A multidisciplinary approach-anatomy, histochemistry and phytochemistry-was used to investigate the leaf structure, the content and the storage location of barbaloin in the leaves of Aloe vera L. var. chinensis (Haw.)...A multidisciplinary approach-anatomy, histochemistry and phytochemistry-was used to investigate the leaf structure, the content and the storage location of barbaloin in the leaves of Aloe vera L. var. chinensis (Haw.) Berg. Xeromorphic characteristics including secondary thickened epidermal cell walls, thicker cuticle, ambiguous differentiation of spongy and palisade tissues in the chlorenchyma, and well-developed aquiferous tissue could be seen in the leaves. Several large parenchymatous cells were observed at the phloem pole of the first ring of vascular bundles. The secondary ring of vascular bundles in the leaf base and the stomata, which are surrounded by five cells, have some classification significance in this species. The density of vascular bundles, the content of barbaloin and the intensity of histochemical reaction differed among leaf numbers Ll (annual leaf), L2 (biennial leaf), L3 (triennial leaf) and L4 (quadrennial leaf), and in different parts of the leaf. These three factors were highest in the youngest leaf, Ll, and top parts of all the leaves and lowest in the basal parts and the oldest leaf, L4. The density of vascular bundles had a positive correlation to the content of barbaloin. The histochemical results revealed that the small sheath cells that surrounded the bundles might be the location of barbaloin synthesis and the large parenchymatous cells beneath the sheath might be the storage places of this metabolite.展开更多

Methods Compounds were separated and purified by macroreticular resin column chromatography and high-performance liquid chromatography. Their structures were elucidated by spectroscopic and hydrolysis analysis. Results Six compounds were isolated from the 70% ethanolic extracts. They were identified as escins IVc, IVd, Ia, Ib, isoescins Ia and Ib, respectively. Conclusion The above compounds were obtained from the seeds of Aesculus chinensis Bunge var.chekiangensis (Hu et Fang) Fang for the first time.展开更多

Berger and Aloe barbadensis Miller, and whether there was a close relationship between the contents o...A reserved-phase HPLC method was developed for the determination of barbaloin in Aloe vera L. var. chinensis (Haw.) Berger and Aloe barbadensis Miller, and whether there was a close relationship between the contents of barbaloin and their environments in which they were growing was decided. A Hypersil ODS column (4.6 mm×200 mm, 5 μm)was used with a mobile phase of methanol-water (40:60, containing 0.1% acetic acid), the flow rate being 1.0 mL·min -1, detection wavelength at 359 nm, and the column temperature being 30℃. The linear range of barbaloin was between 0.0726 and 0.726 μg with a correlation coefficient of 0.9998 and the regression equation being Y=1.9202×10 6X-1801.9. Barbaloin was stable in methanol in 48 h and the instrument precision was 1.2% while the method precision was 4.9%. The contents of barbaloin of 12 samples ranged from 6.160 to 319.1 μg·g -1. The method developed was fast and simple with good reproducibility. There was high correlation between the contents of barbaloin and their growing environments.展开更多

[Method] In the nutrient solutions for growing B. campestris by hydroponics, boric acid and manganese sulfate were added at 0.5, 2.5, and 7.5 mg/L respectively. Another treatment without boron and manganese was prepared as the control. Quality and physiological indices of B. campestris in the 10 treatments were measured. [Result] Boron and manganese shared obvious in-teraction in improving the quality and physiological indices of B. campestris. To cul-tivate B. campestris with high quality and strong resistance, the optimum concentra-tions of boron and manganese in the nutrient solution should be 2.5 mg/L boric acid and 2.5-7.5 mg/L manganese sulfate. [Conclusion] The findings wil provide refer-ence for studying effects of trace elements on nutrient composition of vegetables.展开更多
Amaryllidaceae, a monocot plant family, consists of many important ornamental bulb flower species. Chinese narcissus (Narcissus tazetta var. chinensis Roem), its flowers developed at high temperatures and bloomed at...Amaryllidaceae, a monocot plant family, consists of many important ornamental bulb flower species. Chinese narcissus (Narcissus tazetta var. chinensis Roem), its flowers developed at high temperatures and bloomed at lower temperatures during the Chinese Spring Festival, is a traditional Chinese flower with high economic and ornamental value. To study its flower development, a full length cDNA containing MADS box domain from narcissus was isolated by a reverse transcription polymerase chain reaction (RT-PCR) with degenerate oligo-nucleotide primers derived from a conserved MADS- and K-box domain sequence. The 5' and the 3' regions of the gene were amplified using the PCR protocol for the rapid amplification of cDNA ends (RACE). The 690 bp open reading frame encodes 230 amino acid residues. A comparison of the deduced amino acid sequence of NTAG with the sequence of other MADS box proteins showed 91.3% amino acid identities with HAG (Hyacinthus orientalis). Sequence analysis and alignment showed significant similarity with other AG homologues. RNA blot analysis indicated that the narcissus NTAG gene was expressed only in reproductive organs, especially in stamens and carpels. These results indicated that the NTAG gene was an AG homologue and that the AG genes appeared to be structurally and functionally conserved between dicots and monocots.展开更多

