Seasonal Prediction of Extreme High-Temperature Days in Southwestern China Based on the Physical Precursors 被引量:1

Extreme high temperatures frequently occur in southwestern China,significantly impacting the local ecological system and economic development.However,accurate prediction of extreme high-temperature days(EHDs)in this region is still an unresolved challenge.Based on the spatiotemporal characteristics of EHDs over China,a domain-averaged EHD index over southwestern China(SWC-EHDs)during April-May is defined.The simultaneous dynamic and thermodynamic fields associated with the increased SWC-EHDs are a local upper-level anticyclonic(high-pressure)anomaly and wavy geopotential height anomaly patterns over Eurasia.In tracing the origins of the lower boundary anomalies,two physically meaningful precursors are detected for SWC-EHDs.They are the tripolar SST change tendency from December-January to February-March in the northern Atlantic and the February-March mean snow depth in central Asia.Using these two selected predictors,a physics-based empirical model prediction was applied to the training period of 1961–2005 to obtain a skillful prediction of the EHDs index,attaining a correlation coefficient of 0.76 in the independent prediction period(2006–19),suggesting that 58%of the total SWC-EHDs variability is predictable.This study provides an estimate for the lower bound of the seasonal predictability of EHDs as well as for the hydrological drought over southwestern China. 展开更多

本文利用中国东部1078个测站1961~2003年逐日云量数据资料,揭示了中国东部冬季阴天频次主模态的时空分布特征,探讨了其形成的两条独立影响途径,并根据影响机理建立了季节预测模型。结果表明:(1)中国东部冬季阴天频次的经验正交函数分解... 本文利用中国东部1078个测站1961~2003年逐日云量数据资料,揭示了中国东部冬季阴天频次主模态的时空分布特征,探讨了其形成的两条独立影响途径,并根据影响机理建立了季节预测模型。结果表明:(1)中国东部冬季阴天频次的经验正交函数分解第一模态独立且显著,解释了其59%的总方差。该模态基本呈现空间一致型的分布,表现出显著的年际变率特征。当该模态为正位相时,北太平洋对流层低层存在显著的大尺度反气旋环流异常,其西侧异常偏南风能够将热带海洋的水汽输送至中国东部地区,导致该地区阴天频次增多。(2)前期8月和9月北太平洋副热带持续性海温异常的纬向偶极型分布(NPD)和副热带北大西洋海平面气压9至11月的短期变化(LPA)是该模态的两个主要驱动因子。当NPD中的西极为冷异常时,在局地低层气旋性异常环流的作用下冷海温异常向南平流,发展至热带西太平洋。而当热带西太平洋冷海温异常形成后,皮耶克里斯反馈作用能够发展和维持太平洋“西冷东暖”海表温度异常分布。“西冷东暖”的海表温度异常导致的热带纬向偶极型对流异常能够进一步激发北太平洋“北正南负”的偶极型高度场异常。北部反气旋异常西侧的偏南风有利于水汽输送至中国东部,从而导致阴天频次增多;LPA主要反映的是中高纬准定常罗斯贝波列由秋季至冬季的迅速转变。当副热带北大西洋海表面气压在前秋由正转负时,罗斯贝波列在东北亚上空冬季形成准正压的异常反气旋,其西侧偏南风异常同样能够导致中国东部阴天频次增多。(3)依据以上热带和热带外的两条独立影响机理途径,我们建立了具有物理意义的季节预测模型,利用2004~2013年间数据进行独立预测获得良好预测效果,可供业务预测部门参考和借鉴。 展开更多

The lightness and high strength-to-weight ratio of the magnesium alloy have attracted more interest in various applications.However,micro/nanostructure generation on their surfaces remains a challenge due to the flammability and ignition.Motivated by this,this study proposed a machining process,named the ultraprecision diamond surface texturing process,to machine the micro/nanostructures on magnesium alloy surfaces.Experimental results showed the various microstructures and sawtooth-shaped nanostructures were successfully generated on the AZ31B magnesium alloy surfaces,demonstrating the effectiveness of this proposed machining process.Furthermore,sawtooth-shaped nanostructures had the function of inducing the optical effect and generating different colors on workpiece surfaces.The colorful letter and colorful flower image were clearly viewed on magnesium alloy surfaces.The corresponding cutting force,chip morphology,and tool wear were systematically investigated to understand the machining mechanism of micro/nanostructures on magnesium alloy surfaces.The proposed machining process can further improve the performances of the magnesium alloy and extend its functions to other fields,such as optics. 展开更多

