Clay minerals mainly exist in nano- and micro-meter sized particles, and the pore structure is complex, which leads to its extremely complex wettability. The surface wettability of clay minerals significantly affects the oil and gas-bearing capacity of shale reservoirs. Therefore, studying the wettability of common clay minerals in shale at the nanoscale is of great significance for shale hydrocarbon exploration and development. In this study, the wetting behavior of water in n-hexane and toluene on different clay mineral surfaces at the nanoscale was systematically studied using Molecular dynamics (MD) simulation. And the influencing factors of wettability were analyzed. Through the analysis of the morphological changes of water, relative concentration of water, RDF and interaction energy, it is concluded that the following order of water wettability on the surfaces of clay minerals: montmorillonite > chlorite > kaolinite > illite. Through the analysis of interaction energy, it is concluded that the hydrophilicity of four clay minerals is stronger than that of lipophilicity. And the main interactions between water and oil and the mineral surfaces were van der Waals force and electrostatic force. In addition, the temperature, liquid hydrocarbon type, and mineralization of water affected the wettability of clay minerals. The concentration of water on the surfaces of montmorillonite, kaolinite, and illite decreased with increasing temperature, and the water wettability decreased. At 298 K, the hydrophilicity of the surfaces of the clay minerals in toluene follows the order montmorillonite > chlorite > kaolinite > illite. The higher the NaHCO3 concentration in water, the weaker the wettability of the clay mineral surfaces to water. By comparing the previous experimental results with the MD simulation results, similar wetting characteristics were obtained, and the reliability of the simulation results was verified. MD simulation was used to explore the water wetting of the surfaces of four clay minerals in a shale reservoir from the micro level. This makes up for the lack of experimental means for clarifying the flow and production mechanisms of shale oil and gas and effectively improves the evaluation technology of shale.展开更多
Objective Isletαcells input is essential for insulin secretion fromβcells.The present study aims to investigate the association between 25-hydroxyvitamin D[25(OH)D]and islet function homeostasis in type-2 diabetes(T...Objective Isletαcells input is essential for insulin secretion fromβcells.The present study aims to investigate the association between 25-hydroxyvitamin D[25(OH)D]and islet function homeostasis in type-2 diabetes(T2D)patients.Methods A total of 4670 T2D patients from seven communities in Shanghai,China were enrolled.The anthropometric indices,biochemical parameters,serum 25(OH)D,and islet function[including C-peptide(C-p)and glucagon]were measured.Results The fasting plasma glucose(FPG),glycated hemoglobin(HbA1c),glucagon,and C-p levels exhibited a significantly decreasing trend in T2D patients as the 25(OH)D levels increased.Next,the population was divided into two groups:abdominal obesity and non-abdominal obesity groups.After adjustment,the 25(OH)D level was found to be associated with HbA1c,glucagon,and homeostasis model assessment ofβ(HOMA-β)in the non-abdominal obesity group.There was a significant relationship between 25(OH)D and HbA1c,glucagon,HOMA-IR,baseline insulin or C-p in the abdominal obesity group.In the abdominal obesity group,the ordinary least squares(OLS)regression and quantile regression revealed that 25(OH)D was obviously associated with glucagon and fasting C-p levels.In the abdominal obesity group,the moderate analysis revealed a significant interaction effect of 25(OH)D and glucagon on C-p(P=0.0124).Furthermore,the conditional indirect effect of 25(OH)D on the glucagon/C-p ratio was significantly lower at 1 standard deviation(SD)below the mean(P=0.0002),and lower at the mean of the course of diabetes(P=0.0007).Conclusion 25(OH)D was found to be negatively correlated to glucagon and C-p in T2D patients with abdominal obesity.The 25(OH)D influenced C-p in part by influencing glucagon.The effect of 25(OH)D on the glucagon/C-p ratio in T2D patients with abdominal obesity,in terms of islet homeostasis,is influenced by the course of diabetes.展开更多

