BACKGROUND Visceral obesity is increasingly prevalent among adolescents and young adults and is commonly recognized as a risk factor for type 2 diabetes.Estrogen[17β-estradiol(E2)]is known to offer protection against...BACKGROUND Visceral obesity is increasingly prevalent among adolescents and young adults and is commonly recognized as a risk factor for type 2 diabetes.Estrogen[17β-estradiol(E2)]is known to offer protection against obesity via diverse me-chanisms,while its specific effects on visceral adipose tissue(VAT)remain to be fully elucidated.AIM To investigate the impact of E2 on the gene expression profile within VAT of a mouse model of prediabetes.METHODS Metabolic parameters were collected,encompassing body weight,weights of visceral and subcutaneous adipose tissues(VAT and SAT),random blood glucose levels,glucose tolerance,insulin tolerance,and overall body composition.The gene expression profiles of VAT were quantified utilizing the Whole Mouse Genome Oligo Microarray and subsequently analyzed through Agilent Feature Extraction software.Functional and pathway analyses were conducted employing Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses,respectively.RESULTS Feeding a high-fat diet(HFD)moderately increased the weights of both VAT and SAT,but this increase was mitigated by the protective effect of endogenous E2.Conversely,ovariectomy(OVX)led to a significant increase in VAT weight and the VAT/SAT weight ratio,and this increase was also reversed with E2 treatment.Notably,OVX diminished the expression of genes involved in lipid metabolism compared to HFD feeding alone,signaling a widespread reduction in lipid metabolic activity,which was completely counteracted by E2 adminis-tration.This study provides a comprehensive insight into E2's local and direct protective effects against visceral adiposity in VAT at the gene level.CONCLUSION In conclusion,the present study demonstrated that the HFD-induced over-nutritional challenge disrupted the gene expression profile of visceral fat,leading to a universally decreased lipid metabolic status in E2 deficient mice.E2 treatment effectively reversed this condition,shedding light on the mechanistic role and therapeutic potential of E2 in combating visceral obesity.展开更多

Despite the diverse roles of tripartite motif(Trim)-containing proteins in the regulation of autophagy,the innate immune response,and cell differentiation,their roles in skeletal diseases are largely unknown.We recent...Despite the diverse roles of tripartite motif(Trim)-containing proteins in the regulation of autophagy,the innate immune response,and cell differentiation,their roles in skeletal diseases are largely unknown.We recently demonstrated that Trim21 plays a crucial role in regulating osteoblast(OB)differentiation in osteosarcoma.However,how Trim21 contributes to skeletal degenerative disorders,including osteoporosis,remains unknown.First,human and mouse bone specimens were evaluated,and the results showed that Trim21 expression was significantly elevated in bone tissues obtained from osteoporosis patients.Next,we found that global knockout of the Trim21 gene(KO,Trim2^(1-/-))resulted in higher bone mass compared to that of the control littermates.We further demonstrated that loss of Trim21 promoted bone formation by enhancing the osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs)and elevating the activity of OBs;moreover,Trim21 depletion suppressed osteoclast(OC)formation of RAW264.7 cells.In addition,the differentiation of OCs from bone marrow-derived macrophages(BMMs)isolated from Trim21^(-/-)and Ctsk-cre;Trim21^(f/f)mice was largely compromised compared to that of the littermate control mice.Mechanistically,YAP1/β-catenin signaling was identified and demonstrated to be required for the Trim21-mediated osteogenic differentiation of BMSCs.More importantly,the loss of Trim21 prevented ovariectomy(OVX)-and lipopolysaccharide(LPS)-induced bone loss in vivo by orchestrating the coupling of OBs and OCs through YAP1 signaling.Our current study demonstrated that Trim21 is crucial for regulating OB-mediated bone formation and OC-mediated bone resorption,thereby providing a basis for exploring Trim21 as a novel dual-targeting approach for treating osteoporosis and pathological bone loss.展开更多

Here we report a 54 year old man with acute cavernous sinus syndrome. Brain magnetic resonance imaging identified a clival-based lesion with associated bony erosion. The patient underwent endoscopic endonasal biopsy and partial resection of the clival mass. Because histologic examination of the resected specimen resulted in a diagnosis of RCCC, contrast-enhanced computed tomography scan of the abdomen was performed and showed an enhanced left renal mass. The patient subsequently underwent laparoscopic left radical nephrectomy and gamma knife was planned for the residual clival lesion. We also retrospectively reviewed available published reports on clival metastases, specifically those from RCCC, since 1990.展开更多

