Traditional designs and developments of bone biomaterials mostly concentrate on the positive regulation of osteoblast lineage cells,but often ignore the importance of immune responses and the equilibrium between bone ...Traditional designs and developments of bone biomaterials mostly concentrate on the positive regulation of osteoblast lineage cells,but often ignore the importance of immune responses and the equilibrium between bone resorption mediated by osteoclasts and bone formation mediated by osteoblasts.Immune dysregulation is associated with an imbalance between pro-inflammatory and anti-inflammatory processes,which may influence the efficacy of bone therapy.Therefore,implanted biomaterials should appropriately and precisely modulate subsequent immune responses.Magnesium(Mg)has been used to fabricate various Mg alloys for bone repair because of its favorable attributes such as osteogenic potential,immune regulation characteristics,biodegradability,and biocompatibility.Various basic research and clinical trials have been already conducted in many countries to explore the physical properties of Mg-containing implants and their clinical outcomes in bone fracture and defect repair.Therefore,this review summarizes the immune response to Mg-containing implants,and further organizes the current research and development progress of Mg-containing implants.The review aims to offer an overview of the current knowledge on immunomodulation of Mg-containing implants and future challenges in their clinical application,which could provide further insight in the development of better strategies for the treatment of bone defect and fracture.展开更多

BACKGROUND Post-hepatectomy liver failure(PHLF)is a common consequence of radical partial hepatectomy in hepatocellular carcinoma(HCC).AIMS To investigate the relationship between preoperative antiviral therapy and PH...BACKGROUND Post-hepatectomy liver failure(PHLF)is a common consequence of radical partial hepatectomy in hepatocellular carcinoma(HCC).AIMS To investigate the relationship between preoperative antiviral therapy and PHLF,as well as assess the potential efficacy of hepatitis B virus(HBV)DNA level in predicting PHLF.METHODS A retrospective study was performed involving 1301 HCC patients with HBV who underwent radical hepatectomy.Receiver operating characteristic(ROC)analysis was used to assess the capacity of HBV DNA to predict PHLF and establish the optimal cutoff value for subsequent analyses.Logistic regression analyses were performed to assess the independent risk factors of PHLF.The increase in the area under the ROC curve,categorical net reclassification improvement(NRI),and integrated discrimination improvement(IDI)were used to quantify the efficacy of HBV DNA level for predicting PHLF.The P<0.05 was considered statistically significant.RESULTS Logistic regression analyses showed that preoperative antiviral therapy was independently associated with a reduced risk of PHLF(P<0.05).HBV DNA level with an optimal cutoff value of 269 IU/mL(P<0.001)was an independent risk factor of PHLF.All the reference models by adding the variable of HBV DNA level had an improvement in area under the curve,categorical NRI,and IDI,particularly for the fibrosis-4 model,with values of 0.729(95%CI:0.705-0.754),1.382(95%CI:1.341-1.423),and 0.112(95%CI:0.110-0.114),respectively.All the above findings were statistically significant.CONCLUSION In summary,preoperative antiviral treatment can reduce the incidence of PHLF,whereas an increased preoperative HBV DNA level has a correlative relationship with an increased susceptibility to PHLF.展开更多
Wuyi Mountain,located in the north of Fujian Province,China,is renowned for its abundant medicinal plant resources.In July 2014,the 8th(second team)of Shenyang Pharmaceutical University’s Chinese Medicine Resources S...Wuyi Mountain,located in the north of Fujian Province,China,is renowned for its abundant medicinal plant resources.In July 2014,the 8th(second team)of Shenyang Pharmaceutical University’s Chinese Medicine Resources Scientific Expedition Team conducted field investigation in the area.Through specimen collection and extensive literature review,the team identified and analyzed 223 vascular plant species from 175 genera and 85 families.The most dominant families were Compositae and Rosaceae,and perennial herbs were the predominant species,accounting for 44.39%of the total species identified.Notably,we documented five precious and rare medicinal plants unique to Wuyi Mountain.This study updates the database of plant resources and diversity in the region,providing a valuable reference for future studies.Finally,we put forward some suggestions to enhance the conservation and sustainable utilization of Wuyi Mountain’s plant resources.展开更多
Porous carbon-based materials are promised to be lightweight dielectric microwave absorbents.Deeply understanding the influence of graphitization grade and porous structure on the dielectric parameters is urgently req...Porous carbon-based materials are promised to be lightweight dielectric microwave absorbents.Deeply understanding the influence of graphitization grade and porous structure on the dielectric parameters is urgently required.Herein,utilizing the low boiling point of Zn,porous N-doped carbon was fabricated by carbonization of ZIF-8(Zn)at different temperature,and the microwave absorption performance was investigated.The porous N-doped carbon inherits the high porosity of ZIF-8 precursor.By increasing the carbonization temperature,the contents of Zn and N elements are decreased;the graphitization degree is improved;however,the specific surface area and porosity are increased first and then decreased.When the carbonization temperature is 1000°C,the porous N-doped carbon behaves enhanced microwave absorption.With an ultrathin thickness of 1.29 mm,the ideal RL reaches-50.57 dB at 16.95 GHz and the effective absorption bandwidth is 4.17 GHz.The mechanism of boosted microwave absorption is ascribed to the competition of graphitization and porosity as well as N dopants,resulting in high dielectric loss capacity and good impedance matching.The porous structure can prolong the pathways and raise the contact opportunity between microwaves and porous carbon,causing multiple scattering,interface polarization,and improved impedance matching.Besides,the N dopants can induce electron polarization and defect polarization.These results give a new insight to construct lightweight carbon-based microwave absorbents by regulating the graphitization and porosity.展开更多

