The soil heavy metal pollution degree was evaluated by national environmental quality standard (II class). The results showed that three soil metals of P. massoniana × S. superba were the highest, and the soil metals enrichment ability was strong. The order of single factor pollution index of metal elements was Cu (1.38) > Cr (0.81) > Zn (0.42), and moderately pollution, pollution warning and no pollution, respectively. There was no significant correlation between three soil heavy metals and soil total carbon (TC), total nitrogen (TN) and total phosphorus (TP). These results suggested that the accumulation of heavy metal elements was not derived from the parent material of soil. There was a significant positive correlation between the three metal elements which indicated that the sources of the three elements were similar. The structural equation model showed that the direct and indirect effects among the influencing factors ultimately affected the activity of heavy metals by cascade effects.展开更多
The rapid advancement and broad application of machine learning(ML)have driven a groundbreaking revolution in computational biology.One of the most cutting-edge and important applications of ML is its integration with...The rapid advancement and broad application of machine learning(ML)have driven a groundbreaking revolution in computational biology.One of the most cutting-edge and important applications of ML is its integration with molecular simulations to improve the sampling efficiency of the vast conformational space of large biomolecules.This review focuses on recent studies that utilize ML-based techniques in the exploration of protein conformational landscape.We first highlight the recent development of ML-aided enhanced sampling methods,including heuristic algorithms and neural networks that are designed to refine the selection of reaction coordinates for the construction of bias potential,or facilitate the exploration of the unsampled region of the energy landscape.Further,we review the development of autoencoder based methods that combine molecular simulations and deep learning to expand the search for protein conformations.Lastly,we discuss the cutting-edge methodologies for the one-shot generation of protein conformations with precise Boltzmann weights.Collectively,this review demonstrates the promising potential of machine learning in revolutionizing our insight into the complex conformational ensembles of proteins.展开更多
To provide a scientific basis for controlling mulberry bacterial blight in Bazhong,Sichuan,China(BSC),this study aimed to isolate and purify pathogenic bacteria from diseased branches of mulberry trees in the region a...To provide a scientific basis for controlling mulberry bacterial blight in Bazhong,Sichuan,China(BSC),this study aimed to isolate and purify pathogenic bacteria from diseased branches of mulberry trees in the region and to clarify their taxonomic status using morphological observation,physiological and biochemical detection,molecular-level identification,and the construction of a phylogenetic tree.A total of 218 bacterial strains were isolated from samples of diseased mulberry branches.Of these,7 strains were identified as pathogenic bacteria based on pathogenicity tests conducted in accordance with Koch’s postulates.Preliminary findings from the analysis of the 16S rRNA sequence indicated that the 7 pathogenic bacteria are members of Klebsiella spp.Morphological observation revealed that the pathogenic bacteria were oval-shaped and had capsules but no spores.They could secrete pectinase,cellulase,and protease and were able to utilize D-glucose,D-mannose,D-maltose,and D-Cellobiose.The 7 strains of pathogenic bacteria exhibited the highest homology with Klebsiella oxytoca.This study identifies Klebsiella oxytoca as the causative agent of mulberry bacterial blight in BSC,laying the foundation for the prevention and control of this pathogen and further investigation into its pathogenic mechanism.展开更多