Northeast China(NEC)is China’s national grain production base,and the local precipitation is vital for agriculture during the springtime.Therefore,understanding the dynamic origins of the NEC spring rainfall(NECSR)variability is of socioeconomic importance.This study reveals an interdecadal change in the atmospheric teleconnections associated with the NECSR during a recent 60-year period(1961-2020).Before the mid-1980s,NECSR had been related to a Rossby wave train that is coupled with extratropical North Atlantic sea surface temperature(SST),whereas,since the mid-1980s,NECSR has been linked to a quite different Rossby wave train that is coupled with tropical North Atlantic SST.Both Rossby wave trains could lead to enhanced NECSR through anomalous cyclones over East Asia.The weakening of the westerly jet over North America is found to be mainly responsible for the alternation of the atmospheric teleconnections associated with NECSR during two epochs. 展开更多

【问题】在扶贫对象属地化的当下,公益组织参与扶贫需要与当地政府部门建立良好关系,但现实中公益组织与政府间存在不同的关注点和利益诉求点,公益组织如何入场不同扶贫区域?如何与当地政府部门建立合作关系涉及到公益组织参与扶贫目标... 【问题】在扶贫对象属地化的当下,公益组织参与扶贫需要与当地政府部门建立良好关系,但现实中公益组织与政府间存在不同的关注点和利益诉求点,公益组织如何入场不同扶贫区域?如何与当地政府部门建立合作关系涉及到公益组织参与扶贫目标的实现程度。【方法】以S基金会作为研究对象,采用访谈法、观察法收集相关材料并对材料进行分析。【发现】公益组织为了确保慈善项目在当地的顺利执行以一种互助式嵌入的行动策略与当地政府建立合作关系。这种行动策略的生成先后经历如下阶段:(1)寻找本地组织;(2)与本地组织建立公益共同体;(3)由公益共同体联合处理政府关系。各阶段的组织关系与行为表征存在一定的差异,其内在逻辑既与主体资源拥有量与供给力间的张力有关,也是我国现有的政社关系与扶贫战略共同催化的结果。【结论】扶贫需要转变以往传统的救助扶贫模式,突破和改造原有的规制约束,积极地引入社会力量参与扶贫。创造条件帮助公益组织与当地政府建立良好关系,可以形成官民相融的扶贫共同体。 展开更多

Record-breaking heavy and persistent precipitation occurred over the Yangtze River Valley(YRV)in June-July(JJ)2020.An observational data analysis has indicated that the strong and persistent rainfall arose from the confluence of southerly wind anomalies to the south associated with an extremely strong anomalous anticyclone over the western North Pacific(WNPAC)and northeasterly anomalies to the north associated with a high-pressure anomaly over Northeast Asia.A further observational and modeling study has shown that the extremely strong WNPAC was caused by both La Niña-like SST anomaly(SSTA)forcing in the equatorial Pacific and warm SSTA forcing in the tropical Indian Ocean(IO).Different from conventional central Pacific(CP)El Niños that decay slowly,a CP El Niño in early 2020 decayed quickly and became a La Niña by early summer.This quick transition had a critical impact on the WNPAC.Meanwhile,an unusually large area of SST warming occurred in the tropical IO because a moderate interannual SSTA over the IO associated with the CP El Niño was superposed by an interdecadal/long-term trend component.Numerical sensitivity experiments have demonstrated that both the heating anomaly in the IO and the heating anomaly in the tropical Pacific contributed to the formation and maintenance of the WNPAC.The persistent high-pressure anomaly in Northeast Asia was part of a stationary Rossby wave train in the midlatitudes,driven by combined heating anomalies over India,the tropical eastern Pacific,and the tropical Atlantic. 展开更多

An exceptionally prolonged heavy snow event(PHSE)occurred in southern China from 10 January to 3 February 2008,which caused considerable economic losses and many casualties.To what extent any dynamical model can predict such an extreme event is crucial for disaster prevention and mitigation.Here,we found the three S2S models(ECMWF,CMA1.0 and CMA2.0)can predict the distribution and intensity of precipitation and surface air temperature(SAT)associated with the PHSE at 10-day lead and 10−15-day lead,respectively.The success is attributed to the models’capability in forecasting the evolution of two important low-frequency systems in the tropics and mid-latitudes[the persistent Siberian High and the suppressed phase of the Madden−Julian Oscillation(MJO)],especially in the ECMWF model.However,beyond the 15-day lead,the three models show almost no skill in forecasting this PHSE.The bias in capturing the two critical circulation systems is responsible for the low skill in forecasting the 2008 PHSE beyond the 15-day lead.On one hand,the models cannot reproduce the persistence of the Siberian High,which results in the underestimation of negative SAT anomalies over southern China.On the other hand,the models cannot accurately capture the suppressed convection of the MJO,leading to weak anomalous southerly and moisture transport,and therefore the underestimation of precipitation over southern China.The Singular Value Decomposition(SVD)analyses between the critical circulation systems and SAT/precipitation over southern China shows a robust historical relation,indicating the fidelity of the predictability sources for both regular events and extreme events(e.g.,the 2008 PHSE). 展开更多

