Our previous studies have shown that Lycium barbarum(L. barbarum) polysaccharide(LBP) protects degenerated photoreceptors in rd1, a transgenic mouse model of retinitis pigmentosa. L. barbarum glycopeptide(Lb GP) is an immunoreactive glycoprotein extracted from LBP. In this study, we investigated the potential protective effect of Lb GP on a chemically induced photoreceptor-degenerative mouse model. Wild-type mice received the following: oral administration of Lb GP as a protective pre-treatment on days 1–7;intraperitoneal administration of 40 mg/kg N-methylN-nitrosourea to induce photoreceptor injury on day 7;and continuation of orally administered Lb GP on days 8–14. Treatment with Lb GP increased photoreceptor survival and improved the structure of photoreceptors, retinal photoresponse, and visual behaviors of mice with photoreceptor degeneration. Lb GP was also found to partially inhibit the activation of microglia in N-methyl-N-nitrosourea-injured retinas and significantly decreased the expression of two pro-inflammatory cytokines. In conclusion, Lb GP effectively slowed the rate of photoreceptor degeneration in N-methyl-N-nitrosourea-injured mice, possibly through an anti-inflammatory mechanism, and has potential as a candidate drug for the clinical treatment of photoreceptor degeneration.展开更多
The gradual increase in shipping and drilling activities in the Arctic regions has resulted in the increased importance of studying the structural safety of polar ships in various ice conditions.Rafted ice refers to a...The gradual increase in shipping and drilling activities in the Arctic regions has resulted in the increased importance of studying the structural safety of polar ships in various ice conditions.Rafted ice refers to a type of accumulated and overlapped sea ice;it is driven by external forces,such as wind and waves,and may exert high loads on ships and threaten their structural safety.Therefore,the properties of rafted ice and the construction of numerical models should be studied before exploring the interaction and collision between ships and rafted ice.Based on the nonlinear finite-element method,this paper introduces the cohesive element model for the simulation of rafted ice.The interaction between ships and rafted ice is studied,and the ice force of the hull is obtained.Numerical simulation results are compared with model test findings,and the effectiveness of the cohesive element method in the construction of the model of rafted ice materials is verified.On this basis,a multilayer rafted ice model is constructed,and its interaction with the ship is studied.The research unveils that rafted ice parts impede crack generation and slow down crack propagation to a certain extent.展开更多

It has been confirmed that autophagy promotes chemoresistance and, therefore, novel strategies to reverse chemoresistance by regulating autophagy are desperately needed. Methods: The differentially expressed lncRNAs (DElncRNAs), miRNAs (DEmiRNAs), and mRNAs (DEmRNAs) between A549 and A549/DDP cell lines were identified using the limma package in R, after gene expression profiles were obtained from Gene Expression Omnibus (GEO) database. By combining Autophagy-Related Genes (ARGs) from Human Autophagy Database (HADb), the interactions lncRNA-miRNAs and the interactions miRNAs-mRNAs respectively predicted by miRcode and miRDB/Targetscan database, the autophagy-related ceRNA network was constructed. Then, extraction of ceRNA subnetwork and Cox regression analyses were performed. A prognosis-related ceRNA subnetwork was constructed, and the upstream Transcription Factors (TFs) regulating lncRNAs were predicted by the JASPAR database. Finally, the expression patterns of candidate genes were further verified by quantitative real-time polymerase chain reaction (qRT-PCR) experiments. Results: A total of 3179 DEmRNAs, 180 DEmiRNAs, and 160 DElncRNAs were identified, and 35 DEmRNAs were contained in the HADb. Based on the ceRNA hypothesis, we established a ceRNA network, including 10 autophagy-related DEmRNAs, 9 DEmiRNAs, and 14 DElncRNAs. Then, LINC00520, miR-181d, and BCL2 were identified to construct a risk score model, which was confirmed to be a well-predicting prognostic factor. Furthermore, 5 TF ZNF family members were predicted to regulate LINC00520, whereas the RT-PCR results showed that the 5 ZNFs were consistent with the bioinformatics analysis. Finally, a ZNF regulatory LINC00520/miR-181d/BCL2 ceRNA subnetwork was constructed. Conclusions: An ZNFs/LINC00520/miR-181d/BCL2 axis as a novel network in DDP-resistant LUAD has been constructed successfully, which may provide potential therapeutic targets for LUAD.展开更多
Since its introduction,discontinuous deformation analysis(DDA)has been widely used in different areas of rock mechanics.By dividing large blocks into subblocks and introducing artificial joints,DDA can be applied to r...Since its introduction,discontinuous deformation analysis(DDA)has been widely used in different areas of rock mechanics.By dividing large blocks into subblocks and introducing artificial joints,DDA can be applied to rock fracture simulation.However,parameter calibration,a fundamental issue in discontinuum methods,has not received enough attention in DDA.In this study,the parameter calibration of DDA for intact rock is carefully studied.To this end,a subblock DDA with Voronoi tessellation is presented first.Then,a modified contact constitutive law is introduced,in which the tensile and shear meso-strengths are modified to be independent of the bond lengths.This improvement can prevent the unjustified preferential failure of short edges.A method for imposing confining pressure is also introduced.Thereafter,sensitivity analysis is performed to investigate the influence of the calculated parameters and meso-parameters on the mechanical properties of modeled rock.Based on the sensitivity analysis,a unified calibration procedure is suggested for both cases with and without confining pressure.Finally,the calibration procedure is applied to two examples,including a biaxial compression test.The results show that the proposed Voronoi-based DDA can simulate rock fracture with and without confining pressure very well after careful parameter calibration.展开更多

