Hepatocellular carcinoma(HCC),a prevalent solid carcinoma of significant concern,is an aggressive and often fatal disease with increasing global incidence rates and poor therapeutic outcomes.The etiology and pathologi...Hepatocellular carcinoma(HCC),a prevalent solid carcinoma of significant concern,is an aggressive and often fatal disease with increasing global incidence rates and poor therapeutic outcomes.The etiology and pathological progression of non-alcoholic steatohepatitis(NASH)-related HCC is multifactorial and multistage.However,no single animal model can accurately mimic the full NASH-related HCC pathological progression,posing considerable challenges to transition and mechanistic studies.Herein,a novel conditional inducible wild-type human HRAS overexpressed mouse model(HRAS-HCC)was established,demonstrating 100%morbidity and mortality within approximately one month under normal dietary and lifestyle conditions.Advanced symptoms of HCC such as ascites,thrombus,internal hemorrhage,jaundice,and lung metastasis were successfully replicated in mice.In-depth pathological features of NASH-related HCC were demonstrated by pathological staining,biochemical analyses,and typical marker gene detections.Combined murine anti-PD-1 and sorafenib treatment effectively prolonged mouse survival,further confirming the accuracy and reliability of the model.Based on protein-protein interaction(PPI)network and RNA sequencing analyses,we speculated that overexpression of HRAS may initiate the THBS1-COL4A3 axis to induce NASH with severe fibrosis,with subsequent progression to HCC.Collectively,our study successfully duplicated natural sequential progression in a single murine model over a very short period,providing an accurate and reliable preclinical tool for therapeutic evaluations targeting the NASH to HCC continuum.展开更多
Understanding and monitoring the cross-contamination of food allergens is crucial for safeguarding public health and ensuring food safety.Food allergen risk assessment,derived from classical toxicological principles,c...Understanding and monitoring the cross-contamination of food allergens is crucial for safeguarding public health and ensuring food safety.Food allergen risk assessment,derived from classical toxicological principles,can identify and quantify the risk of allergies.This study aimed to investigate the risk of wheat allergic reactions to prepackaged foods from China through the utilization of food allergen risk assessment.A total of 575 products have been surveyed,wheat/gluten,milk and egg were major allergens labelled on products.According to voluntary incidental trace allergen labelling 3.0(VITAL®3.0)program,the number of products belonged to Action Level 2 were 303.Integration of precautionary allergen labeling(PAL)analysis indicated that 9.57%products would pose a potential risk to wheat allergic individuals.The probabilistic risk assessment results suggest that 7984 allergic reactions may arise among wheat-allergic consumers during 10000 eating occasions due to the consumption of pre-packaged food products with incorrect wheat-related allergen labelling.This study demonstrated that a risk assessment-based approach can support the guidance of allergen labelling and management of food allergen for pre-packaged food products,providing protection for allergic individuals in food consumption and for food manufacturers in food production and trade.展开更多