Bone marrow-derived mesenchymal stem cells(MSCs) is considered to be a potential therapy for I/R injury due to the paracrine effects. High-mobility group box1(HMGB1) is a novel mediator in MSC and regulates the response of inflammation injury. Signal Transduction and Transcription Activator 3(STAT3) is a critical transcription factor and important for release of paracrine factors. However, the relationship between HMGB1 and STAT3 in paracrine effect of MSC remains unknown.METHODS In vitro, hypoxia/reoxygenation injury model was established by Anaero Pack System and examined by Annexin V flow cytometry, CCK8 assay and morphology observation. Detection of apoptotic proteins and protein expression of HMGB1and STAT3 by Western blot.RESULTS The conditioned medium of MSCs with or without LPS pretreatment was cocultured with H9C2 cells for 24 h before hypoxia treatment and MSC showed obvious cardiomyocytes protect role, as evidence by decreased apoptosis rate and improved cells viability, and LPS pretreated MSC exhibited better protect role than untreated MSC. However, such effect was abolished in HMGB1 deficiency group, silencing HMGB1 decreased the secretion of vascular endothelial growth factor(VEGF), hepatocyte growth factor(HGF), insulin growth factor(IGF), cell viability, and the expression of STAT3. Furthermore, STAT3 silence attenuated the protective effect of LPS in MSC.CONCLUSIONS These findings suggested that LPS improved MSC-mediated cardiomyocytes protection by HMGB1/STAT3signaling.展开更多
BACKGROUND Heat-clearing and detoxifying drugs has protective effect on colorectal cancer(CRC).Given the complicated features of Traditional Chinese medicine formulas,network pharmacology is an effective approach for ...BACKGROUND Heat-clearing and detoxifying drugs has protective effect on colorectal cancer(CRC).Given the complicated features of Traditional Chinese medicine formulas,network pharmacology is an effective approach for studying the multiple interactions between drugs and diseases.AIM To systematically explore the anticancer mechanism of heat-clearing and detoxifying drug JC724.METHODS This study obtained the active compounds and their targets in JC724 from Traditional Chinese Medicine System Pharmacology Database.In addition,the CRC targets were obtained from Drugbank,TTD,DisGeNET and GeneCards databases.We performed transcriptome analysis of differentially expressed genes in CRC treated with JC724.Venn diagram was used to screen the JC724-CRC intersection targets as candidate targets.Core targets were selected by proteinprotein interaction network and herb ingredient-target-disease network analysis.The functional and pathway of core targets were analysed by enrichment analysis.RESULTS We found 174 active ingredients and 283 compound targets from JC724.940 CRC-related targets were reserved from the four databases and 304 CRC differentially expressed genes were obtained by transcriptome analysis.We constructed the network and found that the five core ingredients were quercetin,βBeta sitosterol,wogonin,kaempferol and baicalein.The core JC724-CRC targets were CYP1A1,HMOX1,CXCL8,NQO1 and FOSL1.JC724 acts on multiple signaling pathways associated with CRC,including the Nrf2 signaling pathway,oxidative stress,and the IL-17 signaling pathway.CONCLUSION In this study,we systematically analyzed the active ingredients,core targets and main mechanisms of JC724 in the treatment of CRC.This study could bring a new perspective to the heat-clearing and detoxifying therapy of CRC.展开更多