Circular RNA(circ RNA),classified as a type of non-coding RNA,has gained significant attention in the field of biology due to its distinctive ring structure and functional properties.Recent research has provided evide...Circular RNA(circ RNA),classified as a type of non-coding RNA,has gained significant attention in the field of biology due to its distinctive ring structure and functional properties.Recent research has provided evidence that specific circ RNAs have the ability to modulate disease progression through diverse mechanisms,one of which is by regulating cellular ferroptosis.Ferroptosis is a form of regulated cell death that is driven by iron dependency and lipid peroxidation,and extensive investigations have revealed a relationship between ferroptosis and disease development.In addition to evidence that both circ RNAs and ferroptosis exert critical roles in disease progression,circ RNAs have also been shown to actively mediate the process of ferroptosis.The relationship between circ RNAs and ferroptosis therefore influences disease progression and offers novel targets for disease treatment.By directly or indirectly modulating the expression of circ RNAs that regulate the expression of ferroptosisrelated proteins,it may be possible to impact disease progression by promoting or inhibiting ferroptosis.Current research indicates such approaches may hold significant value in a wide variety of common diseases across physiological systems.This review comprehensively summarizes the findings of recent studies investigating the roles of circ RNAs in the regulation of ferroptosis in various diseases.展开更多
To break the narrow diversity bottleneck of the wheat D genome, a set of Aegilops tauschii-wheat introgression(A-WI) lines was developed by crossing Ae. tauschii accession T015 with common wheat elite cultivar Zhoumai...To break the narrow diversity bottleneck of the wheat D genome, a set of Aegilops tauschii-wheat introgression(A-WI) lines was developed by crossing Ae. tauschii accession T015 with common wheat elite cultivar Zhoumai 18(Zhou18). A high-density genetic map was constructed based on Single Nucleotide Polymorphism(SNP) markers and 15 yield-related traits were evaluated in 11 environments for detecting quantitative trait loci(QTL). A total of 27 environmentally stable QTL were identified in at least five environments, 20 of which were derived from Ae. tauschii T015, explaining up to 24.27% of the phenotypic variations. The major QTL for kernel length(KL), QKl-2D.5, was delimited to a physical interval of approximately 2.6 Mb harboring 52 candidate genes. Three Kompetitive Allele Specific PCR(KASP)markers were successfully developed based on nonsynonymous nucleotide mutations of candidate gene AetT093_2Dv1G100900.1 and showed that A-WI lines with the T015 haplotype had significantly longer KL than the Zhou18 haplotype across all 11 environments. Four primary valuable A-WIs with good trait performance and carrying yield-related QTL were selected for breeding improvement. The results will facilitate the efficient transfer of beneficial genes from Ae. tauschii into wheat cultivars to improve wheat yield and other traits.展开更多
BACKGROUND Current research lacks a model of knee extension contracture in rats.AIM To elucidate the formation process of knee extension contracture.METHODS We developed a rat model using an aluminum external fixator....BACKGROUND Current research lacks a model of knee extension contracture in rats.AIM To elucidate the formation process of knee extension contracture.METHODS We developed a rat model using an aluminum external fixator.Sixty male Sprague-Dawley rats with mature bones were divided into the control group(n=6)and groups that had the left knee immobilized with an aluminum external fixator for 1,2,and 3 d,and 1,2,3,4,6,and 8 wk(n=6 in each group).The passive extension range of motion,histology,and expression of fibrosis-related proteins were compared between the control group and the immobilization groups.RESULTS Myogenic contracture progressed very quickly during the initial 2 wk of immobilization.After 2 wk,the contracture gradually changed from myogenic to arthrogenic.The arthrogenic contracture progressed slowly during the 1^(st) week,rapidly progressed until the 3^(rd) week,and then showed a steady progression until the 4^(rd) week.Histological analyses confirmed that the anterior joint capsule of the extended fixed knee became increasingly thicker over time.Correspondingly,the level of transforming growth factor beta 1(TGF-β1)and phosphorylated mothers against decapentaplegic homolog 2(p-Smad2)in the anterior joint capsule also increased with the immobilization time.Over time,the cross-sectional area of muscle fibers gradually decreased,while the amount of intermuscular collagen and TGF-β1,p-Smad2,and p-Smad3 was increased.Unexpectedly,the amount of intermuscular collagen and TGF-β1,p-Smad2,and p-Smad3 was decreased during the late stage of immobilization(6-8 wk).The myogenic contracture was stabilized after 2 wk of immobilization,whereas the arthrogenic contracture was stabilized after 3 wk of immobilization and completely stable in 4 wk.CONCLUSION This rat model may be a useful tool to study the etiology of joint contracture and establish therapeutic approaches.展开更多

