Remyelination failure is one of the main characteristics of multiple sclerosis and is potentially correlated with disease progression.Previous research has shown that the extracellular matrix is associated with remyelination failure because remodeling of the matrix often fails in both chronic and progressive multiple sclerosis.Fibronectin aggregates are assembled and persistently exist in chronic multiple sclerosis,thus inhibiting remyelination.Although many advances have been made in the mechanisms and treatment of multiple sclerosis,it remains very difficult for drugs to reach pathological brain tissues;this is due to the complexity of brain structure and function,especially the existence of the blood-brain barrier.Therefore,herein,we review the effects of fibronectin aggregates on multiple sclerosis and the efficacy of different forms of drug delivery across the blood-brain barrier in the treatment of this disease. 展开更多

The extracellular matrix surrounding oligodendrocytes plays an important role during myelination and remyelination in the brain.In many cases,the microenvironment surrounding demyelination lesions contains inhibitory molecules,which lead to repair failure.Accordingly,blocking the activity of these inhibitory factors in the extracellular matrix should lead to more successful remyelination.In the central nervous system,oligodendrocytes form the myelin sheath.We performed primary cell culture and found that a natural increase in fibronectin promoted the proliferation of oligodendrocyte progenitors during the initial stage of remyelination while inhibiting oligodendrocyte differentiation.Poly-L-ornithine blocked these inhibitory effects without compromising fibronectin’s pro-proliferation function.Experiments showed that poly-L-ornithine activated the Erk1/2 signaling pathway that is necessary in the early stages of differentiation,as well as PI3K signaling pathways that are needed in the mid-late stages.When poly-L-ornithine was tested in a lysolecithin-induced animal model of focal demyelination,it enhanced myelin regeneration and promoted motor function recovery.These findings suggest that poly-L-ornithine has the potential to be a treatment option for clinical myelin sheath injury. 展开更多

本文研究除痰解毒方(Chutan Jiedu Decoction,CJD)联合吉非替尼对肺腺癌H1975荷瘤小鼠移植瘤twist、fibronectin表达的影响,从上皮间质转化(Epithelial-mesenchymal transition,EMT)角度探讨其抗肿瘤作用机制。建立人肺腺癌H1975耐药细... 本文研究除痰解毒方(Chutan Jiedu Decoction,CJD)联合吉非替尼对肺腺癌H1975荷瘤小鼠移植瘤twist、fibronectin表达的影响,从上皮间质转化(Epithelial-mesenchymal transition,EMT)角度探讨其抗肿瘤作用机制。建立人肺腺癌H1975耐药细胞荷瘤裸鼠模型,随机分为模型组,吉非替尼组,除痰解毒方低、中、高剂量组,除痰解毒方中剂量联合吉非替尼组,每组10只,各组予以相应的药物灌胃2周。检测瘤体积和瘤质量,计算抑瘤率,采用免疫组织化学法、Western blot法、荧光定量PCR法检测各组肿瘤组织twist、fibronectin蛋白及基因的表达。结果显示,联合用药组抑瘤率为61.92%,明显高于模型组、吉非替尼组,除痰解毒方低、中、高剂量组(P<0.01)。免疫组织化学法、Western blot法及荧光定量PCR法结果均显示除痰解毒方中剂量、高剂量及联合用药组均能下调twist和fibronectin的表达,且联合用药组下调作用高于单独用药组(P<0.01)。研究结果表明除痰解毒方联合吉非替尼对肺腺癌H1975荷瘤小鼠移植瘤生长及twist、fibronectin表达的抑制作用优于除痰解毒方单药及吉非替尼组,其协同增效作用可能与逆转EMT增强吉非替尼的敏感性有关。 展开更多

