The mitogen-activated protein kinase(MAPK)cascade is crucial to plant growth,development,and stress responses.MAPK kinases(MAPKK)play a vital role in linking upstream MAPKK kinases(MAPKKK)with the downstream MAPK.Black spot is one of the most serious fungal diseases of pear which is an important part of the fruit industry in China.The MAPKK genes have been identified in many plants,however,none has been reported in pear(Pyrus bretschneideri).In order to explore whether MAPK gene of pear is related to black spot disease,we designed this experiment.The present study investigated eight putative PbrMAPKK genes obtained from the Chinese white pear genome.The phylogenetic analysis revealed that PbrMAPKK genes were divided into A,B,C,and D groups.These PbrMAPKK genes are randomly distributed on 7 out of 17 chromosomes and mainly originated from the whole-genome duplication(WGD)event.The expression analysis of PbrMAPKK genes in seven pear tissues and the leaves of susceptible and resistant varieties after Alternaria alternata infection by quantitative real-time PCR(qRT-PCR)identified seven candidate genes associated with resistance.Furthermore,virus-induced gene silencing(VIGS)indicated that PbrMAPKK6 gene enhanced resistance to pear black spot disease in pear. 展开更多

The Spatial Patterns of Road Traffic Crash Black Spots and Emergency Facilities in Federal Capital City

This study aimed at determining the spatial patterns of Road Traffic Crash (RTC) black spots, Federal Road Safety Commission (FRSC) zebra points and emergency health care facilities in Federal Capital City (FCC). The aim was to provide stakeholders with information that will aid their understanding of accident prone locations and accessible rescue possibilities for accident victims on the roads in FCT. GPS Map 76S Mark (GARMIN) was used to locate and pick coordinates of data in the study area. A total of 16 possible emergency health care facilities, seventy (70) RTC black spots and Five Zebra point locations were obtained from FRSC. ArcGIS 10.0 was used to compute the data by plotting the coordinates to produce maps of the spatial relationship and to carry out Nearest Neighbour Analysis (NNA). The result was further used to determine the spatial patterns of RTC black spots as well as patterns of the emergency facilities. Generally, the result shows that the spatial trend is turning towards dispersion. However, there is less than 1% likelihood that the dispersed patterns could be the result of random chance. It was recommended that, the Federal Road Safety Commission should be staffed with trained professionals that can be responsible for accident data surveillance and analysis using geospatial techniques. 展开更多

In general, the location of traffic accidents is described as an address with text, so they are difficult to display on the map. The paper discusses how to utilize the geocoding technology and VRS-GPS positioning technology to record the traffic accidents with Geo-spatial information. Based on the spatial relationship between traffic accidents and road network elements, two-way association relationship is defined by spatial relationship computation. Then the paper presents the method which takes the potential of reducing accidents as an index to extract the black spots. Finally, in the discussion, the association relationship between black spots and traffic attributes is used to analyze the factors that resulting in traffic accidents. 展开更多

The results showed that the base sequence of pathogens collected from five different sites was almost identical with that of Alternaria brassicae,and the similarity degree reached 99. 68%,without base difference greater than 3 bp. It had obvious differences with the sequences of A.brassicicola and A. japonica,and the base difference was greater than 3 bp with a lot of fragment deletions. It was preliminarily determined that the pathogen causing black spot disease on tumorous stem mustard was A. brassicae. 展开更多

This paper describes the development of a hyperspectral imaging approach for identifying fruits infected with citrus black spot(CBS).Hyperspectral images were taken of healthy fruit and those with CBS symptoms or other potentially confounding peel conditions such as greasy spot,wind scar,or melanose.Spectral angle mapper(SAM)and spectral information divergence(SID)hyperspectral analysis approaches were used to classify fruit samples into two classes:CBS or non-CBS.The classification accuracy for CBS with SAM approach was 97.90%,and 97.14% with SID.The combination of hyperspectral images and two classification approaches(SID and SAM)have proven to be effective in recognizing CBS in the presence of other potentially confounding fruit peel conditions.The study result can be a reference for the non-destructive detection of fruits infected with citrus black spot. 展开更多

The identification of accident black spots is of great significance for the prevention of traffic accidents.Commonly used accident black spot identification methods divide road sections for the analysis of accident data,the direct result of which is the splitting of accident black spots,which affects the results.This paper is based on three years of traffic accident data from the Beijing-Harbin Expressway,including the time and location of traffic accidents,form of the accident fatalities,severe injuries,slight injuries,and property damage only(PDO).To avoid road division effects,an identification method based on the accident spacing distribution is established by using quality control theory.The results show that the average number of accidents per kilometer by the method proposed in this paper is 42,which is much higher than 10,identified by other identification methods.The method proposed in this paper improves the accuracy of the identification results.This method avoids the problem of road segmentation found in other common methods and can more accurately reflect the spatial distribution of traffic accidents.Thus making the identification of accidents more scientific and accurate. 展开更多