While most observations of pull-apart basins were from surface mapping or laboratory experiments, we investigated a nascent pull-apart basin in the subsurface. We characterized a nascent pull-apart basin along the strike-slip fault within the Woodford Shale by using seismic attributes analyses, including coherence, dip-azimuth, and curvature. The results indicate a 32 km long, N-S striking strike-slip fault that displays a distinct but young pull-apart basin, which is ~1.6 km by 3.2 km in size and is bounded by two quasi-circular faults. The curvature attribute map reveals two quasi-circular folds, which depart from the main strike-slip fault at ~25°, resulting in an elliptical basin. Inside the basin, a series of echelon quasi-circular normal faults step into the bottom of the basin with ~80 m of total subsidence. We propose that the controls of the shape of pull-apart basin are the brittleness of the shale, and we suggest proper seismic attributes as a useful tool for investigating high fracture intensity in the subsurface for hydrofracturing and horizontal drilling within the shale.展开更多

Methods: A retrospective analysis of COVID-19 patients treated at a tertiary hospital in Mianyang City, Sichuan Province, China, from 1 May to 31 May 2023 was performed. The patients were divided into two groups: 66 cases in the moderate group and 61 cases in the severe group. Additionally, 69 uninfected individuals from the medical examination center during the same period were selected as the control group. Spearman rank correlation was used to determine the correlation between the indices and COVID-19 severity. Multiple logistic regression analysis was performed to identify the factors affecting COVID-19 severity. ROC curves were constructed to assess the predictive value of the TyG and TyG-BMI indices for severe COVID-19. Results: There were significant differences in smoking and diabetes between the three groups (P Conclusion: Smoking, AST, ALB, TyG index, and TyG-BMI index are valuable in assessing the severity of COVID-19, with the TyG-BMI index having a higher predictive value than the TyG index.展开更多

Nucleic acids in exosomes are a group of molecules that can act as biomarkers. Currently, there are many reports on exosomal microRNAs, which are ideal biomarkers for the early diagnosis of cancer. However, there are few reports on the role of exosomal microRNAs in the diagnosis and prognosis of hepatocellular carcinoma(HCC).AIM To understand the mechanism of exosomal microRNA-224(miR-224) in the development of HCC and evaluate its diagnostic and prognostic value.METHODS Cell culture and transfection of exosomal miRNA-224, real-time quantitative PCR, luciferase reporter assay, and other methods were used to find new biomarkers related to the development of HCC that can be used to diagnose HCC and predict HCC prognosis.RESULTSBy targeting glycine N-methyltransferase, incubating exosomes with miR-224 mimic resulted in a significant increase in cell proliferation compared to that of the control group, while incubation with the miR-224 inhibitor significantly reduced cell proliferation. The same results were obtained for the cell invasion assay. Serum exosomal miR-224 did have some ability to differentiate patients with HCC from healthy controls, with an area under the curve of 0.910, and HCC patients with higher serum exosomal miR-224 expression had lower overall survival.CONCLUSION Exosomal miR-224 is a tumor promotor and can be a marker of diagnosis and prognosis of HCC patients, however, its ability to distinguish liver diseases needs further verification.展开更多

METHODS: Total RNA was extracted from 80 colon cancer tissues and matched tumor-adjacent normal colon tissues and reverse transcribed. Quantitative polymerase chain reaction was used to detect the expression of HOTAIR. The relationship between the expression of HOTAIR and clinicopathological parameters of colon cancer was analyzed. RESULTS: The expression of HOTAIR was significantly higher in colon cancer tissues than in matched tumoradjacent normal colon tissues(P < 0.05). HOTAIR expression was significantly higher in cases with lymph node metastasis than in those without metastasis; in lowly differentiated and undifferentiated cases than in highly and moderately differentiated cases; and in stages Ⅲ + Ⅳ cases than in stages?Ⅰ?+ Ⅱ cases(P < 0.05).CONCLUSION: HOTAIR expression is upregulated in colon cancer, suggesting that HOTAIR plays an important role in the tumorigenesis, development and metastasis of colon cancer. HOTAIR may act as an oncogene and represents a new molecular target for the treatment of colon cancer.展开更多
AIM To evaluate whether fish oil(FO) can protect liver injury induced by intestinal ischemia/reperfusion(I/R) via the AMPK/SIRT-1/autophagy pathway.METHODS Ischemia in wistar rats was induced by superior mesenteric ar...AIM To evaluate whether fish oil(FO) can protect liver injury induced by intestinal ischemia/reperfusion(I/R) via the AMPK/SIRT-1/autophagy pathway.METHODS Ischemia in wistar rats was induced by superior mesenteric artery occlusion for 60 min and reperfusion for 240 min. One milliliter per day of FO emulsion or normal saline was administered by intraperitoneal injection for 5 consecutive days to each animal. Animals were sacrificed at the end of reperfusion. Blood andtissue samples were collected for analyses. AMPK, SIRT-1, and Beclin-1 expression was determined in lipopolysaccharide(LPS)-stimulated HepG2 cells with or without FO emulsion treatment.RESULTS Intestinal I/R induced significant liver morphological changes and increased serum alanine aminotransferase and aspartate aminotransferase levels. Expression of p-AMPK/AMPK, SIRT-1, and autophagy markers was decreased whereas tumor necrosis factor-α(TNF-α) and malonaldehyde(MDA) were increased. FO emulsion blocked the changes of the above indicators effectively. Besides, in LPS-stimulated HepG2 cells, small interfering RNA(siRNA) targeting AMPK impaired the FO induced increase of p-AMPK, SIRT-1, and Beclin-1 and decrease of TNF-α and MDA. SIRT-1 siRNA impaired the increase of SIRT-1 and Beclin-1 and the decrease of TNF-α and MDA.CONCLUSION Our study indicates that FO may protect the liver against intestinal I/R induced injury through the AMPK/SIRT-1/autophagy pathway.展开更多