AIM: To investigate the effects of posterior scleral reinforcement(PSR) in the treatment of pathological myopia.·METHODS: The study included 52 eyes in 43 patients with pathological myopia who underwent PSR(PSR g...AIM: To investigate the effects of posterior scleral reinforcement(PSR) in the treatment of pathological myopia.·METHODS: The study included 52 eyes in 43 patients with pathological myopia who underwent PSR(PSR group), and 52 eyes in 36 age- and myopia-matched patients who did not undergo such treatment as control group. Axial length, refraction error, best corrected visual acuity(BCVA), and macular scans by optical coherence tomography(OCT) were recorded at baseline, 6mo, 1, 3and 5y after the surgery, and the complications were noted.·RESULTS: There were no statistical differences in axial length, refractive error, or BCVA between the PSR group and the control group at baseline. At the end of the follow-up, the mean axial length was 29.79 ±1.26 mm in the PSR group, which was significantly shorter than that in the control group(30.78±1.30 mm)(P <0.01), and the mean refractive error was-16.86 ±2.53 D in the PSR group, which was significantly lower than that in the control group(-19.18 ±2.12 D)(P <0.01). A statistically significant difference in BCVA was found between the PSR group(0.51 ±0.25 log MAR) and the control group(0.62±0.26 log MAR) at the postoperative 5-year follow-up(P <0.01). There were no serious complications during the 5-year follow-up period.·CONCLUSION: PSR can prevent axial elongation and myopia progression in eyes with pathological myopia.展开更多

However, imbalances in the microenvironment and adhesion of surrounding tissues can affect the quality of nerve regeneration and repair. Previous studies have shown that human amniotic membrane can promote the healing of a variety of tissues. In this study, the right common peroneal nerve underwent a 5-mm transection in rats. Epineural nerve repair was performed using 10/0 non-absorbable surgical suture. The repair site was wrapped with a two-layer amniotic membrane with α-cyanoacrylate rapid medical adhesive after suture. Hindlimb motor function was assessed using footprint analysis. Conduction velocity of the common peroneal nerve was calculated by neural electrical stimulation. The retrograde axoplasmic transport of the common peroneal nerve was observed using fast blue BB salt retrograde fluorescent staining. Hematoxylin- eosin staining was used to detect the pathological changes of the common peroneal nerve sputum. The mRNA expression of axon regeneration-related neurotrophic factors and inhibitors was measured using real-time polymerase chain reaction. The results showed that the amniotic membrane significantly improved the function of the injured nerve;the toe spread function rapidly recovered, the nerve conduction velocity was restored, and the number of fast blue BB salt particles were increased in the spinal cord. The amniotic membrane also increased the recovery rate of the tibialis anterior muscle and improved the tissue structure of the muscle. Meanwhile, mRNA expression of nerve growth factor, growth associated protein-43, collapsin response mediator protein-2, and brain-derived neurotrophic factor recovered to near-normal levels, while Lingo-1 mRNA expression decreased significantly in spinal cord tissues. mRNA expression of glial-derived neurotrophic factor did not change significantly. Changes in mRNA levels were more significant in amniotic-membrane-wrapping-treated rats compared with model and nerve sutured rats. These results demonstrate that fresh amniotic membrane wrapping can promote the functional recovery of sutured common peroneal nerve via regulation of expression levels of neurotrophic factors and inhibitors associated with axonal regeneration. AIM: To investigate the effects of posterior scleral reinforcement(PSR) combined with vitrectomy for myopic foveoschisis.· METHODS: Thirty-nine highly myopic eyes of 39 patients with myopic foveoschisis underwent...AIM: To investigate the effects of posterior scleral reinforcement(PSR) combined with vitrectomy for myopic foveoschisis.· METHODS: Thirty-nine highly myopic eyes of 39 patients with myopic foveoschisis underwent PSR combined with vitrectomy. Best corrected visual acuity(BCVA), refraction error, and the foveal thickness by optical coherence tomography(OCT) were recorded before and after the surgery, and complications were noted.