For warm/hot and dense plasmas(WDPs),ionization potential depression(IPD)plays a crucial role in determining its ionization balance and understanding the resultant microscopic plasma properties.A sophisticated and uni...For warm/hot and dense plasmas(WDPs),ionization potential depression(IPD)plays a crucial role in determining its ionization balance and understanding the resultant microscopic plasma properties.A sophisticated and unified IPD model is necessary to resolve those existing discrepancies between theoretical and experimental results.However,the applicability of those widely used IPD models nowadays is limited,especially for the nonlocal thermodynamic equilibrium(non-LTE)dense plasma produced by short-pulse laser.In this work,we propose an IPD model that considers inelastic atomic processes,in which three-body recombination and collision ionization processes are found to play a crucial role in determining the electron distribution and IPD for a WDP.This IPD model is validated by reproducing latest experimental results of Al plasmas with a wide-range condition of 70 eV–700 eV temperature and 0.2–3 times solid density,as well as a typical non-LTE system of hollow Al ions.It is demonstrated that the present IPD model has a significant temperature dependence due to the consideration of the inelastic collision processes.With a lower computational cost and wider application range of plasma conditions,the proposed model is expected to provide a promising tool to study the ionization balance and the atomic processes,as well as the related radiation and particle transports properties of the WDP.展开更多
Silkworm pupa is a nourishing food with high nutritional value,but its consumption has been greatly limited given its allergenicity.Enzyme hydrolytic technique is recognized as an effective method to reduce the allerg...Silkworm pupa is a nourishing food with high nutritional value,but its consumption has been greatly limited given its allergenicity.Enzyme hydrolytic technique is recognized as an effective method to reduce the allergenicity of protein.In this study,we aimed to investigate the effect of enzymolysis on the allergenicity of silkworm pupa.Crude silkworm pupa protein was extracted through alkali extraction and acid precipitation,which included 5 proteins with the molecular weights ranging from 34 kDa to 76 kDa,and silkworm pupa were then hydrolyzed by alkaline protease.The allergenicity of silkworm pupa protein and its enzymatic hydrolysates was evaluated by establishing BALB/c mice model,and the mice were immunized via intragastric gavage and intraperitoneal injection,respectively.The results indicated that the intraperitoneal inj ection immunization route induced more by detecting with antibodies,histamine and Th2-related cytokines.Moreover,mice treated with silkworm pupa protein peptide displayed no obvious allergic symptoms,indicating that enzyme hydrolytic technique could significantly reduce the allergenicity of silkworm pupa.展开更多
Milk allergy is a common allergic reaction found in infants and young children,most of them appear tolerance after growing up.In this study,infant formula was digested by simulated in vitro digestion method.The potent...Milk allergy is a common allergic reaction found in infants and young children,most of them appear tolerance after growing up.In this study,infant formula was digested by simulated in vitro digestion method.The potential non-allergenic peptides were further screened from undigested products by exclusion of the known epitopes fromβ-lactoglobulin(BLG)andα-lactalbumin(ALA).These potential non-allergenic peptides were synthesized and their transferability were determined by Caco-2 cell monolayer model.Finally,the potential allergenicity were evaluated by KU812 cell degranulation model.The results showed that 7 peptides were screened as non-allergenic sequences,among which were 3 from ALA and 4 from BLG.The Caco-2 cell model showed that all the synthetic peptides were absorbed and transported well.However,only peptide BLG_(107-118)showed potential allegencity by KU812 model.In conclusion,6 peptides,including ALA_(29-51),ALA_(80-90),ALA_(94-103),BLG_(1-20),BLG_(24-50),and BLG_(123-139)were evaluated as hypoallergenic peptides,which could be used for candidates of peptides inducing immune tolerance for persons with milk allergy.展开更多

In order to improve the performance of UAV's autonomous maneuvering decision-making,this paper proposes a decision-making method based on situational continuity.The algorithm in this paper designs a situation eval...In order to improve the performance of UAV's autonomous maneuvering decision-making,this paper proposes a decision-making method based on situational continuity.The algorithm in this paper designs a situation evaluation function with strong guidance,then trains the Long Short-Term Memory(LSTM)under the framework of Deep Q Network(DQN)for air combat maneuvering decision-making.Considering the continuity between adjacent situations,the method takes multiple consecutive situations as one input of the neural network.To reflect the difference between adjacent situations,the method takes the difference of situation evaluation value as the reward of reinforcement learning.In different scenarios,the algorithm proposed in this paper is compared with the algorithm based on the Fully Neural Network(FNN)and the algorithm based on statistical principles respectively.The results show that,compared with the FNN algorithm,the algorithm proposed in this paper is more accurate and forwardlooking.Compared with the algorithm based on the statistical principles,the decision-making of the algorithm proposed in this paper is more efficient and its real-time performance is better.展开更多

