Background The establishment of a robust gut microbiota in piglets during their early developmental stage holds the potential for long-term advantageous effects.However,the optimal timeframe for introducing probiotics to achieve this outcome remains uncertain.Results In the context of this investigation,we conducted a longitudinal assessment of the fecal microbiota of 63 piglets at three distinct pre-weaning time points.Simultaneously,we gathered vaginal and fecal samples from 23 sows.Employing 16S rRNA gene and metagenomic sequencing methodologies,we conducted a comprehensive analysis of the fluctuation patterns in microbial composition,functional capacity,interaction networks,and colonization resistance within the gut microbiota of piglets.As the piglets progressed in age,discernible modifications in intestinal microbial diversity,composition,and function were observed.A source-tracking analysis unveiled the pivotal role of fecal and vaginal microbiota derived from sows in populating the gut microbiota of neonatal piglets.By D21,the microbial interaction network displayed a more concise and efficient configuration,accompanied by enhanced colonization resistance relative to the other two time points.Moreover,we identified three strains of Ruminococcus sp.at D10 as potential candidates for improving piglets’weight gain during the weaning phase.Conclusions The findings of this study propose that D10 represents the most opportune juncture for the introduction of external probiotic interventions during the early stages of piglet development.This investigation augments our comprehension of the microbiota dynamics in early-life of piglets and offers valuable insights for guiding forthcoming probiotic interventions. 展开更多

Liver cancer is a prevalent malignant cancer,ranking third in terms of mortality rate.Metastasis and recurrence primarily contribute to the high mortality rate of liver cancer.Hepatocellular carcinoma(HCC)has low expression of focal adhesion kinase(FAK),which increases the risk of metastasis and recurrence.Nevertheless,the efficacy of FAK phosphorylation inhibitors is currently limited.Thus,investigating the mechanisms by which FAK affects HCC metastasis to develop targeted therapies for FAK may present a novel strategy to inhibit HCC metastasis.This study examined the correlation between FAK expression and the prognosis of HCC.Additionally,we explored the impact of FAK degradation on HCC metastasis through wound healing experiments,transwell invasion experiments,and a xenograft tumor model.The expression of proteins related to epithelial-mesenchymal transition(EMT)was measured to elucidate the underlying mechanisms.The results showed that FAK PROTAC can degrade FAK,inhibit the migration and invasion of HCC cells in vitro,and notably decrease the lung metastasis of HCC in vivo.Increased expression of E-cadherin and decreased expression of vimentin indicated that EMT was inhibited.Consequently,degradation of FAK through FAK PROTAC effectively suppressed liver cancer metastasis,holding significant clinical implications for treating liver cancer and developing innovative anti-neoplastic drugs. 展开更多

A dual-route optical emission spectroscopy(D-OES)diagnostic is newly developed to monitor the optical emission from the X-point plasma region on the HL-2 A tokamak.This diagnostic is composed of an imaging system,a beam-splitting system for dual-route measurements,fiber bundles,a spectrometer system,and a control and acquisition system.One route is used to obtain wide-spectral-range spectra,and the other route is used to acquire high-wavelengthresolution line shapes.The spectral resolution of the wide-range spectrometers is 0.8 nm with a coverage of 800 nm(@200-1000 nm).The spectral resolution of the high-resolution spectrometer is 0.01 nm with a coverage of 6 nm(@200-660 nm).The spatial resolution of each route of D-OES is about 4 cm with 11 channels.The temporal resolution is 16 ms at maximum in the single-channel mode.Wide-range spectra(containing Balmer series and a Fulcher band)and highly resolved Ha line shapes are obtained by D-OES in the hydrogen glow discharge in the lab.D-OES measurements are carried out in the high-density deuterium experiments of HL-2A.The electron density n_(e)and deuterium temperature T_(D) in the X-point multifaceted asymmetric radiation from the edge(MARFE)region are derived simultaneously by fitting the measured D_(a) shape.The density n_(e)is observed to increase from~8.7×10^(18)m^(-3)to~7.8×10^(19)m^(-3),and the temperature T_(D)drops from~14.4 eV to~2.3 eV after the onset of MARFE in the discharge#38260. 展开更多