However, the majority of studies to date have focused on a single acupoint at a time. In the present study, we observed activity changes in the brains of healthy volunteers before and after acupuncture at Taichong(LR3) and Taixi(KI3) using resting-state functional magnetic resonance imaging. Fifteen healthy volunteers underwent resting-state functional magnetic resonance imaging of the brain 15 minutes before acupuncture, then received acupuncture at Taichong and Taixi using the nail-pressing needle insertion method, after which the needle was retained in place for 30 minutes. Fifteen minutes after withdrawal of the needle, the volunteers underwent a further session of resting-state functional magnetic resonance imaging, which revealed that the amplitude of low-frequency fluctuation, a measure of spontaneous neuronal activity, increased mainly in the cerebral occipital lobe and middle occipital gyrus(Brodmann area 18/19), inferior occipital gyrus(Brodmann area 18) and cuneus(Brodmann area 18), but decreased mainly in the gyrus rectus of the frontal lobe(Brodmann area 11), inferior frontal gyrus(Brodmann area 44) and the center of the posterior lobe of the cerebellum. The present findings indicate that acupuncture at Taichong and Taixi specifically promote blood flow and activation in the brain areas related to vision, emotion and cognition, and inhibit brain areas related to emotion, attention, phonological and semantic processing, and memory.展开更多

The pressure curves of cartridge chamber are measured in experiment, and the tensile test data of cartridge are loaded into the numerical calculation to compare with the experimental data. The conclusion was obtained that the calculated results match better with the experimental results by considering strain rate bilinear kinematic hardening material constitutive model. The forces on the various parts of the cartridge during launching and their deformation rules are achieved, in which the equivalent plastic strain decreases and the cylinder ring withstands the maximum equivalent stress when the cartridge case clings to the bore from the mouth to the bottom.展开更多
As an important evolutionary innovation and unique organ,the rumen has played a crucial role in ruminant adaptation to complex ecological environments.However,the cellular basis of its complex morphology and function ...As an important evolutionary innovation and unique organ,the rumen has played a crucial role in ruminant adaptation to complex ecological environments.However,the cellular basis of its complex morphology and function remains largely unknown.In this study,we identified eight major cell types from seven representative prenatal and postnatal rumen samples using~56600 single-cell transcriptomes.We captured the dynamic changes and high heterogeneity in cellular and molecular profiles before,during,and after the appearance of keratinized stratified squamous epithelium with neatly arranged papillae and functional maturity.Basal cells,keratinocytes,differentiating keratinocytes,terminally differentiated keratinocytes,and special spinous cells provided the cellular basis for rumen epithelium formation.Notably,we obtained clear evidence of two keratinization processes involved in early papillogenesis and papillae keratinization and identified TBX3 as a potential marker gene.Importantly,enriched stratum spinosum cells played crucial roles in volatile fatty acid(VFA)metabolism and immune response.Our results provide a comprehensive transcriptional landscape of rumen development at single-cell resolution,as well as valuable insight into the interactions between dietary metabolism and the rumen.展开更多

Integration of thin films with soft substrate is an effective way to fabricate flexible electronic devices. Ga_2O_3 thin films deposited directly on soft substrates would be amorphous mostly. However, the thickness of the thin film obtained by mechanical exfoliation method is difficult to control and the edge of the film is fragile and easy to be damaged. In this work, we fabricated free-standing Ga_2O_3 thin films using the water-soluble perovskite Sr_3Al_2O_6 as a sacrificial buffer layer. The obtained Ga_2O_3 thin films were polycrystalline. The thickness and dimension of the films were controllable. A flexible Ga_2O_3solar-blind UV photodetector was fabricated by transferring the free-standing Ga_2O_3 film on a flexible polyethylene terephthalate substrate. The results displayed that the photoelectric performances of the flexible Ga_2O_3 photodetector were not sensitive to bending of the device. The free-standing Ga_2O_3 thin films synthesized through the method described here can be transferred to any substrates or integrated with other thin films to fabricate electronic devices.展开更多
Entanglement is the key resource in quantum information processing,and an entanglement witness(EW)is designed to detect whether a quantum system has any entanglement.However,prior knowledge of the target states should...Entanglement is the key resource in quantum information processing,and an entanglement witness(EW)is designed to detect whether a quantum system has any entanglement.However,prior knowledge of the target states should be known first to design a suitable EW,which weakens this method.Nevertheless,a recent theory shows that it is possible to design a universal entanglement witness(UEW)to detect negative-partial-transpose(NPT)entanglement in unknown bipartite states with measurement-device-independent(MDI)characteristic.The outcome of a UEW can also be upgraded to be an entanglement measure.In this study,we experimentally design and realize an MDI UEW for two-qubit entangled states.All of the tested states are well-detected without any prior knowledge.We also show that it is able to quantify entanglement by comparing it with concurrence estimated through state tomography.The relation between them is also revealed.The entire experimental framework ensures that the UEW is MDI.展开更多

At the same time, adult females secrete androgens, and females synthesizeandrogens that are mainly produced by the adrenal cortex. 文摘The original article by Yuichi et al explored whether the Japan Narrow-Band Imaging Expert Team classification and the pit pattern classification are suitable for diagnosing neoplastic lesions in patients with ulcerative colitis.In this letter,we offer some other perspectives.Risk factors for colorectal tumors include type 2 diabetes.Among genetic factors,the deletion or mutation of some genes,such as the p53 gene,can lead to colorectal tumors.There are significant gender differences in the occurrence and development of colorectal tumors.Some nongenetic factors,such as smoking,are also associated with the development of colorectal tumors.These all suggest that colorectal tumors are not only caused by ulcerative colitis,and we suggest further exploration and differentiation between colitis and colorectal tumors.