Nuclear receptors(NRs), especially the ecdysone receptor(EcR) and ultras...Lycosidae females demonstrate meticulous maternal care of offspring by carrying egg sacs and juvenile spiderlings during the reproductive stage. Nuclear receptors(NRs), especially the ecdysone receptor(EcR) and ultraspiracle(USP), have attracted considerable attention in the regulation of arthropod development and reproduction due to their pivotal roles in ecdysteroid signaling cascades. In the present study, 23 NRs, including one EcR and two USPs, were identified in the genome of the predatory wolf spider Pardosa pseudoannulata. RNA interference(RNAi) targeting EcR and USP-1inhibited spiderling development and resulted in nonviable eggs in the egg sacs. EcR and USP-1responded to changes in ecdysteroid levels, and interference in ecdysteroid biosynthesis led to similar phenotypes as ds EcR and ds USP-1 treatments.These findings suggest that EcR/USP-1-mediated ecdysteroid signaling regulates P. pseudoannulata development and reproduction. The P.pseudoannulata females with suppressed ecdysteroid signaling proactively consumed their non-viable egg sacs, resulting in a 7.19 d shorter first reproductive cycle than the controls. Termination of the failed reproductive cycle enabled the spiders to produce a new egg sac more rapidly. This reproductive strategy may partially rescue the reduction in population growth due to non-viable eggs and compensate for the physiological expenditure of wasted maternal care, which would be beneficial for the conservation of P.pseudoannulata populations and their natural control of insect pests.展开更多

In order to reduce the titanium layer thickness, the explosive welding of TA1 titanium foil to Q235 steel plate was carried out. The interfacial bonding performance was analyzed by micromorphology analysis and mechanical property test, and the formation process of interfacial wave and molten block in the vortex was simulated by smoothed particle hydrodynamics(SPH) method. The results showed that salt as pressure transfer layer used in explosive welding could play a good buffer effect on the collision between flyer and base layers. Regular waveforms were formed on the bonding interface, and the titanium foil/steel clad plate exhibited good welding quality and bonding property. The crest of the observed interfacial wave moved 200 μm from the beginning to the final formation, and it was important of jet on the formation of interfacial waveform. The interface was mainly bonded in the form of molten layer, and the grains near the interface were streamlined. Molten block containing intermetallic compounds and metal oxides appeared in the vortex of wave crest.展开更多
The low-energy mutual neutralization(MN)reactions Na^(+)+H^(-)→Na(nl)+H have been studied by employing the full quantum-mechanical molecular-orbital close-coupling(QMOCC)method over a wide energy range of 10^(-3)-10^...The low-energy mutual neutralization(MN)reactions Na^(+)+H^(-)→Na(nl)+H have been studied by employing the full quantum-mechanical molecular-orbital close-coupling(QMOCC)method over a wide energy range of 10^(-3)-10^(3) e V/u.Total and state-selective cross sections have been investigated and compared with the available theoretical and experimental data,and the state-selective rate coefficients for the temperature range of 100-10000 K have been obtained.In the present work,all the necessary highly excited states are included,and the influences of rotational couplings and 10 active electrons are considered.It is found that in the energy below 10 e V/u,the Na(4s)state is the most dominant exit state with a contribution of approximately 78%to the branch fraction,which is in best agreement with the experimental data.For energies above 10 e V/u,the MN total cross section is larger than those obtained in other theoretical calculations and shows a slow decreasing trend because the main exit states change,when the energy is above 100 e V/u,the dominant exit state becomes the Na(3p)state,while the Na(4s)state becomes the third most important exit state.The datasets presented in this paper,including the potential energy curve,the radial and rotational couplings,the total and state-selective cross sections,are openly available at https://doi.org/10.57760/sciencedb.j00113.00112.展开更多
Spectral line widths produced by collisions between charged particles and emitters are of special interest for precise plasma spectroscopy.The highly charged Ar XV ion is demonstrated to have strong intrashell electro...Spectral line widths produced by collisions between charged particles and emitters are of special interest for precise plasma spectroscopy.The highly charged Ar XV ion is demonstrated to have strong intrashell electron interactions,which manifest as an atomic system with many resonance structures,due to the quasi-degeneracy of orbital energies.In this paper we use the relativistic R-matrix method to investigate the electron-impact broadening of highly charged Ar XV ion spectral lines under the impact approximation.It is found that the results considering resonance structures are significantly different from those of the distorted wave approach.Furthermore,we propose a new empirical formula with a correction term to take into account the effect of resonances for electron-impact widths over a relatively wide range of plasma conditions.The corresponding fitting parameters of the new empirical formula for all 47 calculated transitions are also given with an estimated accuracy within 1%,which should be convenient for practical applications.The dataset that supported the findings of this study is available in Science Data Bank,with the link https://doi.org/10.57760/sciencedb.j00113.00101.展开更多
Background Methamphetamine(METH)addiction causes a huge burden on society.The prefrontal cortex(PFC),associated with emotion and cognitive behaviours,is also involved in addiction neurocircuitry.Although bulk RNA sequ...Background Methamphetamine(METH)addiction causes a huge burden on society.The prefrontal cortex(PFC),associated with emotion and cognitive behaviours,is also involved in addiction neurocircuitry.Although bulk RNA sequencing has shown METH-induced gene alterations in the mouse PFC,the impact on different cell types remains unknown.Aims To clarify the effects of METH treatment on different cell types of the PFC and the potential pathways involved in METH-related disorders.Methods We performed single-nucleus RNA sequencing(snRNA-seq)to examine the transcriptomes of 20465 nuclei isolated from the PFC of chronic METH-treated and control mice.Main cell types and differentially expressed genes(DEGs)were identified and confirmed by RNA fluorescence in situ hybridization(FISH).Results Six main cell types were identified depending on the single-cell nucleus sequencing;of particular interest were the mature oligodendrocytes in the PFC.The DEGs of mature oligodendrocytes were enriched in the myelin sheath,adenosine triphosphate(ATP)metabolic process,mitochondrial function and components,and so on.The messenger RNA levels of Aldoc and Atp5l(FISH)and the protein level of the mitochondrial membrane pore subunit TOM40(immunofluorescence)decreased in the mature oligodendrocytes.Fast blue staining and transmission electron microscopy image indicated myelin damage,and the myelin thickness decreased in METH brains.Conclusions snRNA-seq reveals altered transcriptomes of different cell types in mouse PFC induced by chronic METH treatment,underscoring potential relationships with psychiatric disorders.展开更多