橄榄黑斑病病原菌的分离鉴定及防治药剂筛选

为明确引起潮州市橄榄黑斑病的病原菌种类,筛选有效防治药剂,对发病果实采用组织分离法进行了病原菌的分离与纯化,并结合形态学和分子生物学技术对病原菌进行鉴定。鉴定结果表明,引起橄榄黑斑病的病原菌为链格孢菌(Alternaria alternata... 为明确引起潮州市橄榄黑斑病的病原菌种类,筛选有效防治药剂,对发病果实采用组织分离法进行了病原菌的分离与纯化,并结合形态学和分子生物学技术对病原菌进行鉴定。鉴定结果表明,引起橄榄黑斑病的病原菌为链格孢菌(Alternaria alternata)。通过菌丝生长速率法测定了14种杀菌剂对病原菌的抑菌效果,结果表明14种杀菌剂对链格孢菌均有抑制作用,其中3%中生菌素可湿性粉剂、80%克菌丹水分散粒剂、8%氟硅唑微乳剂和40%苯甲·嘧菌酯悬浮剂的抑菌效果较好,EC_(50)分别为1.267、1.207、1.100、0.944 mg·L^(-1)。田间防效测定结果表明,40%苯甲·嘧菌酯悬浮剂、8%氟硅唑微乳剂的防治效果显著,防效为68.58%、65.44%;而30%王铜、2%春雷霉素的防效较差,仅为10%左右。综上,推荐将8%氟硅唑微乳剂和40%苯甲·嘧菌酯悬浮剂2种药剂作为防控橄榄黑斑病的首选药剂。本研究可为橄榄黑斑病高效杀菌剂的筛选提供数据参考,并为其防控工作提供科学依据。 展开更多

为解析菌株ZL397的分类地位及其对黑斑蛙源米尔伊丽莎白菌(Elizabethkingia miri-cola)的抑菌活性,利用形态分析、生理生化检测和16S rDNA序列分析,对ZL397进行菌种鉴定;采用细菌浓度测定法和扫描电镜观察法,考察ZL397对米尔伊丽莎白菌... 为解析菌株ZL397的分类地位及其对黑斑蛙源米尔伊丽莎白菌(Elizabethkingia miri-cola)的抑菌活性,利用形态分析、生理生化检测和16S rDNA序列分析,对ZL397进行菌种鉴定;采用细菌浓度测定法和扫描电镜观察法,考察ZL397对米尔伊丽莎白菌的抑菌活性;采用滤纸片法测定ZL397对12种病原菌的抑菌活性。攻毒保护试验选取成年黑斑蛙360只,随机分成4个组(每个处理3个重复,每个重复30只),即预防组、治疗组、攻毒对照组和对照组。试验期14 d。菌种鉴定、抑菌活性和抗菌谱试验结果表明,ZL397为特基拉芽孢杆菌(B.tequilensis);ZL397发酵上清液的50倍稀释液对米尔伊丽莎白菌的抑制率为90.29%,并可致使靶标菌细胞壁和膜破裂、菌体断裂;ZL397对多种病原细菌有抑制作用,尤其对藤黄八叠球菌(Micrococcus luteus)、鸡沙门氏菌(Salmonella gullina-rum)、金黄色葡萄球菌(Staphyloccocus aureus)、米尔伊丽莎白菌(Elizabethkingia miricola)、乙酸钙不动杆菌(Acinetobacter caleoaceticus)的抑菌活性较强。攻毒保护试验结果表明,7 d后攻毒对照组黑斑蛙存活率仅为41.11%,而添加0.1‰ZL397粉剂饲喂的预防组黑斑蛙存活率高达92.22%,两者差异显著(P<0.05);同时预防组体重增重率为35.15%,与不攻毒对照组接近,显著高于攻毒对照组(P<0.05)。综上结果得出,特基拉芽孢杆菌对黑斑蛙有抗病促生长功效,具有良好的应用前景。 展开更多

