This study investigates the effects of ginsenoside Rbl, a major constituent of ginseng,on H<sub>2</sub>O<sub>2</sub>-induced endothelial senescence.Methods Primary human umbilical vein endothelial cells(HUVECs) senescence was induced by H<sub>2</sub>O<sub>2</sub> as judged by senescence-associated P-galactosidase assay (SA-P-gal).Fntracellur superoxide dismutase(S0D1) activity and malondialdehyde(MDA) level were determined by commercial kit.S0D1 mRNA and protein expression were analyzed by real time PCR and Western blot.Reactive oxygen species(ROS) were determined by flow cytometry.Results Rb1 was found to reverse endothelial senescence,as witnessed by a significant decrease of senescent cell numbers. Rbl could markedly increase intracellular SOD activity, decrease the MDA level,and suppress the generation of intracellular ROS in H<sub>2</sub>O<sub>2</sub>-treated HUVECs.Consistent with these findings,Rbl can effectively restore SOD1 mRNA and protein expression which decreased in H<sub>2</sub>O<sub>2</sub> treated cells. Conclusions Our report demonstrates thatRbl can exert reversal effects on H<sub>2</sub>O<sub>2</sub>-induced cellular senescence through modulating cellular redox status.展开更多

Objective:To investigate the effects and mechanisms of tetrahydroberberine(THB)on vascular endothelial cell injury induced by oxygen glucose deprivation(OGD)and LPS-induced cell injury model.Methods:MTT assay was used...Objective:To investigate the effects and mechanisms of tetrahydroberberine(THB)on vascular endothelial cell injury induced by oxygen glucose deprivation(OGD)and LPS-induced cell injury model.Methods:MTT assay was used to detect the effects of THB final concentration on the survival rate of EA.hy926 cells,which were 0μg/mL,2.5μg/mL,5μg/mL,10μg/mL,20μg/mL and 40μg/mL,respectively.Group I of the experiment:normal control group,OGD group,OGD+THB-10μg/mL group,OGD+THB-20μg/mL group,OGD+THB-40μg/mL group;Experimental group II group:normal Control group,LPS group,OGD+LPS group and OGD+LPS+THB-20μg/mL group.The relative expression of inflammation-related factors such as Caspase-1,ASC,IL-1βand IL-6 was detected by fluorescence quantitative PCR,and the expression of IL-1βand HO-1 was detected by Western Blot analysis.Results:The results of MTT assay showed that THB had no effect on the survival of EA.hy926 cells.The qPCR results of the experimental group I and II showed that the OGD model could play an anti-inflammatory protective role by down-regulating the expression of Caspase-1,ASC,IL-1βand IL-6 genes.The anti-inflammatory effect of THB on the OGD model was general,but it was found that THB also exerted an anti-inflammatory effect by down-regulating the expression of inflammatory genes by OGD+LPS group compared with OGD+LPS+THB-20μg/mL group.Western Blot analysis showed that the expression of IL-1βprotein was consistent with the results of qPCR.The expression of HO-1 protein as a protective factor was contrary to that of inflammatory factor fluorescence quantitative analysis.Conclusion:OGD model can improve the protective effect of cell stress.THB plays an anti-inflammatory role by down-regulating the expression of Caspase-1,ASC,IL-1βand IL-6 genes,up-regulating the expression of HO-1 protein and inhibiting the activation of NLRP3 inflammatory bodies.展开更多
Acupuncture is widely used in the treatment of cerebral hemorrhage,and it improves outcomes in experimental animal models and patients.However,the mechanisms underlying the effectiveness of acupuncture treatment for c...Acupuncture is widely used in the treatment of cerebral hemorrhage,and it improves outcomes in experimental animal models and patients.However,the mechanisms underlying the effectiveness of acupuncture treatment for cerebral hemorrhage are still unclear.In this study,a model of intracerebral hemorrhage was produced by injecting 50μL autologous blood into the caudate nucleus in Wistar rats.Acupuncture at Baihui(DU20)and Qubin(GB7)acupoints was performed at a depth of 1.0 inch,12 hours after blood injection,once every 24 hours.The needle was rotated at 200 r/min for 5 minutes,For each 30-minute session,needling at 200 r/min was performed for three sessions,each lasting 5 minutes.For the positive control group,at 6 hours,and 1,2,3 and 7 days after induction of hemorrhage,the rats were intraperitoneally injected with 1 mL aniracetam(0.75 mg/mL),three times a day.The Bederson behavioral test was used to assess palsy in the contralateral limbs.Western blot assay was used to examine the expression levels of Nestin and basic fibroblast growth factor in the basal ganglia.Immunohistochemistry was performed to count the number of Nestin-and glial cell line-derived neurotrophic factor-positive cells in the basal ganglia.Acupuncture effectively reduced hemorrhage and brain edema,elevated the expression levels of Nestin and basic fibroblast growth factor in the basal ganglia,and increased the number of Nestin-and glial cell line-derived neurotrophic factor-positive cells in the basal ganglia.Together,these findings suggest that acupuncture promotes functional recovery after cerebral hemorrhage by increasing the expression of neurotrophic factors.The study was approved by the Committee for Experimental Animals of Heilongjiang Medical Laboratory Animal Center(approval No.2017061001)on June 10,2017.展开更多