Therefore, how to regenerate new neurons to restore vision is an important issue. NeuroD1 is a neural transcription factor with the ability to reprogram brain astrocytes into neurons in vivo. Here, we demonstrate that in adult mice, NeuroD1 can reprogram Müller cells, the principal glial cell type in the retina, to become retinal neurons. Most strikingly, ectopic expression of NeuroD1 using two different viral vectors converted Müller cells into different cell types. Specifically, AAV7 m8 GFAP681::GFP-ND1 converted Müller cells into inner retinal neurons, including amacrine cells and ganglion cells. In contrast, AAV9 GFAP104::ND1-GFP converted Müller cells into outer retinal neurons such as photoreceptors and horizontal cells, with higher conversion efficiency. Furthermore, we demonstrate that Müller cell conversion induced by AAV9 GFAP104::ND1-GFP displayed clear dose-and time-dependence. These results indicate that Müller cells in adult mice are highly plastic and can be reprogrammed into various subtypes of retinal neurons.展开更多

Although most studies to date have typically focused on the lysosome, a single o...Cerebral ischemia is a serious disease that triggers sequential pathological mechanisms, leading to significant morbidity and mortality. Although most studies to date have typically focused on the lysosome, a single organelle, current evidence supports that the function of lysosomes cannot be separated from that of the endolysosomal system as a whole. The associated membrane fusion functions of this system play a crucial role in the biodegradation of cerebral ischemia-related products. Here, we review the regulation of and the changes that occur in the endolysosomal system after cerebral ischemia, focusing on the latest research progress on membrane fusion function. Numerous proteins, including N-ethylmaleimide-sensitive factor and lysosomal potassium channel transmembrane protein 175, regulate the function of this system. However, these proteins are abnormally expressed after cerebral ischemic injury, which disrupts the normal fusion function of membranes within the endolysosomal system and that between autophagosomes and lysosomes. This results in impaired “maturation” of the endolysosomal system and the collapse of energy metabolism balance and protein homeostasis maintained by the autophagy-lysosomal pathway. Autophagy is the final step in the endolysosomal pathway and contributes to maintaining the dynamic balance of the system. The process of autophagosome-lysosome fusion is a necessary part of autophagy and plays a crucial role in maintaining energy homeostasis and clearing aging proteins. We believe that, in cerebral ischemic injury, the endolysosomal system should be considered as a whole rather than focusing on the lysosome. Understanding how this dynamic system is regulated will provide new ideas for the treatment of cerebral ischemia.展开更多
In response to spinal surgery,neurons secrete a large amount of substance P into the epidural area.Substance P is involved in macrophage differentiation and fibrotic disease.However,the specific roles and mechanisms o...In response to spinal surgery,neurons secrete a large amount of substance P into the epidural area.Substance P is involved in macrophage differentiation and fibrotic disease.However,the specific roles and mechanisms of substance P in epidural fibrosis remain unclear.In this study,we established a mouse model of L1–L3 laminectomy and found that dorsal root ganglion neurons and the macrophages infiltrating into the wound area released sphingolipids.In vitro experiments revealed that type 1 macrophages secreted substance P,which promoted differentiation of type 1 macrophages towards a type 2 phenotype.High-throughput mRNA-seq analysis revealed that the sphingolipid metabolic pathway may be involved in the regulation of type 2 macrophages by substance P.Specifically,sphingomyelin synthase 2,a component of the sphingolipid metabolic pathway,promoted M2 differentiation in substance P-treated macrophages,while treating the macrophages with LY93,a sphingomyelin synthase 2 inhibitor,suppressed M2 differentiation.In addition,substance P promoted the formation of neutrophil extracellular traps,which further boosted M2 differentiation.Blocking substance P with the neurokinin receptor 1 inhibitor RP67580 decreased the number of M2 macrophages in the wound area after spinal surgery and alleviated epidural fibrosis,as evidenced by decreased fibronectin,α-smooth muscle actin,and collagen I in the scar tissue.These results demonstrated that substance P promotes M2 macrophage differentiation in epidural fibrosis via sphingomyelin synthase 2 and neutrophil extracellular traps.These findings provide a novel strategy for the treatment of epidural fibrosis.展开更多