The relationship among microstructure,mechanical properties and texture of TA32 titanium alloy sheets during hot tensile deformation at 800℃was investigated.In the test,the original sheet exhibited relatively low flo...The relationship among microstructure,mechanical properties and texture of TA32 titanium alloy sheets during hot tensile deformation at 800℃was investigated.In the test,the original sheet exhibited relatively low flow stress and sound plasticity,and increasing the heat treatment temperature resulted in an increased ultimate tensile strength(UTS)and a decreased elongation(EL).The deformation mechanism of TA32 alloy was dominated by high angle grain boundaries sliding and coordinated by dislocation motion.The coarsening of grains and the annihilation of dislocations in heat-treated specimens weakened the deformation ability of material,which led to the increase in flow stress.Based on the high-temperature creep equation,the quantitative relationship between microstructure and flow stress was established.The grain size exponent andαphase strength constant of TA32 alloy were calculated to be 1.57 and 549.58 MPa,respectively.The flow stress was accurately predicted by combining with the corresponding phase volume fraction and grain size.Besides,the deformation behavior of TA32 alloy was also dependent on the orientation of predominantαphase,and the main slip mode was the activation of prismaticslip system.The decrease of near prism-oriented texture in heat-treated specimens resulted in the enhancement of strength of the material.展开更多