Background:Shengxuebao mixture(SXBM)is a novel herbal drug approved by China State Food and Drug Administration for the treatment of Leukopenia and iron deficiency anemia caused by radiotherapy and chemotherapy.Methods:To explore the mechanism of SXBM in treating blood deficiency syndrome(BDS).Firstly,network pharmacology and in vivo experiments were used to screen candidate targets and important signaling pathways of SXBM,GO functional enrichment and KEGG pathway analysis were performed.Secondly,a BDS rat model was established to verify the results of the analysis of network pharmacological enrichment.Histopathology and routine peripheral blood examination were observed.The expressions of tumor necrosis factor-α,interleukin(IL)-6,HIF-1αand NF-κB were detected by Western blot,and the expressions of IL-6,IL-1βwere detected by ELISA.Results:62 bioactive components,66 potential targets and 131 signaling pathways of BDS were successfully identified by network pharmacology.Molecular docking simulation techniques showed that key targets tumor necrosis factor-α,IL-6,IL-1βcan dock well with crucial components,and the BDS-related signaling pathways HIF-1 and JAK-STAT play a vital role.The combined model experiment of acetylphenylhydrazine and cyclophosphamide showed that the model group had obvious blood deficiency,and the histopathology and blood routine were effectively restored after administration.Our findings indicate that SXBM’s therapeutic effect on BDS primarily involves the mediation of the HIF-1α/NF-κB signaling pathway and the regulation of hematopoietic factor expression.Conclusion:This study not only affirmed the protective properties of SXBM against BDS but also provided insights into a potential mechanism for blood replenishment in the treatment of BDS using SXBM. 展开更多

为有效改善蒸馏膜(MD)的高能耗问题,提出一种低能耗的太阳纳米光子蒸馏膜(NESMD)组件,对其热力学性能开展研究,并分析关键操作参数对其热力学特性的影响规律。研究结果表明,温度极化是导致MD技术高能耗的主要内因,NESMD组件采用面加热... 为有效改善蒸馏膜(MD)的高能耗问题,提出一种低能耗的太阳纳米光子蒸馏膜(NESMD)组件,对其热力学性能开展研究,并分析关键操作参数对其热力学特性的影响规律。研究结果表明,温度极化是导致MD技术高能耗的主要内因,NESMD组件采用面加热方式进行料液加热,不存在温度极化现象,因此具有显著的节能优势。在相同进料温度和进料速度下,NESMD组件相较于MD组件,温度极化系数和热效率均有显著提升,NESMD组件热效率受潜热影响最大,可采取低进料速度和高进料温度来提高热效率;当将聚乙烯醇(PVA)涂层作为功率输入段时,NESMD和MD组件均存在最佳输入段长度使得热效率最大;NESMD组件相较于MD组件具有更好的拓展性和更长的活性长度。上述性能规律为进一步优化结构提供了科学依据。 展开更多

喷水射流预冷对加力燃烧室特性影响分析

为了探究采用射流预冷技术之后加力燃烧室性能,开展了不同喷嘴布置方案、喷水量和来流温度对预冷效果的影响研究。对射流预冷发动机工作过程进行了简化,建立了加力燃烧室进口前段射流预冷喷水特性计算的数学模型。同时搭建了小型试验台... 为了探究采用射流预冷技术之后加力燃烧室性能,开展了不同喷嘴布置方案、喷水量和来流温度对预冷效果的影响研究。对射流预冷发动机工作过程进行了简化,建立了加力燃烧室进口前段射流预冷喷水特性计算的数学模型。同时搭建了小型试验台,通过与试验结果的比对验证了该模型的准确性,并利用该模型对射流预冷效果进行了仿真预测。结果表明:提高喷嘴数量与布置均匀性能够小幅度改善预冷效果;当来流温度不变时,射流预冷喷射腔室出口处的液态水蒸发量随着喷水量的增加而提高,但蒸发率却处于下降的趋势;当喷水量达到2%时,加力燃烧室燃烧效率对比不喷水工况会有一定的提升;喷水量达到4%以后,加力燃烧室出口温度及燃烧效率随着喷水量的提高而降低;喷水量大于8%以后,恶化了加力燃烧室(V型火焰稳定器)贫油熄火极限与燃烧效率;喷水量达到最大10%时,油气比需从原来设计工况的0.052上升到0.064才能保持稳定点火且对比不喷水时工况,加力燃烧室出口温度由1860K下降到1373K,燃烧效率由80.2%下降到69.2%。 展开更多