The Ordos Basin is the largest continental multi-energy mineral basin in China,which is rich in coal,oil and gas,and uranium resources.The exploitation of mineral resources is closely related to reservoir water.The ch...The Ordos Basin is the largest continental multi-energy mineral basin in China,which is rich in coal,oil and gas,and uranium resources.The exploitation of mineral resources is closely related to reservoir water.The chemical properties of reservoir water are very important for reservoir evaluation and are significant indicators of the sealing of reservoir oil and gas resources.Therefore,the caprock of the Chang 6 reservoir in the Yanchang Formation was evaluated.The authors tested and analyzed the chemical characteristics of water samples selected from 30 wells in the Chang 6 reservoir of Ansai Oilfield in the Ordos Basin.The results show that the Chang 6 reservoir water in Ansai Oilfield is dominated by calcium-chloride water type with a sodium chloride coefficient of generally less than 0.5.The chloride magnesium coefficients are between 33.7 and 925.5,most of which are greater than 200.The desulfurization coefficients range from 0.21 to 13.4,with an average of 2.227.The carbonate balance coefficients are mainly concentrated below 0.01,with an average of 0.008.The calcium and magnesium coefficients are between 0.08 and 0.003,with an average of 0.01.Combined with the characteristics of the four-corner layout of the reservoir water,the above results show that the graphics are basically consistent.The study indicates that the Chang 6 reservoir in Ansai Oilfield in the Ordos Basin is a favorable block for oil and gas storage with good sealing properties,great preservation conditions of oil and gas,and high pore connectivity.展开更多

Most studies hav...Strabismus and amblyopia are common ophthalmologic developmental diseases caused by abnormal visual experiences. However, the underlying pathogenesis and visual defects are still not fully understood. Most studies have used experimental interference to establish diseaseassociated animal models, while ignoring the natural pathophysiological mechanisms. This study was designed to investigate whether natural strabismus and amblyopia are associated with abnormal neurological defects. We screened one natural strabismic monkey(Macaca fascicularis) and one natural amblyopic monkey from hundreds of monkeys, and retrospectively analyzed one human strabismus case. Neuroimaging, behavioral,neurophysiological, neurostructural, and genovariation features were systematically evaluated using magnetic resonance imaging(MRI), behavioral tasks, flash visual evoked potentials(FVEP),electroretinogram(ERG), optical coherence tomography(OCT), and whole-genome sequencing(WGS), respectively. Results showed that the strabismic patient and natural strabismic and amblyopic monkeys exhibited similar abnormal asymmetries in brain structure, i.e., ipsilateral impaired right hemisphere. Visual behavior, visual function, retinal structure, and fundus of the monkeys were impaired. Aberrant asymmetry in binocular visual function and structure between the strabismic and amblyopic monkeys was closely related, with greater impairment of the left visual pathway.Several similar known mutant genes for strabismus and amblyopia were also identified. In conclusion,natural strabismus and amblyopia are accompanied by abnormal asymmetries of the visual system,especially visual neurophysiological and neurostructural defects. Our results suggest that future therapeutic and mechanistic studies should consider defects and asymmetries throughout the entire visual system.展开更多