·RESULTS: The follow-up period was 12 mo, and the main focus was on the results of the 12-month follow-up visit. The mean preoperative BCVA was 0.96±0.43 log MAR.At the final follow-up visit, the mean BCVA was 0.46 ±0.28 log MAR, which significantly improved compared with the preoperative one(P =0.003). The BCVA improved in33 eyes(84.62%), and unchanged in 6 eyes(15.38%). At the end of follow- up, the mean refractive error was-15.13 ±2.55 D, and the improvement was significantly compared with the preoperative one(-17.53±4.51 D)(P =0.002). Twelve months after surgery, OCT showed complete resolution of the myopic foveoschisis and a reattachment of the fovea in 37 eyes(94.87%) and partial resolution in the remained two eyes(5.13%). The foveal thickness was obviously reduced at 12-month follow-up visit(196.45 ±36.35 μm) compared with the preoperative one(389.32±75.56 μm)(P =0.002). There were no serious complications during the 12 mo follow-up period.·CONCLUSION: PSR combined with vitrectomy is a safe and effective procedure for myopic foveoschisis with both visual and anatomic improvement.展开更多
AIM:To investigate the effects of hepatocyte growth factor(HGF)on MMP-2 expression in scleral fibroblasts from guinea pig with LIM.METHODS:Sixty 1-week-old guinea pigs were chosen for the study.The right eyes were tre...AIM:To investigate the effects of hepatocyte growth factor(HGF)on MMP-2 expression in scleral fibroblasts from guinea pig with LIM.METHODS:Sixty 1-week-old guinea pigs were chosen for the study.The right eyes were treated with-10.0 D lenses as the LIM group;the left eyes remained untreated as the control group.The refraction and axial length were measured by streak retinoscopy and A-scan ultrasonography respectively prior to and 4wk after the experiment.Four weeks later,the guinea pigs were sacrificed and primary scleral fibroblasts were taken for tissue culture.The 3rd-5th generation scleral fibroblasts were chosen for the experiments.The expression levels of HGF and MMP-2 protein in the scleral fibroblasts were analyzed by Western blotting.After HGF with different doses acted on the scleral fibroblasts of the control group,MMP-2 protein expression in the scleral fibroblasts was analyzed by Western blotting.HGF siRNA was transfected into the scleral fibroblasts of the LIM group and the protein expressions of HGF and MMP-2were analyzed by Western blotting.RESULTS:The LIM group became myopic with a significant increase in axial length(7.97±0.29 mm vs7.01±0.26 mm,P<0.05),and a significant decrease in refraction(-5.06±0.31 D vs 0.55±0.25 D,P<0.05)compared with the control group.The protein expression of HGF in the scleral fibroblasts of the LIM group was significantly higher compared with the control group(1.26±0.04 vs 0.32±0.04,P<0.05).The protein expression of MMP-2 in the scleral fibroblasts of the LIM group wassignificantly higher compared with the control group(0.89±0.06 vs 0.42±0.05,P<0.05).In the scleral fibroblasts of the control group,HGF(0,0.1,1,10 ng/mL)upregulated MMP-2 protein expression in a dose-dependent manner(0.35±0.03,0.44±0.02,0.91±0.03,1.33±0.04,all P<0.05).In the scleral fibroblasts of the LIM group transfected with HGF siRNA,MMP-2 protein expressions were significantly decreased compared with the negative control group(0.29±0.03 vs 0.81±0.05,P<0.05).CONCLUSION:HGF is a upstream mediator of MMP-2in scleral fibroblasts from guinea pig.展开更多
To assess the effectiveness of complementary food supplements with protein and multi-micronutrients on hemoglobin and anemia in infants and young children.Methods In 5 poor counties of Gansu,984 children aged 6-12 mon...To assess the effectiveness of complementary food supplements with protein and multi-micronutrients on hemoglobin and anemia in infants and young children.Methods In 5 poor counties of Gansu,984 children aged 6-12 months were enrolled and divided into two groups.In addition to the usual home-made complementary food,all the children were fed one sachet of either Formula I or Formula II supplements each day.Protein and micronutrients were provided in Formula I,while the same energy intake was secured in Formula II as in Formula I.A massive dose of vitamin A was supplemented to all the children every 6 months.Hemoglobin test was done at the same time.Results Prevalence of anemia was about 35% in both Formula