Astrocytic endothelin-1 overexpression impairs learning and memory ability in ischemic stroke via altered hippocampal neurogenesis and lipid metabolism 被引量:1

Vascular etiology is the second most prevalent cause of cognitive impairment globally.Endothelin-1,which is produced and secreted by endothelial cells and astrocytes,is implicated in the pathogenesis of stroke.However,the way in which changes in astrocytic endothelin-1 lead to poststroke cognitive deficits following transient middle cerebral artery occlusion is not well understood.Here,using mice in which astrocytic endothelin-1 was overexpressed,we found that the selective overexpression of endothelin-1 by astrocytic cells led to ischemic stroke-related dementia(1 hour of ischemia;7 days,28 days,or 3 months of reperfusion).We also revealed that astrocytic endothelin-1 overexpression contributed to the role of neural stem cell proliferation but impaired neurogenesis in the dentate gyrus of the hippocampus after middle cerebral artery occlusion.Comprehensive proteome profiles and western blot analysis confirmed that levels of glial fibrillary acidic protein and peroxiredoxin 6,which were differentially expressed in the brain,were significantly increased in mice with astrocytic endothelin-1 overexpression in comparison with wild-type mice 28 days after ischemic stroke.Moreover,the levels of the enriched differentially expressed proteins were closely related to lipid metabolism,as indicated by Kyoto Encyclopedia of Genes and Genomes pathway analysis.Liquid chromatography-mass spectrometry nontargeted metabolite profiling of brain tissues showed that astrocytic endothelin-1 overexpression altered lipid metabolism products such as glycerol phosphatidylcholine,sphingomyelin,and phosphatidic acid.Overall,this study demonstrates that astrocytic endothelin-1 overexpression can impair hippocampal neurogenesis and that it is correlated with lipid metabolism in poststroke cognitive dysfunction. 展开更多

背景与目的自计算机断层扫描(computed tomography,CT)技术普及以来,影像学随访为主要管理方法的磨玻璃结节(ground glass nodules,GGNs)检出率明显增加,本研究旨在应用三维重建技术定量分析GGNs随访过程中的变化情况,探索GGNs自然进展... 背景与目的自计算机断层扫描(computed tomography,CT)技术普及以来,影像学随访为主要管理方法的磨玻璃结节(ground glass nodules,GGNs)检出率明显增加,本研究旨在应用三维重建技术定量分析GGNs随访过程中的变化情况,探索GGNs自然进展规律,为临床指导患者合理地进行结节管理提供有效依据。方法纳入2015年3月-2022年11月舟山医院肺结节联合门诊中规律随访的GGNs患者共115例。利用3DSlicer软件半自动分割提取结节的定量影像学特征,评估随访过程中的结节增长情况及临床干预情况。结果患者平均基线年龄为(56.9±10.1)岁;平均随访时间为(48.8±18.9)个月。CT二维直径为(7.9±2.9)mm,三维最大径为(10.1±3.4)mm。末次CT扫描二维直径为(9.9±4.7)mm,三维最大径为(11.4±5.1)mm。共27例(23.5%)随访期间出现增长,中位体积倍增时间为822 d,中位质量倍增时间为1,007 d。手术切除32例,其中6例浸润性腺癌(invasive adenocarcinoma,IAC),16例微浸润腺癌(minimally invasive adenocarcinoma,MIA),8例原位腺癌(adenocarcinoma in situ,AIS),2例非典型腺瘤样增生(atypical adenomatous hyperplasia,AAH)。5例结节因二维直径显示进展而行手术干预,病理证实为浸润前病变,但其三维最大径提示无明显变化。单因素分析结果显示形态不规则、边缘欠光整,具有分叶、毛刺、空泡征均为促进结节增长的因素;增长组与稳定组年龄、基线直径和平均CT值、中位CT值、10%位数CT值、90%位数CT值差异具有统计学意义(P<0.05)。多因素Logistic回归分析结果表明年龄和平均CT值是影响结节增长的危险因素(P<0.05)。受试者工作特征(receiver-operating characteristic,ROC)曲线分析结果提示年龄≥63岁、基线三维最大径≥9.2 mm、平均CT值≥-507.8 HU的GGNs出现增长的可能性更大;三维最大径≥14.4 mm、平均CT值≥-495.7 HU时恶性概率更高。结论GGNs呈惰性生长过程,随访过程中应用三维测量值意义更大,对于持续存在的GGNs年龄≥63岁、基线三维最大径≥9.2 mm、平均CT值≥-507.8 HU出现增长的可能性更大,但多数结节出现进展后仍预后良好,长期随访是安全的。 展开更多