The microstructures on electrode level are crucial for battery performance, but the ambiguous understanding of both electrode microstructures and their structuring process causes critical challenges in controlling and evaluating the electrode quality during fabrication. In this review, analogous to the cell microenvironment well-known in biology, we introduce the concept of ‘‘active material microenvironment”(ME@AM)that is built by the ion/electron transport structures surrounding the AMs, for better understanding the significance of the electrode microstructures. Further, the scientific significance of electrode processing for electrode quality control is highlighted by its strong links to the structuring and quality control of ME@AM. Meanwhile, the roles of electrode rheology in both electrode structuring and structural characterizations involved in the entire electrode manufacturing process(i.e., slurry preparation, coating/printing/extrusion, drying and calendering) are specifically detailed. The advantages of electrode rheology testing on in-situ characterizations of the electrode qualities/structures are emphasized. This review provides a glimpse of the electrode rheology engaged in electrode manufacturing process and new insights into the understanding and effective regulation of electrode microstructures for future high-performance batteries. 展开更多

The demand on low-carbon emission fabrication technologies for energy storage materials is increasing dramatically with the global interest on carbon neutrality.As a promising active material for metal-sulfur batteries,sulfur is of great interest due to its high-energy-density and abundance.However,there is a lack of industry-friendly and low-carbon fabrication strategies for high-performance sulfur-based active particles,which,however,is in critical need by their practical success.Herein,based on a hail-inspired sulfur nano-storm(HSN)technology developed in our lab,we report an energy-saving,solvent-free strategy for producing core-shell sulfur/carbon electrode particles(CNT@AC-S)in minutes.The fabrication of the CNT@AC-S electrode particles only involves low-cost sulfur blocks,commercial carbon nanotubes(CNT)and activated carbon(AC)micro-particles with high specific surface area.Based on the above core-shell CNT@AC-S particles,sulfur cathode with a high sulfur-loading of 9.2 mg cm^(-2) delivers a stable area capacity of 6.6 mAh cm^(-2) over 100 cycles.Furthermore,even for sulfur cathode with a super-high sulfur content(72 wt%over the whole electrode),it still delivers a high area capacity of 9 mAh cm^(-2) over50 cycles in a quasi-lean electrolyte condition.In a nutshell,this study brings a green and industryfriendly fabrication strategy for cost-effective production of rationally designed S-rich electrode particles. 展开更多

The South Mid-Atlantic Ridge(SMAR)19°S segment,approximately located along the line of Saint Helena volcanic chain(created by Saint Helena mantle plume),is an ideal place to investigate the issue whether the ridgehotpot interaction process affected the whole MAR.In this study,we present major and trace elemental compositions and Sr-Nd-Pb isotopic ratios of twenty fresh lava samples concentrated in a relatively small area in the SMAR 19°S segment.Major oxides compositions show that all samples are tholeiite.Low contents of compatible trace elements(e.g.,Ni=239-594 ppm and Cr=456-1010 ppm)and low Fe/Mn(54-67)and Ce/Yb(0.65-1.5)ratios of these lavas show that their parental magmas are partially melted by a spinel lherzolite mantle source.Using software PRIMELT3,this study obtained mantle potential temperatures(Tp)beneath the segment of1321-1348℃,which is lower relative to those ridges influenced by mantle plumes.The asthenospheric mantle beneath the SMAR 19°S segment starts melting at a depth of^63 km and ceases melting at^43 km with a final melting temperature of^1265℃.The extent of partial melting is up to 16%-17.6%with an average adiabatic decompression value of 2.6%/kbar.The correlations of major oxides(CaO/Al2 O3)and trace elements(Cr,Co,V)with MgO and Zr show that the parental magma experienced olivine and plagioclase fractional crystallization during its ascent to the surface.87Sr/86Sr(0.702398-0.702996),143 Nd/144 Nd(0.513017-0.513177)and 206Pb/204Pb(18.444-19.477)ratios of these lavas indicate the mantle source beneath the SMAR 19°S segment is composed of a three-component mixture of depleted MORB mantle,PREMA mantle,and HIMU mantle materials.The simple,binary mixing results among components from plume-free SMAR MORB,Saint Helena plume and Tristan plume show that asthenospheric mantle beneath the SMAR 19°S segment may be polluted by both Saint Helena and Tristan plume enriched materials.The abovementioned mantle potential temperatures,together with the low Saint Helena(<10%)and Tristan(<5%)components remaining in the asthenospheric mantle at present,show that the physically ridge-hotspot interactions at SMAR 19°S segment may have ceased.However,the trace element and SrNd-Pb isotopically binary mixing calculation results imply that these lavas tapped some enriched pockets left when Saint Helena and/or Tristan plume were once on the SMAR during earlier Atlantic rifted history. 展开更多

