Long non-coding RNA H19 regulates neurogenesis of induced neural stem cells in a mouse model of closed head injury 被引量:3

Stem cell-based therapies have been proposed as a potential treatment for neural regeneration following closed head injury.We previously reported that induced neural stem cells exert beneficial effects on neural regeneration via cell replacement.However,the neural regeneration efficiency of induced neural stem cells remains limited.In this study,we explored differentially expressed genes and long non-coding RNAs to clarify the mechanism underlying the neurogenesis of induced neural stem cells.We found that H19 was the most downregulated neurogenesis-associated lnc RNA in induced neural stem cells compared with induced pluripotent stem cells.Additionally,we demonstrated that H19 levels in induced neural stem cells were markedly lower than those in induced pluripotent stem cells and were substantially higher than those in induced neural stem cell-derived neurons.We predicted the target genes of H19 and discovered that H19 directly interacts with mi R-325-3p,which directly interacts with Ctbp2 in induced pluripotent stem cells and induced neural stem cells.Silencing H19 or Ctbp2 impaired induced neural stem cell proliferation,and mi R-325-3p suppression restored the effect of H19 inhibition but not the effect of Ctbp2 inhibition.Furthermore,H19 silencing substantially promoted the neural differentiation of induced neural stem cells and did not induce apoptosis of induced neural stem cells.Notably,silencing H19 in induced neural stem cell grafts markedly accelerated the neurological recovery of closed head injury mice.Our results reveal that H19 regulates the neurogenesis of induced neural stem cells.H19 inhibition may promote the neural differentiation of induced neural stem cells,which is closely associated with neurological recovery following closed head injury. 展开更多

Microglial activation that occurs rapidly after closed head injury may play important and complex roles in neuroinflammation-associated neuronal damage and repair.We previously reported that induced neural stem cells can modulate the behavior of activated microglia via CXCL12/CXCR4 signaling,influencing their activation such that they can promote neurological recovery.However,the mechanism of CXCR4 upregulation in induced neural stem cells remains unclear.In this study,we found that nuclear factor-κB activation induced by closed head injury mouse serum in microglia promoted CXCL12 and tumor necrosis factor-αexpression but suppressed insulin-like growth factor-1 expression.However,recombinant complement receptor 2-conjugated Crry(CR2-Crry)reduced the effects of closed head injury mouse serum-induced nuclear factor-κB activation in microglia and the levels of activated microglia,CXCL12,and tumor necrosis factor-α.Additionally,we observed that,in response to stimulation(including stimulation by CXCL12 secreted by activated microglia),CXCR4 and Crry levels can be upregulated in induced neural stem cells via the interplay among CXCL12/CXCR4,Crry,and Akt signaling to modulate microglial activation.In agreement with these in vitro experimental results,we found that Akt activation enhanced the immunoregulatory effects of induced neural stem cell grafts on microglial activation,leading to the promotion of neurological recovery via insulin-like growth factor-1 secretion and the neuroprotective effects of induced neural stem cell grafts through CXCR4 and Crry upregulation in the injured cortices of closed head injury mice.Notably,these beneficial effects of Akt activation in induced neural stem cells were positively correlated with the therapeutic effects of induced neural stem cells on neuronal injury,cerebral edema,and neurological disorders post–closed head injury.In conclusion,our findings reveal that Akt activation may enhance the immunoregulatory effects of induced neural stem cells on microglial activation via upregulation of CXCR4 and Crry,thereby promoting induced neural stem cell–mediated improvement of neuronal injury,cerebral edema,and neurological disorders following closed head injury. 展开更多

Effects of cement-enhanced soil on the ultimate lateral resistance of composite pile in clayey soil

The composite pile consisting of core-pile and surrounding cement-enhanced soil is a promising pile foundation in recent years.However,how and to what extent the cement-enhanced soil influences the ultimate lateral resistance has not been fully investigated.In this paper,the ultimate lateral resistance of the composite pile was studied by finite element limit analysis(FELA)and theoretical upper-bound analysis.The results of FELA and theoretical analysis revealed three failure modes of laterally loaded composite piles.The effects of the enhanced soil thickness,strength,and pile-enhanced soil interface characteristics on the ultimate lateral resistance were studied.The results show that increasing the enhanced soil thickness leads to a significant improvement on ultimate lateral resistance factor(N P),and there is a critical thickness beyond which the thickness no longer affects the N P.Increasing the enhanced soil strength induced 6.2%-232.6%increase of N P.However,no noticeable impact was detected when the enhanced soil strength was eight times higher than that of the natural soil.The maximum increment of N P is only 30.5%caused by the increase of interface adhesion factor(a).An empirical model was developed to calculate the N P of the composite pile,and the results show excellent agreement with the analytical results. 展开更多