Halide perovskites with excellent piezoelectric properties,but their poor stability hinders their largescale application.Herein,a sandwich-structured halide perovskite flexible sensor with good stability was developed according to a three-step procedure as follows:(ⅰ) in-situ growth of wool keratinCsPbBr_(3)(WK-CsPbBr_(3)) using wool keratin in interfacial passivation and coating,(ⅱ) electrospinning of a wool keratin-CsPbBr_(3)/polyacrylonitrile(WCP) nanofiber film,and(ⅲ) coating of the WCP nanofiber with polydimethylsiloxane(PDMS) to obtain a sensor(WCPP).The sensor could generate a piezoelectric voltage of 7.8 V at a pressure of 6 kPa in the stages of pressing and releasing,and the output characteristics did not decline even after 10,000 cycles.Compared to the 4-month stability of the perovskite sensor,WCPP sensor exhibited the output performance even after 16 months,which indicated that wool keratin as a multidentate improved the stability of the halide perovskite.Additionally,the sensor displayed a self-cleaning property and could also light up 14 commercial LEDs.The close-loop recycling of the lead halide perovskite was achieved by dissolving the WCP nanofiber film in DMF and then reelectrospinning.Therefore,the method proposed is a step forward for achieving the commercialization of WK-CsPbBr_(3) and providing new avenues for further utilization of wool waste. 展开更多

Impact of Energy Efficiency and Financial Support on Green Upgrading in China’s Industrial Sector

The report from the 20th National Congress of China emphasizes the importance of focusing on the clean,low-carbon,and efficient use of energy,increasing financial support,and promoting green upgrading within the industrial sector.This paper,based on annual data,employs the entropy weight method to construct a comprehensive index reflecting the impact of green upgrading in industrial sectors.To delve deeper,it utilizes the DEA model to measure energy efficiency and its subdivision BCC model to break down energy efficiency into technical and scale efficiency.The financial support landscape is examined from the vantage points of both direct and indirect financing.Using a multivariate time series model,this paper thoroughly investigates the influence of energy efficiency and financial support on the green upgrading of the industrial sector.The findings reveal a significant positive impact of both energy efficiency and financial support on green upgrading in industrial industries.Notably,scale efficiency emerges as the primary driver of energy efficiency.Moreover,indirect financing proves to be more effective in promoting financial support than direct financing.The empirical results retain their robustness even after substituting explanatory variables.The study concludes by contextualizing the research findings within the current real-world scenario,offering practical insights,and proposing specific recommendations. 展开更多

The enhancement of industrial green total factor productivity is pivotal for achieving high-quality and sustainable economic development.This study assesses China’s performance using the SBM-GML model,employing province-level panel data spanning from 2004 to 2020.Furthermore,we examine the influence of green finance and technological progress on industrial green total factor productivity using a spatial econometric model.The findings uncover that the relationship between the level of green financial development and industrial green total factor productivity follows a U-shaped curve.Initially,low levels of green financial development exert a suppressive effect on industrial green total factor productivity,proving ineffective in the short term.However,with the progression of green finance development,a positive and significant long-term impact on industrial green total factor productivity emerges.Moreover,technological progress demonstrates a noteworthy promotional effect on industrial green total factor productivity.The analysis delves deeper into revealing that industrial structure and environmental regulation intensity exhibit a significant negative relationship with industrial green total factor productivity.In contrast,both energy structure and education level showcase a substantial positive relationship with industrial green total factor productivity. 展开更多

Metastasis and paclitaxel(PTX)resistance are the main reason for the poor prognosis of ovarian cancer(OC).Evidence showed that RNA-binding proteins(RBPs)and long noncoding RNAs(lncRNAs)can modulate post-transcriptional regulation.The aim of this study was to determine the relationship among RBP,lncRNA and OC and to further guide clinical therapy.Immunohistochemistry revealed that pre-mRNA processing factor 6(PRPF6)was upregulated in OC chemoresistant tissues and was closely related to advanced(Federation of International of Gynecologists and Obstetricians)FIGO stages and chemo-resistance.PRPF6 promoted progression,and PTX resistance in vitro and in vivo.And the transcripts of small nucleolar RNA host gene SNHG16-L/S were differentially expressed in OC cells and tissues as detected through real-time PCR(RT-PCR).SNHG16-L/S had opposite effects on progression and PTX resistance in OC.Mechanistically,SNHG16-L inhibited GATA-binding protein 3(GATA3)transcription by binding to CCAAT/enhancer-binding protein B(CEBPB).Moreover,PRPF6 induced the alternative splicing of SNHG16,causing downregulation of SNHG16-L and,leading to the upregulation of GATA3 expression to further promote metastasis and PTX-resistance in OC.Totally,these data unveiled that PRPF6 promotes metastasis and PTX resistance of OC through SNHG16-L/CEBPB/GATA3 axis,which provides a new direction for OC treatment. 展开更多

We divide the whole insurance market into two parts, namely property insurance and personal insurance.By constructing and analyzing minimum spanning trees of insurance market, we obtain the results as follows:(i) The connections between provinces are much closer than those of firms, and there are regional links between neighboring provinces in the minimum spanning tree(MST); and(ii) the domestic funded firms and foreign funded firms form two explicit clusters in the MSTs of property and personal insurance market. 展开更多