【目的】蜜蜂球囊菌(Ascosphaera apis,简称球囊菌)是一种专性侵染蜜蜂幼虫的真菌病原,导致的白垩病是严重影响养蜂生产的顽疾,每年给养蜂业造成较大损失。本研究旨在基于已获得的第三代长读段测序数据对球囊菌菌丝(Aam)和孢子(Aas)中... 【目的】蜜蜂球囊菌(Ascosphaera apis,简称球囊菌)是一种专性侵染蜜蜂幼虫的真菌病原,导致的白垩病是严重影响养蜂生产的顽疾,每年给养蜂业造成较大损失。本研究旨在基于已获得的第三代长读段测序数据对球囊菌菌丝(Aam)和孢子(Aas)中基因的可变剪切(alternative splicing,AS)和可变多聚腺苷酸化(alternative polyadenylation,APA)进行深入分析。【方法】利用Astalavista软件鉴定Aam和Aas中基因的AS事件类型。利用IGV浏览器对部分剪切异构体(isoform)的结构进行可视化。通过TAPIS pipeline对Aam和Aas中基因的APA位点进行鉴定。通过MEME软件对Aam和Aas中全长转录本的APA位点上游50bp的序列特征进行分析并对motif进行鉴定。【结果】在Aam中共鉴定到286次AS事件,包括162次RI(Retained intron),87次A3(Alternative 3'splice-site)、32次A5(Alternative 5'splice-site)和5次SE (Skipping exon);在Aas中共鉴定到559次AS事件,包括305次RI、155次A3、85次A5、13次SE和1次MEE (Mutually exclusive exon)。进一步分析发现,现有参考基因组上的多数注释基因结构并不完整;部分注释基因在菌丝和孢子中转录形成的isoform在数量和结构方面均存在差异;部分isoform在参考基因组中没有对应的注释基因。Aam中共鉴定到2748个基因含有1个及以上的APA位点,其中含有1个APA位点的基因数量最多(726,26.42%);Aas中共鉴定到2768个基因含有1个及以上的APA位点,其中含有5个以上APA位点的基因数量最多(1180,42.63%)。部分基因在菌丝和孢子中含有不同的APA位点数。序列特征分析结果显示,球囊菌全长转录本的3'UTR的上下游表现出明显的碱基倾向性,U和A分别富集在3'UTR的上游和下游。此外,在球囊菌全长转录本的APA位点上游鉴定到4个motif,分别是UCUCCU、UCUUCU、CCCACC和CCCCCU。【结论】本研究通过对球囊菌菌丝和孢子中基因的AS和APA进行深入分析,揭示了球囊菌转录组的复杂性,为完善现有的基因组和转录组注释提供了宝贵信息,也为探究AS和APA在球囊菌的基因表达调控中的作用提供了关键基础。 展开更多