Engineering disasters(e.g.rock slabbing and rockburst)of the tunnel groups induced by the transient excavation of an adjacent tunnel threaten the stability of the existing tunnel,especially for those excavated by usin...Engineering disasters(e.g.rock slabbing and rockburst)of the tunnel groups induced by the transient excavation of an adjacent tunnel threaten the stability of the existing tunnel,especially for those excavated by using the drill and blast tunneling(D&B).However,the dynamic response and failure mechanism of surrounding rocks of the existing tunnel caused by adjacent transient excavation are not clear due to the difficulty in conducting field tests and laboratory experiments.Therefore,a novel transient unloading experimental system for deep tunnel excavation was proposed in this study.The real stress path and the unloading rate can be reproduced by using this proposed system.The experiments were conducted for observing the dynamic response of the existing tunnel induced by adjacent transient excavation under different lateral pressure coefficients l(?0.4,0.6,0.8,1,1.2,1.4,1.6,1.8)with a polymethyl methacrylate(PMMA)specimen.The propagation of the impact wave and unloading surface wave was detected through the digital image correlation(DIC)analysis.The reflection of the unloading surface wave on the incident side of the existing tunnel(tunnel-E)was observed and analyzed.Moreover,the dynamic characteristics of the stress redistribution,the particle displacement and vibration velocity of surrounding rocks of tunnel-E were analyzed and summarized.In addition,the Mohr-Coulomb(MeC)failure criterion with tension cut-off was adopted to evaluate the stability of the existing tunnel under adjacent transient excavation.The results indicate that the incident side of the existing tunnel under the dynamic disturbance of transient excavation of an adjacent tunnel was more prone to fail,followed by the shadow side and the top/bottom side.展开更多
Based on 20 models from phase 6 of the Coupled Model Intercomparison Project(CMIP6),this article explored possible reasons for differences in simulation biases and projected changes in precipitation in northern China ...Based on 20 models from phase 6 of the Coupled Model Intercomparison Project(CMIP6),this article explored possible reasons for differences in simulation biases and projected changes in precipitation in northern China among the allmodel ensemble(AMME),“highest-ranked”model ensemble(BMME),and“lowest-ranked”model ensemble(WMME),from the perspective of atmospheric circulations and moisture budgets.The results show that the BMME and AMME reproduce the East Asian winter circulations better than the WMME.Compared with the AMME and WMME,the BMME reduces the overestimation of evaporation,thereby improving the simulation of winter precipitation.The three ensemble simulated biases for the East Asian summer circulations are generally similar,characterized by a stronger zonal pressure gradient between the mid-latitudes of the North Pacific and East Asia and a northward displacement of the East Asian westerly jet.However,the simulated vertical moisture advection is improved in the BMME,contributing to the slightly higher performance of the BMME than the AMME and WMME on summer precipitation in North and Northeast China.Compared to the AMME and WMME,the BMME projects larger increases in precipitation in northern China during both seasons by the end of the 21st century under the Shared Socioeconomic Pathway 5-8.5(SSP5-8.5).One of the reasons is that the increase in evaporation projected by the BMME is larger.The projection of a greater dynamic contribution by the BMME also plays a role.In addition,larger changes in the nonlinear components in the BMME projection contribute to a larger increase in winter precipitation in northern China.展开更多
The Global Navigation Satellite System(GNSS)has been widely used in various fields.To achieve positioning,the receiver must first lock the satellite signal.This is a complicated and expensive process that consumes a l...The Global Navigation Satellite System(GNSS)has been widely used in various fields.To achieve positioning,the receiver must first lock the satellite signal.This is a complicated and expensive process that consumes a lot of resources of the receiver.For this reason,this paper proposes a new fast acquisition algorithm with High Signal-tonoise ratio(SNR)performance based on sparse fast Fourier transform(HSFFT).The algorithm first replaces the IFFT process of the traditional parallel code phase capture algorithm with inverse sparse fast Fourier transform(ISFFT)with better computing performance,and then uses linear search combined with code phase discrimination to replace the positioning loop and the estimation loop with poor noise immunity in ISFFT.Theoretical analysis and simulation results show that,compared with the existing SFFT parallel code phase capture algorithm,the calculation amount of this algorithm is reduced by 19%,and the SNR performance is improved by about 5dB.Compared with the classic FFT parallel code phase capture algorithm,the calculation amount of the algorithm in this paper is reduced by 43%,and when the capture probability is greater than 95%,the SNR performance of the two is approximately the same.展开更多
BACKGROUND Hysteromyoma is not a rare tumor among pregnant women.During pregnancy,the symptoms caused by hysteromyoma can be improved through conservative treatment in most cases.However,in order to ensure the safety ...BACKGROUND Hysteromyoma is not a rare tumor among pregnant women.During pregnancy,the symptoms caused by hysteromyoma can be improved through conservative treatment in most cases.However,in order to ensure the safety of mothers and children,surgeries are necessary in some special cases.CASE SUMMARY We report a case of pregnancy complicated with hysteromyoma red degeneration.The patient had peritonitis after sudden abdominal pain during the 20th week of pregnancy.Laparoscopic exploration suggested rupture and bleeding of hysteromyoma,which were improved after drainage and an anti-inflammatory treatment.A cesarean section was performed after full term.This case shows the complications of rupture after red degeneration of hysteromyoma during pregnancy.CONCLUSION We should be alert to rupture of hysteromyoma during pregnancy,and active laparoscopic exploration is essential to improve the prognosis of such patients.展开更多

