PLATZ is a novel zinc finger DNA-binding protein that plays an important role in regulating plant growth and development and resisting abiotic stress.However,there has been very little research on the function of this family gene in tomatoes,which limits its application in germplasm resource improvement.Therefore,the PLATZ gene family was identified and analyzed in tomato,and its roles were predicted and verified to provide a basis for in-depth research on SlPLATZ gene function.In this study,the PLATZ family members of tomato were identified in the whole genome,and 19 SlPLATZ genes were obtained.Functional prediction was conducted based on gene and promoter structure analysis and RNA-seq-based expression pattern analysis.SlPLATZ genes that responded significantly under different abiotic stresses or were significantly differentially expressed among multiple tissues were screened as functional gene resources.SlPLATZ17 was selected for functional verification by experiment-based analysis.The results showed that the downregulation of SlPLATZ17 gene expression reduced the drought and salt tolerance of tomato plants.Tomato plants overexpressing SlPLATZ17 had larger flower sizes and long,thin petals,adjacent petals were not connected at the base,and the stamen circumference was smaller.This study contributes to understanding the functions of the SlPLATZ family in tomato and provides a reference for functional gene screening. 展开更多

Effect of perioperative chemotherapy on resection of isolated pulmonary metastases from colorectal cancer:A single center experience

BACKGROUND Numerous studies have assessed surgical resection as a standard treatment option for patients with colorectal cancer(CRC)and resectable pulmonary metastases(PM).However,the role of perioperative chemotherapy after complete resection of isolated PM from patients with CRC patients remains controversial.We hypothesize that perioperative chemotherapy does not provide significant survival benefits for patients undergoing resection of PM from CRC.AIM To determine whether perioperative chemotherapy affects survival after radical resection of isolated PM from CRC.METHODS We retrospectively collected demographic,clinical,and pathologic data on patients who underwent radical surgery for isolated PM from CRC.Cancerspecific survival(CSS)and disease-free survival were calculated using Kaplan-Meier analysis.Inter-group differences were compared using the log-rank test.For multivariate analysis,Cox regression was utilized when indicated.RESULTS This study included 120 patients with a median age of 61.6 years.The 5-year CSS rate was 78.2%,with 36.7% experiencing recurrence.Surgical resection for isolated PM resulted in a 5-year CSS rate of 50.0% for second metastases.Perioperative chemotherapy(P=0.079)did not enhance survival post-resection.Factors associated with improved survival included fewer metastatic lesions[hazard ratio(HR):2.51,P=0.045],longer disease-free intervals(HR:0.35,P=0.016),and wedge lung resections(HR:0.42,P=0.035).Multiple PM predicted higher recurrence risk(HR:2.22,P=0.022).The log-rank test showed no significant difference in CSS between single and repeated metastasectomy(P=0.92).CONCLUSION Perioperative chemotherapy shows no survival benefit post-PM resection in CRC.Disease-free intervals and fewer metastatic lesions predict better survival.Repeated metastasectomy is warranted for eligible patients. 展开更多

BACKGROUND The benefit of adjuvant chemotherapy(ACT)for patients with no evidence of disease after pulmonary metastasis resection(PM)from colorectal cancer(CRC)remains controversial.AIM To assess the efficacy of ACT in patients after PM resection for CRC.METHODS This study included 96 patients who underwent pulmonary metastasectomy for CRC at a single institution between April 2008 and July 2023.The primary end-point was overall survival(OS);secondary endpoints included cancer-specific survival(CSS)and disease-free survival(DFS).An inverse probability of treat-ment-weighting(IPTW)analysis was conducted to address indication bias.Sur-vival outcomes compared using Kaplan-Meier curves,log-rank test,Cox regre-ssion and confirmed by propensity score-matching(PSM).RESULTS With a median follow-up of 27.5 months(range,18.3-50.4 months),the 5-year OS,CSS and DFS were 72.0%,74.4%and 51.3%,respectively.ACT had no significant effect on OS after PM resection from CRC[original cohort:P=0.08;IPTW:P=0.15].No differences were observed for CSS(P=0.12)and DFS(P=0.68)between the ACT and non-ACT groups.Multivariate analysis showed no association of ACT with better survival,while sublobar resection(HR=0.45;95%CI:0.20-1.00,P=0.049)and longer disease-free interval(HR=0.45;95%CI:0.20-0.98,P=0.044)were associated with improved survival.CONCLUSION ACT does not improve survival after PM resection for CRC.Further well-designed randomized controlled trials are needed to determine the optimal ACT regimen and duration. 展开更多