I and Formula II group at baseline,and there were no differences in hemoglobin concentration between the two groups.During the 6-month and 12-month supplementation,hemoglobin of children in Formula I group was higher than that in Formula II group(P<0.05),and hemoglobin increase in Formula I group was significantly higher than that in Formula II group(P<0.001).After 6-and 12-month supplementation,the prevalence of anemia in Formula I group dropped to 19.1% and 8.2% respectively,and it was 28.0% and 12.4% in Formula 2 group.The prevalence of anemia in Formula I group was significantly lower than that in Formula II group(P<0.05).After adjusting age and hemoglobin level at baseline,the hemoglobin increase at age of 24 months in formula 1 group was higher(10.7 g/L vs 7.9 g/L,P<0.0001).Conclusion Micronutrient fortified complementary food supplements,with large-dose vitamin A,is effective for children aged 6-12 months in terms of iron deficiency prevention.展开更多
Background:Non-communicable chronic diseases have become the leading causes of disease burden worldwide.The trends and burden of“metabolic associated fatty liver disease”(MAFLD)are unknown.We aimed to investigate th...Background:Non-communicable chronic diseases have become the leading causes of disease burden worldwide.The trends and burden of“metabolic associated fatty liver disease”(MAFLD)are unknown.We aimed to investigate the cardiovascular and renal burdens in adults with MAFLD and non-alcoholic fatty liver disease(NAFLD).Methods:Nationally representative data were analyzed including data from 19,617 non-pregnant adults aged≥20years from the cross-sectional US National Health and Nutrition Examination Survey periods,1999 to 2002,2003 to 2006,2007 to 2010,and 2011 to 2016.MAFLD was defined by the presence of hepatic steatosis plus general overweight/obesity,type 2 diabetes mellitus,or evidence of metabolic dysregulation.Results:The prevalence of MAFLD increased from 28.4%(95% confidence interval 26.3–30.6)in 1999 to 2002 to 35.8%(33.8–37.9)in 2011 to 2016.In 2011 to 2016,among adults with MAFLD,49.0%(45.8–52.2)had hypertension,57.8%(55.2–60.4)had dyslipidemia,26.4%(23.9–28.9)had diabetes mellitus,88.7%(87.0–80.1)had central obesity,and 18.5%(16.3–20.8)were current smokers.The 10-year cardiovascular risk ranged from 10.5%to 13.1%;19.7%(17.6–21.9)had chronic kidney diseases(CKDs).Throughthe fourperiods,adults withMAFLD showedanincrease in obesity;increase intreatment to lower bloodpressure(BP),lipids,and hemoglobin A1c;and increase in goal achievements for BP and lipids but not in goal achievement for glycemic control in diabetes mellitus.Patients showed a decreasing 10-year cardiovascular risk over time but no change in the prevalence of CKDs,myocardial infarction,or stroke.Generally,although participants with NAFLD and those with MAFLD had a comparable prevalence of cardiovascular disease and CKD,the prevalence of MAFLD was significantly higher than that of NAFLD.Conclusions:From 1999 to 2016,cardiovascular and renal risks and diseases have become highly prevalent in adults with MAFLD.The absolute cardiorenal burden may be greater for MAFLD than for NAFLD.These data call for early identification and risk stratification of MAFLD and close collaboration between endocrinologists and hepatologists.展开更多

Detection of apoptotic proteins and protein expression of HMGB1and STAT3 by Western blot.RESULTS The conditioned medium of MSCs with or without LPS pretreatment was cocultured with H9C2 cells for 24 h before hypoxia treatment and MSC showed obvious cardiomyocytes protect role, as evidence by decreased apoptosis rate and improved cells viability, and LPS pretreated MSC exhibited better protect role than untreated MSC. However, such effect was abolished in HMGB1 deficiency group, silencing HMGB1 decreased the secretion of vascular endothelial growth factor(VEGF), hepatocyte growth factor(HGF), insulin growth factor(IGF), cell viability, and the expression of STAT3. Furthermore, STAT3 silence attenuated the protective effect of LPS in MSC.CONCLUSIONS These findings suggested that LPS improved MSC-mediated cardiomyocytes protection by HMGB1/STAT3signaling.