文摘As positioning sensors,edge computation power,and communication technologies continue to develop,a moving agent can now sense its surroundings and communicate with other agents.By receiving spatial information from both its environment and other agents,an agent can use various methods and sensor types to localize itself.With its high flexibility and robustness,collaborative positioning has become a widely used method in both military and civilian applications.This paper introduces the basic fundamental concepts and applications of collaborative positioning,and reviews recent progress in the field based on camera,LiDAR(Light Detection and Ranging),wireless sensor,and their integration.The paper compares the current methods with respect to their sensor type,summarizes their main paradigms,and analyzes their evaluation experiments.Finally,the paper discusses the main challenges and open issues that require further research.
文摘Background: Resistance to cisplatin (DDP) leads to poor prognosis in patients with Lung Adenocarcinoma (LUAD) and limits its clinical application. It has been confirmed that autophagy promotes chemoresistance and, therefore, novel strategies to reverse chemoresistance by regulating autophagy are desperately needed. Methods: The differentially expressed lncRNAs (DElncRNAs), miRNAs (DEmiRNAs), and mRNAs (DEmRNAs) between A549 and A549/DDP cell lines were identified using the limma package in R, after gene expression profiles were obtained from Gene Expression Omnibus (GEO) database. By combining Autophagy-Related Genes (ARGs) from Human Autophagy Database (HADb), the interactions lncRNA-miRNAs and the interactions miRNAs-mRNAs respectively predicted by miRcode and miRDB/Targetscan database, the autophagy-related ceRNA network was constructed. Then, extraction of ceRNA subnetwork and Cox regression analyses were performed. A prognosis-related ceRNA subnetwork was constructed, and the upstream Transcription Factors (TFs) regulating lncRNAs were predicted by the JASPAR database. Finally, the expression patterns of candidate genes were further verified by quantitative real-time polymerase chain reaction (qRT-PCR) experiments. Results: A total of 3179 DEmRNAs, 180 DEmiRNAs, and 160 DElncRNAs were identified, and 35 DEmRNAs were contained in the HADb. Based on the ceRNA hypothesis, we established a ceRNA network, including 10 autophagy-related DEmRNAs, 9 DEmiRNAs, and 14 DElncRNAs. Then, LINC00520, miR-181d, and BCL2 were identified to construct a risk score model, which was confirmed to be a well-predicting prognostic factor. Furthermore, 5 TF ZNF family members were predicted to regulate LINC00520, whereas the RT-PCR results showed that the 5 ZNFs were consistent with the bioinformatics analysis. Finally, a ZNF regulatory LINC00520/miR-181d/BCL2 ceRNA subnetwork was constructed. Conclusions: An ZNFs/LINC00520/miR-181d/BCL2 axis as a novel network in DDP-resistant LUAD has been constructed successfully, which may provide potential therapeutic targets for LUAD.

文摘Since its introduction,discontinuous deformation analysis(DDA)has been widely used in different areas of rock mechanics.By dividing large blocks into subblocks and introducing artificial joints,DDA can be applied to rock fracture simulation.However,parameter calibration,a fundamental issue in discontinuum methods,has not received enough attention in DDA.In this study,the parameter calibration of DDA for intact rock is carefully studied.To this end,a subblock DDA with Voronoi tessellation is presented first.Then,a modified contact constitutive law is introduced,in which the tensile and shear meso-strengths are modified to be independent of the bond lengths.This improvement can prevent the unjustified preferential failure of short edges.A method for imposing confining pressure is also introduced.Thereafter,sensitivity analysis is performed to investigate the influence of the calculated parameters and meso-parameters on the mechanical properties of modeled rock.Based on the sensitivity analysis,a unified calibration procedure is suggested for both cases with and without confining pressure.Finally,the calibration procedure is applied to two examples,including a biaxial compression test.The results show that the proposed Voronoi-based DDA can simulate rock fracture with and without confining pressure very well after careful parameter calibration.

