during frroptosis thereby inhibiting ferroptosis in vascular endothelial cells and reducing the eficiency of Apatinib. In conclusion, inhibition of miR-214-3p can increase the sensitivity of vascular endothelial cells to Apatinib, thereby promoting the antiangiogenic efect of Apatinib, suggesting a potential combination therapy for advanced gastric cancer.

文摘Human normal flora is a source of probiotics. The safety characteristics of a specific isolate determine its application in foods or drugs. The food-borne-pathogen antagonist strain Lactobacillus gasseri HMV18 is one of the isolates from normal human flora. In this work,we assessed the in vitro pH tolerance, bile tolerance, biogenic amine production, mucin utilization, and safety of in vivo administration to mice to evaluate general health,organ-body weight index, organ histopathological change, whether L.gasseri HMV18 can colonize in the gut or modulate the gut microbiota after oral administration. The results suggest that L.gasseri HMV18 can tolerate pH 3 for 2 h,3%bile for 3 h,biogenic amine negative, mucin usage negative, does not encode verified toxins, and cause no visible change in mice's organs.L.gasseri HMV18 might not colonize in mice's gut,but can significantly affect the structure of gut microbiota.A bibliographical survey suggested that there were as few as 8 opportunistic infection cases from 1984 to 2022 and that the possibility for L.gasseri to cause infection is relatively low. Therefore, this work provides a basis for the foods or drugs application of L.gasseri HMV18 and gives a map of experiments for the safety assessment of probiotics.

文摘Josephson junction plays a key role not only in studying the basic physics of unconventional iron-based superconductors but also in realizing practical application of thin-film based devices, therefore the preparation of high-quality iron pnictide Josephson junctions is of great importance. In this work,we have successfully fabricated Josephson junctions from Co-doped BaFe_(2)As_(2)thin films using a direct junction fabrication technique which utilizes high energy focused helium ion beam(FHIB).The electrical transport properties were investigated for junctions fabricated with various He^(+)irradiation doses. The junctions show sharp superconducting transition around 24 K with a narrow transition width of 2.5 K,and a dose correlated foot-structure resistance which corresponds to the effective tuning of junction properties by He^(+)irradiation.Significant J_c suppression by more than two orders of magnitude can be achieved by increasing the He^(+)irradiation dose, which is advantageous for the realization of low noise ion pnictide thin film devices.Clear Shapiro steps are observed under 10 GHz microwave irradiation. The above results demonstrate the successful fabrication of high quality and controllable Co-doped BaFe_(2)As_(2)Josephson junction with high reproducibility using the FHIB technique, laying the foundation for future investigating the mechanism of iron-based superconductors, and also the further implementation in various superconducting electronic devices.

文摘The measurement of nuclear magnetic resonance(NMR)porosity is affected by temperature.Without considering the impact of NMR logging tools, this phenomenon is mainly caused by variations in magnetization intensity of the measured system due to temperature fluctuations and difference between the temperature of the porous medium and calibration sample. In this study, the effect of temperature was explained based on the thermodynamic theory, and the rules of NMR porosity responses to temperature changes were identified through core physics experiments. In addition,a method for correcting the influence of temperature on NMR porosity measurement was proposed, and the possible factors that may affect its application were also discussed.

文摘The gut microbiota is a complex and dynamic ecosystem known as the second brain'.Composing the microbiota-gut-brain axis, the gut microbiota and its metabolites regulate the central nervous system through neural, endocrine and immune pathways to ensure the normal functioning of the organism, tuning individuals'health and disease status.Short-chain fatty acids(SCFAs),the main bioactive metabolites of the gut microbiota, are involved in several neuropsychiatric disorders, including depression.SCFAs have essential effects on each component of the microbiota-gut-brain axis in depression. In the present review, the roles of major SCFAs(acetate, propionate and butyrate)in the pathophysiology of depression are summarised with respect to chronic cerebral hypoperfusion, neuroinflammation, host epigenome and neuroendocrine alterations.Concluding remarks on the biological mechanisms related to gut microbiota will hopefully address the clinical value of microbiota-related treatments for depression.

文摘The microscopic characteristics and mechanical properties of rocks change after the action of acid on deep shale, which affects the fracturing effect.Accordingly,we designed and conducted indoor experiments related to the changes in macro and microscopic characteristics after the interaction of acid with the shale of Wujiaping Formation, based on which the characteristic law of fracture volume modification after acid fracturing was studied using numerical simulation. The results demonstrate that the pores and fractures are enlarged and the structure is significantly loosened after the acid immersion. And a 15%concentration of hydrochloric acid can effectively dissolve shale.Furthermore, the degree of acid-etching reaction is highly variable because of the different carbonate content, which reveals the strong inhomogeneity of the shale system in the Wujiaping Group reservoir section. After the acid interacted with the shale rock samples, the triaxial compressive strength, elastic modulus, and Poisson’s ratio of shale decreased. Moreover, the evaluation of the effect after acid fracturing simulated by fracturing software revealed that the smaller the value of elastic modulus in shale-based reservoirs, the more favorable the fracture volume modification. This discovery not only provides a theoretical basis for the expansion and extension patterns of acid-fracturing in carbonaceous shale formations but also offers research methods and theoretical insights for the fundamental exploration of other deep-seated oil and gas resources.