背景:类风湿关节炎是常见的自身免疫性疾病,主要侵犯机体关节部位,特征为持续性滑膜炎症,研究提示促炎性的细胞焦亡与类风湿关节炎疾病的发生发展密切相关。目的:明确类风湿关节炎患者血清和关节液中细胞焦亡相关因子NOD样受体蛋白3、... 背景:类风湿关节炎是常见的自身免疫性疾病,主要侵犯机体关节部位,特征为持续性滑膜炎症,研究提示促炎性的细胞焦亡与类风湿关节炎疾病的发生发展密切相关。目的:明确类风湿关节炎患者血清和关节液中细胞焦亡相关因子NOD样受体蛋白3、半胱氨酸天冬氨酸蛋白酶1、白细胞介素18及白细胞介素1β的表达水平及其临床意义。方法:选择2021年1月至2022年1月河南省洛阳正骨医院(河南省骨科医院)风湿病科收治的类风湿关节炎患者24例(类风湿关节炎组),同期选取伴膝关节积液的骨关节炎患者30例(骨关节炎组),健康受试者40例(对照组)。检测并记录3组受试者的C-反应蛋白、红细胞沉降率、抗环瓜氨酸肽抗体、类风湿因子水平和类风湿关节炎28个关节疾病活动度评分。采用酶联免疫吸附法测定血清和关节液中NOD样受体蛋白3、半胱氨酸天冬氨酸蛋白酶1、白细胞介素18及白细胞介素1β的表达水平,分析上述4项因子表达水平与类风湿关节炎患者临床指标之间的相关性。结果与结论:①类风湿关节炎组患者血清中的NOD样受体蛋白3、半胱氨酸天冬氨酸蛋白酶1、白细胞介素18及白细胞介素1β水平显著高于对照组(P<0.0001),关节液中NOD样受体蛋白3、半胱氨酸天冬氨酸蛋白酶1、白细胞介素18及白细胞介素1β水平显著高于骨关节炎组(P<0.05-0.01);②类风湿关节炎患者血清和关节液中NOD样受体蛋白3、半胱氨酸天冬氨酸蛋白酶1、白细胞介素18及白细胞介素1β的表达水平分别与类风湿关节炎患者的类风湿关节炎28个关节疾病活动度评分、C-反应蛋白和红细胞沉降率水平呈正相关;③提示细胞焦亡相关因子在类风湿关节炎患者中明显升高,且与疾病活动度相关,提示细胞焦亡可能在类风湿关节炎发病中发挥作用。 展开更多

M2 macrophages mediate fibrotic scar formation in the early stages after cerebral ischemia in rats 被引量:1

In the central nervous system, the formation of fibrotic scar after injury inhibits axon regeneration and promotes repair. However, the mechanism underlying fibrotic scar formation and regulation remains poorly understood. M2 macrophages regulate fibrotic scar formation after injury to the heart, lung, kidney, and central nervous system. However, it remains to be clarified whether and how M2 macrophages regulate fibrotic scar formation after cerebral ischemia injury. In this study, we found that, in a rat model of cerebral ischemia induced by middle cerebral artery occlusion/reperfusion, fibrosis and macrophage infiltration were apparent in the ischemic core in the early stage of injury(within 14 days of injury). The number of infiltrated macrophages was positively correlated with fibronectin expression. Depletion of circulating monocyte-derived macrophages attenuated fibrotic scar formation. Interleukin 4(IL4) expression was strongly enhanced in the ischemic cerebral tissues, and IL4-induced M2 macrophage polarization promoted fibrotic scar formation in the ischemic core. In addition, macrophage-conditioned medium directly promoted fibroblast proliferation and the production of extracellular matrix proteins in vitro. Further pharmacological and genetic analyses showed that sonic hedgehog secreted by M2 macrophages promoted fibrogenesis in vitro and in vivo, and that this process was mediated by secretion of the key fibrosis-associated regulatory proteins transforming growth factor beta 1 and matrix metalloproteinase 9. Furthermore, IL4-afforded functional restoration on angiogenesis, cell apoptosis, and infarct volume in the ischemic core of cerebral ischemia rats were markedly impaired by treatment with an sonic hedgehog signaling inhibitor, paralleling the extent of fibrosis. Taken together, our findings show that IL4/sonic hedgehog/transforming growth factor beta 1 signaling targeting macrophages regulates the formation of fibrotic scar and is a potential therapeutic target for ischemic stroke. 展开更多