After 6 wk course of diabetes, the treatment groups were given β-elemene through periocular and intravitreous injection separately and the control groups were given blank emulsion injection. HE staining was used to observe the morphology of retina. The m RNA expressions of HIF-1α, VEGF and i NOS was assayed by real-time polymerase chain reaction(PCR) and the protein expression was measured by Western blot and immunocytochemistry methods.RESULTS: The results indicated that the protein and m RNA expressions of HIF-1α, VEGF and i NOS after treated by β-elemene periocularly and intravitreally injections were all found to be reduced compared with the levels in the diabetic rats group(P<0.05). The inhibitory effect of intravitreal injection was more remarkable.CONCLUSION: The results show β-elemene protect the retina of diabetic rats from high glucose damage by downregulating the expression of HIF-1α, VEGF and iNOS.展开更多

To solve this prob...With the increase in the power receiving proportion and an insufficient peak regulation capacity of the local units, the receiving-end power grid struggles to achieve peak regulation in valley time. To solve this problem while considering the potential of the large-scale charge load of electric vehicles(EVs), an aggregator-based demand response(DR) mechanism for EVs that are participating in the peak regulation in valley time is proposed in this study. In this aggregator-based DR mechanism, the profits for the power grid’s operation and the participation willingness of the EV owners are considered. Based on the characteristics of the EV charging process and the day-ahead unit generation scheduling, a rolling unit commitment model with the DR is established to maximize the social welfare. In addition, to improve the efficiency of the optimization problem solving process and to achieve communication between the independent system operator(ISO) and the aggregators, the clustering algorithm is utilized to extract typical EV charging patterns. Finally, the feasibility and benefits of the aggregator-based DR mechanism for saving the costs and reducing the peak-valley difference of the receiving-end power grid are verified through case studies.展开更多

文摘Circular RNA(circ RNA),classified as a type of non-coding RNA,has gained significant attention in the field of biology due to its distinctive ring structure and functional properties.Recent research has provided evidence that specific circ RNAs have the ability to modulate disease progression through diverse mechanisms,one of which is by regulating cellular ferroptosis.Ferroptosis is a form of regulated cell death that is driven by iron dependency and lipid peroxidation,and extensive investigations have revealed a relationship between ferroptosis and disease development.In addition to evidence that both circ RNAs and ferroptosis exert critical roles in disease progression,circ RNAs have also been shown to actively mediate the process of ferroptosis.The relationship between circ RNAs and ferroptosis therefore influences disease progression and offers novel targets for disease treatment.By directly or indirectly modulating the expression of circ RNAs that regulate the expression of ferroptosisrelated proteins,it may be possible to impact disease progression by promoting or inhibiting ferroptosis.Current research indicates such approaches may hold significant value in a wide variety of common diseases across physiological systems.This review comprehensively summarizes the findings of recent studies investigating the roles of circ RNAs in the regulation of ferroptosis in various diseases.

文摘To break the narrow diversity bottleneck of the wheat D genome, a set of Aegilops tauschii-wheat introgression(A-WI) lines was developed by crossing Ae. tauschii accession T015 with common wheat elite cultivar Zhoumai 18(Zhou18). A high-density genetic map was constructed based on Single Nucleotide Polymorphism(SNP) markers and 15 yield-related traits were evaluated in 11 environments for detecting quantitative trait loci(QTL). A total of 27 environmentally stable QTL were identified in at least five environments, 20 of which were derived from Ae. tauschii T015, explaining up to 24.27% of the phenotypic variations. The major QTL for kernel length(KL), QKl-2D.5, was delimited to a physical interval of approximately 2.6 Mb harboring 52 candidate genes. Three Kompetitive Allele Specific PCR(KASP)markers were successfully developed based on nonsynonymous nucleotide mutations of candidate gene AetT093_2Dv1G100900.1 and showed that A-WI lines with the T015 haplotype had significantly longer KL than the Zhou18 haplotype across all 11 environments. Four primary valuable A-WIs with good trait performance and carrying yield-related QTL were selected for breeding improvement. The results will facilitate the efficient transfer of beneficial genes from Ae. tauschii into wheat cultivars to improve wheat yield and other traits.