为找出核桃黑斑病的发生与核桃品种、生长环境以及栽培措施之间的关系,调查了17个核桃品种的病情指数,生长地的海拔、地形坡度、降雨量、温度、郁闭度、地面覆盖率等环境因子,以及灌溉次数、防治次数、是否清园等栽培措施,采用主成分分... 为找出核桃黑斑病的发生与核桃品种、生长环境以及栽培措施之间的关系,调查了17个核桃品种的病情指数,生长地的海拔、地形坡度、降雨量、温度、郁闭度、地面覆盖率等环境因子,以及灌溉次数、防治次数、是否清园等栽培措施,采用主成分分析法和相关性方法分析了环境因子、栽培措施与核桃细菌性黑斑病发生的关系。结果表明:(1)根据17个核桃品种的病情指数得出中强特勒、辽核1号、西扶为抗病品种;(2)发现核桃黑斑病病情指数与郁闭度、防治次数和是否清园具有强相关性,郁闭度为正相关,防治次数和是否清园为负相关,平均温度、地面覆盖率与病情指数为负相关。(3)核桃黑斑病发病率与雨水为极显著正相关,与海拔高度为极显著负相关。建议在推广中应结合立地条件,合理布局,两树的间距需在6 m以上,郁闭度控制在0.6以下,并做好清园、灌溉和防治等树体管理工作,以减少核桃病害的发生。 展开更多

【目的】筛选对刺五加黑斑病具有生防潜力的细菌菌株。【方法】采用平板对峙法从健康刺五加根际土壤中分离筛选对刺五加黑斑病菌具有拮抗作用的菌株,基于形态学、生理生化鉴定及16S rDNA结合gyrB测序分析进行菌种分类鉴定,测定其抗菌谱... 【目的】筛选对刺五加黑斑病具有生防潜力的细菌菌株。【方法】采用平板对峙法从健康刺五加根际土壤中分离筛选对刺五加黑斑病菌具有拮抗作用的菌株,基于形态学、生理生化鉴定及16S rDNA结合gyrB测序分析进行菌种分类鉴定,测定其抗菌谱;并通过叶面定殖及盆栽防效试验进一步评价该菌的生防潜力。【结果】分离得到419株细菌,其中,菌株YZ-375鉴定为贝莱斯芽孢杆菌Bacillus velezensis。菌株YZ-375对刺五加黑斑病菌的拮抗效果较好,无菌滤液的抑制率为65.85%,且对人参立枯丝核菌Rhizoctonia solani、人参链格孢菌Alternaria panax、人参茄腐皮镰孢菌Fusarium solani、人参核盘菌Sclerotinia ginseng、人参毁灭柱孢菌Cylindrocarpon destructans、五味子尖镰孢菌Fusarium oxysporum均具有拮抗效果,抑制率为69.21%~77.44%。在鲜叶定殖35 d后,菌株YZ-375菌量仍能保持在每克鲜叶103CFU。菌株YZ-375对刺五加黑斑病防效达72.31%,与枯草芽孢杆菌、苯醚甲环唑防效无显著差异。【结论】贝莱斯芽孢杆菌YZ-375具有良好的生防潜力,具有一定的开发应用前景。 展开更多

超声波联合水杨酸处理对冬枣采后黑斑病的防控研究

为探究超声波(ultrasound,US)联合水杨酸(salicylic acid,SA)处理对冬枣采后黑斑病的防控效果及其机理,该文使用US单独或联合SA处理冬枣果实,测定发病率、自然腐烂率,筛选出最佳的处理条件,并从其对病原菌的直接作用、诱导枣果实抗病性... 为探究超声波(ultrasound,US)联合水杨酸(salicylic acid,SA)处理对冬枣采后黑斑病的防控效果及其机理,该文使用US单独或联合SA处理冬枣果实,测定发病率、自然腐烂率,筛选出最佳的处理条件,并从其对病原菌的直接作用、诱导枣果实抗病性等方面对其防治机制进一步研究。结果表明,US单独或联合SA处理均能有效控制冬枣采后黑斑病,并显著降低自然腐烂率,且联合处理的防治效果显著优于单独处理(P<0.05)。对其作用机制进一步探究,US联合SA处理能显著抑制Alternaria alternata的菌丝生长(P<0.05)。US处理不仅能降低A.alternata在冬枣伤口处的附着量,还能使冬枣果皮表面形成微裂纹,从而利于SA与果实接触,促进SA向果实渗透。此外,US与SA联合处理可以显著提高果实过氧化物酶、多酚氧化酶、苯丙氨酸解氨酶、4-香豆酸辅酶A连接酶活性(P<0.05),促进冬枣中的总酚、黄酮、游离酚、结合酚的积累(P<0.05),诱导枣果实抗病性,提高枣果实对病害的防御能力,从而降低果实腐烂率。综上所述,US联合SA作为一种绿色安全的处理方式对于控制冬枣采后黑斑病具有良好的研究潜力和开发前景。 展开更多