The equilibrium optimizer(EO)represents a new,physics-inspired metaheuristic optimization approach that draws inspiration from the principles governing the control of volume-based mixing to achieve dynamic mass equilibrium.Despite its innovative foundation,the EO exhibits certain limitations,including imbalances between exploration and exploitation,the tendency to local optima,and the susceptibility to loss of population diversity.To alleviate these drawbacks,this paper introduces an improved EO that adopts three strategies:adaptive inertia weight,Cauchy mutation,and adaptive sine cosine mechanism,called SCEO.Firstly,a new update formula is conceived by incorporating an adaptive inertia weight to reach an appropriate balance between exploration and exploitation.Next,an adaptive sine cosine mechanism is embedded to boost the global exploratory capacity.Finally,the Cauchy mutation is utilized to prevent the loss of population diversity during searching.To validate the efficacy of the proposed SCEO,a comprehensive evaluation is conducted on 15 classical benchmark functions and the CEC2017 test suite.The outcomes are subsequently benchmarked against both the conventional EO,its variants,and other cutting-edge metaheuristic techniques.The comparisons reveal that the SCEO method provides significantly superior results against the standard EO and other competitors.In addition,the developed SCEO is implemented to deal with a mobile robot path planning(MRPP)task,and compared to some classical metaheuristic approaches.The analysis results demonstrate that the SCEO approach provides the best performance and is a prospective tool for MRPP. 展开更多

Human umbilical mesenchymal stem cells from Wharton's jelly of the umbilical cord were induced to differentiate into oligodendrocyte precursor-like cells in vitro. Oligodendrocyte precursor cells were transplanted into contused rat spinal cords. Immunofluorescence double staining indicated that transplanted cells survived in injured spinal cord, and differentiated into mature and immature oligodendrocyte precursor cells. Biotinylated dextran amine tracing results showed that cell transplantation promoted a higher density of the corticospinal tract in the central and caudal parts of the injured spinal cord. Luxol fast blue and toluidine blue staining showed that the volume of residual myelin was significantly increased at 1 and 2 mm rostral and caudal to the lesion epicenter after cell transplantation. Furthermore, immunofluorescence staining verified that the newly regenerated myelin sheath was derived from the central nervous system. Basso, Beattie and Bresnahan testing showed an evident behavioral recovery. These results suggest that human umbilical mesenchymal stem cell-derived oligodendrocyte precursor cells promote the regeneration of spinal axons and myelin sheaths. 展开更多

Background: To evaluate the short-term and long-term outcomes after laparoscopic hysterectomy (LH) compared with abdominal hysterectomy (AH) in case of benign gynecological disease. Methods: A multi-center cohort retrospective comparative study of population among 4,895 hysterectomies (3,539 LH vs.1,356 AH) between 2007 and 2013 was involved. Operative time (OT), estimated blood loss (EBL), intra-operative and post-operative complications, passing flatus; days with indwelling catheter, questionnaires covering pelvic floor functions and sexual functions were assessed. Results: The EBL (174.1±157.4 vs. 263.1±183.2 cc, LH and AH groups, respectively), passing flatus (38.7±14.1 vs. 48.1±13.2 hours), days with indwelling catheter (1.5±0.6 vs. 2.2±0.8 days), use of analgesics (6.5% vs. 73.1%), intra-operative complication rate (2.4% vs. 4.1%), post-operative complication rate (2.3 % vs. 5.7%), post-operative constipation (12.1% vs. 24.6%), mild and serious stress urinary incontinence (SUI) post-operative (P〈0.001; P=0.014), and proportion of Female Sexual Functioning Index (FSFI) total score 〈26.55 post-operative (P〈0.001) of the LH group were significantly less than those of AH group. There were no significant differences in OT (106.5±34.5 vs. 106.2±40.3 min) between the two groups. Conclusions: LH is a safe and efficient operation for improving patients' long-term quality of life (QoL), and LH is a cost-effectiveness procedure for treating benign gynecological disease. LH is superior to AH due to reduced EBL, reduced post-operative pain and earlier passing flatus. 展开更多