在现有对辣椒炒制和糊辣椒舂捣的研究中,存在着辣椒炒制温度及舂捣效果不佳等问题。为了解决上述问题,对炒制和舂捣集成一体的辣椒加工装置进行结构设计及仿真分析,得到了满足炒制温度和捣碎均匀的糊辣椒加工装置。本文利用电磁加热原... 在现有对辣椒炒制和糊辣椒舂捣的研究中,存在着辣椒炒制温度及舂捣效果不佳等问题。为了解决上述问题,对炒制和舂捣集成一体的辣椒加工装置进行结构设计及仿真分析,得到了满足炒制温度和捣碎均匀的糊辣椒加工装置。本文利用电磁加热原理对干辣椒进行翻炒加热,通过内部设置温度探头实时监测温度变化,滚筒翻炒辣椒装置内部设计有4叶绞片一体化炒制干辣椒机构,设计可360°旋转滚筒翻炒辣椒装置,可保证内部辣椒均匀受热以达到表面呈现焦糊状;利用凸轮机构原理,设计糊辣椒舂捣机构模型,实现糊辣椒的舂捣目的;利用ADAMS、ANSYS、ABAQUS等软件进行仿真验证分析。仿真结果显示:电磁线圈加热能满足炒制干辣椒的温度需求;凸轮传递到筛网上的冲撞力足以达到捣碎糊辣椒的效果需求。 展开更多

In this paper,an integrated substrate gap waveguide(ISGW)filtering antenna is proposed at millimeter wave band,whose surface wave and spurious modes are simultaneously suppressed.A secondorder filtering response is obtained through a coupling feeding scheme using one uniform impedance resonator(UIR)and two stepped-impedance resonators(SIRs).To increase the stopband width of the antenna,the spurious modes are suppressed by selecting the appropriate sizes of the ISGW unit cell.Furthermore,the ISGW is implemented to improve the radiation performance of the antenna by alleviating the propagation of surface wave.And an equivalent circuit is investigated to reveal the working principle of ISGW.To demonstrate this methodology,an ISGW filtering antenna operating at a center frequency of 25 GHz is designed,fabricated,and measured.The results show that the antenna achieves a stopband width of 1.6f0(center frequency),an out-of-band suppression level of 21 dB,and a peak realized gain of 8.5 dBi. 展开更多

Osteoporosis(OP),a systemic and chronic bone disease,is distinguished by low bone mass and destruction of bone microarchitecture.Ginsenoside Compound-K(CK),one of the metabolites of ginsenoside Rb1,has anti-aging,anti-inflammatory,anti-cancer,and hypolipidemic activities.We have demonstrated CK could promote osteogenesis and fracture healing in our previous study.However,the contribution of CK to osteoporosis has not been examined.In the present study,we investigated the effect of CK on osteoclastogenesis and ovariectomy(OVX)-induced osteoporosis.The results showed that CK inhibited receptor activator for nuclear factor-κB ligand(RANKL)-mediated osteoclast differentiation and reactive oxygen species(ROS)activity by inhibiting the phosphorylation of NF-κB p65 and oxidative stress in RAW264.7 cells.In addition,we also demonstrated that CK could inhibit bone resorption using bone marrow-derived macrophages.Furthermore,we demonstrated that CK attenuated bone loss by suppressing the activity of osteoclast and alleviating oxidative stress in vivo.Taken together,these results showed CK could inhibit osteoclastogenesis and prevent OVX-induced bone loss by inhibiting NF-κB signaling pathway. 展开更多