文摘BACKGROUND Heat-clearing and detoxifying drugs has protective effect on colorectal cancer(CRC).Given the complicated features of Traditional Chinese medicine formulas,network pharmacology is an effective approach for studying the multiple interactions between drugs and diseases.AIM To systematically explore the anticancer mechanism of heat-clearing and detoxifying drug JC724.METHODS This study obtained the active compounds and their targets in JC724 from Traditional Chinese Medicine System Pharmacology Database.In addition,the CRC targets were obtained from Drugbank,TTD,DisGeNET and GeneCards databases.We performed transcriptome analysis of differentially expressed genes in CRC treated with JC724.Venn diagram was used to screen the JC724-CRC intersection targets as candidate targets.Core targets were selected by proteinprotein interaction network and herb ingredient-target-disease network analysis.The functional and pathway of core targets were analysed by enrichment analysis.RESULTS We found 174 active ingredients and 283 compound targets from JC724.940 CRC-related targets were reserved from the four databases and 304 CRC differentially expressed genes were obtained by transcriptome analysis.We constructed the network and found that the five core ingredients were quercetin,βBeta sitosterol,wogonin,kaempferol and baicalein.The core JC724-CRC targets were CYP1A1,HMOX1,CXCL8,NQO1 and FOSL1.JC724 acts on multiple signaling pathways associated with CRC,including the Nrf2 signaling pathway,oxidative stress,and the IL-17 signaling pathway.CONCLUSION In this study,we systematically analyzed the active ingredients,core targets and main mechanisms of JC724 in the treatment of CRC.This study could bring a new perspective to the heat-clearing and detoxifying therapy of CRC.

文摘AIM: To investigate the effects of posterior scleral reinforcement(PSR) in the treatment of pathological myopia.·METHODS: The study included 52 eyes in 43 patients with pathological myopia who underwent PSR(PSR group), and 52 eyes in 36 age- and myopia-matched patients who did not undergo such treatment as control group. Axial length, refraction error, best corrected visual acuity(BCVA), and macular scans by optical coherence tomography(OCT) were recorded at baseline, 6mo, 1, 3and 5y after the surgery, and the complications were noted.·RESULTS: There were no statistical differences in axial length, refractive error, or BCVA between the PSR group and the control group at baseline. At the end of the follow-up, the mean axial length was 29.79 ±1.26 mm in the PSR group, which was significantly shorter than that in the control group(30.78±1.30 mm)(P <0.01), and the mean refractive error was-16.86 ±2.53 D in the PSR group, which was significantly lower than that in the control group(-19.18 ±2.12 D)(P <0.01). A statistically significant difference in BCVA was found between the PSR group(0.51 ±0.25 log MAR) and the control group(0.62±0.26 log MAR) at the postoperative 5-year follow-up(P <0.01). There were no serious complications during the 5-year follow-up period.·CONCLUSION: PSR can prevent axial elongation and myopia progression in eyes with pathological myopia.

文摘Suture and autologous nerve transplantation are the primary therapeutic measures for completely severed nerves. However, imbalances in the microenvironment and adhesion of surrounding tissues can affect the quality of nerve regeneration and repair. Previous studies have shown that human amniotic membrane can promote the healing of a variety of tissues. In this study, the right common peroneal nerve underwent a 5-mm transection in rats. Epineural nerve repair was performed using 10/0 non-absorbable surgical suture. The repair site was wrapped with a two-layer amniotic membrane with α-cyanoacrylate rapid medical adhesive after suture. Hindlimb motor function was assessed using footprint analysis. Conduction velocity of the common peroneal nerve was calculated by neural electrical stimulation. The retrograde axoplasmic transport of the common peroneal nerve was observed using fast blue BB salt retrograde fluorescent staining. Hematoxylin- eosin staining was used to detect the pathological changes of the common peroneal nerve sputum. The mRNA expression of axon regeneration-related neurotrophic factors and inhibitors was measured using real-time polymerase chain reaction. The results showed that the amniotic membrane significantly improved the function of the injured nerve;the toe spread function rapidly recovered, the nerve conduction velocity was restored, and the