This study aimed to evaluate the feasibility and safety of a novel stent manufactured by metal injection molding(MIM)in clinical practice through animal experiments.Vessel stents were prepared using powder injection m...This study aimed to evaluate the feasibility and safety of a novel stent manufactured by metal injection molding(MIM)in clinical practice through animal experiments.Vessel stents were prepared using powder injection molding technology to considerably improve material utilization.The influence of MIM carbon impurity variation on the mechanical properties and corrosion resistance of 316L stainless steel was studied.In vitro cytotoxicity and animal transplantation tests were also carried out to evaluate the safety of MIM stents.The results showed that the performance of 316L stainless steel was very sensitive to the carbon content.Carbon fluctuations should be precisely controlled during MIM.All MIM stents were successfully implanted into the aortas of the dogs,and the MIM 316L stents had no significant cytotoxicity.The novel intravascular stent manufactured using MIM can maintain a stable form and structure with fast endothelialization of the luminal surface of the stent and ensure long-term patency in an animal model.The novel intravascular stent manufactured using MIM demonstrates favorable structural,physical,and chemical stability,as well as biocompatibility,offering promising application in clinical practice.展开更多
BACKGROUND Alphaherpesvirus belongs to the Herpesviridae family and has large,monopartite double-stranded linear DNA.It mainly infects the skin,mucosa,and nerves,and can affect various hosts,including humans and other...BACKGROUND Alphaherpesvirus belongs to the Herpesviridae family and has large,monopartite double-stranded linear DNA.It mainly infects the skin,mucosa,and nerves,and can affect various hosts,including humans and other animals.Here,we present a case of a patient seen by the gastroenterology department at our hospital who experienced an oral and perioral herpes infection following treatment with a ven-tilator.The patient was treated with oral and topical antiviral drugs,furacilin,oral and topical antibiotics,local epinephrine injection,topical thrombin powder,and nutritional and supportive care.A wet wound healing approach was also implemented with good response.CASE SUMMARY A 73-year-old woman presented to the hospital with a chief complaint of"abdominal pain for 3 d with dizziness for 2 d."She was admitted to the intensive care unit for septic shock and spontaneous peritonitis secondary to cirrhosis and was given antiinflammatory and symptomatic supportive treatment.A ventilator was used to assist breathing for acute respiratory distress syndrome,which developed during her admission.A large area of herpes infection appeared in the perioral region 2 d following noninvasive ventilation.The patient was transferred to the gastroenterology department,at which time she had a body temperature of 37.8 C and a respiratory rate of 18/min.The patient's con-sciousness was intact,and she no longer had abdominal pain or distension,chest tightness,or asthma.At this point,the infected perioral region changed in appearance and was now accompanied by local bleeding with crusting of blood at the wounds.The surface area of the wounds measured approximately 10 cm×10 cm.A cluster blisters appeared on the patient’s right neck,and ulcers developed in her mouth.On a subjective numerical pain scale,the patient reported a pain level of 2.Overall,her diagnoses other than the oral and perioral herpes infection included:(1)Septic shock;(2)spontaneous peritonitis;(3)abdominal infection;(4)decompensated cirrhosis;and(5)hypoproteinemia.Dermatology was consulted regarding the treatment of the patient’s wounds;they suggested treatment with oral antiviral drugs,an intramuscular injection of nutritious nerve drugs,and the application of topical penciclovir and mupirocin around the lips.Stomatology was also consulted and suggested the use of nitrocilin in a local wet application around the lips.CONCLUSION Through multidisciplinary consultation,the patient’s oral and perioral herpes infection was successfully treated with the following combined approach:(1)Application of topical antviral and antibiotic treatments;(2)keeping the wound moist with a wet wound healing strategy;(3)systemic use of oral antiviral drugs;and(4)symptomatic and nutritional supportive care.The patient was discharged from the hospital after successful wound healing.展开更多

HPLC fingerprints were...Using Dachengqi Tang(DCQT) as a model, high performance liquid chromatography(HPLC)fingerprints were applied to optimize machine extracting process with the Box–Behnken experimental design. HPLC fingerprints were carried out to investigate the chemical ingredients of DCQT; synthetic weighing method based on analytic hierarchy process(AHP) and criteria importance through intercriteria correlation(CRITIC) was performed to calculate synthetic scores of fingerprints; using the mark ingredients contents and synthetic scores as indicators, the Box–Behnken design was carried out to optimize the process parameters of machine decocting process under high pressure for DCQT. Results of optimal process showed that the herb materials were soaked for 45 min and extracted with 9 folds volume of water in the decocting machine under the temperature of 140 1C till the pressure arrived at 0.25 MPa;then hot decoction was excreted to soak Dahuang and Mangxiao for 5 min. Finally, obtained solutions were mixed, filtrated and packed. It concluded that HPLC fingerprints combined with the Box–Behnken experimental design could be used to optimize extracting process of traditional Chinese medicine(TCM).展开更多
Objective To investigate the prevalence and levels of anti-HEV IgG in the population of Jiangsu Province.Methods Total of 2 656 samples from Qindong and 11 463 samples from Anfeng were colleted.The antiHEV antibody wa...Objective To investigate the prevalence and levels of anti-HEV IgG in the population of Jiangsu Province.Methods Total of 2 656 samples from Qindong and 11 463 samples from Anfeng were colleted.The antiHEV antibody was qualitatively and quantitatively detected using ELISA kits and the references had been established.Results The positive rates of anti-HEV IgG in male and female were 55.6%and 40.1%,respectively.The positive rate of anti-HEV IgM in male and female were both 3.4%.In opposite to anti-HEV IgG,the positive rate of anti-HEV IgM in Anfeng was significant higher than that in Qindong.The mean anti-HEV IgG titers for6 age groups were 0.94,0.92,1.07,1.46,1.27,1.19 and 0.68,1.31,1.08,1.14,1.31,1.68 IU/ml,in Qindong and Anfeng region,respectively.The positive rate of anti-HEV IgG tended to increase with age and the titer of antiHEV IgG was associated with age(R>0.90).Conclusions The results in this study showed that HEV was widely prevalent in both Qindong and Anfeng of Jiansu Province and the prevalence and the anti-HEV IgG titer were associated with gender and age.展开更多