在线化学分析需要实现开放环境下的样品取样和电离/激发。相比于激光切削或者激光诱导击穿,大气压微等离子体系统结构简单,更利于小型化。因而基于大气压微等离子体的在线化学分析技术引起行业的广泛关注。为了确定合适的微等离子体源... 在线化学分析需要实现开放环境下的样品取样和电离/激发。相比于激光切削或者激光诱导击穿,大气压微等离子体系统结构简单,更利于小型化。因而基于大气压微等离子体的在线化学分析技术引起行业的广泛关注。为了确定合适的微等离子体源进行样品的在线元素检测,需要进一步了解各放电模式及工作参数下微等离子体的自身特性以及取样效果。该工作主要研究了电弧及辉光放电微等离子体在大气压下对样品铁取样发射光谱的特性。实现了在开放环境下对高熔点金属样品的在线检测,并发现电弧放电微等离子体与光谱分析源联用具有更高的取样效率。高采样效率的电弧放电微等离子体源为实现金属及难解离样品的检测提供了一种新的方法。同时,相较于传统的取样装置,避免了复杂的样品制备、样品传输过程。实验装置采取简单的针对板放电结构,分别利用高压脉冲电源、直流电源获得电弧放电和辉光放电。实验的结果表明,在放电功率近似相等的条件下,电弧放电产生的微等离子体对样品铁取样的光学发射谱中,样品元素的特征谱线占据主导地位,同时伴随有空气中氮气的谱线,而且铁离子(FeⅡ)谱线的相对强度显著高于氮气分子谱线的相对强度。而在直流辉光放电中,样品铁原子(FeⅠ)谱线相对强度非常不明显。由此说明,电弧放电产生的微等离子体具有更高的采样效率。放电在样品表面留下的溅射坑也得出了相同的结论。增加辉光放电电流到25mA,发现样品元素铁的谱线仍然没有明显的增强。同时,也研究了采样间距对两种采样模式的影响。实验结果表明,间距对两种模式的采样光谱没有显著的影响。采用主要成分为铝的合金铝箔进行了上述对比实验,得出相同的结论,即电弧放电微等离子体更适合作为光谱分析源来实现对金属样品的实时快速检测。 展开更多

Due to the requirements for mobile robots to search or rescue in unknown environments,reactive navigation which plays an essential role in these applications has attracted increasing interest.However,most existing reactive methods are vulnerable to local minima in the absence of prior knowledge about the environment.This paper aims to address the local minimum problem by employing the proposed boundary gap(BG)based reactive navigation method.Specifically,the narrowest gap extraction algorithm(NGEA)is proposed to eliminate the improper gaps.Meanwhile,we present a new concept called boundary gap which enables the robot to follow the obstacle boundary and then get rid of local minima.Moreover,in order to enhance the smoothness of generated trajectories,we take the robot dynamics into consideration by using the modified dynamic window approach(DWA).Simulation and experimental results show the superiority of our method in avoiding local minima and improving the smoothness. 展开更多

Detection of circulating tumor-specific DNA, RNA or proteins can be difficult due to relative scarcity. Exosomes are extracellular vesicles, 30 - 150 nm in diameter derived from fusion of multivesicular bodies with the plasma membrane. They are composed of a lipid bilayer membrane and contain proteins, mRNA and miRNA. Exosomes are secreted by multiple cell types, including cancer cells. However, there is a relative lack of information concerning the contents of exosomes secreted by various tumor cell types. To examine exosomes in cancer, we collected blood plasma samples from patients with breast, ovarian, prostate, hepatic, gastric, colon, and pancreatic cancers. Exosomes were isolated from plasma and confirmed by AchE assay, transmission electron microscopy and expression of the CD63 exosomal marker. Expression of AFP, CA724, CA153, CEA, CA125, CA199 and PSA antigens were determined using an automated electro-chemiluminescence assay. Expression of the tumor-related chaperone protein, mortalin, was determined by Western blot analysis. Levels of exosome secretion were variable among the different tumor types. Both exosome levels and mortalin expression within tumor cell exosomes were higher than in healthy donors, except in pancreatic carcinoma, where exosomes were elevated but mortalin expression was not significantly different from healthy donors. Exosomes provide unique opportunities for the enrichment of tumor-specific materials and may be useful as biomarkers and possibly as tools of cancer therapies. Mortalin, which has been linked to cell proliferation and induction of epithelial-mesenchymal transition of cancer cells, may be useful as a prognostic biomarker and as a possible therapeutic target. 展开更多