文摘BACKGROUND Current research lacks a model of knee extension contracture in rats.AIM To elucidate the formation process of knee extension contracture.METHODS We developed a rat model using an aluminum external fixator.Sixty male Sprague-Dawley rats with mature bones were divided into the control group(n=6)and groups that had the left knee immobilized with an aluminum external fixator for 1,2,and 3 d,and 1,2,3,4,6,and 8 wk(n=6 in each group).The passive extension range of motion,histology,and expression of fibrosis-related proteins were compared between the control group and the immobilization groups.RESULTS Myogenic contracture progressed very quickly during the initial 2 wk of immobilization.After 2 wk,the contracture gradually changed from myogenic to arthrogenic.The arthrogenic contracture progressed slowly during the 1^(st) week,rapidly progressed until the 3^(rd) week,and then showed a steady progression until the 4^(rd) week.Histological analyses confirmed that the anterior joint capsule of the extended fixed knee became increasingly thicker over time.Correspondingly,the level of transforming growth factor beta 1(TGF-β1)and phosphorylated mothers against decapentaplegic homolog 2(p-Smad2)in the anterior joint capsule also increased with the immobilization time.Over time,the cross-sectional area of muscle fibers gradually decreased,while the amount of intermuscular collagen and TGF-β1,p-Smad2,and p-Smad3 was increased.Unexpectedly,the amount of intermuscular collagen and TGF-β1,p-Smad2,and p-Smad3 was decreased during the late stage of immobilization(6-8 wk).The myogenic contracture was stabilized after 2 wk of immobilization,whereas the arthrogenic contracture was stabilized after 3 wk of immobilization and completely stable in 4 wk.CONCLUSION This rat model may be a useful tool to study the etiology of joint contracture and establish therapeutic approaches.