文摘The onset of retinal degenerative disease is often associated with neuronal loss. Therefore, how to regenerate new neurons to restore vision is an important issue. NeuroD1 is a neural transcription factor with the ability to reprogram brain astrocytes into neurons in vivo. Here, we demonstrate that in adult mice, NeuroD1 can reprogram Müller cells, the principal glial cell type in the retina, to become retinal neurons. Most strikingly, ectopic expression of NeuroD1 using two different viral vectors converted Müller cells into different cell types. Specifically, AAV7 m8 GFAP681::GFP-ND1 converted Müller cells into inner retinal neurons, including amacrine cells and ganglion cells. In contrast, AAV9 GFAP104::ND1-GFP converted Müller cells into outer retinal neurons such as photoreceptors and horizontal cells, with higher conversion efficiency. Furthermore, we demonstrate that Müller cell conversion induced by AAV9 GFAP104::ND1-GFP displayed clear dose-and time-dependence. These results indicate that Müller cells in adult mice are highly plastic and can be reprogrammed into various subtypes of retinal neurons.

文摘By introducing the dimensional splitting(DS)method into the multiscale interpolating element-free Galerkin(VMIEFG)method,a dimension-splitting multiscale interpolating element-free Galerkin(DS-VMIEFG)method is proposed for three-dimensional(3D)singular perturbed convection-diffusion(SPCD)problems.In the DSVMIEFG method,the 3D problem is decomposed into a series of 2D problems by the DS method,and the discrete equations on the 2D splitting surface are obtained by the VMIEFG method.The improved interpolation-type moving least squares(IIMLS)method is used to construct shape functions in the weak form and to combine 2D discrete equations into a global system of discrete equations for the three-dimensional SPCD problems.The solved numerical example verifies the effectiveness of the method in this paper for the 3D SPCD problems.The numerical solution will gradually converge to the analytical solution with the increase in the number of nodes.For extremely small singular diffusion coefficients,the numerical solution will avoid numerical oscillation and has high computational stability.

文摘Cerebral ischemia is a serious disease that triggers sequential pathological mechanisms, leading to significant morbidity and mortality. Although most studies to date have typically focused on the lysosome, a single organelle, current evidence supports that the function of lysosomes cannot be separated from that of the endolysosomal system as a whole. The associated membrane fusion functions of this system play a crucial role in the biodegradation of cerebral ischemia-related products. Here, we review the regulation of and the changes that occur in the endolysosomal system after cerebral ischemia, focusing on the latest research progress on membrane fusion function. Numerous proteins, including N-ethylmaleimide-sensitive factor and lysosomal potassium channel transmembrane protein 175, regulate the function of this system. However, these proteins are abnormally expressed after cerebral ischemic injury, which disrupts the normal fusion function of membranes within the endolysosomal system and that between autophagosomes and lysosomes. This results in impaired “maturation” of the endolysosomal system and the collapse of energy metabolism balance and protein homeostasis maintained by the autophagy-lysosomal pathway. Autophagy is the final step in the endolysosomal pathway and contributes to maintaining the dynamic balance of the system. The process of autophagosome-lysosome fusion is a necessary part of autophagy and plays a crucial role in maintaining energy homeostasis and clearing aging proteins. We believe that, in cerebral ischemic injury, the endolysosomal system should be considered as a whole rather than focusing on the lysosome. Understanding how this dynamic system is regulated will provide new ideas for the treatment of cerebral ischemia.

文摘In response to spinal surgery,neurons secrete a large amount of substance P into the epidural area.Substance P is involved in macrophage differentiation and fibrotic disease.However,the specific roles and mechanisms of substance P in epidural fibrosis remain unclear.In this study,we established a mouse model of L1–L3 laminectomy and found that dorsal root ganglion neurons and the macrophages infiltrating into the wound area released sphingolipids.In vitro experiments revealed that type 1 macrophages secreted substance P,which promoted differentiation of type 1 macrophages towards a type 2 phenotype.High-throughput mRNA-seq analysis revealed that the sphingolipid metabolic pathway may be involved in the regulation of type 2 macrophages by substance P.Specifically,sphingomyelin synthase 2,a component of the sphingolipid metabolic pathway,promoted M2 differentiation in substance P-treated macrophages,while treating the macrophages with LY93,a sphingomyelin synthase 2 inhibitor,suppressed M2 differentiation.In addition,substance P promoted the formation of neutrophil extracellular traps,which further boosted M2 differentiation.Blocking substance P with the neurokinin receptor 1 inhibitor RP67580 decreased the number of M2 macrophages in the wound area after spinal surgery and alleviated epidural fibrosis,as evidenced by decreased fibronectin,α-smooth muscle actin,and collagen I in the scar tissue.These results demonstrated that substance P promotes M2 macrophage differentiation in epidural fibrosis via sphingomyelin synthase 2 and neutrophil extracellular traps.These findings provide a novel strategy for the treatment of epidural fibrosis.