High-speed and precision positioning are fundamental requirements for high-acceleration low-load mechanisms in integrated circuit (IC) packaging equipment. In this paper, we derive the transient nonlinear dynamicresponse equations of high-acceleration mechanisms, which reveal that stiffness, frequency, damping, and driving frequency are the primary factors. Therefore, we propose a new structural optimization and velocity-planning method for the precision positioning of a high-acceleration mechanism based on optimal spatial and temporal distribution of inertial energy. For structural optimization, we first reviewed the commonly flexible multibody dynamic optimization using equivalent static loads method (ESLM), and then we selected the modified ESLM for optimal spatial distribution of inertial energy; hence, not only the stiffness but also the inertia and frequency of the real modal shapes are considered. For velocity planning, we developed a new velocity-planning method based on nonlinear dynamic-response optimization with varying motion conditions. Our method was verified on a high-acceleration die bonder. The amplitude of residual vibration could be decreased by more than 20% via structural optimization and the positioning time could be reduced by more than 40% via asymmetric variable velocity planning. This method provides an effective theoretical support for the precision positioning of high-acceleration low-load mechanisms. 展开更多

PEGylation has been widely used to improve the pharmacokinetic properties of prodrug self-assembled nanoparticles(prodrug-SANPs).However,the impacts of the amount of PEG on the self-assemble stability,cellular uptake,pharmacokinetics,and antitumor efficacy of prodrug-SANPs are still unknown.Herein,selenoether bond bridged docetaxel dimeric prodrug was synthesized as the model prodrug.Five prodrug-SANPs were designed by using different mass ratios of prodrugs to PEG(W_(prodrug)/W_(DSPE-mPEG2000)=10:0,9:1,8:2,7:3 and 6:4),and defined as Pure drug NPs,9:1NPs,8:2NPs,7:3 NPs and 6:4 NPs,respectively.Interestingly,8:2 NPs formed the most compact nanostructure,thus improving the self-assemble stability and pharmacokinetics behavior.In addition,the difference of these prodrug-SANPs in cellular uptake was investigated,and the influence of PEG on cytotoxicity and antitumor efficacy was also clarified in details.The 8:2 NPs exhibited much better antitumor efficacy than other prodrug-SANPs and even commercial product.Our findings demonstrated the pivotal role of the amount of PEG on prodrug-SANPs. 展开更多

The development of wireless communication technology has become increasingly important in the communications industry.How to allocate limited channel resources reasonably and reliably to each competing user is a problem that the access protocol of the MAC(Multiple Access Control)layer needs to solve.As an important way of random access,NP-CSMA(Non Persistent Carrier Sense Multiple Access)has a higher network throughput rate when the arrival rate is higher.This paper analyzes and improves the implemented NP-CSMA model,and obtains a three-slot NP-CSMA model.The mathematical tool MATLAB is used to analyze the network throughput,delay and energy efficiency of the model,and compare several random multiple access protocols adheres to the network throughput,they are NP-CSMA,Three-slot NP-CSMA,1-persistent CSMA,P-persistent CSMA,and concludes that the three-slot NP-CSMA is significantly letter than several other multiple access protocols.Finally,using Quartus to design the three-slot NP-CSMA circuit,the statistical value of the circuit system is compared with the theoretical value of the model,and the error is less than 0.01,which proves that the system is reasonable in design and can be applied to wireless communication networks. 展开更多

A giant cell tumor occurs mainly in the proximal tibia,humerus,distal radius bone and the pelvic bone.It is rarely observed in such sites as the ribs and the temporal bone.The condition is primarily treated with surgical excision and functional reconstruction.The effect of chemotherapy on lung metastases and locally advanced giant cell tumors has remained unknown.We collected and analyzed the data of six patients with rare giant cell tumors located in the head and neck patients.After an average follow-up of 42.6 months after surgery (14 to 90 months),no local recurrence or metastasis was observed.We also collected and analyzed the data of five patients with metastatic giant cell tumors who were undergoing surgery for the primary tumor before; of three patients who had experienced multiple chemotherapy cycles,one had spontaneous regression,and one survived for long timer despite progression.The other two patients had their major metastatic lesions resected by surgery,and presented long-term survival during the follow up.In addition,this study reports one patient with locally advanced giant cell tumor of the rib,who has undergone successful surgical resection following two cycles of chemotherapy with ifosfamide and liposomal doxorubicin.Complete resection of the lesion at the head and neck is the key to relapse-free survival.The prognosis of lung metastases in patients with giant cell tumors is relatively satisfying.Neoadjuvant chemotherapy is also conducive to the surgery for locally advanced lesions and improvement of the quality of life. 展开更多