文摘A minimum-modified Debye-Hückel(DH)theory for electrolytes with size asymmetry is developed.Com-pared with the conventional DH theory,the minimum-modified DH theory only introduces an extra surface charge density to capture the electrostatic effect of the size asymmetry of the electrolytes and hence facilitates a boundary element method for electrostatic potential calculation.This theory can distinguish the electrostat-ic energies and excess chemical potentials of ions with the same sizes but opposite charges,and is applied to a binary primitive electrolyte solution with moderate electrostatic coupling.Compared with the hyper-netted chain theory,the validity of this modified DH theory demonstrates significant improvement over the conventional DH theory.
文摘Pull-apart basins are faulting and folding zones with high intensity of fractures that strongly affect the production in unconventional shale gas. While most observations of pull-apart basins were from surface mapping or laboratory experiments, we investigated a nascent pull-apart basin in the subsurface. We characterized a nascent pull-apart basin along the strike-slip fault within the Woodford Shale by using seismic attributes analyses, including coherence, dip-azimuth, and curvature. The results indicate a 32 km long, N-S striking strike-slip fault that displays a distinct but young pull-apart basin, which is ~1.6 km by 3.2 km in size and is bounded by two quasi-circular faults. The curvature attribute map reveals two quasi-circular folds, which depart from the main strike-slip fault at ~25°, resulting in an elliptical basin. Inside the basin, a series of echelon quasi-circular normal faults step into the bottom of the basin with ~80 m of total subsidence. We propose that the controls of the shape of pull-apart basin are the brittleness of the shale, and we suggest proper seismic attributes as a useful tool for investigating high fracture intensity in the subsurface for hydrofracturing and horizontal drilling within the shale.
文摘Objective: The objective of this study was to investigate the relationship between the triglyceride-glucose (TyG) index, triglyceride-glucose-BMI (TyG-BMI) index, laboratory indices, and disease severity in patients with COVID-19. Methods: A retrospective analysis of COVID-19 patients treated at a tertiary hospital in Mianyang City, Sichuan Province, China, from 1 May to 31 May 2023 was performed. The patients were divided into two groups: 66 cases in the moderate group and 61 cases in the severe group. Additionally, 69 uninfected individuals from the medical examination center during the same period were selected as the control group. Spearman rank correlation was used to determine the correlation between the indices and COVID-19 severity. Multiple logistic regression analysis was performed to identify the factors affecting COVID-19 severity. ROC curves were constructed to assess the predictive value of the TyG and TyG-BMI indices for severe COVID-19. Results: There were significant differences in smoking and diabetes between the three groups (P Conclusion: Smoking, AST, ALB, TyG index, and TyG-BMI index are valuable in assessing the severity of COVID-19, with the TyG-BMI index having a higher predictive value than the TyG index.
文摘In recent years,the study of production-living-ecological space has progressively expanded from urban to rural areas.With the onset of a new era in rural development,diverse requirements for rural landscapes have emerged.Consequently,rural landscape planning in this new era is incorporating the three-pillar conception of sustainability.Island villages,with their distinct natural ecology and marine resources compared to inland villages,offer unique conditions,resulting in the production of diversified landscape types with significant development potential.Despite the limited attention given to the domestic development of island villages,this paper delves into the analysis of the three-pillar conception of sustainability and explores landscape protection strategies and renewal modes specific to island villages.To illustrate these concepts,East Xiaoqing Island Village in Rushan City,Weihai City serves as a case study.
文摘Globally,digital technology and the digital economy have propelled technological revolution and industrial change,and it has become one of the main grounds of international industrial competition.It was estimated that the scale of China’s digital economy would reach 50 trillion yuan in 2022,accounting for more than 40%of GDP,presenting great market potential and room for the growth of the digital economy.With the rapid development of the digital economy,the state attaches great importance to the construction of digital infrastructure and has introduced a series of policies to promote the systematic development and large-scale deployment of digital infrastructure.In 2022 the Chinese government planned to build 8 arithmetic hubs and 10 national data center clusters nationwide.To proactively address the future demand for AI across various scenarios,there is a need for a well-structured computing power infrastructure.The data center,serving as the pivotal hub for computing power,has evolved from the conventional cloud center to a more intelligent computing center,allowing for a diversified convergence of computing power supply.Besides,the data center accommodates a diverse array of arithmetic business forms from customers,reflecting the multi-industry developmental trend.The arithmetic service platform is consistently broadening its scope,with ongoing optimization and innovation in the design scheme of machine room processes.The widespread application of submerged phase-change liquid cooling technology and cold plate cooling technology introduces a series of new challenges to the construction of digital infrastructure.This paper delves into the design objectives,industry considerations,layout,and other dimensions of a smart computing center and proposes a new-generation data center solution that is“flexible,resilient,green,and low-carbon.”

文摘BACKGROUND Exosomes contain proteins, lipids, and biological molecules such as DNA and RNA. Nucleic acids in exosomes are a group of molecules that can act as biomarkers. Currently, there are many reports on exosomal microRNAs, which are ideal biomarkers for the early diagnosis of cancer. However, there are few reports on the role of exosomal microRNAs in the diagnosis and prognosis of hepatocellular carcinoma(HCC).AIM To understand the mechanism of exosomal microRNA-224(miR-224) in the development of HCC and evaluate its diagnostic and prognostic value.METHODS Cell culture and transfection of exosomal miRNA-224, real-time quantitative PCR, luciferase reporter assay, and other methods were used to find new biomarkers related to the development of HCC that can be used to diagnose HCC and predict HCC prognosis.RESULTSBy targeting glycine N-methyltransferase, incubating exosomes with miR-224 mimic resulted in a significant increase in cell proliferation compared to that of the control group, while incubation with the miR-224 inhibitor significantly reduced cell proliferation. The same results were obtained for the cell invasion assay. Serum exosomal miR-224 did have some ability to differentiate patients with HCC from healthy controls, with an area under the curve of 0.910, and HCC patients with higher serum exosomal miR-224 expression had lower overall survival.CONCLUSION Exosomal miR-224 is a tumor promotor and can be a marker of diagnosis and prognosis of HCC patients, however, its ability to distinguish liver diseases needs further verification.