Currently, no previous study explored the role of Th22 cells in the p...Objective: IL-22-producing CD4+ T helper cells (Th22 cells) have been identified as major inducers of tissue inflammation and immune responses. Currently, no previous study explored the role of Th22 cells in the pathogenesis of hepatocellular carcinoma (HCC). The study aimed to determine the biological function of Th22 cells and its effector IL-22 in HCC patients. Methods: Forty-five HCC patients and 19 healthy controls were recruited and their peripheral blood was collected. The flesh HCC tissues, adjacent HCC tissues and ten normal liver tissues were also collected. Flow cytometry analysis was used to determine the frequencies of circulating Th22 cells and Thl7 cells. Serum IL-22 levels were tested by enzyme-linked immtmosorbent assay (ELISA). Immunohistochemical staining and real-time polymerase chain reaction (PCR) were used to detect IL-22 protein and mRNA in tissues specimens, respectively. Results: Circulating Th22 cells, Thl7 cells and serum IL-22 levels were significantly elevated in HCC patients compared with those of healthy controls (P〈0.001). Th22 cells were showed to be positively correlated with IL-22 in HCC patients (P〈0.05), but not in healthy controls. No significant differences were found in HCC patients with HBeAg positivity or negativity in term of Th22 cells and serum IL-22 levels. The expression of IL-22 protein and mRNA was highest in HCC tissues, followed by adjacent HCC tissues and normal liver tissues. Furthermore, Th22 cells, serum IL-22 levels and IL-22 mRNA were elevated at stage III-IV compared with stage I-II of HCC (P〈0.05). Conclusions: Elevation of circulating Th22 cells and IL-22 may be implicated in the pathogenesis of HCC, and potentially be cellular targets for therapeutic intervention.展开更多
AIM:To clarify the current understanding of the association between interleukin-10(IL-10)polymorphisms and the risk of irritable bowel syndrome(IBS).METHODS:We searched for studies in any language recorded in PubMed,E...AIM:To clarify the current understanding of the association between interleukin-10(IL-10)polymorphisms and the risk of irritable bowel syndrome(IBS).METHODS:We searched for studies in any language recorded in PubMed,Embase and Cochrane library before August 2013.The associations under allele contrast model,codominant model,dominant model,and recessive model were analyzed.The strengths of the association between IL-10 polymorphisms and IBS risk were estimated using odds ratios(OR)with 95%confidence interval(CI).Fixed effects model was used to pool the result if the test of heterogeneity was not significant,otherwise the random-effect model was selected.RESULTS:Eight case-control studies analyzing three single-nucleotide polymorphisms rs1800870(-1082 A/G),rs1800871(-819C/T),and rs1800872(-592A/C)of the IL-10 gene,which involved 928 cases and 1363 controls,were eligible for our analysis.The results showed that rs1800870 polymorphisms were associated with a decreased risk of IBS(GG+GA vs AA:OR=0.80,95%CI:0.66-0.96),(AA+GA vs GG:OR=0.68,95%CI:0.52-0.90).Subgroup analysis revealed such association only existed in Caucasian ethnicity(AA+GA vs GG,OR=0.70,95%CI:0.55-0.89).The rs1800872 polymorphisms were associated with an increased risk of IBS in Asian ethnicity(CC vs GG:OR=1.29,95%CI:1.01-1.16).There were no associations between rs1800871 polymorphisms and the IBS risk.CONCLUSION:The results suggest that IL-10 rs1800870confers susceptibility to the risk of IBS in Caucasian ethnicity,and the rs1800872 may associate with IBS risk in Asians.However,no significant associations are found between rs1800871 and IBS risk.展开更多