目的:评估地塞米松玻璃体内植入剂(DEX-I)治疗视网膜静脉阻塞(RVO)继发不同类型黄斑水肿的疗效。方法:回顾性研究。收集2019-07/2020-06在大连市第三人民医院诊断为RVO-ME并接受DEX-I(0.7mg)为初始治疗的患者46例46眼,根据光学相干断层... 目的:评估地塞米松玻璃体内植入剂(DEX-I)治疗视网膜静脉阻塞(RVO)继发不同类型黄斑水肿的疗效。方法:回顾性研究。收集2019-07/2020-06在大连市第三人民医院诊断为RVO-ME并接受DEX-I(0.7mg)为初始治疗的患者46例46眼,根据光学相干断层扫描成像(OCT)形态特征分为弥漫性视网膜增厚型(DRT型,13例13眼)、黄斑囊样水肿型(CME型,22例22眼)、浆液性视网膜脱离型(SRD型,11例11眼)。分别于治疗前、治疗后1、2、3mo观察并比较3种黄斑水肿分型患者最佳矫正视力(BCVA)和中心凹视网膜厚度(CRT)情况。结果:与治疗前比较,DRT型、CME型、SRD型患者BCVA治疗后1、2、3mo均显著改善,CRT均显著降低(均P<0.017)。治疗后3mo,DRT型和SRD型患者BCVA(0.21±0.12、0.22±0.10LogMAR)均显著优于CME型(0.45±0.14LogMAR,均P<0.017),CRT(254.08±49.07、248.92±44.19μm)均显著低于CME型(314.70±92.66μm,均P<0.017)。结论:DEX-I治疗RVO继发各种OCT类型黄斑水肿均有效,CME型治疗反应相较DRT型和SRD型差。 展开更多

目的:观察糖尿病合并年龄相关性白内障患者晶状体上皮细胞(LECs)中色素上皮衍生因子(PEDF)、血管内皮细胞生长因子(VEGF)的表达水平,探讨糖尿病合并年龄相关性白内障的发病机制。方法:回顾性研究。收集2020-08/2021-04在蚌埠医学院第一... 目的:观察糖尿病合并年龄相关性白内障患者晶状体上皮细胞(LECs)中色素上皮衍生因子(PEDF)、血管内皮细胞生长因子(VEGF)的表达水平,探讨糖尿病合并年龄相关性白内障的发病机制。方法:回顾性研究。收集2020-08/2021-04在蚌埠医学院第一附属医院眼科就诊的年龄相关性白内障和2型糖尿病合并年龄相关性白内障患者各30例。所有患者白内障超声乳化术中收取术眼晶状体中央区5.5~6.0mm直径的前囊膜标本,采用蛋白免疫印迹法(Western-blot)检测LECs中PEDF、VEGF蛋白表达水平。实时荧光定量PCR(qRT-PCR)方法检测PEDF、VEGF mRNA的相对表达量。结果:两组患者LECs中均存在PEDF、VEGF的表达,2型糖尿病合并年龄相关性白内障组患者VEGF mRNA相对表达量为1.364±0.062,高于年龄相关性白内障组的1.000±0.0(P<0.01),PEDF mRNA的相对表达量为0.398±0.053,明显低于年龄相关性白内障组的1.000±0.0(P<0.001)。2型糖尿病合并年龄相关性白内障组患者LECs中VEGF和PEDF蛋白表达量为2.053±0.026、0.579±0.045,年龄相关性白内障组为1.680±0.064、1.058±0.007(均P<0.01)。结论:2型糖尿病合并年龄相关性白内障患者LECs中PEDF和VEGF表达水平发生变化,可能与糖尿病患者白内障的发生和发展有关。 展开更多

