Despite the diverse roles of tripartite motif(Trim)-containing proteins in the regulation of autophagy,the innate immune response,and cell differentiation,their roles in skeletal diseases are largely unknown.We recent...Despite the diverse roles of tripartite motif(Trim)-containing proteins in the regulation of autophagy,the innate immune response,and cell differentiation,their roles in skeletal diseases are largely unknown.We recently demonstrated that Trim21 plays a crucial role in regulating osteoblast(OB)differentiation in osteosarcoma.However,how Trim21 contributes to skeletal degenerative disorders,including osteoporosis,remains unknown.First,human and mouse bone specimens were evaluated,and the results showed that Trim21 expression was significantly elevated in bone tissues obtained from osteoporosis patients.Next,we found that global knockout of the Trim21 gene(KO,Trim2^(1-/-))resulted in higher bone mass compared to that of the control littermates.We further demonstrated that loss of Trim21 promoted bone formation by enhancing the osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs)and elevating the activity of OBs;moreover,Trim21 depletion suppressed osteoclast(OC)formation of RAW264.7 cells.In addition,the differentiation of OCs from bone marrow-derived macrophages(BMMs)isolated from Trim21^(-/-)and Ctsk-cre;Trim21^(f/f)mice was largely compromised compared to that of the littermate control mice.Mechanistically,YAP1/β-catenin signaling was identified and demonstrated to be required for the Trim21-mediated osteogenic differentiation of BMSCs.More importantly,the loss of Trim21 prevented ovariectomy(OVX)-and lipopolysaccharide(LPS)-induced bone loss in vivo by orchestrating the coupling of OBs and OCs through YAP1 signaling.Our current study demonstrated that Trim21 is crucial for regulating OB-mediated bone formation and OC-mediated bone resorption,thereby providing a basis for exploring Trim21 as a novel dual-targeting approach for treating osteoporosis and pathological bone loss.展开更多
We will introduce the construction and design of a new simultaneous three-channel multicolor CCD photometer.This photometer has been mounted on the 1.2 m telescope at Jilin Astronomical Observatory,and is applied to s...We will introduce the construction and design of a new simultaneous three-channel multicolor CCD photometer.This photometer has been mounted on the 1.2 m telescope at Jilin Astronomical Observatory,and is applied to study space debris,gamma-ray burst afterglows,asteroids and other rapidly variable objects.As one of the ground follow-up telescopes of the Chinese-French Space-based multi-band astronomical Variable Objects Monitor(SVOM)mission,it appears very essential to evaluate the performance of the photometry system of this photometer,which can achieve simultaneous imaging within a field of view of 21’.5×21’.5,21’.5×21’.5 and21’.3×21’.3 in the Sloan Digital Sky Survey(SDSS)g’,r’,i’bands,respectively.Photometric calibrations were carried out by using plenty of SDSS standard stars,and the relationship between the photometric system and the Johnson-Bessel filter system was also studied.The results of the performance evaluation from observing open cluster M67 are presented.展开更多
Comprehensive characterization of metabolites and metabolic profiles in plasma has considerable significance in determining the efficacy and safety of traditional Chinese medicine(TCM)in vivo.However,this process is u...Comprehensive characterization of metabolites and metabolic profiles in plasma has considerable significance in determining the efficacy and safety of traditional Chinese medicine(TCM)in vivo.However,this process is usually hindered by the insufficient characteristic fragments of metabolites,ubiquitous matrix interference,and complicated screening and identification procedures for metabolites.In this study,an effective strategy was established to systematically characterize the metabolites,deduce the metabolic pathways,and describe the metabolic profiles of bufadienolides isolated from Venenum Bufonis in vivo.The strategy was divided into five steps.First,the blank and test plasma samples were injected into an ultra-high performance liquid chromatography/linear trap quadrupole-orbitrap-mass spectrometry(MS)system in the full scan mode continuously five times to screen for valid matrix compounds and metabolites.Second,an extension-mass defect filter model was established to obtain the targeted precursor ions of the list of bufadienolide metabolites,which reduced approximately 39%of the interfering ions.Third,an acquisition model was developed and used to trigger more tandem MS(MS/MS)fragments of precursor ions based on the targeted ion list.The acquisition mode enhanced the acquisition capability by approximately four times than that of the regular data-dependent acquisition mode.Fourth,the acquired data were imported into Compound Discoverer software for identification of metabolites with metabolic network prediction.The main in vivo metabolic pathways of bufadienolides were elucidated.A total of 147 metabolites were characterized,and the main biotransformation reactions of bufadienolides were hydroxylation,dihydroxylation,and isomerization.Finally,the main prototype bufadienolides in plasma at different time points were determined using LC-MS/MS,and the metabolic profiles were clearly identified.This strategy could be widely used to elucidate the metabolic profiles of TCM preparations or Chinese patent medicines in vivo and provide critical data for rational drug use.展开更多