BACKGROUND Coronary microembolization(CME)is a complicated problem that commonly arises in the context of coronary angioplasty.MicroRNAs play crucial roles in cardiovascular diseases.However,the role and mechanism of ...BACKGROUND Coronary microembolization(CME)is a complicated problem that commonly arises in the context of coronary angioplasty.MicroRNAs play crucial roles in cardiovascular diseases.However,the role and mechanism of miR-181a-5p in CME-induced myocardial injury remains unclear.METHODS We established CME rat models.Cardiac function was detected by echocardiography.Haematoxylin-basic fuchsin-picric acid staining was used to measure micro-infarction size.Serum samples and cell culture supernatants were evaluated via enzyme-linked immunosorbent assay.Cellular reactive oxygen species were determined by dichloro-dihydro-fluorescein diacetate assay,and the other oxidative stress related parameters were assayed by spectrophotometry.The dual-luciferase reporter(DLR)assay and RNA pulldown were conducted to validate the association between miR-181a-5p and X-linked inhibitor of apoptosis protein(XIAP).The expression of miR-181a-5p and XIAP mRNA were determined by quantitative reverse transcription polymerase chain reaction.Proteins were evaluated via immunoblotting.The viability of the cell was evaluated via cell counting kit-8 assay.RESULTS The miR-181a-5p level was significantly increased in CME myocardial tissues.Downregulation of miR-181a-5p improved CME-induced cardiac dysfunction and alleviated myocardial oxidative stress and inflammatory injury,whereas miR-181a-5p exhibited the opposite effects.Then,the DLR assay and RNA pulldown results revealed that miR-181a-5p directly targeting on XIAP.The XIAP level was found to be remarkably decreased after CME.XIAP overexpression attenuated CME-induced myocardial oxidative stress and inflammatory injury.Finally,in vitro rescue experiments revealed that knockdown of XIAP could abolish the protective effects of miR-181a-5p knockdown on hypoxia-induced cardiomyocyte oxidative stress and inflammatory injury.CONCLUSIONS Downregulation of miR-181a-5p alleviates CME-induced myocardial damage by suppressing myocardial oxidative stress and inflammation through directly targeting XIAP.展开更多

The use of metoprolol before PCI can significan...BACKGROUND: Coronary microembolization (CME) is a serious complication following percutaneous coronary intervention (PCI) in patients with acute coronary syndromes. The use of metoprolol before PCI can significantly protect ischemic myocardium from myocardial damage, but the function of metoprolol in the treatment of CME is not entirely clear. This study was to explore the effect and significance of metoprolol on myocardial apoptosis and caspase-3 activation after CME in rats. METHODS: Thirty rats were randomly divided into three groups including sham-operation (control group), CME plus saline (CME group), CME plus metoprolol (metoprolol group), 10 rats for each group. The CME group was induced by injecting 3 000 polyethylene microspheres (42 tJm) into the left ventricle during a 10-second occlusion of the ascending aorta; the control group was injected with physiological saline instead of microembolization ball; the metoprolol or saline group was given three intravenous bolus injections before CME. Echocardiography, TUNEL staining, and Western blotting were used to evaluate cardiac function, proportion of apoptotic cells and activation of caspase-3 respectively at 6 hours after operation. RESULTS: Echocardiographic parameters displayed that the metoprolol group improved cardiac function significantly compared with the CME group (P〈0.05). The myocardial apoptotic rate of the CME group as well as the contents of activated caspase-3 increased significantly (P〈0.05), both of which were ameliorated significantly by metoprolol treatment (P〈0.05). CONCLUSIONS: This study demonstrates that metoprolol can protect the myocardium during CME in rats by inhibiting apoptosis and improving cardiac function. These results suggest that the inhibition of apoptosis can be a potential therapeutic strategy for the treatment of CME.展开更多

