CRISPR/Cas9 technology is a powerful genome manipulation tool in insects.However,little is known about whether mRNA and protein of a target gene are completely cleared in homozygous mutants.This study generated homozy...CRISPR/Cas9 technology is a powerful genome manipulation tool in insects.However,little is known about whether mRNA and protein of a target gene are completely cleared in homozygous mutants.This study generated homozygous mutants of the insulin receptor gene 2(NlInR2)in the brown planthopper(Nilaparvata lugens)using CRISPR/Cas9 genome editing.Both frameshift mutants,E5_D17 and E6_I7,differentiated towards long wings,but there were differences in wing morphology,with E5_D17 showing wing deformities.Subsequent investigations revealed the presence of residual expression of NlInR2 mRNA in both mutants,as well as the occurrence of spliceosomes featuring exon skipping splicing in E5_D17.Additionally,the E5_D17 exhibited the detection of N-terminally truncated NlInR2 protein.RNA interference experiments indicated that the knockdown of NlInR2 expression in the E5_D17 mutant line increased the proportion of wing deformities from 11.1 to 65.6%,suggesting that the residual NlInR2 mRNA of the E5_D17 mutant might have retained some genetic functions.Our results imply that systematic characterization of residual protein expression or function in CRISPR/Cas9-generated mutant lines is necessary for phenotypic interpretation.展开更多

Hippocampal and cortical...The present study established a rat model of focal brain ischemia by occlusion of the middle cerebral artery covered with FeCl3, and investigated the protective effect of 3'-methoxy-puerarin. Hippocampal and cortical c-fos gene expression was determined using in situ hybridization. Results showed that 3'-methoxy-puerarin reduced neurological deficit scores, cerebral infarcted zone and water content of brain tissues, dramatically increased the activity of catalase and glutathione peroxidase in the ischemia zone of the hippocampus, increased the activity of catalase in the cortex, decreased lipid peroxide and lactic acid contents in the hippocampus and cerebral cortex, and down-regulated c-fos gene expression in brain ischemic rats. Results demonstrated that 3'-methoxy-puerarin exhibited cerebroprotective effects against focal brain ischemia, which involved c-fos gene expression.展开更多
Enhancing saturation magnetic flux density(Bs)while reducing high-frequency core loss in Finemet-type nanocrystalline alloys is of great significance in achieving the miniaturization,high-frequency,and energy-saving o...Enhancing saturation magnetic flux density(Bs)while reducing high-frequency core loss in Finemet-type nanocrystalline alloys is of great significance in achieving the miniaturization,high-frequency,and energy-saving of modern power electronic devices.In this work,we first designed a high-Bs Fe_(77.2)Si_(11)B_(8.5)Cu_(0.8)Nb_(2.5)alloy by appropriately reducing the non-magnetic elements in typical Finemet nanocrystalline alloys,and subsequently alloyed 2 at%Co,Al,and Mo,respectively.The effects of alloying elements on structure and static and high-frequency magnetic properties were studied.The results reveal that,alloying Al or Mo reduces the averageα-Fe grain size(Dα-Fe)in the nanocrystalline alloys,while Co exhibits a slight influence.The added Al or Mo results in decreases in both the Bs and coercivity(Hc)of the nanocrystalline alloys,whereas Co increases the Bs without changing Hc,and meanwhile,all alloying elements show minimal effects on effective permeability(μe).Furthermore,the addition of Co,Al,or Mo lowers the core loss(Pcv)at 0.2 T/100 kHz of the based nanocrystalline alloy with reductions of 10.9%,29.6%,and 26.8%,respectively.A Fe_(75.2)Si_(11)B_(8.5)Cu_(0.8)Nb_(2.5)Al_(2)nanocrystalline alloy exhibits outstanding soft magnetic properties with Bs,Hc,μe at 10 kHz and 100 kHz,and Pcv at 0.2 T/100 kHz of 1.34 T,0.8 A/m,27,400,18,000,and 350 kW/m3,respectively.The reduction in Pcv is primarily attributed to the decreased eddy current losses,originating from the increased electrical resistivity by elements alloying.展开更多
In this work,a traditional solid-state method was adopted to prepare dense Ba_(0.85)Ca_(0.15)[(Zr_(0.1)Ti_(0.9))_(1-x)–(Nb_(0.5)La_(0.5))_(x)]O_(3)(BCZT–xNL)lead-free relaxation ferroelectrics with excellent energy ...In this work,a traditional solid-state method was adopted to prepare dense Ba_(0.85)Ca_(0.15)[(Zr_(0.1)Ti_(0.9))_(1-x)–(Nb_(0.5)La_(0.5))_(x)]O_(3)(BCZT–xNL)lead-free relaxation ferroelectrics with excellent energy storage performance(ESP).Remarkably,a large breakdown field strength of BDS(-410 kV/cm)resulting from decreased grain size by double ions doping at the B-site was achieved in BCZT–xNL ceramics.Especially,the BCZT–0.12NL ceramic displays a good recoverable energy density(W_(rec))of 3.1 J/cm^(3),a high efficiency of 86.7%(η)and a superior fatigue resistance,as well as a superior charge–discharge performance(C_(D)-1623.14 A/cm^(2),P_(D)-162.31 MW/cm^(3),W_(D)-1.30 J/cm^(3),t_(0.9)-49.6 ns)and good thermal stability.Besides,upon NL doping,the FE to RFE phase transition results in a dielectric behavior traversing the relaxation phase(γ-1.84)accompanied by frequency dispersion,which is beneficial to improve ESP.The superior ESP in this work indicates that BCZT–0.12NL ceramics are a promising candidate used in energy storage capacitors.展开更多