【目的】本研究旨在探究长链非编码RNA(long non-coding RNA,lncRNA)在中华蜜蜂(Apis cerana cerana,简称中蜂)6日龄幼虫应答蜜蜂球囊菌(Ascosphaera apis,简称球囊菌)侵染过程中的差异表达谱及调控作用。【方法】利用链特异性cDNA建库... 【目的】本研究旨在探究长链非编码RNA(long non-coding RNA,lncRNA)在中华蜜蜂(Apis cerana cerana,简称中蜂)6日龄幼虫应答蜜蜂球囊菌(Ascosphaera apis,简称球囊菌)侵染过程中的差异表达谱及调控作用。【方法】利用链特异性cDNA建库的RNA-seq技术对未被侵染及球囊菌侵染的中蜂6日龄幼虫肠道(AcCK和AcT)进行深度测序。通过相关生物信息学软件分析lncRNA的结构特征和表达谱。筛选并分析差异表达lncRNA(differentially expressed lncRNA,DElncRNA)的顺式(cis)作用及竞争性内源RNA(competing endogenous RNA,ceRNA)调控网络。采用RT-qPCR验证测序数据及DElncRNA差异变化趋势的可靠性。【结果】AcCK和AcT共预测出642个已知lncRNA和487个新lncRNA。与蛋白编码基因相比,上述中蜂lncRNA外显子数更少、长度更短且表达量更低。43个antisense lncRNA与40个正义链mRNA之间互补配对。AcCK和AcT比较组包含367个上调lncRNA和268个下调lncRNA。有194个DElncRNA潜在调控461个上下游基因,并涉及细胞进程、代谢进程和催化活性等38个功能条目以及氨基酸代谢、内吞作用和MAPK等191条通路。此外,有180个DElncRNA可靶向结合50个DEmiRNA,进而调控6365个mRNA;三者之间形成较为复杂的ceRNA调控网络。【结论】中蜂6日龄幼虫肠道的部分lncRNA可作为antisense lncRNA参与应答球囊菌侵染;部分DElncRNA可通过cis作用调节物质代谢和免疫途径相关的上下游基因,从而介导宿主的侵染应答;TCONS_00010661和TCONS_00003104等DElncRNA可通过ceRNA网络调控Jak-STAT和氧化磷酸化等通路及富集基因,进而参与宿主的侵染应答。 展开更多

【目的】本研究旨在明确蜜蜂球囊菌(Ascosphaera apis,简称球囊菌)菌丝和孢子中环状RNA(circular RNA,circRNA)的数量、种类和表达谱差异,并探讨共有circRNA、特有circRNA和差异表达circRNA(differentially expressed circRNA,DEcircRNA... 【目的】本研究旨在明确蜜蜂球囊菌(Ascosphaera apis,简称球囊菌)菌丝和孢子中环状RNA(circular RNA,circRNA)的数量、种类和表达谱差异,并探讨共有circRNA、特有circRNA和差异表达circRNA(differentially expressed circRNA,DEcircRNA)在菌丝与孢子中的潜在作用。【方法】基于前期获得的球囊菌菌丝(AaM)和孢子(AaS)的高质量RNA-seq数据,利用find_circ软件预测circRNA。通过Venn分析筛选AaM和AaS的共有circRNA和特有circRNA。根据P≤0.05且|log2 fold change|≥1的标准筛选AaM vs.AaS比较组的DEcircRNA。通过比对GO和KEGG数据库对circRNA的来源基因进行功能和通路注释。利用TargetFinder软件预测circRNA靶向结合的miRNA及miRNA靶向结合的mRNA。采用Cytoscape软件对竞争性内源RNA(competing endogenous RNA,ceRNA)调控网络进行可视化。通过RT-qPCR对DEcircRNA进行验证。【结果】AaM和AaS中分别含有13210156和19011000条短序列读段(anchors reads),其中分别有6124922和11392886条能够比对上球囊菌参考基因组。在AaM和AaS中分别鉴定到1868个和2225个circRNA,二者共有的circRNA为1098个,AaM的特有circRNA为770个,AaS的特有circRNA为1127个。AaM和AaS的circRNA长度主要介于1000–2000 nt,基因间区circRNA为主要环化类型。AaM vs.AaS比较组包含456个上调circRNA和97个下调circRNA。共有circRNA的来源基因可注释到29个功能条目和14类通路;AaM的特有circRNA的来源基因可注释到31个功能和17类通路;AaS的特有circRNA的来源基因可注释到34个功能条目和16类通路;DEcircRNA的来源基因可注释到29个功能条目和40条通路。调控网络分析结果显示,36个共有circRNA靶向4个miRNA进而调控6个与内吞作用通路相关的靶mRNA;4(255)个AaM(AaS)的特有circRNA靶向2(2)个miRNA进而调控8(2)个次级代谢产物生物合成通路相关的靶mRNA;9个DEcircRNA靶向2个DEmiRNA进而调控3个MAPK信号通路相关的DEmRNA。RT-qPCR结果显示10个DEcircRNA的表达趋势与测序数据一致,证实了测序数据的可靠性。【结论】球囊菌菌丝和孢子的共有circRNA、特有circRNA和DEcircRNA可能通过调控来源基因表达和充当ceRNA的方式调节球囊菌的物质和能量代谢、内吞作用、次级代谢产物生物合成和MAPK信号通路,进而影响球囊菌菌丝生长、孢子萌发和致病性。 展开更多