number of fast blue BB salt particles were increased in the spinal cord. The amniotic membrane also increased the recovery rate of the tibialis anterior muscle and improved the tissue structure of the muscle. Meanwhile, mRNA expression of nerve growth factor, growth associated protein-43, collapsin response mediator protein-2, and brain-derived neurotrophic factor recovered to near-normal levels, while Lingo-1 mRNA expression decreased significantly in spinal cord tissues. mRNA expression of glial-derived neurotrophic factor did not change significantly. Changes in mRNA levels were more significant in amniotic-membrane-wrapping-treated rats compared with model and nerve sutured rats. These results demonstrate that fresh amniotic membrane wrapping can promote the functional recovery of sutured common peroneal nerve via regulation of expression levels of neurotrophic factors and inhibitors associated with axonal regeneration. 文摘AIM: To investigate the effects of posterior scleral reinforcement(PSR) combined with vitrectomy for myopic foveoschisis.· METHODS: Thirty-nine highly myopic eyes of 39 patients with myopic foveoschisis underwent PSR combined with vitrectomy. Best corrected visual acuity(BCVA), refraction error, and the foveal thickness by optical coherence tomography(OCT) were recorded before and after the surgery, and complications were noted.·RESULTS: The follow-up period was 12 mo, and the main focus was on the results of the 12-month follow-up visit. The mean preoperative BCVA was 0.96±0.43 log MAR.At the final follow-up visit, the mean BCVA was 0.46 ±0.28 log MAR, which significantly improved compared with the preoperative one(P =0.003). The BCVA improved in33 eyes(84.62%), and unchanged in 6 eyes(15.38%). At the end of follow- up, the mean refractive error was-15.13 ±2.55 D, and the improvement was significantly compared with the preoperative one(-17.53±4.51 D)(P =0.002). Twelve months after surgery, OCT showed complete resolution of the myopic foveoschisis and a reattachment of the fovea in 37 eyes(94.87%) and partial resolution in the remained two eyes(5.13%). The foveal thickness was obviously reduced at 12-month follow-up visit(196.45 ±36.35 μm) compared with the preoperative one(389.32±75.56 μm)(P =0.002). There were no serious complications during the 12 mo follow-up period.·CONCLUSION: PSR combined with vitrectomy is a safe and effective procedure for myopic foveoschisis with both visual and anatomic improvement.

文摘AIM:To investigate the effects of hepatocyte growth factor(HGF)on MMP-2 expression in scleral fibroblasts from guinea pig with LIM.METHODS:Sixty 1-week-old guinea pigs were chosen for the study.The right eyes were treated with-10.0 D lenses as the LIM group;the left eyes remained untreated as the control group.The refraction and axial length were measured by streak retinoscopy and A-scan ultrasonography respectively prior to and 4wk after the experiment.Four weeks later,the guinea pigs were sacrificed and primary scleral fibroblasts were taken for tissue culture.The 3rd-5th generation scleral fibroblasts were chosen for the experiments.The expression levels of HGF and MMP-2 protein in the scleral fibroblasts were analyzed by Western blotting.After HGF with different doses acted on the scleral fibroblasts of the control group,MMP-2 protein expression in the scleral fibroblasts was analyzed by Western blotting.HGF siRNA was transfected into the scleral fibroblasts of the LIM group and the protein expressions of HGF and MMP-2were analyzed by Western blotting.RESULTS:The LIM group became myopic with a significant increase in axial length(7.97±0.29 mm vs7.01±0.26 mm,P<0.05),and a significant decrease in refraction(-5.06±0.31 D vs 0.55±0.25 D,P<0.05)compared with the control group.The protein expression of HGF in the scleral fibroblasts of the LIM group was significantly higher compared with the control group(1.26±0.04 vs 0.32±0.04,P<0.05).The protein expression of MMP-2 in the scleral fibroblasts of the LIM group wassignificantly higher compared with the control group(0.89±0.06 vs 0.42±0.05,P<0.05).In the scleral fibroblasts of the control group,HGF(0,0.1,1,10 ng/mL)upregulated MMP-2 protein expression in a dose-dependent manner(0.35±0.03,0.44±0.02,0.91±0.03,1.33±0.04,all P<0.05).In the scleral fibroblasts of the LIM group transfected with HGF siRNA,MMP-2 protein expressions were significantly decreased compared with the negative control group(0.29±0.03 vs 0.81±0.05,P<0.05).CONCLUSION:HGF is a upstream mediator of MMP-2in scleral fibroblasts from guinea pig.