Myelinated nerves also carry out plasticity in which microglia remove myelin debris by phagocytosis. However, it remains unknown whether microglia explore structures on nerve fibers, such as Ranvier’s node(RN) or myelin sheath, before they become debris. By double or triple staining RNs or myelin sheathes and microglia in healthy rat corpus callosum, this study unveiled direct contacts of microglia pseudopodia with RNs and with para-and inter-nodal myelin sheathes, which was then verified by electron microscopic observations. Our data indicated that microglia also explore unmyelinated nerve fibers. Furthermore, we used the animals with matured white matter;therefore, microglia may be actively involved in plasticity of matured white matter tracts as it does for synapse pruning, instead of only passively phagocytize myelin debris.展开更多

Furthermore, Th22 cells, serum IL-22 levels and IL-22 mRNA were elevated at stage III-IV compared with stage I-II of HCC (P〈0.05). Conclusions: Elevation of circulating Th22 cells and IL-22 may be implicated in the pathogenesis of HCC, and potentially be cellular targets for therapeutic intervention.

文摘AIM:To clarify the current understanding of the association between interleukin-10(IL-10)polymorphisms and the risk of irritable bowel syndrome(IBS).METHODS:We searched for studies in any language recorded in PubMed,Embase and Cochrane library before August 2013.The associations under allele contrast model,codominant model,dominant model,and recessive model were analyzed.The strengths of the association between IL-10 polymorphisms and IBS risk were estimated using odds ratios(OR)with 95%confidence interval(CI).Fixed effects model was used to pool the result if the test of heterogeneity was not significant,otherwise the random-effect model was selected.RESULTS:Eight case-control studies analyzing three single-nucleotide polymorphisms rs1800870(-1082 A/G),rs1800871(-819C/T),and rs1800872(-592A/C)of the IL-10 gene,which involved 928 cases and 1363 controls,were eligible for our analysis.The results showed that rs1800870 polymorphisms were associated with a decreased risk of IBS(GG+GA vs AA:OR=0.80,95%CI:0.66-0.96),(AA+GA vs GG:OR=0.68,95%CI:0.52-0.90).Subgroup analysis revealed such association only existed in Caucasian ethnicity(AA+GA vs GG,OR=0.70,95%CI:0.55-0.89).The rs1800872 polymorphisms were associated with an increased risk of IBS in Asian ethnicity(CC vs GG:OR=1.29,95%CI:1.01-1.16).There were no associations between rs1800871 polymorphisms and the IBS risk.CONCLUSION:The results suggest that IL-10 rs1800870confers susceptibility to the risk of IBS in Caucasian ethnicity,and the rs1800872 may associate with IBS risk in Asians.However,no significant associations are found between rs1800871 and IBS risk.

文摘The methanol-to-olefin induction reaction over the SAPO-34 was performed using a fluidized-bed system.We found that the whole induction period could be divided into three reaction stages.Further investigation of the reaction kinetics revealed that this induction reaction behavior was different from that over H-ZSM-5 catalyst.Compared with the H-ZSM-5,the generation of initial active centers is easier over SAPO-34 because of its limited diffusivity and the spatial confinement effect of the cages.However,the autocatalysis reaction stage is difficult over SAPO-34 because of the continuous formation of inactive methyladamantanes.