Synthesis and Anti-Tumor Activities of Fluoride-Containing Gossypol Derivatives Compounds

We report herein the design and synthesis of a series of novel fluoride-containing gossypol derivatives by the condensation reaction of gossypol with fluoride-containing aromatic amine. These fluoride-containing gossypol derivatives were characterized by IR, 1H-NMR and high resolution mass spectral data, then screened as antitumor agents against three human cancer cell lines (HeLa, A-549 and BGC-823) and a normal cell line (VEC) in vitro by using MTT cell proliferation assays. Results revealed that compounds 3a, 3c and 3f exhibited superior anticancer activity against HeLa, compounds 3b,3c, 3e and 3f exhibited superior anticancer activity against A-549, compounds 3b, 3c and 3f exhibited superior anticancer activity against BGC-823 compared to gossypol. In particular, fluorine substituent at the para positions of the phenyl ring showed remarkable inhibitory effects on HeLa?(3c: IC50 = 14.2 μM, 3f:?IC50 = 8.34 μM), A-549(3c: IC50 = 6.32 μM, 3f: IC50 = 9.76 μM) and BGC-823 cells?(3c: IC50 = 8.62 μM, 3f: IC50 = 4.36 μM). Furthermore, all the compounds 3a-3f exhibited lessened cytotoxicity against VEC compared to gossypol. 展开更多

Synthesizing orientated liquid crystal elastomers(LCEs)via the two-stage thiol-acrylate Michael addition and photopolymerization(TAMAP)reaction is extensively used.However,excess acrylates,initiators,and strong stimuli are inevitably involved in the second stage crosslinking.Herein,we simplify the strategy through taking advantage of a volatile alkaline(originally added to catalyze the thiol-acrylate addition in the first crosslinking stage).Without excess functional groups,the residual catalyst after annealing is still enough to trigger reactions of dynamic covalent bonds at a relatively mild temperature(80℃)to program the alignment of LCEs.The reversible reaction switches off by itself after this process since the catalyst gradually but totally evaporates upon heating.The obtained soft actuators exhibit robust actuation during repeated deformation(over 1000 times).Many shape-morphing modes can be achieved by rationally designing orientation patterns.This strategy not only facilitates the practical synthesis of LCE actuators,but also balances the intrinsic conflict between stability and reprogrammability of exchangeable LCEs.Moreover,the method of applying volatile catalysts has the potential to be extended to other dynamic covalent bonds(DCBs)applied to crosslinked polymer systems. 展开更多

Alzheimer’s disease(AD)is a neurodegenerative disorder characterized by the predominant impairment of neurons in the hippocampus and the formation of amyloid plaques,hyperphosphorylated tau protein,and neurofibrillary tangles in the brain.The overexpression of amyloid-βprecursor protein(APP)in an AD brain results in the binding of APP intracellular domain(AICD)to Fe65 protein via the C-terminal Fe65-PTB2 interaction,which then triggers the secretion of amyloid-βand the consequent pathogenesis of AD.Apparently,targeting the interaction between APP and Fe65 can offer a promising therapeutic approach for AD.Recently,exosome,a type of extracellular vesicle with diameter around 30–200 nm,has gained much attention as a potential delivery tool for brain diseases,including AD,due to their ability to cross the blood–brain barrier,their efficient uptake by autologous cells,and their ability to be surface-modified with target-specific receptor ligands.Here,the engineering of hippocampus neuron cell-derived exosomes to overexpress Fe65,enabled the development of a novel exosome-based targeted drug delivery system,which carried Corynoxine-B(Cory-B,an autophagy inducer)to the APP overexpressed-neuron cells in the brain of AD mice.The Fe65-engineered HT22 hippocampus neuron cell-derived exosomes(Fe65-EXO)loaded with Cory-B(Fe65-EXO-Cory-B)hijacked the signaling and blocked the natural interaction between Fe65 and APP,enabling APP-targeted delivery of Cory-B.Notably,Fe65-EXO-Cory-B induced autophagy in APP-expressing neuronal cells,leading to amelioration of the cognitive decline and pathogenesis in AD mice,demonstrating the potential of Fe65-EXO-Cory-B as an effective therapeutic intervention for AD. 展开更多

