The cancer cell metastasis is a major death reason for patients with non-small cell lung cancer(NSCLC).Although researchers have disclosed that interleukin 17(IL-17)can increase matrix metalloproteinases(MMPs)inductio...The cancer cell metastasis is a major death reason for patients with non-small cell lung cancer(NSCLC).Although researchers have disclosed that interleukin 17(IL-17)can increase matrix metalloproteinases(MMPs)induction causing NSCLC cell metastasis,the underlying mechanism remains unclear.In the study,we found that IL-17 receptor A(IL-17RA),p300,p-STAT3,Ack-STAT3,and MMP19 were up-regulated both in NSCLC tissues and NSCLC cells stimulated with IL-17.p300,STAT3 and MMP19 overexpression or knockdown could raise or reduce IL-17-induced p-STAT3,Ack-STAT3 and MMP19 level as well as the cell migration and invasion.Mechanism investigation revealed that STAT3 and p300 bound to the same region(−544 to−389 nt)of MMP19 promoter,and p300 could acetylate STAT3-K631 elevating STAT3 transcriptional activity,p-STAT3 or MMP19 expression and the cell mobility exposed to IL-17.Meanwhile,p300-mediated STAT3-K631 acetylation and its Y705-phosphorylation could interact,synergistically facilitating MMP19 gene transcription and enhancing cell migration and invasion.Besides,the animal experiments exhibited that the nude mice inoculated with NSCLC cells by silencing p300,STAT3 or MMP19 gene plus IL-17 treatment,the nodule number,and MMP19,Ack-STAT3,or p-STAT3 production in the lung metastatic nodules were all alleviated.Collectively,these outcomes uncover that IL-17-triggered NSCLC metastasis involves up-regulating MMP19 expression via the interaction of STAT3-K631 acetylation by p300 and its Y705-phosphorylation,which provides a new mechanistic insight and potential strategy for NSCLC metastasis and therapy.展开更多
Two-dimensional(2D)nonlinear optical mediums with high and tunable light modulation capability can significantly stimulate the development of ultrathin,compact,and integrated optoelectronics devices and photonic eleme...Two-dimensional(2D)nonlinear optical mediums with high and tunable light modulation capability can significantly stimulate the development of ultrathin,compact,and integrated optoelectronics devices and photonic elements.2D carbides and nitrides of transition metals(MXenes)are a new class of 2D materials with excellent intrinsic and strong light-matter interaction characteristics.However,the current understanding of their photo-physical properties and strategies for improving optical performance is insufficient.To address this issue,we rationally designed and in situ synthesized a 2D Nb_(2)C/MoS_(2) heterostructure that outperforms pristine Nb2C in both linear and nonlinear optical performance.Excellent agreement between experimental and theoretical results demonstrated that the Nb_(2)C/MoS_(2) inherited the preponderance of Nb_(2)C and MoS_(2) in absorption at different wavelengths,resulting in the broadband enhanced optical absorption characteristics.In addition to linear optical modulation,we also achieved stronger near infrared nonlinear optical modulation,with a nonlinear absorption coefficient of Nb_(2)C/MoS_(2) being more than two times that of the pristine Nb_(2)C.These results were supported by the band alinement model which was determined by the X-ray photoelectron spectroscopy(XPS)experiment and first-principal theory calculation.The presented facile synthesis approach and robust light modulation strategy pave the way for broadband optoelectronic devices and optical modulators.展开更多
Mycobacterium tuberculosis 6-kDa early secretory antigenic target(ESAT-6)is a dominant target antigen for cell-mediated immunity in the early phase of tuberculosis.The fms-like tyrosine kinase 3 ligand(FL)that induces...Mycobacterium tuberculosis 6-kDa early secretory antigenic target(ESAT-6)is a dominant target antigen for cell-mediated immunity in the early phase of tuberculosis.The fms-like tyrosine kinase 3 ligand(FL)that induces potent immune response has been used as an adjuvant in vaccine development.In this study,a new recombinant plasmid(pIRES-epitope-peptides-FL)encoding three T cell epitopes of ESAT-6 and FL was constructed,and the immunogenicity of the DNA vaccine was assessed in C57BL/6 mice immunized with the plasmid DNA vaccine.Additionally,a strategy of intramuscular injection with the DNA vaccine(prime)and intranasal administration of the epitope peptides(boost)was employed to induce higher immune reaction of the mice.The results showed that mice vaccinated with the recombinant plasmid DNA vaccine and boosted with the peptides not only increased the levels of Th1 cytokines(IFN-γand IL-12),the number of IFN-γ+T cells and activities of cytotoxic T lymphocytes as well as IgG,but also enhanced protection against Mycobacterium tuberculosis challenge.In conclusion,these data indicate that the novel recombinant pIRES-epitope-peptides-FL plasmid is a useful DNA vaccine for preventing Mycobacterium tuberculosis infection.展开更多

Our previous studies have demonstrated that the thrombospondin-1 (TSP-1) gene was involved in mediating rat GMC proliferation and ECM synthesis induced by sublytic C5b-9 in vitro. In the present study, the roles of the TSP-1 gene in GMC proliferation, ECM production, and urinary protein secretion in Thy-1N rats were determined by using TSP-1 small hairpin RNA, and the results revealed that silencing of the TSP-1 gene in rat renal tissues could diminish GMC proliferation (P < 0.01) and ECM secretion (P < 0.01) as well as urinary protein secretion (P < 0.05) in Thy-1N rats. Together, the current findings suggested that TSP-1 gene expression was required for GMC proliferation and ECM production in Thy-1N rats.展开更多
This study focuses on the characters of public perceptions on climate and cryosphere change,which are based on a questionnaire survey in the(U|¨)r(u|¨)mqi River Basin.In comparison with scientific observatio...This study focuses on the characters of public perceptions on climate and cryosphere change,which are based on a questionnaire survey in the(U|¨)r(u|¨)mqi River Basin.In comparison with scientific observation results of climate and cryosphere change,this paper analyzes the possible impact of the change on water resources and agriculture production in the area.Perceptions of most respondents on climate and cryosphere changes confirm the main objective facts.For the selection of adaptation measures addressing the shortage of water resource,the results are as follows:most people preferred to choose the measures like "policy change" and "basic facility construction" which are mostly implemented by the government and the policy-making department;some people showed more preference to the measures of avoiding unfavorable natural environment,such as finding job in or migrating to other places.The urgency of personal participation in the adaptation measures is still inadequate.Some adaptation measures should be implemented in line with local conditions and require the organic combination of "resource-development" with "water-saving".展开更多

For this purpose, mobile gravi...This study adopted the Euler deconvolution method to conduct an inversion and interpretation of the depth and spatial distribution pattern of field source that lead to gravity variation. For this purpose, mobile gravity data from four periods in the Hexi region between 2011 and 2015 were obtained from an observation network. With a newly established theoretical model, we acquired the optimum inversion parameters and conducted calculation and analysis with the actual data. The results indicate that one is the appropriate value of the structure index for the inversion of the mobile gravity data. The inversion results of the actual data showed a comparable spatial distribution of the field source and a consistent structural trend with observations from the Qilian-Haiyuan Fault zone between 2011 and 2015. The distribution was in a blocking state at the epicenter of the Menyuan earthquake in 2016. Our quantitative study of the field source provides new insights into the inversion and interpretation of signals of mobile gravity variation.展开更多