Methods Qualitative analysis of coumarins in C. monnieri was detected by UHPLC-ESI-Q-TOF/MS. Quadrupole TOF/MS in either full scan mode or extracted ion mode was used for the qualitative analysis of the constituents. Relative peak area of each component was used for the hierarchical cluster analysis. Results According to UHPLC-ESI-QTOF-MS data, chemical structures of 28 coumarins in the fruits of C. monnieri were identified, including 19 simple coumarins, seven linear coumarins, and two angular coumarins. Among these constituents, ten coumarins were firstly identified in C. monnieri. In addition, hierarchical cluster analysis suggested that C. monnieri from different regions could be classified into four groups, and this clustering was correlated to the distribution significantly, Xuzhou could be regarded as the genuine producing area. Conclusion UHPLC-ESI-Q-TOF-MS is a viable method for qualitative analysis and quality evaluation of coumarins from the fruit of C. monnieri. Coumarins in C. monnieri exists intra-species variance, which indicates significant meaning for the quality control and choice of famous region drug for C. monnieri in the clinic medication.展开更多
Vitellin(Vn)homeostasis is central to the fecundity of oviparous insects.Most studies have focused on the synthesis and transportation of Vn as a building block for developing eggs during vitellogenesis;however,less i...Vitellin(Vn)homeostasis is central to the fecundity of oviparous insects.Most studies have focused on the synthesis and transportation of Vn as a building block for developing eggs during vitellogenesis;however,less is known about how the utilization of this nutrient reserve affects embryonic development.Here,we show that the single ortholog of the knirps and knirps-like nuclear receptors,KNRL,negatively regulates Vn breakdown by suppressing the expression of hydrolase genes in the brown planthopper,Nilaparvata lugens.KNRL was highly expressed in the ovary of adult females,and knockdown of KNRL by RNA interference resulted in the acceleration of Vn breakdown and the inhibition of embryonic development.Transcriptome sequencing analysis revealed that numerous hydrolase genes,including cathepsins and trypsins were up-regulated after KNRL knockdown.At least eight of the nine significantly enriched Gene Ontology terms for the up-regulated genes were in proteolysis-related categories.The expression levels of five selected trypsin genes and the enzymatic activities of trypsin in the embryos were significantly increased after KNRL knockdown.Moreover,trypsin injection prolonged egg duration,delayed embryonic development,accelerated Vn breakdown and severely reduced egg hatchability,a pattern similar to that observed in A7V/?L-silenced N.lugens.These observations suggest that KNRL controls Vn breakdown in embryos via the transcriptional inhibition of hydrolases.Generally,this study provides a foundation for understanding how embryo nutrient reserves are mobilized during embryogenesis and identifies several genes and pathways that may prove valuable targets for pest control.展开更多

In this article, using Da-cheng-qi Decoction(DCQD) as a model of formula, traditional decoction(TD), machinery decoction under high pressure(MDHP), and machinery decoction under normal pressure(MDNP) were compared. Methods For chemical components, HPLC fingerprints were established and evaluated using AHP combined with CRITIC weighing method; For animals' effects, the experiments of small intestinal propulsion were conducted; For clinical effects, a randomized clinical trial(RCT) was designed and performed. Results Although there were some differences between TD and MDNP in chemical ingredients, there was no significant difference in animal experiments and clinical trials(P 〉 0.05). Background:Psoriatic arthritis(PsA)is an inflammatory arthropathy characterized by psoriasis and bone erosion on radiology.Dickkopf-1(Dkk-1)is considered to be the main inhibitor of the Wnt signaling pathway and resul...Background:Psoriatic arthritis(PsA)is an inflammatory arthropathy characterized by psoriasis and bone erosion on radiology.Dickkopf-1(Dkk-1)is considered to be the main inhibitor of the Wnt signaling pathway and results in reduced osteoblast proliferation.The aim of this study was to investigate the serum level of Dkk-1 and its association with bone erosion in PsA patients.Methods:Serum Dkk-1 levels were measured by enzyme-linked immunosorbent assay(ELISA)in 69 patients with PsA and 60 controls,including 39 rheumatoid arthritis(RA)patients,and 21 healthy controls(HCs).Rheumatoid factor and anti-cyclic citrullinated peptide levels were also determined by ELISA.The association of Dkk-1 level with clinical and laboratory features of PsA was analyzed.Logistic regression analysis was used to analyze the risk factors for bone erosion in PsA.Results:Dkk-1 was elevated in 68.1%(47/69)of the patients with PsA,46.2%(18/39)of RA patients,and 9.5%(2/21)of HCs.Serum Dkk-1 concentration was significantly higher in PsA patients compared with that in HCs.The level of serum Dkk-1 was correlated with a swollen joint count,and levels of complement components 3 and 4.Elevated Dkk-1 level(odds ratio=4.440,95%confidence interval:1.246-15.817,P=0.021)was identified as the risk factor for bone erosion in PsA.Conclusions:The serum level of Dkk-1 is abnormally elevated in PsA patients.The elevation of Dkk-1 might be involved in the mechanism of bone erosion in patients with PsA.展开更多
