A new algorithm for segmentation of suspected lung ROI(regions of interest)by mean-shift clustering and multi-scale HESSIAN matrix dot filtering was proposed.Original image was firstly filtered by multi-scale HESSIAN matrix dot filters, round suspected nodular lesions in the image were enhanced, and linear shape regions of the trachea and vascular were suppressed. Then, three types of information, such as,shape filtering value of HESSIAN matrix, gray value, and spatial location, were introduced to feature space. The kernel function of mean-shift clustering was divided into product form of three kinds of kernel functions corresponding to the three feature information. Finally, bandwidths were calculated adaptively to determine the bandwidth of each suspected area, and they were used in mean-shift clustering segmentation.Experimental results show that by the introduction of HESSIAN matrix of dot filtering information to mean-shift clustering, nodular regions can be segmented from blood vessels, trachea,or cross regions connected to the nodule,non-nodular areas can be removed from ROIs properly, and ground glass object(GGO)nodular areas can also be segmented. For the experimental data set of 127 different forms of nodules, the average accuracy of the proposed algorithm is more than 90%. 展开更多

The urgency of early lung cancer(LC)diagnosis and treatment has been more and more significant.Exhaled breath analysis using gas sensors is a promising way to find out if someone has LC due to its low-cost,non-invasive,and real-time monitoring compared with traditional invasive diagnostic techniques.Among sensor-based gas detection techniques, metal oxide semiconductor’s gas sensors are one of the most important types. This review presents the-state-of-art in metal oxide gas sensors for the diagnosis of early LC.First, the exhaled breath biomarkers are described with emphasis on the concentration of abnormal volatile organic compounds(VOCs)caused by the metabolic process of LC cells. Then, the research status of metal oxide gas sensors in LC diagnosis is summarized. The sensing performance and enhancement strategy of biomarkers provided by metal oxide semiconductor materials are reviewed.Another effective way to improve VOC detection performance is to build a gas sensor array. At the same time, various gas sensors combined with self-powered techniques are mentioned to display a broad development prospect in breath diagnosis. Finally, metal oxide gas sensor-based LC diagnosis is prospected. 展开更多

Objectives To evaluate the value of detecting p53 gene point mutations in sputum samples and its validity and reliability as a surveillance index in the early diagnosis of lung cancer in suspicious patients. Methods Sputum samples were collected from 54 cases identified as lung cancer and 114 cases identified as pulmonary benign disease. The polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) was performed for the detection of point mutations at exons 5-8 of the p53 gene, and sputum smears were also used for each sample. Conclusion Detection of p53 gene alterations in sputum samples by PCR-SSCP-silver stain can be used as a follow-up surveillance index for the early diagnosis of lung cancer in suspicious patients. 展开更多

To evaluate the early diagnostic value of circulating miRNA-21 in diagnosis of lung cancer, databases such as Wan Fang, VIP, PubMed, and Elsevier were systematically searched from 2005 to 2013 to collect relevant references in which the diagnostic value had been evaluated. The statistics were consolidated and the qualities of the studies were classified. The data were analyzed using Meta Disc1.4 software. The diagnostic value of circulating miRNA-21 in lung cancer was assessed by pooling sensitivity, specificity, the likelihood ratio, and the Summary Receiver Operating Characteristic(SROC) curve. Publication biases of the studies involved were analyzed using Stata 11.0 software. A total of 143 papers were collected of which 8 were included, which contained 600 cases and440 controls. A heterogeneity test proved the existence of homogeneity in this study. Upon analysis using random effects models, the weighted sensitivity was 0.68, the specificity 0.77, the positive likelihood ratio 2.84, the negative likelihood ratio 0.40, and the SROC Area Under the Curve(AUC) was 0.8133. Further analysis by subgroup showed that the 5 indicators mentioned above were 0.72, 0.84, 4.50, 0.27, and 0.8987, respectively, for the serum group and 0.63, 0.70, 1.95, 0.53, and 0.7318, respectively, for the plasma group. We conclude that circulating miRNA-21can be regarded a valuable reference in diagnosis of lung cancer. This research showed that in lung cancer the early diagnostic value of miRNA-21 in serum was better than that in plasma. 展开更多

Screening and early diagnosis of lung cancer relies mainly on chest X-ray, low-dose computed tomography, bronchoscopy, sputum cytology, and measurement of tumor markers such as carcinoembryonic antigen (CEA), cytokeratin-19 fragments (Cyfra21-1), and neuron-specific enolase (NSE). However, all these methods lack adequate sensitivity and/or specificity. 1-3 Better methods are therefore urgently needed in screening and early diagnosis of lung cancer, and proteomic fingerprinting represents a promising approach. Surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS), an innovative proteomic technology, has overcome many of the limitations of two-dimensional electrophoresis (2D) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). 4,5 Recently, this technology has been successfully used to distinguish pancreatic, ovarian and prostate cancer patients from healthy control subjects. 5-7 The aim of the current study was to investigate the application of serum SELDI protein profiling in distinguishing lung cancer patients from healthy persons. 展开更多

Aim:A significant fraction of mortalities from non-small cell lung cancer could be prevented,if the cancer would be diagnosed earlier.Nanobiosensors for the ultrasensitive detection of active proteases in serum were designed to detect a significant protease activity signature of non-small cell lung cancer(stage I and higher). Methods:We determined the activity of nine protease biomarkers in the sera of non-small cell lung cancer patients and compared them with the protease activities of a control group of healthy human subjects using optical nanobiosensors. They consist of a central Fe/Fe3O4 core/shell nanoparticle with an attached Fluorescence resonance energy transfer-pair[tetrakis-carboxyphenyl porphyrin(TCPP)and cyanine 5.5].TCPP is attached to the central nanoparticle via a protease-cleavable tether, whereas cyanine 5.5 is tethered permanently to the dopamine-layer surrounding the nanoparticle. Results:Based on the activity pattern of urokinase plasminogen activator, matrix metalloproteinases 1,2,3,7,9,and 13,and cathepsins B and L as well,non-small cell lung cancer could be detected at stage I by means of a liquid biopsy. Conclusion:This feasibility study, comprising 33 non-small cell lung cancer patients and 20 apparently healthy subjects, clearly demonstrated the feasibility of minimally invasive early diagnosis of non-small cell lung cancer, starting with stage I. 展开更多
