Down-regulation of histone deacetylase 7 reduces biological activities of retinal microvascular endothelial cells under high glucose condition and related mechanism

AIM:To investigate the expression and effect of histone deacetylase 7(HDAC7)in human retinal microvascular endothelial cells(HRMECs)under high glucose condition and related mechanism,and the expression of HDAC7 in the retinal tissue in diabetic rats.METHODS:The expression of HDAC7 in HRMECs under high glucose and the retinal tissue from normal or diabetic rats were detected with immunohistochemistry and Western blot.LV-shHDAC7 HRMECs were used to study the effect of HDAC7 on cell activities.Cell count kit-8(CCK-8),5-ethynyl2’-deoxyuridine(EdU),flow cytometry,scratch test,Transwell test and tube formation assay were used to examine the ability of cell proliferation,migration,and angiogenesis.Finally,a preliminary exploration of its mechanism was performed by Western blot.RESULTS:The expression of HDAC7 was both upregulated in retinal tissues of diabetic rats and high glucosetreated HRMECs.Down-regulation of HDAC7 expression significantly reduced the ability of proliferation,migration,and tube formation,and reversed the high glucose-induced high expression of CDK1/Cyclin B1 and vascular endothelial growth factor in high glucose-treated HRMECs.CONCLUSION:High glucose can up-regulate the expression of HDAC7 in HRMECs.Down-regulation of HDAC7 can inhibit HRMECs activities.HDAC7 is proposed to be involved in pathogenesis of diabetic retinopathy and a therapeutic target. 展开更多

BACKGROUND Gait is influenced by race,age,and diseases type.Reference values for gait are closely related to numerous health outcomes.To gain a comprehensive understanding of gait patterns,particularly in relation to race-related pathologies and disorders,it is crucial to establish reference values for gait in daily life considering sex and age.Therefore,our objective was to present sex and age-based reference values for gait in daily life,providing a valuable foundation for further research and clinical applications.AIM To establish reference values for lower extremity joint kinematics and kinetics during gait in asymptomatic adult women and men.METHODS Spatiotemporal,kinematics and kinetics parameters were measured in 171 healthy adults(70 males and 101 females)using the computer-aided soft tissue foot model.Full curve statistical parametric mapping was performed using independent and paired-samples t-tests.RESULTS Compared with females,males required more time(cycle time,double-limb support time,stance time,swing time,and stride time),and the differences were statistically significant.In addition,the step and stride lengths of males were longer.Compared to males,female cadence was faster,and statures-per-second and stride-per-minute were higher.There were no statistical differences in speed and stride width between the two groups.After adjusting for height,it was observed that women walked significantly faster than men,and they also had a higher cadence.However,in terms of step length,stride length,and stride width,both genders exhibited similarities.CONCLUSION We established reference values for gait speed and spatiotemporal gait parameters in Chinese university students.This contributes to a valuable database for gait assessment and evaluation of preventive or rehabilitative programs. 展开更多

目的:探讨不同分期下的非增殖期糖尿病视网膜病变微血管直径的变化。方法:前瞻性病例对照研究。选取2020-09/2021-03于本院内分泌科室住院的2型糖尿病并糖尿病视网膜病变患者,据我国中华医学会于1985年制定的“糖尿病视网膜病变分期标... 目的:探讨不同分期下的非增殖期糖尿病视网膜病变微血管直径的变化。方法:前瞻性病例对照研究。选取2020-09/2021-03于本院内分泌科室住院的2型糖尿病并糖尿病视网膜病变患者,据我国中华医学会于1985年制定的“糖尿病视网膜病变分期标准”将其分为糖尿病无视网膜病变组(50例50眼)、糖尿病视网膜病变Ⅰ期组(50例50眼)、糖尿病视网膜病变Ⅱ期组(50例50眼)、糖尿病视网膜病变Ⅲ期组(50例50眼),另取体检正常的对照组50例50眼,共计250例250眼。摄取眼底彩照图,录入ARIA1.0自动分析软件,记录不同范围内动静脉与毛细血管直径,每个范围取值4段,每组为200段,进行统计学分析。结果:不同分期的非增殖期糖尿病视网膜病变的视网膜动脉直径均无差异(P>0.05),不同分期的非增殖期糖尿病视网膜病变的视网膜静脉直径、黄斑周围毛细血管直径均有差异(P<0.05),视网膜静脉直径与黄斑周围毛细血管直径的不同组别事后多重比较均有差异(P<0.01),0~<0.5PD和0.5~1.0PD范围的视网膜静脉直径组别平均值得分对比结果为“视网膜病变Ⅲ期组>视网膜病变Ⅱ期组>视网膜病变I期组>糖尿病无视网膜病变组>正常对照组”,黄斑周围毛细血管直径组别平均值得分对比结果为“视网膜病变Ⅲ期组>视网膜病变Ⅰ期组>正常对照组;视网膜病变Ⅲ期组>糖尿病无视网膜病变组;视网膜病变Ⅱ期组>视网膜病变Ⅰ期组>正常对照组;视网膜病变Ⅱ期组>糖尿病无视网膜病变组;糖尿病无视网膜病变组>正常对照组”,而视网膜病变Ⅲ期组与视网膜病变Ⅱ期组,视网膜病变Ⅰ期组与糖尿病无视网膜病变组均无显著差异(P>0.05)。结论:视网膜动脉直径在非增殖期糖尿病视网膜病变不同分期中无明显变化;糖尿病患者视网膜静脉及黄斑周围毛细血管扩张,在非增殖期糖尿病视网膜病变不同分期中,静脉与黄斑周围毛细血管的直径呈逐渐增宽的趋势。 展开更多

We applied the Koch snowflake fractal antenna in planning calibration of the Five-hundred-meter Aperture Spherical radio Telescope(FAST), hypothesizing second-order fractal primary reflectors can optimize the orientated sensitivity of the telescope. Meanwhile, on the grounds of NASA Science Working Group Report in 1984, we reexamine the strategy of Search for Extraterrestrial Intelligence(SETI).A mathematical analysis of the radar equation will be performed in the first section, aiming to make it convenient to design a receiver system that can detect activities of an extraterrestrial civilization, according to the observable region of the narrowband. Taking advantage of the inherent potential of FAST, we simulate the theoretical detection of a Kardashev Type I civilization by a snowflake-selected reflecting area(Drake et al.). 展开更多