文摘The Ordos Basin is the largest continental multi-energy mineral basin in China,which is rich in coal,oil and gas,and uranium resources.The exploitation of mineral resources is closely related to reservoir water.The chemical properties of reservoir water are very important for reservoir evaluation and are significant indicators of the sealing of reservoir oil and gas resources.Therefore,the caprock of the Chang 6 reservoir in the Yanchang Formation was evaluated.The authors tested and analyzed the chemical characteristics of water samples selected from 30 wells in the Chang 6 reservoir of Ansai Oilfield in the Ordos Basin.The results show that the Chang 6 reservoir water in Ansai Oilfield is dominated by calcium-chloride water type with a sodium chloride coefficient of generally less than 0.5.The chloride magnesium coefficients are between 33.7 and 925.5,most of which are greater than 200.The desulfurization coefficients range from 0.21 to 13.4,with an average of 2.227.The carbonate balance coefficients are mainly concentrated below 0.01,with an average of 0.008.The calcium and magnesium coefficients are between 0.08 and 0.003,with an average of 0.01.Combined with the characteristics of the four-corner layout of the reservoir water,the above results show that the graphics are basically consistent.The study indicates that the Chang 6 reservoir in Ansai Oilfield in the Ordos Basin is a favorable block for oil and gas storage with good sealing properties,great preservation conditions of oil and gas,and high pore connectivity.

文摘Strabismus and amblyopia are common ophthalmologic developmental diseases caused by abnormal visual experiences. However, the underlying pathogenesis and visual defects are still not fully understood. Most studies have used experimental interference to establish diseaseassociated animal models, while ignoring the natural pathophysiological mechanisms. This study was designed to investigate whether natural strabismus and amblyopia are associated with abnormal neurological defects. We screened one natural strabismic monkey(Macaca fascicularis) and one natural amblyopic monkey from hundreds of monkeys, and retrospectively analyzed one human strabismus case. Neuroimaging, behavioral,neurophysiological, neurostructural, and genovariation features were systematically evaluated using magnetic resonance imaging(MRI), behavioral tasks, flash visual evoked potentials(FVEP),electroretinogram(ERG), optical coherence tomography(OCT), and whole-genome sequencing(WGS), respectively. Results showed that the strabismic patient and natural strabismic and amblyopic monkeys exhibited similar abnormal asymmetries in brain structure, i.e., ipsilateral impaired right hemisphere. Visual behavior, visual function, retinal structure, and fundus of the monkeys were impaired. Aberrant asymmetry in binocular visual function and structure between the strabismic and amblyopic monkeys was closely related, with greater impairment of the left visual pathway.Several similar known mutant genes for strabismus and amblyopia were also identified. In conclusion,natural strabismus and amblyopia are accompanied by abnormal asymmetries of the visual system,especially visual neurophysiological and neurostructural defects. Our results suggest that future therapeutic and mechanistic studies should consider defects and asymmetries throughout the entire visual system.

文摘Using allelochemicals to suppress cyanobacteria growth is a prospective method for its high efficiency and ecological safety.However,the suppression efficiency is affected inevitably by the extracellular polymeric substances(EPS)produced by cyanobacteria,and the knowledge about the roles of EPS in resistance to allelochemical stress is scarce.For the study,two typical anti-cyanobacterial allelochemicals were adopted to investigate the role of EPS in resistance to allelochemical stress on Microcystis aeruginosa.Results show that EPS was crucial in alleviating the toxicity of allelochemicals to algae,especially in stabilizing the metabolism and photosynthetic activity of algal cells.The aggregation rate of algal cells increased with the increase of EPS secretion,which alleviated the stress of allelopathy.Tryptophan proteins and humic acids in EPS provided a binding site for allelochemicals,and the EPS-allelochemicals complex were formed by chemical bonding.This study improved our comprehension of the role of EPS in algal inhibition by allelochemicals.