文摘AIM: To detect the expression of the long noncoding RNA HOTAIR in colon cancer and analyze its relationship with clinicopathological parameters of colon cancer. METHODS: Total RNA was extracted from 80 colon cancer tissues and matched tumor-adjacent normal colon tissues and reverse transcribed. Quantitative polymerase chain reaction was used to detect the expression of HOTAIR. The relationship between the expression of HOTAIR and clinicopathological parameters of colon cancer was analyzed. RESULTS: The expression of HOTAIR was significantly higher in colon cancer tissues than in matched tumoradjacent normal colon tissues(P < 0.05). HOTAIR expression was significantly higher in cases with lymph node metastasis than in those without metastasis; in lowly differentiated and undifferentiated cases than in highly and moderately differentiated cases; and in stages Ⅲ + Ⅳ cases than in stages?Ⅰ?+ Ⅱ cases(P < 0.05).CONCLUSION: HOTAIR expression is upregulated in colon cancer, suggesting that HOTAIR plays an important role in the tumorigenesis, development and metastasis of colon cancer. HOTAIR may act as an oncogene and represents a new molecular target for the treatment of colon cancer.

文摘AIM To evaluate whether fish oil(FO) can protect liver injury induced by intestinal ischemia/reperfusion(I/R) via the AMPK/SIRT-1/autophagy pathway.METHODS Ischemia in wistar rats was induced by superior mesenteric artery occlusion for 60 min and reperfusion for 240 min. One milliliter per day of FO emulsion or normal saline was administered by intraperitoneal injection for 5 consecutive days to each animal. Animals were sacrificed at the end of reperfusion. Blood andtissue samples were collected for analyses. AMPK, SIRT-1, and Beclin-1 expression was determined in lipopolysaccharide(LPS)-stimulated HepG2 cells with or without FO emulsion treatment.RESULTS Intestinal I/R induced significant liver morphological changes and increased serum alanine aminotransferase and aspartate aminotransferase levels. Expression of p-AMPK/AMPK, SIRT-1, and autophagy markers was decreased whereas tumor necrosis factor-α(TNF-α) and malonaldehyde(MDA) were increased. FO emulsion blocked the changes of the above indicators effectively. Besides, in LPS-stimulated HepG2 cells, small interfering RNA(siRNA) targeting AMPK impaired the FO induced increase of p-AMPK, SIRT-1, and Beclin-1 and decrease of TNF-α and MDA. SIRT-1 siRNA impaired the increase of SIRT-1 and Beclin-1 and the decrease of TNF-α and MDA.CONCLUSION Our study indicates that FO may protect the liver against intestinal I/R induced injury through the AMPK/SIRT-1/autophagy pathway.
文摘AIM: To evaluate the effect of β-elemene on the expressions of hypoxia-inducible factor(HIF)-lα, vascular endothelial growth factor(VEGF) and inducible nitric oxide synthase(i NOS) in a streptozotocin(STZ) induced diabetic SpragueDawley(SD) rat model.METHODS: SD rats were administered an abdominal injection of STZ and induced to a diabetic model. After 6 wk course of diabetes, the treatment groups were given β-elemene through periocular and intravitreous injection separately and the control groups were given blank emulsion injection. HE staining was used to observe the morphology of retina. The m RNA expressions of HIF-1α, VEGF and i NOS was assayed by real-time polymerase chain reaction(PCR) and the protein expression was measured by Western blot and immunocytochemistry methods.RESULTS: The results indicated that the protein and m RNA expressions of HIF-1α, VEGF and i NOS after treated by β-elemene periocularly and intravitreally injections were all found to be reduced compared with the levels in the diabetic rats group(P<0.05). The inhibitory effect of intravitreal injection was more remarkable.CONCLUSION: The results show β-elemene protect the retina of diabetic rats from high glucose damage by downregulating the expression of HIF-1α, VEGF and iNOS.

文摘With the increase in the power receiving proportion and an insufficient peak regulation capacity of the local units, the receiving-end power grid struggles to achieve peak regulation in valley time. To solve this problem while considering the potential of the large-scale charge load of electric vehicles(EVs), an aggregator-based demand response(DR) mechanism for EVs that are participating in the peak regulation in valley time is proposed in this study. In this aggregator-based DR mechanism, the profits for the power grid’s operation and the participation willingness of the EV owners are considered. Based on the characteristics of the EV charging process and the day-ahead unit generation scheduling, a rolling unit commitment model with the DR is established to maximize the social welfare. In addition, to improve the efficiency of the optimization problem solving process and to achieve communication between the independent system operator(ISO) and the aggregators, the clustering algorithm is utilized to extract typical EV charging patterns. Finally, the feasibility and benefits of the aggregator-based DR mechanism for saving the costs and reducing the peak-valley difference of the receiving-end power grid are verified through case studies.