文摘Bohr’s principle of complementarity has a long history and it is an important topic in quantum theory,among which the famous example is the duality relation.The relation between visibilityC and distinguishability D,C2+D2≤1,has long been recognized as the only representative of the duality relation.However,recent researches have shown that this inequality is not good enough because it is not tight for multipath interferometers.Meanwhile,a tight bound for the multipath interferometer has been put forward.Here we design and experimentally implement a three-path interferometer coupling with path indicator states.The wave property of photons is characterized by l1-norm coherence measure,and the particle property is based on distinguishability of the indicator states.The new duality relation of the three-path interferometer is demonstrated in our experiment,which bounds the union of a right triangle and a part of elliptical area inside the quadrant of a unit circle.Data analysis confirms that the new bound is tight for photons in three-path interferometers.

文摘Acupuncture can induce changes in the brain. However, the majority of studies to date have focused on a single acupoint at a time. In the present study, we observed activity changes in the brains of healthy volunteers before and after acupuncture at Taichong(LR3) and Taixi(KI3) using resting-state functional magnetic resonance imaging. Fifteen healthy volunteers underwent resting-state functional magnetic resonance imaging of the brain 15 minutes before acupuncture, then received acupuncture at Taichong and Taixi using the nail-pressing needle insertion method, after which the needle was retained in place for 30 minutes. Fifteen minutes after withdrawal of the needle, the volunteers underwent a further session of resting-state functional magnetic resonance imaging, which revealed that the amplitude of low-frequency fluctuation, a measure of spontaneous neuronal activity, increased mainly in the cerebral occipital lobe and middle occipital gyrus(Brodmann area 18/19), inferior occipital gyrus(Brodmann area 18) and cuneus(Brodmann area 18), but decreased mainly in the gyrus rectus of the frontal lobe(Brodmann area 11), inferior frontal gyrus(Brodmann area 44) and the center of the posterior lobe of the cerebellum. The present findings indicate that acupuncture at Taichong and Taixi specifically promote blood flow and activation in the brain areas related to vision, emotion and cognition, and inhibit brain areas related to emotion, attention, phonological and semantic processing, and memory.
文摘In order to explore the rules of the deformation force during the launching of a small-diameter steel cartridge, the semi-closed bomb test method is used to test the greatest strains on chamber outer wall under the different chamber pressures. The pressure curves of cartridge chamber are measured in experiment, and the tensile test data of cartridge are loaded into the numerical calculation to compare with the experimental data. The conclusion was obtained that the calculated results match better with the experimental results by considering strain rate bilinear kinematic hardening material constitutive model. The forces on the various parts of the cartridge during launching and their deformation rules are achieved, in which the equivalent plastic strain decreases and the cylinder ring withstands the maximum equivalent stress when the cartridge case clings to the bore from the mouth to the bottom.

文摘As an important evolutionary innovation and unique organ,the rumen has played a crucial role in ruminant adaptation to complex ecological environments.However,the cellular basis of its complex morphology and function remains largely unknown.In this study,we identified eight major cell types from seven representative prenatal and postnatal rumen samples using~56600 single-cell transcriptomes.We captured the dynamic changes and high heterogeneity in cellular and molecular profiles before,during,and after the appearance of keratinized stratified squamous epithelium with neatly arranged papillae and functional maturity.Basal cells,keratinocytes,differentiating keratinocytes,terminally differentiated keratinocytes,and special spinous cells provided the cellular basis for rumen epithelium formation.Notably,we obtained clear evidence of two keratinization processes involved in early papillogenesis and papillae keratinization and identified TBX3 as a potential marker gene.Importantly,enriched stratum spinosum cells played crucial roles in volatile fatty acid(VFA)metabolism and immune response.Our results provide a comprehensive transcriptional landscape of rumen development at single-cell resolution,as well as valuable insight into the interactions between dietary metabolism and the rumen.

