Familial amyloidotic polyneuropathy(FAP)is a type of systemic amyloidosis characterized by peripheral and autonomic neuropathy.Although FAP is a typical autosomal dominant disorder caused by a point mutation in the TT...Familial amyloidotic polyneuropathy(FAP)is a type of systemic amyloidosis characterized by peripheral and autonomic neuropathy.Although FAP is a typical autosomal dominant disorder caused by a point mutation in the TTR gene,the average age at onset varies significantly among different countries.This discrepancy clearly suggests that a combination of intrinsic factors as well as extrinsic(environmental)factors shapes the development of FAP.However,these factors are difficult to analyze in humans,because detailed pathologic tissue analysis is only possible at autopsy.Thus,mouse models have been produced and used to disentangle these factors.This review covers the mouse models produced thus far and how these models are applied to analyze intrinsic and extrinsic factors involved in disease development and to test drug efficacy.展开更多

Cryptotanshinone(CTN) is one of t...The clinical manifestations of variant angina is unevenly distributed during the 24 h, thusthe in vivo performance of drugs should be tailored according to the angina circadianrhythm. Cryptotanshinone(CTN) is one of the representative bioactive lipid-soluble com-ponents of Danshen which has been commonly used for cardiovascular diseases such asangina pectoris. The aim of this study was to develop a novel CTN sustained-released pel-lets(CTN-SRPs) to precisely synchronize the CTN plasma concentrations with predictedoccurrence of angina pectoris for angina chronotherapy. A deconvolution-based methodwas applied to develop and optimize the CTN-SRPs. The plasma concentration-time curveof CTN immediate-released formulation after oral administration in rats was used as theweight function. The predicted plasma concentration-time curve of CTN-SRPs simulatedaccording to the incidence of variant angina during 24 h was used as the response func-tion. Then the desired drug release profile of CTN-SRPs was calculated based on deconvo-lution using weight function and response function, and subsequently used for guiding theformulation optimization. CTN-SRPs were prepared with the combinations of PVP, polox-amer 127 and EC as matrix using fluidized bed technology. An orthogonal design was em-ployed to obtain the optimal formulation with its release profile similar with the desiredone. Pharmacokinetic studies validated that the actual plasma concentration-time curve ofthese optimized CTN-SRPs was similar with the predicted one. In addition, the percent er-rors(%PE) of CTN plasma concentrations in 8–12 h were less than 10%. In conclusion, thisdeconvolution-based method could be applied to adjust the in vivo performance of drugs forangina chronotherapy.展开更多
This study attempted to compare the performance of local polynomial interpolation,inverse distance weighted interpolation,and ordinary kriging in studying distribution patterns of swimming crabs.Cross-validation was u...This study attempted to compare the performance of local polynomial interpolation,inverse distance weighted interpolation,and ordinary kriging in studying distribution patterns of swimming crabs.Cross-validation was used to select the optimum method to get distribution results,and kriging was used for making spatial variability analysis.Data were collected from 87 sampling stations in November of 2015(autumn)and February(winter),May(spring)and August(summer)of 2016.Results indicate that swimming crabs widely distributed in autumn and summer:in the summer,they were more spatially independent,and resources in each sampling station varied a lot;in the winter and spring,the abundance of crabs was much lower,but the individual crab size was bigger,and they showed the patchy and more concentrative distribution pattern,which means they were more spatially dependent.Distribution patterns were in accordance with ecological migration features of swimming crabs,which were affected by the changing marine environment.This study could infer that it is applicable to study crab fishery or even other crustacean species using geostatistical analysis.It not only helps practitioners have a better understanding of how swimming crabs migrate from season to season,but also assists researchers in carrying out a more comprehensive assessment of the fishery.Therefore,it may facilitate advancing the implementation in the pilot quota management program of swimming crabs in northern Zhejiang fishing grounds.展开更多
