In this study, five genetic similarity coefficients were compared for analysis of phylogenetic relationship among 31 hot pepper inbred lines based on SRAP. The applicability of different genetic similarity coefficient was investigated by means of SRAP data of hot pepper inbred lines. According to the experimental results, the variation ranges of Nei & Li, Jaceard, Sorensen, Simple matching and Yule coefficients were 0. 598 - 0. 973, 0. 427 - 0. 947, 0. 598 - 0. 973, 0.427 - 0. 947 and 0. 133 - 0. 997, respectively. Results of cluster analysis based on different similarity coefficients varied greatly. To be specific, clustering results based on Nei & Li, Jaccard and Sorensen coefficients were consistent; clustering with Simple matching and Yule coef ficients led to consistent classification of category in different order and slightly different classification of subcategory. Comprehensively comparing the results of cluster analysis and the dendrograms of hot pepper inbred lines, Yule coefficient is suitable for SRAP analysis of hot pepper.展开更多

[ Method] Tissue culture seedlings of...Abstract [ Objective] This study was conducted to investigate the variation degree of plant traits of Dendrobium radiated by 60 Coγ ray to determine the optimum radiation dosage. [ Method] Tissue culture seedlings of Dendrobium sonia ' 166' treated by different doses of 60 Coγ, ray were analyzed with SRAP molecular marker technique, so as to investigate its mutagenic effect. [ Result3 Genomic DNA of Dendrobium treated with 60 Coγ ray at different doses was amplified with 12 pairs of primers screened out from 100 pairs primers, and 216 bands were obtained totally. Among them, there were 41 polymorphic bands, showing a polymorphie rate of 19.0%- Clustering analysis was performed to amplified bands of different treatments and the CK. It was found that populations of different radiation treatments mu- tated to different degrees, the 20 Gy treatment mutated to the highest degree, while the 5 Gy treatment mutated to the lowest degree. [ Conclusion] It was indicated that 20 Gy is the optimum dose for radiation mutagenesis of tissue culture seedlings of D. Sonia " 166'. This study provides fundamental basis and technical support for mutation breeding by radiation and selection of mutation progenies.展开更多