文摘Engineering disasters(e.g.rock slabbing and rockburst)of the tunnel groups induced by the transient excavation of an adjacent tunnel threaten the stability of the existing tunnel,especially for those excavated by using the drill and blast tunneling(D&B).However,the dynamic response and failure mechanism of surrounding rocks of the existing tunnel caused by adjacent transient excavation are not clear due to the difficulty in conducting field tests and laboratory experiments.Therefore,a novel transient unloading experimental system for deep tunnel excavation was proposed in this study.The real stress path and the unloading rate can be reproduced by using this proposed system.The experiments were conducted for observing the dynamic response of the existing tunnel induced by adjacent transient excavation under different lateral pressure coefficients l(?0.4,0.6,0.8,1,1.2,1.4,1.6,1.8)with a polymethyl methacrylate(PMMA)specimen.The propagation of the impact wave and unloading surface wave was detected through the digital image correlation(DIC)analysis.The reflection of the unloading surface wave on the incident side of the existing tunnel(tunnel-E)was observed and analyzed.Moreover,the dynamic characteristics of the stress redistribution,the particle displacement and vibration velocity of surrounding rocks of tunnel-E were analyzed and summarized.In addition,the Mohr-Coulomb(MeC)failure criterion with tension cut-off was adopted to evaluate the stability of the existing tunnel under adjacent transient excavation.The results indicate that the incident side of the existing tunnel under the dynamic disturbance of transient excavation of an adjacent tunnel was more prone to fail,followed by the shadow side and the top/bottom side.

文摘Based on 20 models from phase 6 of the Coupled Model Intercomparison Project(CMIP6),this article explored possible reasons for differences in simulation biases and projected changes in precipitation in northern China among the allmodel ensemble(AMME),“highest-ranked”model ensemble(BMME),and“lowest-ranked”model ensemble(WMME),from the perspective of atmospheric circulations and moisture budgets.The results show that the BMME and AMME reproduce the East Asian winter circulations better than the WMME.Compared with the AMME and WMME,the BMME reduces the overestimation of evaporation,thereby improving the simulation of winter precipitation.The three ensemble simulated biases for the East Asian summer circulations are generally similar,characterized by a stronger zonal pressure gradient between the mid-latitudes of the North Pacific and East Asia and a northward displacement of the East Asian westerly jet.However,the simulated vertical moisture advection is improved in the BMME,contributing to the slightly higher performance of the BMME than the AMME and WMME on summer precipitation in North and Northeast China.Compared to the AMME and WMME,the BMME projects larger increases in precipitation in northern China during both seasons by the end of the 21st century under the Shared Socioeconomic Pathway 5-8.5(SSP5-8.5).One of the reasons is that the increase in evaporation projected by the BMME is larger.The projection of a greater dynamic contribution by the BMME also plays a role.In addition,larger changes in the nonlinear components in the BMME projection contribute to a larger increase in winter precipitation in northern China.
文摘The Global Navigation Satellite System(GNSS)has been widely used in various fields.To achieve positioning,the receiver must first lock the satellite signal.This is a complicated and expensive process that consumes a lot of resources of the receiver.For this reason,this paper proposes a new fast acquisition algorithm with High Signal-tonoise ratio(SNR)performance based on sparse fast Fourier transform(HSFFT).The algorithm first replaces the IFFT process of the traditional parallel code phase capture algorithm with inverse sparse fast Fourier transform(ISFFT)with better computing performance,and then uses linear search combined with code phase discrimination to replace the positioning loop and the estimation loop with poor noise immunity in ISFFT.Theoretical analysis and simulation results show that,compared with the existing SFFT parallel code phase capture algorithm,the calculation amount of this algorithm is reduced by 19%,and the SNR performance is improved by about 5dB.Compared with the classic FFT parallel code phase capture algorithm,the calculation amount of the algorithm in this paper is reduced by 43%,and when the capture probability is greater than 95%,the SNR performance of the two is approximately the same.
文摘BACKGROUND Hysteromyoma is not a rare tumor among pregnant women.During pregnancy,the symptoms caused by hysteromyoma can be improved through conservative treatment in most cases.However,in order to ensure the safety of mothers and children,surgeries are necessary in some special cases.CASE SUMMARY We report a case of pregnancy complicated with hysteromyoma red degeneration.The patient had peritonitis after sudden abdominal pain during the 20th week of pregnancy.Laparoscopic exploration suggested rupture and bleeding of hysteromyoma,which were improved after drainage and an anti-inflammatory treatment.A cesarean section was performed after full term.This case shows the complications of rupture after red degeneration of hysteromyoma during pregnancy.CONCLUSION We should be alert to rupture of hysteromyoma during pregnancy,and active laparoscopic exploration is essential to improve the prognosis of such patients.