文摘Flexible electronic devices have attracted much attention due to their practical and commercial value. Integration of thin films with soft substrate is an effective way to fabricate flexible electronic devices. Ga_2O_3 thin films deposited directly on soft substrates would be amorphous mostly. However, the thickness of the thin film obtained by mechanical exfoliation method is difficult to control and the edge of the film is fragile and easy to be damaged. In this work, we fabricated free-standing Ga_2O_3 thin films using the water-soluble perovskite Sr_3Al_2O_6 as a sacrificial buffer layer. The obtained Ga_2O_3 thin films were polycrystalline. The thickness and dimension of the films were controllable. A flexible Ga_2O_3solar-blind UV photodetector was fabricated by transferring the free-standing Ga_2O_3 film on a flexible polyethylene terephthalate substrate. The results displayed that the photoelectric performances of the flexible Ga_2O_3 photodetector were not sensitive to bending of the device. The free-standing Ga_2O_3 thin films synthesized through the method described here can be transferred to any substrates or integrated with other thin films to fabricate electronic devices.

文摘Entanglement is the key resource in quantum information processing,and an entanglement witness(EW)is designed to detect whether a quantum system has any entanglement.However,prior knowledge of the target states should be known first to design a suitable EW,which weakens this method.Nevertheless,a recent theory shows that it is possible to design a universal entanglement witness(UEW)to detect negative-partial-transpose(NPT)entanglement in unknown bipartite states with measurement-device-independent(MDI)characteristic.The outcome of a UEW can also be upgraded to be an entanglement measure.In this study,we experimentally design and realize an MDI UEW for two-qubit entangled states.All of the tested states are well-detected without any prior knowledge.We also show that it is able to quantify entanglement by comparing it with concurrence estimated through state tomography.The relation between them is also revealed.The entire experimental framework ensures that the UEW is MDI.

文摘An experimental study was carried out to investigate the flame propagation and thermal hazard of the premixed N2O/fuel mixtures,including NH3,C3H8 and C2H4.The study provided the high speed video images and data about the flame locations,propagation patterns,overpressures and the quenching diameters during the course of combustion in different channels to elucidate the dynamics of various combustion processes.The onset decomposition temperature was determined using high-performance adiabatic calorimetry.It was shown that the order of the flame acceleration rate and thermal hazard was N2O/C2H4>N2O/C3H8>N2O/NH3.

文摘Dysbiosis of the gastric microbiome is involved in the development of gastric cancer(GC).A number of studies have demonstrated an increase in the relative abundance of Lactobacillus in GC.In this review,we present data that support the overgrowth of Lactobacillus in GC from studies on molecular and bacterial culture of the gastric microbiome,discuss the heterogenic effects of Lactobacillus on the health of human stomach,and explore the potential roles of the overgrowth of Lactobacillus in gastric carcinogenesis.Further studies are required to examine the association between Lactobacillus and GC at strain and species levels,which would facilitate to elucidate its role in the carcinogenic process.

文摘The effect of Fuzhuan brick tea(FBT)on metabolism in obese mice is mediated by regulation of N-methyltransferase by aryl hydrocarbon receptor.The expression of the phosphatidylethanolamine N-methyltransferase gene is regulated by many transcription factors,and those specific to this effect need further investigation.Experimental animal studies have been designed to observe the effects of a single drug or the sequential effects of drugs.A washout period should be included if different drugs(e.g.,antibiotics and FBT)are given to avoid or reduce additive effects or synergy.Currently,most experimental studies performed in mice used only male animals.However,experience has revealed that the results of using only male mice are very likely to have sex differences.

文摘The original study by Alessio et al reported that skinny people (SP) serum canpromote the formation of brown adipocytes, but not the differentiation of whiteadipocytes. This finding may explain why SP do not often become obese, despiteconsuming more calories than the body needs. More importantly, theydemonstrated that circulating factors in SP serum can promote the expression ofUCP-1 protein, thereby reducing fat accumulation. In this study, only male serumsamples were evaluated to avoid the interference of sex hormones in experiments,but adult males also synthesize estrogen, which is produced by the cells of thetestes. At the same time, adult females secrete androgens, and females synthesizeandrogens that are mainly produced by the adrenal cortex. We believe that theapproach of excluding sex hormone interference by sex selection alone may beflawed, so we comment on the article and debate the statistical analysis of thearticle.
文摘Intestinal dysbiosis and small intestinal bacterial overgrowth(SIBO)are common in patients with liver cirrhosis.Existing studies have not explored the association between gut dysbiosis and SIBO.We propose some suggestions for the authors’experimental methods and concepts,and we hope these suggestions can be adopted.The hydrogen breath test is worthy of recommendation due to its high accuracy and convenient operation.We suggest changing the substrate of the hydrogen breath test from lactulose to glucose to improve the accuracy of each parameter.SIBO is a small subset of gut dysbiosis,and we propose clarifying the concept of both.SIBO may be caused by liver cirrhosis or one of the pathogeneses of gastrointestinal diseases.Therefore,interference from other gastrointestinal diseases should be excluded from this study.