【目的】蜜蜂球囊菌(Ascosphaeraapis,简称球囊菌)是专性侵染蜜蜂幼虫的致死性真菌病原。MicroRNA(miRNA)作为一类重要的基因表达调控因子,能够广泛参与真菌及其宿主的相互作用过程。本研究通过比较分析球囊菌孢子(AaCK)和侵染中华蜜蜂(... 【目的】蜜蜂球囊菌(Ascosphaeraapis,简称球囊菌)是专性侵染蜜蜂幼虫的致死性真菌病原。MicroRNA(miRNA)作为一类重要的基因表达调控因子,能够广泛参与真菌及其宿主的相互作用过程。本研究通过比较分析球囊菌孢子(AaCK)和侵染中华蜜蜂(Apis cerana cerana,简称中蜂)6日龄幼虫肠道内的球囊菌(AaT)的smallRNA(sRNA)组学数据对球囊菌的差异表达miRNA(differentiallyexpressed miRNA,DEmiRNA)、靶mRNA及二者间的调控网络进行全面解析,旨在揭示miRNA介导的球囊菌对中蜂幼虫的侵染机制。【方法】对于球囊菌侵染的中蜂6日龄幼虫肠道的small RNA-seq(sRNA-seq)数据,利用BLAST工具连续比对东方蜜蜂(Apiscerana)和球囊菌的参考基因组筛滤得到AaT的sRNA组学数据。分别将AaCK和AaT的sRNA组学数据比对miRBase数据库,对球囊菌侵染宿主前后miRNA的数量和结构特征进行分析。联用RNAhybrid+svm_light、Miranda和TargetScan软件预测AaCK vs AaT比较组中DEmiRNA的靶mRNA,进而利用相关生物信息学软件对上述靶mRNA进行GO分类和KEGG代谢通路富集分析。通过Cytoscape软件对DEmiRNA-mRNA调控网络进行可视化。利用Stem-loop RT-PCR、RT-qPCR和分子克隆验证测序结果的可靠性。【结果】在AaCK和AaT中分别鉴定到380和387个miRNA。结构特征分析结果显示,AaCK和AaT的mi RNA皆集中分布在18–25 nt,且首位碱基主要偏向于U。AaCKvsAaT比较组共有270个DEmiRNA,包含155个上调miRNA和115个下调miRNA,分别靶向结合6091和6145个mRNA。GO分类结果显示,上述靶mRNA主要涉及代谢进程、细胞进程、应激反应等15个生物学进程;细胞、细胞组分、细胞器等12个细胞组分;催化活性、结合、转运子活性等11个分子功能。KEGG代谢通路富集分析结果显示,上述靶mRNA富集在123条代谢通路,参与对氨基酸代谢、碳水化合物代谢以及核苷酸代谢等物质代谢,氧化磷酸化、硫代谢、氮代谢等能量代谢,以及MAPK和Hippo等信号通路的调控。球囊菌DEmiRNA与靶mRNA之间存在复杂的调控关系,其中miR-29-x、miR-250-x、miR-4968-y、miR-11200-x、novel-m0023-5p、novel-m0130-5p和novel-m0135-5p等DEmiRNA可靶向结合与球囊菌的半胱氨酸蛋白酶、DNA甲基化转移酶以及几丁质酶相关的mRNA;此外,miR-7-x、miR-9-z、miR-319-y和miR-5951-y等同时参与调控MAPK信号通路;进一步分析发现,miR-250-x同时参与对DNA甲基化转移酶、MAPK信号通路及其他酶类合成与代谢途径的调控,并可能参与球囊菌与中蜂6日龄幼虫之间的跨界调控。通过Stem-loopRT-PCR和RT-qPCR验证了4个DEmiRNA的差异表达,并利用分子克隆和Sanger测序证实miR-7-x的序列与测序结果一致。【结论】本研究解析了侵染中蜂6日龄幼虫的球囊菌的miRNA差异表达谱及DEmiRNA的调控网络,揭示了球囊菌DEmiRNA可能通过调控病原的物质和能量代谢、增殖、毒力、信号通路及相关mRNA参与对中蜂幼虫的侵染过程。miR-7-x、miR-250-x、novel-m0023-5p等关键DEmiRNA有望作为白垩病治疗的新型分子靶点。 展开更多