At present,developing a simple strategy to effectively solve the shackles of volume expansion,poor conductivity and interface compatibility faced by Si-C anode in lithium batteries(LIBs)is the key to its commercialization.Here,low-cost nano-Si powders were prepared from Si-waste of solar-cells by sanding treatment,which can effectively reduce the commercialization cost for Si-C anode.Furthermore,micro-nano structured Gr@Si/C/TiO_(2) anode materials with graphite(Gr)as the inner core,TiO_(2)-doped and carbon-coated Si as the outer coating-layer,were synthesized at kilogram-scale per milling batch.Comprehensive characterization results indicate that TiO_(2)-doped carbon layer can improve the interface compatibility with the electrolyte,further promote the reduction of electrode polarization,and finally enhance the battery performance for the Gr@Si/C/TiO_(2) anodes.Accordingly,Gr@Si/C/TiO_(2) composites can output excellent LIB performance,especially with high initial coulombic efficiency(ICE)of 82.51%and large average reversible capacity of~810 mA h g^(-1) at 0.8 A g^(-1) after 1000 cycles.Moreover,Gr@Si/C/TiO_(2)‖NCM811 pouch full cells deliver impressive performance especially with high energy density of~489.3 W h kg^(-1) based on the total weight of active materials,suggesting its promising application in the high performance LIBs. 展开更多

We investigate the two-mode quantum Rabi model(QRM)describing the interaction between a two-level atom and a two-mode cavity field.The quantum phase transitions are found when the ratioηof transition frequency of atom to frequency of cavity field approaches infinity.We apply the Schrieffer–Wolff(SW)transformation to derive the low-energy effective Hamiltonian of the two-mode QRM,thus yielding the critical point and rich phase diagram of quantum phase transitions.The phase diagram consists of four regions:a normal phase,an electric superradiant phase,a magnetic superradiant phase and an electromagnetic superradiant phase.The quantum phase transition between the normal phase and the electric(magnetic)superradiant phase is of second order and associates with the breaking of the discrete Z_(2) symmetry.On the other hand,the phase transition between the electric superradiant phase and the magnetic superradiant phase is of first order and relates to the breaking of the continuous U(1)symmetry.Several important physical quantities,for example the excitation energy and average photon number in the four phases,are derived.We find that the excitation spectra exhibit the Nambu–Goldstone mode.We calculate analytically the higher-order correction and finite-frequency exponents of relevant quantities.To confirm the validity of the low-energy effective Hamiltonians analytically derived by us,the finite-frequency scaling relation of the averaged photon numbers is calculated by numerically diagonalizing the two-mode quantum Rabi Hamiltonian. 展开更多

The structural transformations,oxygen releasing and side reactions with electrolytes on the surface are considered as the main causes of the performance degradation of Li-rich layered oxides(LROs)cathodes in Li-ion batteries.Thus,stabilizing the surfaces of LROs is the key to realize their practical application in high energy density Li-ion batteries.Surface coating is regarded as one of the most effective strategies for high voltage cathodes.The ideal coating materials should prevent cathodes from electrolyte corrosion and possess both electronic and Li-ionic conductivities simultaneously.However,commonly reported coating materials are unable to balance these functions well.Herein,a new type of coating material,La_(2)CuO_(4)was introduced to mitigate the surface issues of LROs for the first time,due to its superb electronic conductivity(26-35 mS·cm^(-1))and lithium-ionic diffusion coefficient(10^(-12)-10^(-13)cm^(2)·s^(-1)).After coating with the La_(2)CuO_(4),the capacity retention of Li_(1.2)Ni_(0.54)Co_(0.13)Mn_(0.13)O_(2)cathode was increased to 85.9%(compared to 79.3%of uncoated cathode)after 150 cycles in the voltage range of 2.0-4.8 V.In addition,only negligible degradations on the deliverable capacity and rate capability were observed. 展开更多