文摘For warm/hot and dense plasmas(WDPs),ionization potential depression(IPD)plays a crucial role in determining its ionization balance and understanding the resultant microscopic plasma properties.A sophisticated and unified IPD model is necessary to resolve those existing discrepancies between theoretical and experimental results.However,the applicability of those widely used IPD models nowadays is limited,especially for the nonlocal thermodynamic equilibrium(non-LTE)dense plasma produced by short-pulse laser.In this work,we propose an IPD model that considers inelastic atomic processes,in which three-body recombination and collision ionization processes are found to play a crucial role in determining the electron distribution and IPD for a WDP.This IPD model is validated by reproducing latest experimental results of Al plasmas with a wide-range condition of 70 eV–700 eV temperature and 0.2–3 times solid density,as well as a typical non-LTE system of hollow Al ions.It is demonstrated that the present IPD model has a significant temperature dependence due to the consideration of the inelastic collision processes.With a lower computational cost and wider application range of plasma conditions,the proposed model is expected to provide a promising tool to study the ionization balance and the atomic processes,as well as the related radiation and particle transports properties of the WDP.

文摘Silkworm pupa is a nourishing food with high nutritional value,but its consumption has been greatly limited given its allergenicity.Enzyme hydrolytic technique is recognized as an effective method to reduce the allergenicity of protein.In this study,we aimed to investigate the effect of enzymolysis on the allergenicity of silkworm pupa.Crude silkworm pupa protein was extracted through alkali extraction and acid precipitation,which included 5 proteins with the molecular weights ranging from 34 kDa to 76 kDa,and silkworm pupa were then hydrolyzed by alkaline protease.The allergenicity of silkworm pupa protein and its enzymatic hydrolysates was evaluated by establishing BALB/c mice model,and the mice were immunized via intragastric gavage and intraperitoneal injection,respectively.The results indicated that the intraperitoneal inj ection immunization route induced more by detecting with antibodies,histamine and Th2-related cytokines.Moreover,mice treated with silkworm pupa protein peptide displayed no obvious allergic symptoms,indicating that enzyme hydrolytic technique could significantly reduce the allergenicity of silkworm pupa.

文摘Milk allergy is a common allergic reaction found in infants and young children,most of them appear tolerance after growing up.In this study,infant formula was digested by simulated in vitro digestion method.The potential non-allergenic peptides were further screened from undigested products by exclusion of the known epitopes fromβ-lactoglobulin(BLG)andα-lactalbumin(ALA).These potential non-allergenic peptides were synthesized and their transferability were determined by Caco-2 cell monolayer model.Finally,the potential allergenicity were evaluated by KU812 cell degranulation model.The results showed that 7 peptides were screened as non-allergenic sequences,among which were 3 from ALA and 4 from BLG.The Caco-2 cell model showed that all the synthetic peptides were absorbed and transported well.However,only peptide BLG_(107-118)showed potential allegencity by KU812 model.In conclusion,6 peptides,including ALA_(29-51),ALA_(80-90),ALA_(94-103),BLG_(1-20),BLG_(24-50),and BLG_(123-139)were evaluated as hypoallergenic peptides,which could be used for candidates of peptides inducing immune tolerance for persons with milk allergy.

文摘By introducing the dimensional splitting(DS)method into the multiscale interpolating element-free Galerkin(VMIEFG)method,a dimension-splitting multiscale interpolating element-free Galerkin(DS-VMIEFG)method is proposed for three-dimensional(3D)singular perturbed convection-diffusion(SPCD)problems.In the DSVMIEFG method,the 3D problem is decomposed into a series of 2D problems by the DS method,and the discrete equations on the 2D splitting surface are obtained by the VMIEFG method.The improved interpolation-type moving least squares(IIMLS)method is used to construct shape functions in the weak form and to combine 2D discrete equations into a global system of discrete equations for the three-dimensional SPCD problems.The solved numerical example verifies the effectiveness of the method in this paper for the 3D SPCD problems.The numerical solution will gradually converge to the analytical solution with the increase in the number of nodes.For extremely small singular diffusion coefficients,the numerical solution will avoid numerical oscillation and has high computational stability.