Seedlings of Taxus chinensis var.mairei were used as experimental materials to study the adaptation of Piriformospora indica to this plant under water stress.The materials were divided into two groups,namely,with or w...Seedlings of Taxus chinensis var.mairei were used as experimental materials to study the adaptation of Piriformospora indica to this plant under water stress.The materials were divided into two groups,namely,with or without inoculation with P.indica.Each group was subjected to four different levels of water stress.Vitality and physiological and biochemical indexes of the roots of T.chinensis var.mairei were regularly measured.Under water stress,T.chinensis var.mairei had significantly decreased root vitality;root vitality was higher in inoculated roots than in uninoculated roots.Under intense water stress,the inoculated roots had a higher soluble sugar content than the uninoculated roots.Under water stress,T.chinensis var.mairei experienced decreased activity of aerobic respiratory metabolic enzymes.The activity of anaerobic respiratory metabolic enzymes and alcohol dehydrogenase initially increased and then decreased,whereas that of lactate dehydrogenase increased.The inoculated roots had a higher activity of respiratory metabolic enzymes than the uninoculated roots.As water stress was further intensified,the roots had significantly decreased activity of aerobic respiratory metabolic enzymes and significantly increased activity of anaerobic respiratory metabolic enzymes.The activity of respiratory metabolic enzymes decreased faster in the uninoculated roots than in the inoculated roots.This study demonstrated that Piriformospora indica plays a positive role in enhancing the antihypoxic ability of T.chinensis var.mairei,thereby alleviating plant damage due to water stress.展开更多

The plantlets should be transplanted from test-tube to soil when they grew ' to 2.5 ~ 4.0 cm high and have 3 ~ 5 strips short roots. That is, Ce4+ of low concentration improves the soluble protein synthetic strength and cell activity, while Ce4+ of high concentration is harmful to protein synthesis and cell activity. In order to learn dormancy mechanism of Taxtts chinensis var. mairei seed, effects of stratification and hormone treatments on physiological and biochemical indexes of the seeds after stratifying for eight months were...In order to learn dormancy mechanism of Taxtts chinensis var. mairei seed, effects of stratification and hormone treatments on physiological and biochemical indexes of the seeds after stratifying for eight months were researched. The content of amylase activity and soluble sugar gradually increased at first stratification, and then to the maximum for the end of after-ripening period. With the dormancy of seed relieving, some soluble sugar of endosperm was used or consumed by respiration and that led to their content decreased. Seed soluble protein was gradually accumulated at the beginning of stratification and then the initial raw material of protein synthesis of embryo during development had been used or in part converted into carbohydrates for energy consumption, so that the soluble protein content decreased. The fat and starch content slowly decreased during stratification, which showed that the storage of material had continually carrying out catabolism and conversion, which might provide energy sources for seed germination in the stratification process. The peroxidase activity of Taxus seeds increased gradually at the beginning of stratification, and then reached maximum point during stratification for 4 months in A1 and A2. The G-6-PDH and 6-PGDH joint activity was rapidly increased to reach the maximum during stratification for 4 months in A1, which showed the pentose phosphate pathway was activated at this time. The peroxidase activity rapidly increased might also be related to respiratory metabolic pathway during stratification. In plants, acid phosphatase hydrolyze PAM metabolites, namely phosphate material was related to energy metabolism, especially to high-energy phosphate, and phosphate enzyme activity changes affected the energy metabolism. The acid phosphatase activity of Taxus seeds in different treatments has no significant changes during stratification process, but there were two maximums. Energy metabolism might get benefit from the increase of acid phosphatase activity. There were significant differences in amylase activity, peroxidase activity, soluble sugar and soluble protein content with different treatments. Effect of promoting seed germination soaked by GA before stratification was obvious, but the concentration of GA should not be too high. The high concentration of GA caused seed to rot and reduce the seed germination rate. In this study, the highest germination rate of seed was 71.8% and soaked in 200 mg·L-1 gibberellin.展开更多

[Conclusions] Diseases significantly affected composition and content of total saponins and steroidal saponins from P. polyphylla var. chinensis,further affecting quality of P. polyphylla var. chinensis.

