METHODS:Seventy-two Sprague-Dawley rats were randomly divided into a saline control group(group S),an SAP group (group P),and a Bindarit group(group T).The SAP model was induced by retrograde infusion of 4%sodium taurocholate into the bilio-pancreatic duct.Based on the SAP model,Bindarit was injected intraperitoneally in group T,and 0.5% methyl cellulose was injected intraperitoneally in groups S and P.In group S,saline was retrogradely infused into the bili- pancreatic duct.Serum amylase levels and the histological changes in the pancreas were assessed at different time- points in each group.Expression of MCP-1 in serum was measured by enzyme-linked immunoadsorbent assay(ELISA). MCP-1 protein and mRNA expression levels were detected by immunohistochemistry,Western blotting,and semi- quantitative reverse transcriptase-polymerase chain reaction (RT-PCR). RESULTS:Serum amylase levels in groups P and T were higher than those in group S.Serum amylase levels were significantly lower in group T than in group P at 6 and 12 hours after operation.The levels of MCP-1 in serum at 6 and 12 hours after operation in group P were significantly higher than in group S,and significantly lower in group T than in group P at 6 and 12 hours after operation.The pathological damage in the pancreas was milder in group T than in group P.MCP-1 protein and mRNA expression levels in the pancreas were higher in groups P and T than in group S.These expression levels were positively correlated with the pathological damage of pancreatic tissues.The activity of MCP-1 in group T was significantly lower than in group P.CONCLUSION:MCP-1 may play important roles in the pathogenesis of SAP.The data suggest that Bindarit ameliorates SAP by inhibiting the activity of MCP-1 in vivo.展开更多

METHODS: The data about 75 OGIB patients who underwent DBE in January 2007-June 2009 in our hospi...AIM: To evaluate the diagnostic value of double-balloon enteroscopy (DBE) for obscure gastrointestinal bleeding (OGIB). METHODS: The data about 75 OGIB patients who underwent DBE in January 2007-June 2009 in our hospital were retrospectively analyzed. RESULTS: DBE was successfully performed in all 75 patients without complication. Of the 75 patients, 44 (58.7%) had positive DBE findings, 22 had negative DBE findings but had potential bleeding at surgery and capsule endoscopy, etc . These 66 patients were finally diagnosed as OGIB which was most commonly caused by small bowel tumor (28.0%), angiodysplasia (18.7%) and Crohn’s disease (10.7%). Lesions occurred more frequently in proximal small bowel than in distal small bowel (49.3% vs 33.3%, P = 0.047). CONCLUSION: DBE is a safe, effective and accurate procedure for the diagnosis of OGIB.展开更多

There are currently four known TIMP family members- TIMP-1,2,3,4.We determined whether increasing levels of TIMP-3 expression could suppress the malignant phenotype of human hepatocarcinoma cell line HCC-7721. METHODS:A recombinant expression vector,which contained full-length cDNA of human TIMP-3, was constructed and transfected into the human hepatocarcinoma cell line HCC-7721 by the lipofectamine technique.In vitro and in vivo tests such as Western blotting,immunohistochemistry as well as xenografting in nude mice were used to analyze expression levels of TIMP and MMP,and changes in malignant phenotype after the gene transfection. RESULTS:TIMP-3 expression in TIMP-3 gene-transfected HCC-7721 cells was upregulated as assessed by Western blotting.The ability of in vitro invasion through a Boyden chamber was significantly decreased compared to controls.Following subcutaneous injection into nude mice,the TIMP-3 transfected cells suppressed primary tumor growth,as characterized by reduced tumor weight, size and microvasculature as well as maintaining the extracellular matrix. CONCLUSION:The results suggest that upregulation of TIMP-3 expression in HCC-7721 cells inhibits invasion capacity in vitro as well as tumorigenic and metastatic potential in nude mice.展开更多
A 83-year-old woman was admitted to our hospital because of intermittent abdominal colicky pain and vomiting for 26 h.The pain localized over the periumbilical area with radiation along the medial side of the thigh.Co...A 83-year-old woman was admitted to our hospital because of intermittent abdominal colicky pain and vomiting for 26 h.The pain localized over the periumbilical area with radiation along the medial side of the thigh.Computed tomography scan with three-dimensional reconstruction revealed a loop of small bowel protruding into the left obturator canal. Incarcerated obturator hernia was diagnosed and emergency laparotomy was arranged immediately. Unfortunately,her family refused surgery because of her worsening condition.On the third evening after admission,the patient developed peritonitis and sepsis. Perforation of small bowel due to the incarceration was noted during laparotomy.Bowel resection and an endileostomy were performed.She recovered well despite of the complication of multiple organ dysfunction syndrome.Literature is reviewed,and the pathogenesis,clinical manifestation,imaging features and treatment are discussed.展开更多
AIM:To investigate the relation between gastric cancer and microsatellite instability(MSI),loss of heterozygosity(LOH)and promoter region methylation.METHODS:Fifty primary gastric carcinoma specimens were collected fr...AIM:To investigate the relation between gastric cancer and microsatellite instability(MSI),loss of heterozygosity(LOH)and promoter region methylation.METHODS:Fifty primary gastric carcinoma specimens were collected from patients with no family history of cancer.In addition,normal tissues were also collected from patients as controls.DNA was extracted by polymerase chain reaction for single-strand conformation polymorphism,bisulfite DNA sequencing,and methylation-specific band analysis.RESULTS:The positive rate for MSI and LOH in gastric carcinoma was 16%and 20%,respectively.According to the tumor,node and metastasis staging system,the LOH frequency was higher in gastric carcinoma at stages Ⅲ and Ⅳ than in gastric carcinoma at stagesⅠ and Ⅱ(P=0.01),which was also significantly correlated with lymph node metastasis and clinicopathological characteristics of gastric carcinoma.Methylation of bone morphogenetic protein 3(BMP3)gene promoter was detected in 64.44% of gastric carcinoma tissue samples.However,no statistical significance was observed between promoter region methylation and carcinoma differentiation.Interestingly,the BMP3 gene methylation rate was 71.05% and 28.58%,respectively,in MSI positive and negative cases(P=0.031),suggesting that BMP3 genetic instability and promoter methylation are initiated during gastric carcinogenesis.LOH was detected mostly in the late stages of gastric carcinoma,indicating that gastric carcinoma at late stages has a higher infiltration and a poorer prognosis.CONCLUSION:Promotor region methylation of the BMP3 gene may cause gastric carcinoma in Chinese people.展开更多

METHODS:Exfoliated cells obtained by pancreatic duct brushing during ERCP from 21 patients(18 with pancreatic cancer,3 with chronic pancreatitis) were examined.Telomerase activity was detected by polymerase chain reaction and telomeric repeat amplification protocol assay(PCR-TRAP-ELISA). RESULTS:D450 values of telomerase activity were 0.446± 0.2700 in pancreatic cancer and 0.041±0.0111 in chronic pancreatitis.77.8%(14/18)of patients with pancreatic cancer had cells with telomerase activity.None of the samples from patients with chronic pancreatitis showed telomerase activity,when the cutoff value of telomerase activity was set at 2.0.Cytological examination showed cancer cells in 66.7%(12/18)of the patients. CONCLUSIONS:Telomerase activity may be an early malignant event in pancreatic cancer development Cytology and telomerase activity in cells obtained bypancreatic duct brushing may complement each other for the diagnosis of pancreatic cancer.展开更多