Silicon/carbon composites are promising alternatives to current graphite anodes in commercial lithiumion batteries(LIBs)because of their high capacity and excellent safety.Nevertheless,the unsatisfactory fastcharging ...Silicon/carbon composites are promising alternatives to current graphite anodes in commercial lithiumion batteries(LIBs)because of their high capacity and excellent safety.Nevertheless,the unsatisfactory fastcharging capability and cycle stability of Si/C composites caused by slow charge transport capability and huge volume change under industrial electrode conditions severely hamper their development.Here,a novel Si/C anode was fabricated by homogeneously depositing amorphous C-Si nanolayers on graphite(C-Si@graphite).C-Si nanolayers with uniformly dispersed sub-nanometer Si particles in 3D carbon skeleton significantly boost electron and Li-ion transport and efficiently relieve Si's agglomeration and volume change.As a result,the tailored C-Si@graphite electrodes show an excellent rate capacity(760.3 mAh·g^(-1)at 5.0C)and long cycle life of over 1000 cycles at 1.0C and800 cycles at 2.0C under industrial electrode conditions.In addition,the assembled full cells(C-Si@graphite,anode;Li[Ni_(0.8)Co_(0.1)Mn_(0.1)]O_(2),cathode)present superior fastcharging capability(240.4 Wh·kg^(-1),charging for16.2 min,3.0C)and long cycle life(80.7%capacity retention after 500 cycles at 1.0C),demonstrating the massive potential of C-Si@graphite for practical application.展开更多

Kernel density estimation is a frequently used method, but to select an optimal bandwidth o...It is a common practice to evaluate probability density function or matter spatial density function from statistical samples. Kernel density estimation is a frequently used method, but to select an optimal bandwidth of kernel estimation, which is completely based on data samples, is a long-term issue that has not been well settled so far. There exist analytic formulae of optimal kernel bandwidth, but they cannot be applied directly to data samples,since they depend on the unknown underlying density functions from which the samples are drawn. In this work, we devise an approach to pick out the totally data-based optimal bandwidth. First, we derive correction formulae for the analytic formulae of optimal bandwidth to compute the roughness of the sample's density function. Then substitute the correction formulae into the analytic formulae for optimal bandwidth, and through iteration we obtain the sample's optimal bandwidth. Compared with analytic formulae, our approach gives very good results, with relative differences from the analytic formulae being only 2%~3% for sample size larger than 10~4. This approach can also be generalized easily to cases of variable kernel estimations.展开更多
Objective:The objective of this study was to characterize the chemical compounds of a Hanshi-Yufei formulation(HSYF;a modified formulation of a traditional Chinese medicine used for treating COVID-19)to elucidate the ...Objective:The objective of this study was to characterize the chemical compounds of a Hanshi-Yufei formulation(HSYF;a modified formulation of a traditional Chinese medicine used for treating COVID-19)to elucidate the mechanism of action and to evaluate potential anti-inflammatory effects of HSYF.Materials and Methods:The chemical constituents of HSYF extract were characterized using UPLC-Q-TOF/MS.Subsequently,a set of TCM network pharmacology methods was applied to identify disease-associated genes and to predict target profiles and pharmacological actions associated with the constituents of HSYF.Then,the antiviral effects of HSYF on H1N1 were assessed in RAW264.7 cells using MTT assays.Expression levels of pro-inflammatory cytokines interleukin(IL)-6 and tumor necrosis factor(TNF)-αfollowing infection of RAW264.7 cells with H1N1 were measured using an enzyme-linked immune sorbent assay(ELISA),and expression levels of inflammatory-related factors were detected using western blotting.Results:In total,165 chemical constituents(including glycosides,tannins,volatile oils,amino acids,triterpenoids,polyphenols,phenylpropanoids,sesquiterpenes,alkaloids,and flavonoids,among others)were tentatively identified in HSYF.Network pharmacology demonstrated that HSYF can regulate immunomodulatory-and anti-inflammatory-related targets of multiple pathways through its active ingredients,suggesting potential anti-COVID-19 effects.Furthermore,cell viability assays and ELISA showed that HSYF significantly inhibited H1N1 replication in RAW64.7 cells and markedly reduced expression of pro-inflammatory cytokines TNF-αand IL-6 at the proteins level.Conclusions:The results of the present study help improve our understanding of the therapeutic effects of HSYF in COVID-19 treatment from multi-level perspectives.展开更多