Objective:To explore the intervention effects and mechanism of Simiao Yongan(SMYA)Decoction on carotid atherosclerotic plaque in ApoE knockout mice by antagonizing lipid metabolism of oxidation low density lipoprotein(Ox-LDL).Methods:ApoE-/-mice were fed high-fat feeding for 2 weeks,combined with perivascular collar placement(PCCP)and high fat feeding for 8 weeks to establish carotid AS plaque model.All the mouses were divided into 5 groups,after successful AS model preparation.The sham operation as well as model group intragastrically administered with deionized water.The rest mouses were intragastrically administered with SMYA decoction or Pioglitazone,or treated with Atorvastatin for 8 weeks.Then adopting automatic biochemical analyzer to test total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)content.The content of ox-LDL in serum,which was tested by enzyme-linked immunosorbent assay(ELISA).The pathological changes of carotid artery were observed by hematoxylin-eosin(HE)staining and the vascular parameters were measured.Immunohistochemical method and Western blot assay was used to detect the manifestations of protein include fatty acid binding protein 4(FABP4)and peroxisome proliferator-activated receptorγ(PPARγ)as well as matrix metalloproteinase 2(MMP 2).Results:Compared with the sham operation group,the serum ox-LDL content of the model group was increased,and the intima thickness(IT),intima-media thickness(IT/MT),plaque area(PA)as well as lumen area ratio of plaque vessels(PA/LA)were enlarged(P<0.05).Immunohistochemistry and Western blot results showed that the relative expression of FABP4 and MMP2 in the model group increased markedly,while the relative expression of PPARγsignificantly decreased.Compared SMYA decoction group and Atorvastatin group as well as Pioglitazone with model group,the serum ox-LDL content was decreased(P<0.05).In the mean time,the IT,PA,IT/MT and PA/LA in carotid artery were reduced(P<0.05).Immunohistochemistry and Western blot results showed that the expression of FABP4 and MMP2 was reduced,while PPARγprotein expression increased of mouses in each administration group.Conclusion:SMYA decoction is able to antagonize Ox-LDL,increase the expression of PPARγ,decrease the level of FABP4,reduce MMP2 in carotid arteries of ApoE-/-atherosclerotic mice,and increase the stability of plaque. 展开更多

The biological toxicity of its leachate was determined by luminescent bacteria method. The leaching toxicity of the leachate was determined by atomic fluorescence spectrometry. The results showed that the leaching with Aspergillus niger had good removal effect on As and Hg, and the leached fly ash and leachate could meet the safe discharge standard. 37 ℃ was the best temperature for A. niger leaching. The concentrations of As and Hg in fly ash leachate were reduced to 0.438 and 0.053 g/L, respectively after 5 d of leaching by A. niger at 37 ℃. The luminous intensity of leachate increased with the increase of the number of days. Therefore, the leaching with A. niger can effectively remove As and Hg in fly ash from municipal solid waste incineration, and the leachate can also meet the safe discharge standard. 展开更多

Academic English reading requires deeper cognitive involvement in addition to language proficiency and plays a vital role in moulding a critical mindset. However, cognitive progress is seldom targeted in classroom teaching. This study proposed a teaching method of three reading phases in terms of cognitive mechanism and communicative teaching approach.Ninety university freshmen of non-English major participated in the study, who were taking the course of Academic English Reading in the 2nd semester. The data were collected from self-report Research Skills Assessment twice and semi-structured interview, and analyzed with paired sample t test and Pearson correlation. The results showed that 1) “critical thinking”, “organizing ideas”, and “finding information” were believed to improve significantly at the end of the course; 2) reading skill was associated with writing skill and the sense of “big picture” in self perception; 3) the cognition-based interactive reading instruction was acknowledged to be conducive to academic reading. It complements Academic English reading pedagogy by juxtaposing the need for both individual cognition and group interaction in classroom teaching. 展开更多