文摘BACKGROUND Coronary microembolization(CME)is a complicated problem that commonly arises in the context of coronary angioplasty.MicroRNAs play crucial roles in cardiovascular diseases.However,the role and mechanism of miR-181a-5p in CME-induced myocardial injury remains unclear.METHODS We established CME rat models.Cardiac function was detected by echocardiography.Haematoxylin-basic fuchsin-picric acid staining was used to measure micro-infarction size.Serum samples and cell culture supernatants were evaluated via enzyme-linked immunosorbent assay.Cellular reactive oxygen species were determined by dichloro-dihydro-fluorescein diacetate assay,and the other oxidative stress related parameters were assayed by spectrophotometry.The dual-luciferase reporter(DLR)assay and RNA pulldown were conducted to validate the association between miR-181a-5p and X-linked inhibitor of apoptosis protein(XIAP).The expression of miR-181a-5p and XIAP mRNA were determined by quantitative reverse transcription polymerase chain reaction.Proteins were evaluated via immunoblotting.The viability of the cell was evaluated via cell counting kit-8 assay.RESULTS The miR-181a-5p level was significantly increased in CME myocardial tissues.Downregulation of miR-181a-5p improved CME-induced cardiac dysfunction and alleviated myocardial oxidative stress and inflammatory injury,whereas miR-181a-5p exhibited the opposite effects.Then,the DLR assay and RNA pulldown results revealed that miR-181a-5p directly targeting on XIAP.The XIAP level was found to be remarkably decreased after CME.XIAP overexpression attenuated CME-induced myocardial oxidative stress and inflammatory injury.Finally,in vitro rescue experiments revealed that knockdown of XIAP could abolish the protective effects of miR-181a-5p knockdown on hypoxia-induced cardiomyocyte oxidative stress and inflammatory injury.CONCLUSIONS Downregulation of miR-181a-5p alleviates CME-induced myocardial damage by suppressing myocardial oxidative stress and inflammation through directly targeting XIAP.

文摘BACKGROUND: Coronary microembolization (CME) is a serious complication following percutaneous coronary intervention (PCI) in patients with acute coronary syndromes. The use of metoprolol before PCI can significantly protect ischemic myocardium from myocardial damage, but the function of metoprolol in the treatment of CME is not entirely clear. This study was to explore the effect and significance of metoprolol on myocardial apoptosis and caspase-3 activation after CME in rats. METHODS: Thirty rats were randomly divided into three groups including sham-operation (control group), CME plus saline (CME group), CME plus metoprolol (metoprolol group), 10 rats for each group. The CME group was induced by injecting 3 000 polyethylene microspheres (42 tJm) into the left ventricle during a 10-second occlusion of the ascending aorta; the control group was injected with physiological saline instead of microembolization ball; the metoprolol or saline group was given three intravenous bolus injections before CME. Echocardiography, TUNEL staining, and Western blotting were used to evaluate cardiac function, proportion of apoptotic cells and activation of caspase-3 respectively at 6 hours after operation. RESULTS: Echocardiographic parameters displayed that the metoprolol group improved cardiac function significantly compared with the CME group (P〈0.05). The myocardial apoptotic rate of the CME group as well as the contents of activated caspase-3 increased significantly (P〈0.05), both of which were ameliorated significantly by metoprolol treatment (P〈0.05). CONCLUSIONS: This study demonstrates that metoprolol can protect the myocardium during CME in rats by inhibiting apoptosis and improving cardiac function. These results suggest that the inhibition of apoptosis can be a potential therapeutic strategy for the treatment of CME.

文摘Over the recent years, it has been found that microglia pseudopodia contact synapses, detect sick ones and prune them, even in adult animals. Myelinated nerves also carry out plasticity in which microglia remove myelin debris by phagocytosis. However, it remains unknown whether microglia explore structures on nerve fibers, such as Ranvier’s node(RN) or myelin sheath, before they become debris. By double or triple staining RNs or myelin sheathes and microglia in healthy rat corpus callosum, this study unveiled direct contacts of microglia pseudopodia with RNs and with para-and inter-nodal myelin sheathes, which was then verified by electron microscopic observations. Our data indicated that microglia also explore unmyelinated nerve fibers. Furthermore, we used the animals with matured white matter;therefore, microglia may be actively involved in plasticity of matured white matter tracts as it does for synapse pruning, instead of only passively phagocytize myelin debris.
