Objective To investigate the clinical application of Real-Time PCR for rapid detection of methicillin-resistant Staphylococcus aureus(MRSA) directly from nasopharyngeal swab specimens.Methods We collected the nasal and throat swab specimens from patients or medical staffs in 3 intensive care units,blood laminar flow ward and respiratory ward in Beijing Hospital,Ministry of Health from December 2010to April 2011.Each sample was tested by RT-PCR and conventional culture-based method for the presence of MRSA.Results The total number of the specimens was 206.Compared with the conventional culture-based method,we demonstrated the diagnostic values for Real-Time PCR were 96.4%sensitivity,96.6%specificity,81.8%positive predictive rate,and 99.4%negative predictive rate.And the limit of detection was 10~2CFU/ml.Conclusions This Real-Time PCR is a simple,rapid,sensitive and specific method.With the high negative predictive value,it can be used for the exclusion of MRSA colonization or infection.However,the application of its low positive predictive value should be further evaluated. 展开更多

Mosquitoes are of great medical significance as vectors of many deadly diseases.Mitogenomes have been widely used in phylogenetic studies,but mitogenome knowledge within the family Culicidae is limited,and Culicidae phylogeny is far from resolved.In this study,we surveyed the mitogenomes of 149 Culicidae species,including 7 newly sequenced species.Comparative analysis of 149 mosquito mitogenomes shows gene composition and order to be identical to that of an ancestral insect,and the AT bias,length variation,and codon usage are all consistent with that of other reported Dipteran mitogenomes.Phylogenetic analyses based on the DNA sequences of the 13 protein-coding genes from the 149 species robustly support the monophyly of the subfamily Anophelinae and the tribes Aedini,Culicini,Mansonini,Sabethini,and Toxorhynchitini.To resolve ambiguous relationships between clades within the subfamily Culicinae,we performed topological tests and show that Aedini is a sister to Culicini and that Uranotaenini is a sister to(Mansonini)+(Toxorhynchitini+Sabethini).In addition,we estimated divergence times using a Bayesian relaxation clock based on the sequence data and 3 fossil calibration points.The results show mosquitoes diverged during the Early Jurassic with massive Culicinae radiations during the Cretaceous,coincident with the emergence of angiosperms and the burst of mammals and birds.Overall,this study,which uses the largest number of Culicidae mitogenomes sequenced to date,comprehensively reveals the mitogenome characteristics and mitogenome-based phylogeny and divergence times of Culicidae,providing information for further studies on the mitogenome,phylogeny,evolution,and taxonomic revision of Culicidae. 展开更多

To solve the coupling relationship between the strip automatic gauge control and the looper control in traditional control strategy of tandem hot rolling,a distributed model predictive control(DMPC)strategy for the tandem hot rolling was explored,and a series of simulation experiments were carried out.Firstly,based on the state space analysis method,the multivariable dynamic transition process of hot strip rolling was studied,and the state space model of a gauge-looper integrated system in tandem hot rolling was established.Secondly,DMPC strategy based on neighborhood optimization was proposed,which fully considered the coupling relationship in this integrated system.Finally,a series of experiments simulating disturbances and emergency situations were completed with actual rolling data.The experimental results showed that the proposed DMPC control strategy had better performance compared with the traditional proportional-integral control and centralized model predictive control,which is applicable for the gauge-looper integrated system. 展开更多

Roll gap prediction in acceleration and deceleration process of cold rolling based on a data-driven method

Severe fluctuation of the effective roll gap in the acceleration and deceleration section of the cold rolling process is a significant factor causing thickness deviation.However,the conventional roll gap compensation method and control strategy do not meet the stringent strip quality requirements.The roll gap model in the acceleration and deceleration process is studied to increase the thickness control precision.In order to improve model accuracy,a roll gap prediction method based on data-driven is proposed.Given the complexities of the cold rolling process,the extreme gradient boosting(XGBoost)method is used to predict the roll gap model as the rolling speed changes.Meanwhile,support vector regression and neural network-based methods are taken to evaluate and compare the prediction performances.Based on the field data,the simulation experiments are carried out.It demonstrated that the prediction performance of the proposed method outperformed the other two methods.The values of root mean square error,determination coefficient value,mean absolute percentage error and mean absolute error obtained from the XGBoost model were equal to 0.000346,0.952,7.02,and 0.00028,respectively.In addition,the proposed method analyzed the contribution rates of the rolling affecting parameters on the roll gap.The data showed that in the controllable rolling parameters,the rolling speed is the most impacting factor that disturbs the roll gap model in the acceleration and deceleration process,which can provide a useful direction for actual roll gap adjustment. 展开更多