文摘To assess the effectiveness of complementary food supplements with protein and multi-micronutrients on hemoglobin and anemia in infants and young children.Methods In 5 poor counties of Gansu,984 children aged 6-12 months were enrolled and divided into two groups.In addition to the usual home-made complementary food,all the children were fed one sachet of either Formula I or Formula II supplements each day.Protein and micronutrients were provided in Formula I,while the same energy intake was secured in Formula II as in Formula I.A massive dose of vitamin A was supplemented to all the children every 6 months.Hemoglobin test was done at the same time.Results Prevalence of anemia was about 35% in both Formula I and Formula II group at baseline,and there were no differences in hemoglobin concentration between the two groups.During the 6-month and 12-month supplementation,hemoglobin of children in Formula I group was higher than that in Formula II group(P<0.05),and hemoglobin increase in Formula I group was significantly higher than that in Formula II group(P<0.001).After 6-and 12-month supplementation,the prevalence of anemia in Formula I group dropped to 19.1% and 8.2% respectively,and it was 28.0% and 12.4% in Formula 2 group.The prevalence of anemia in Formula I group was significantly lower than that in Formula II group(P<0.05).After adjusting age and hemoglobin level at baseline,the hemoglobin increase at age of 24 months in formula 1 group was higher(10.7 g/L vs 7.9 g/L,P<0.0001).Conclusion Micronutrient fortified complementary food supplements,with large-dose vitamin A,is effective for children aged 6-12 months in terms of iron deficiency prevention.

文摘Background:Non-communicable chronic diseases have become the leading causes of disease burden worldwide.The trends and burden of“metabolic associated fatty liver disease”(MAFLD)are unknown.We aimed to investigate the cardiovascular and renal burdens in adults with MAFLD and non-alcoholic fatty liver disease(NAFLD).Methods:Nationally representative data were analyzed including data from 19,617 non-pregnant adults aged≥20years from the cross-sectional US National Health and Nutrition Examination Survey periods,1999 to 2002,2003 to 2006,2007 to 2010,and 2011 to 2016.MAFLD was defined by the presence of hepatic steatosis plus general overweight/obesity,type 2 diabetes mellitus,or evidence of metabolic dysregulation.Results:The prevalence of MAFLD increased from 28.4%(95% confidence interval 26.3–30.6)in 1999 to 2002 to 35.8%(33.8–37.9)in 2011 to 2016.In 2011 to 2016,among adults with MAFLD,49.0%(45.8–52.2)had hypertension,57.8%(55.2–60.4)had dyslipidemia,26.4%(23.9–28.9)had diabetes mellitus,88.7%(87.0–80.1)had central obesity,and 18.5%(16.3–20.8)were current smokers.The 10-year cardiovascular risk ranged from 10.5%to 13.1%;19.7%(17.6–21.9)had chronic kidney diseases(CKDs).Throughthe fourperiods,adults withMAFLD showedanincrease in obesity;increase intreatment to lower bloodpressure(BP),lipids,and hemoglobin A1c;and increase in goal achievements for BP and lipids but not in goal achievement for glycemic control in diabetes mellitus.Patients showed a decreasing 10-year cardiovascular risk over time but no change in the prevalence of CKDs,myocardial infarction,or stroke.Generally,although participants with NAFLD and those with MAFLD had a comparable prevalence of cardiovascular disease and CKD,the prevalence of MAFLD was significantly higher than that of NAFLD.Conclusions:From 1999 to 2016,cardiovascular and renal risks and diseases have become highly prevalent in adults with MAFLD.The absolute cardiorenal burden may be greater for MAFLD than for NAFLD.These data call for early identification and risk stratification of MAFLD and close collaboration between endocrinologists and hepatologists.