In this paper, we propose a two-class priority- based polling system that uses gated and exhaustive services to achieve the priority-based scheme. This model is set up according to the method of the imbedded Markov chain theory and the generation function and explicitly analyzes key system performance characteristics including mean queue length, cyclic time, and throughput. CSF rhinorrhea may cause pneumocephalus and intracranial infection. Rupture of the aneurysm may cause fatal consequence. The authors report such a rare case to draw more attentions. A 55-year-old man presented with sexual dysfunction for 2 years. The serum prolactin was tested as 1,600 ng/ml(normal range, 1.39–24.2). Enhanced cranial MR showed an evident lesion at the sellar area, invading the right cavernous sinus. Prolactinoma was diagnosed. He took bromocriptine for one year and received gamma knife therapy thereafter. Four months after the treatment of gamma knife, he got CSF rhinorrhea and nasal bleeding. The endoscopic transnasalsphenoidal approach was performed to resect the tumor and repair the dura defect.The CSF rhinorrhea stopped after the surgery, however his nasal bleeding continued. The digital subtraction angiography(DSA) showed an aneurysm at the right cavernous internal carotid. The endovascular coil embolization was performed to treat the aneurysm. The patient recovered well. The coexistence of CSF rhinorrhea and pituitary adenoma is a high risk factor for the rupture of cavernous internal carotid aneurysm. When treating patients with pituitary adenoma and CSF rhinorrhea, doctors should exclude the aneurysm. When nasal bleeding occurs, the hemorrhage of internal carotid should be considered, and appropriate measures should be taken immediately. 展开更多

Objective: Primary intracranial malignant fibrous histiocytoma(MFH) is rare. We describe the detailed clinical features of 8 cases and fully review the literature to evaluate several prognostic factors. Methods: Eight patients with pathologically confirmed primary intracranial MFH were retrospectively reviewed. We searched Pub Med for relevant articles with the term "intracranial malignant fibrous histiocytoma". Results: Of the 8 patients, 4 were men and 4 were women. Three patients had received previous radiotherapy. The age of the patients ranged from 19 to 69 years, with a median age of 48 years. Most tumors could be totally resected; and only 1 tumor was subtotally resected. Six patients received postoperative radiotherapy and 3 patients received postoperative chemotherapy. Most patients died within the first year after surgery; and only 1 patient was still alive on the date of the last follow-up. We reviewed the literature and included a total of 46 patients in the Kaplan-Meier survival analysis. Young patients(less than 30 years old) seemed to have a better prognosis and survival rate than older patients(more than 30 years old)(log-rank test, P = 0.008).However, sex(P = 0.675), extent of resection(P = 0.934), postoperative radiotherapy(P = 0.592), and postoperative chemotherapy(P = 0.424) did not affect patient prognosis.Conclusions: The prognosis of MFH is usually poor, and most patients die within the first year after surgery. Younger MFH patients(less than 30 years old) seem to have a better prognosis and improved survival compared to older patients. 展开更多

When facial and cranial bones are involved in FD, it is termed craniofacial fibrous dysplasia(CFD). Although several reports have reported that CFD has a tendency for spontaneous cerebrospinal fluid(CSF) leakage, there have been no related English-language case reports. We present the first case of post-traumatic CSF rhinorrhea associated with CFD. Methods: A 30-year-old man presented with CSF rhinorrhea after a mild head trauma. Computed tomography cisternogram located a defect in the posterior wall of the right frontal sinus. Imaging examination also showed the evident expansion of multiple skull bones, spinal scoliosis, and multiple local enlargements of ribs.Without café-au-lait cutaneous spots and endocrine abnormalities, polyostotic FD was diagnosed instead of Mc Cune-Albright syndrome(MAS). The patient underwent craniotomy fistula repair surgery. The excised bone was contoured to be thinner to increase the cranial cavity. The patient recovered well and CSF leakage did not recur.But during a nineteen-month follow up, sight in the patient's left eye was decreased.MAS was suspected. Unfortunately the patient refused to take the proposed decompression surgery and laboratory tests of serum hormones. Conclusions: CFD, if the wall of the paranasal sinus is involved and the cranial cavity is decreased, may increase the risk of CSF rhinorrhea after head trauma.Expectant management is recommended in asymptomatic CFD patients even in the presence of optic nerve compression. As MAS may cause more problems, it should be precluded before polyostotic FD is diagnosed. 展开更多