文摘In order to improve the performance of UAV's autonomous maneuvering decision-making,this paper proposes a decision-making method based on situational continuity.The algorithm in this paper designs a situation evaluation function with strong guidance,then trains the Long Short-Term Memory(LSTM)under the framework of Deep Q Network(DQN)for air combat maneuvering decision-making.Considering the continuity between adjacent situations,the method takes multiple consecutive situations as one input of the neural network.To reflect the difference between adjacent situations,the method takes the difference of situation evaluation value as the reward of reinforcement learning.In different scenarios,the algorithm proposed in this paper is compared with the algorithm based on the Fully Neural Network(FNN)and the algorithm based on statistical principles respectively.The results show that,compared with the FNN algorithm,the algorithm proposed in this paper is more accurate and forwardlooking.Compared with the algorithm based on the statistical principles,the decision-making of the algorithm proposed in this paper is more efficient and its real-time performance is better.

文摘Lycosidae females demonstrate meticulous maternal care of offspring by carrying egg sacs and juvenile spiderlings during the reproductive stage. Nuclear receptors(NRs), especially the ecdysone receptor(EcR) and ultraspiracle(USP), have attracted considerable attention in the regulation of arthropod development and reproduction due to their pivotal roles in ecdysteroid signaling cascades. In the present study, 23 NRs, including one EcR and two USPs, were identified in the genome of the predatory wolf spider Pardosa pseudoannulata. RNA interference(RNAi) targeting EcR and USP-1inhibited spiderling development and resulted in nonviable eggs in the egg sacs. EcR and USP-1responded to changes in ecdysteroid levels, and interference in ecdysteroid biosynthesis led to similar phenotypes as ds EcR and ds USP-1 treatments.These findings suggest that EcR/USP-1-mediated ecdysteroid signaling regulates P. pseudoannulata development and reproduction. The P.pseudoannulata females with suppressed ecdysteroid signaling proactively consumed their non-viable egg sacs, resulting in a 7.19 d shorter first reproductive cycle than the controls. Termination of the failed reproductive cycle enabled the spiders to produce a new egg sac more rapidly. This reproductive strategy may partially rescue the reduction in population growth due to non-viable eggs and compensate for the physiological expenditure of wasted maternal care, which would be beneficial for the conservation of P.pseudoannulata populations and their natural control of insect pests.
文摘Ti/Fe clad plate had attracted extensive attention because of its important application. In order to reduce the titanium layer thickness, the explosive welding of TA1 titanium foil to Q235 steel plate was carried out. The interfacial bonding performance was analyzed by micromorphology analysis and mechanical property test, and the formation process of interfacial wave and molten block in the vortex was simulated by smoothed particle hydrodynamics(SPH) method. The results showed that salt as pressure transfer layer used in explosive welding could play a good buffer effect on the collision between flyer and base layers. Regular waveforms were formed on the bonding interface, and the titanium foil/steel clad plate exhibited good welding quality and bonding property. The crest of the observed interfacial wave moved 200 μm from the beginning to the final formation, and it was important of jet on the formation of interfacial waveform. The interface was mainly bonded in the form of molten layer, and the grains near the interface were streamlined. Molten block containing intermetallic compounds and metal oxides appeared in the vortex of wave crest.

文摘The low-energy mutual neutralization(MN)reactions Na^(+)+H^(-)→Na(nl)+H have been studied by employing the full quantum-mechanical molecular-orbital close-coupling(QMOCC)method over a wide energy range of 10^(-3)-10^(3) e V/u.Total and state-selective cross sections have been investigated and compared with the available theoretical and experimental data,and the state-selective rate coefficients for the temperature range of 100-10000 K have been obtained.In the present work,all the necessary highly excited states are included,and the influences of rotational couplings and 10 active electrons are considered.It is found that in the energy below 10 e V/u,the Na(4s)state is the most dominant exit state with a contribution of approximately 78%to the branch fraction,which is in best agreement with the experimental data.For energies above 10 e V/u,the MN total cross section is larger than those obtained in other theoretical calculations and shows a slow decreasing trend because the main exit states change,when the energy is above 100 e V/u,the dominant exit state becomes the Na(3p)state,while the Na(4s)state becomes the third most important exit state.The datasets presented in this paper,including the potential energy curve,the radial and rotational couplings,the total and state-selective cross sections,are openly available at https://doi.org/10.57760/sciencedb.j00113.00112.

