Huntington病(HD)是一种常染色体显性遗传性神经退行性疾病,主要组织病理学特征是纹状体运动神经元进行性死亡,目前尚无有效治疗方法,本实验探讨了nortriptyline(去甲替林)的潜在疗效。采用N548mu[1955-128]huntingtin ST14A细胞系进行体外培养,观察不同浓度nortriptyline对细胞存活的影响。选用R6/2转基因鼠,从第5周开始每天腹腔注射nortriptyline,直至21周,对照组腹腔注射与nortriptyline组等剂量的生理盐水。结果发现,nortriptyline对移入不同温度环境的ST14A细胞具有保护作用。nortriptyline可推迟R6/2转基因鼠的发病,由102d推迟到127d,与对照组比较具有显著性差异。nortriptyline对R6/2转基因鼠死亡率的影响,与对照组比较无显著性差异。作为检测nortriptyline毒性的指标,nortriptyline对R6/2转基因鼠体重的影响,与对照组比较无显著性差异。结果表明,nortriptyline可推迟Huntington转基因鼠的发病,是目前毒性低、能够延长Huntington转基因鼠存活的有效药物。 展开更多

This work evaluates intercalation of Nortriptyline(NT)and Venlafaxine(VFX)in an interlayer gallery of Na^(+)-MMT(Montmorillonite),which was further compounded with Poly(LLactide)(PLLA)to form microcomposite spheres(MPs)for oral controlled drug delivery.The XRD patterns,thermal and spectroscopic analyses indicated intercalation of drugs into the MMT interlayer that was stabilized by electrostatic interaction.No significant changes in structural and functional properties of drugs were found in the MMT layers.In vitro drug release studies showed controlled release pattern. 展开更多

Background: Melatonin is a safe and effective treatment for patients with functional pain and gastrointestinal disorders. Objective: To evaluate the efficacy and safety of melatonin for 3 months in patients with functional heartburn. Methods: In the randomized, placebo-controlled pilot study, patients with functional heartburn were treated with omeprazole 20 mg before breakfast and randomized to receive either melatonin 6 mg (n = 20), nortripty line 25 mg (n = 20), or placebo (n = 20) at bedtime for 3 months. Gastroesophageal Reflux Disease-Health-Related Quality of Life (GERD-HRQOL) scores were assessed at baseline and at the end of the study. Results: Melatonin improved GERD-HRQOL scores compared with nortriptyline (P = 0.0015) and placebo (P < 0.0001). Fewer adverse events were reported by patients receiving melatonin compared with those receiving nortriptyline or placebo. Conclusions: Melatonin was a safe and effective treatment for functional heartburn. 展开更多