文摘The production of paclitaxel from suspension culture of Taxus chinensis var,mairei was improved by in situ extraction with organic solvents to avoid feedback repression and product degradation.Oleic acid and dibutyl phthalate were proved to be suitable solvents .The optimal volumetric percentage of organic solvents in the culture medium was found to be around 8%,and the favorable time for their introduction was at the exponential phase of cell growth,Paclitaxel production with the in situ extraction was ca 3-fold of that without extraction.
文摘The tissue cullurc of Schloss Mannheim (Rosa Chinensus , var. Florihunda ) with full and unsprouting bud of stem segments as the explants was experimented The result shows that the buds sprouted best on MS medium with the addition of 6-HA 1 .0 mg/L,. and differentiation was best on MS medium with addition of 6-BA 1 .5 + NAA0.05 + ZT0.1 mg/L, or KT 1,0 + NAA0,05 + ZT0.1mg/L. The MS emdium with addition of 6-BA 0.3 + NAA 0.0 5+ ZT0.1 mg/L, or KT 0.3 + NAA0.05 +ZT0. 1 mg/L showed a good result for developing strong shoots. 1/2 MS medium with the addition ofIBA 0.1 mg/L, or IBA0.1 + NAA0.02 mg/L, had best result for rooting. The plantlets should be transplanted from test-tube to soil when they grew ' to 2.5 ~ 4.0 cm high and have 3 ~ 5 strips short roots. Ahigher survival rate was obtained under the conditions of conterolling humidity and temperature
文摘The dynamic effects of Ce4+ on the syntheses of soluble protein and taxol in suspension cultures of Taxus chinensis var. mairei cells were studied. The phenomena of 'partition' and 'bifurcation' were observed in studying the dynamic effect of Ce4+ on soluble protein synthesis and cell activity. That is, Ce4+ of low concentration improves the soluble protein synthetic strength and cell activity, while Ce4+ of high concentration is harmful to protein synthesis and cell activity. In addition, Ce4+ of appropriate concentration enhances taxol synthesis.
文摘In order to learn dormancy mechanism of Taxtts chinensis var. mairei seed, effects of stratification and hormone treatments on physiological and biochemical indexes of the seeds after stratifying for eight months were researched. The content of amylase activity and soluble sugar gradually increased at first stratification, and then to the maximum for the end of after-ripening period. With the dormancy of seed relieving, some soluble sugar of endosperm was used or consumed by respiration and that led to their content decreased. Seed soluble protein was gradually accumulated at the beginning of stratification and then the initial raw material of protein synthesis of embryo during development had been used or in part converted into carbohydrates for energy consumption, so that the soluble protein content decreased. The fat and starch content slowly decreased during stratification, which showed that the storage of material had continually carrying out catabolism and conversion, which might provide energy sources for seed germination in the stratification process. The peroxidase activity of Taxus seeds increased gradually at the beginning of stratification, and then reached maximum point during stratification for 4 months in A1 and A2. The G-6-PDH and 6-PGDH joint activity was rapidly increased to reach the maximum during stratification for 4 months in A1, which showed the pentose phosphate pathway was activated at this time. The peroxidase activity rapidly increased might also be related to respiratory metabolic pathway during stratification. In plants, acid phosphatase hydrolyze PAM metabolites, namely phosphate material was related to energy metabolism, especially to high-energy phosphate, and phosphate enzyme activity changes affected the energy metabolism. The acid phosphatase activity of Taxus seeds in different treatments has no significant changes during stratification process, but there were two maximums. Energy metabolism might get benefit from the increase of acid phosphatase activity. There were significant differences in amylase activity, peroxidase activity, soluble sugar and soluble protein content with different treatments. Effect of promoting seed germination soaked by GA before stratification was obvious, but the concentration of GA should not be too high. The high concentration of GA caused seed to rot and reduce the seed germination rate. In this study, the highest germination rate of seed was 71.8% and soaked in 200 mg·L-1 gibberellin.