文摘Spectral line widths produced by collisions between charged particles and emitters are of special interest for precise plasma spectroscopy.The highly charged Ar XV ion is demonstrated to have strong intrashell electron interactions,which manifest as an atomic system with many resonance structures,due to the quasi-degeneracy of orbital energies.In this paper we use the relativistic R-matrix method to investigate the electron-impact broadening of highly charged Ar XV ion spectral lines under the impact approximation.It is found that the results considering resonance structures are significantly different from those of the distorted wave approach.Furthermore,we propose a new empirical formula with a correction term to take into account the effect of resonances for electron-impact widths over a relatively wide range of plasma conditions.The corresponding fitting parameters of the new empirical formula for all 47 calculated transitions are also given with an estimated accuracy within 1%,which should be convenient for practical applications.The dataset that supported the findings of this study is available in Science Data Bank,with the link https://doi.org/10.57760/sciencedb.j00113.00101.

文摘Background Methamphetamine(METH)addiction causes a huge burden on society.The prefrontal cortex(PFC),associated with emotion and cognitive behaviours,is also involved in addiction neurocircuitry.Although bulk RNA sequencing has shown METH-induced gene alterations in the mouse PFC,the impact on different cell types remains unknown.Aims To clarify the effects of METH treatment on different cell types of the PFC and the potential pathways involved in METH-related disorders.Methods We performed single-nucleus RNA sequencing(snRNA-seq)to examine the transcriptomes of 20465 nuclei isolated from the PFC of chronic METH-treated and control mice.Main cell types and differentially expressed genes(DEGs)were identified and confirmed by RNA fluorescence in situ hybridization(FISH).Results Six main cell types were identified depending on the single-cell nucleus sequencing;of particular interest were the mature oligodendrocytes in the PFC.The DEGs of mature oligodendrocytes were enriched in the myelin sheath,adenosine triphosphate(ATP)metabolic process,mitochondrial function and components,and so on.The messenger RNA levels of Aldoc and Atp5l(FISH)and the protein level of the mitochondrial membrane pore subunit TOM40(immunofluorescence)decreased in the mature oligodendrocytes.Fast blue staining and transmission electron microscopy image indicated myelin damage,and the myelin thickness decreased in METH brains.Conclusions snRNA-seq reveals altered transcriptomes of different cell types in mouse PFC induced by chronic METH treatment,underscoring potential relationships with psychiatric disorders.

文摘The relationship among microstructure,mechanical properties and texture of TA32 titanium alloy sheets during hot tensile deformation at 800℃was investigated.In the test,the original sheet exhibited relatively low flow stress and sound plasticity,and increasing the heat treatment temperature resulted in an increased ultimate tensile strength(UTS)and a decreased elongation(EL).The deformation mechanism of TA32 alloy was dominated by high angle grain boundaries sliding and coordinated by dislocation motion.The coarsening of grains and the annihilation of dislocations in heat-treated specimens weakened the deformation ability of material,which led to the increase in flow stress.Based on the high-temperature creep equation,the quantitative relationship between microstructure and flow stress was established.The grain size exponent andαphase strength constant of TA32 alloy were calculated to be 1.57 and 549.58 MPa,respectively.The flow stress was accurately predicted by combining with the corresponding phase volume fraction and grain size.Besides,the deformation behavior of TA32 alloy was also dependent on the orientation of predominantαphase,and the main slip mode was the activation of prismaticslip system.The decrease of near prism-oriented texture in heat-treated specimens resulted in the enhancement of strength of the material.
