Because of the high affinity of lectin for saccharides, the lectin conjugated GNPs are able to employ as indicators for monitoring the antibiotic induced changes of glycosyl complexes. The interactions of a well known antibiotic, tunicamycin, with two different cell lines, HeLa and SHG-44, were selected to establish this assay. In the presence of tunicamycin, the dose- and time-dependence on the decreasing of binding affinity of lectin conjugated GNPs with living cells were demonstrated by conventional microscopic and UV-Vis spectroscopic studies. The experimental result demonstrates that our approach can be used to identify antibiotic induced expression difference of glycosyl complexes on different cellular surfaces and determine drug activity quantitatively. For further confirming the capability of the GNP-based assay, the system was also studied by confocal laser scanning microscopy(CLSM) and classic flow cytometry(FCM) assay, and satisfactory results were obtained.展开更多
According to the differences in ore-controlling structural systems and the characteristics of host rocks, textures and structures of ores and mineral associations of ores, quartz vein-type gold deposits in the Rushan ...According to the differences in ore-controlling structural systems and the characteristics of host rocks, textures and structures of ores and mineral associations of ores, quartz vein-type gold deposits in the Rushan area can be divided into the Rushan and Tongling styles. Rushan style gold deposits, occurring in the Kunyushan complex, include Rushan, Tangjiagou and Tongxishan gold mines. They are distributed along four NNE-trending and sinistral, compresso-shear faults with a right stepping array. A prominent characteristic of the gold mineralization is that the orebodies in neighbouring gold deposits distributed in a single ore-controlling fault zone take opposite pitches. Study of the locating structures of the quartz vein gold deposits shows that the Rushan-style gold deposits are characterized by NNE and NE zoning. Therefore, the intersections of the NE direction of the known gold deposit and the neighbouring NNE-trending fault zones are favourable for looking for gold deposits, and the ends of the curving segments of sinistral and right-stepping faults are favourable for looking for large gold deposits. Tongling-style gold deposits occur in the Sanfosan porphyritic monzonitic granite. Emplacement of gold deposits is controlled by arcuate and radiate structures formed during the intrusion of the Sanfoshan porphyritic monzonitic granite; the radiate faults controlled the distribution of orebodies and the arcuate faults controlled the pitch of the orebodies.展开更多
Studies on the influence of one critical parameter (e.g., size), targeting a specific disease, while keeping other factors unchanged, are important for improving understanding and application of the molecular design...Studies on the influence of one critical parameter (e.g., size), targeting a specific disease, while keeping other factors unchanged, are important for improving understanding and application of the molecular design of biomedical nanomaterials. In this study, we used doxorubicin (Dox)-conjugated gold nanoparticles (GNPs) to investigate the effects of the size of the gold core (10, 20, or 60 nm) on the performance of their conjugates. We found that all three conjugates differed slightly in their physicochemical properties, facilitating a direct and accurate assessment of the size effects of GNP-Dox conjugates on their in vitro and in vivo performance. The cytological properties (the cell penetration rate and efficiency, as well as the cytotoxicity) and antitumor performance (the intratumoral penetration, treatment efficacy, and biodistribution) were highly correlated to the size of the inorganic core. Among all test groups, although the conjugate with a 60-nm gold core had the highest drug loading and release efficiency, the conjugate with a 10-nm gold core displayed the best antitumor efficacy toward the liver cancer models. This was because it showed the deepest tumor permeability and the highest tumor cell-killing ability of Dox transported by the relatively small GNPs. This study provides important evidence for better understanding the effect of size on in vitro and in vivo properties of potential therapeutic nanosystems and their structure design.展开更多

[Method] Based on bio- layer interferometry technology, ampicillin-BSA conjugate was fixed on the bottom of APS fiber optic biosensor probe through hydrophobic interaction and bound to 40 mn colloidal gold-labeled/3-1actam antibiotic receptor, to detect β-lactam antibiotics in milk. [ Result] The sensitivity of colloidal gold-labeled BLI method was twice as high as that of immunechromatographic test strip in detection of β-1actam antibiotic residue in milk. Colloidal gold-labeled BLI method exhibited good speci- ficity and had no cross-reaction with 1 000 ng/ml aflatoxin M1, gentamicin, kanamycin, streptomycin, tylosin, chloromycetin and melamine. [Condusion] The colloidal gold-labeled BLI method is not suitable for quantitative detection in actual production due to its small quantitative range in detection of β-lactam antibiot- ics, but it is a simple and rapid qualitative detection method that can be used in rapid detection of β-1actam antibiotic residue in milk.展开更多