目的:研究fibronectin 1(FN1)在甲状腺癌的表达情况,探讨FN1与临床参数的关系,预测其在肿瘤发生发展中的可能机制。方法:从癌症基因组图谱(TCGA)数据库下载甲状腺癌中FN1的RNA SEqV2资料和临床信息,分别采用SPSS进行数据统计比较甲状腺... 目的:研究fibronectin 1(FN1)在甲状腺癌的表达情况,探讨FN1与临床参数的关系,预测其在肿瘤发生发展中的可能机制。方法:从癌症基因组图谱(TCGA)数据库下载甲状腺癌中FN1的RNA SEqV2资料和临床信息,分别采用SPSS进行数据统计比较甲状腺癌组织和非肿瘤甲状腺组织中FN1的表达,利用linkedomics在线分析系统总结fibronectin 1与临床病理参数的关系,使用Kaplan-Meier Plotter进行无复发生存期比较,通过linkedomics在线系统中的基因富集分析(GSEA)分析预测可能相关的信号通路。结果:FN1在甲状腺癌组织中的mRNA表达显著增多(16.663 0±0.378 6 vs 12.182 4±0.139 6,P <0.000 1;16.724 4±0.124 1 vs 12.182 4±0.139 6,P <0.000 1),在乳头状甲状腺癌中的表达偏高( P =0.197×10^-33 ),随T分期增加表达逐渐升高( P =3.321×10^-8 ),在淋巴结转移N 1中表达显著增高( P =9.963×10^-16 ),随病理分级增加表达升高( P =2.780×10^-9 )和随残余增多时表达量升高( P =7.481×10^-3 )。高表达FN1的患者无复发生存期显著短于低表达患者(HR 高表达=2;P HR =0.026<0.05)。FN1高表达样本富集了NF-κB通路,肿瘤坏死因子(TNF)信号通路中激活的PI3K/AKT途径,细胞黏附分子(cell adhesion molecules)中激活的Claudin-1-Notch pathway-EMT途径和甲状腺激素合成(thyroid hormone synthesis)中抑制的PAX8/PPARγ途径等基因集。验证实验结果发现甲状腺癌的肿瘤基质中FN1表达、mRNA转录水平和蛋白表达显著升高。结论:甲状腺癌中FN1上调表达,符合肿瘤研究的相关通路,可以作为预测淋巴结转移和判断预后的重要因子。 展开更多

目的通过Fibronectin介导进行差别黏附筛选软骨终板干细胞,观察筛选的效果及其初步生物学性能。方法从脊柱融合术中获取椎间盘软骨终板标本,机械-酶消化法获取原代软骨终板细胞,体外扩增至第2代后接种于Fibronectin包被过的培养瓶,孵育2... 目的通过Fibronectin介导进行差别黏附筛选软骨终板干细胞,观察筛选的效果及其初步生物学性能。方法从脊柱融合术中获取椎间盘软骨终板标本,机械-酶消化法获取原代软骨终板细胞,体外扩增至第2代后接种于Fibronectin包被过的培养瓶,孵育20 min后将未贴壁细胞及培养液转移至新培养瓶再孵育40 min,吸弃未贴壁细胞及培养液,所得2瓶细胞进行体外扩增,流式细胞仪检测细胞表面标志物。对筛选后的细胞进行向多系细胞诱导分化。结果第2代细胞筛选前,细胞形态个体差异较大,呈三角形或多角形,融合时呈铺路石样外观,筛选后细胞形态比较均匀,形成细胞克隆群。流式细胞仪检测发现:经Fibronectin筛选后所获软骨终板干细胞CD90、CD73表达阳性率>95%,CD105表达阳性率>85%;经诱导能向骨、软骨及脂肪细胞方向分化。结论 Fibronectin筛选所得的软骨终板细胞基本符合国际细胞治疗协会对间充质干细胞的定义标准。Fibronectin差别黏附筛选法得够有效筛选、纯化软骨终板干细胞。 展开更多

Many studies have shown that fibronectin type III domain-containing protein 5(FDNC5) and brain-derived neurotrophic factor(BDNF) play vital roles in plasticity after brain injury. An enriched environment refers to an environment that provides animals with multi-sensory stimulation and movement opportunities. An enriched environment has been shown to promote the regeneration of nerve cells, synapses, and blood vessels in the animal brain after cerebral ischemia;however, the exact mechanisms have not been clarified. This study aimed to determine whether an enriched environment could improve neurobehavioral functions after the experimental inducement of cerebral ischemia and whether neurobehavioral outcomes were associated with the expression of FDNC5 and BDNF. This study established ischemic mouse models using permanent middle cerebral artery occlusion(pMCAO) on the left side. On postoperative day 1, the mice were randomly assigned to either enriched environment or standard housing condition groups. Mice in the standard housing condition group were housed and fed under standard conditions. Mice in the enriched environment group were housed in a large cage, containing various toys, and fed with a standard diet. Sham-operated mice received the same procedure, but without artery occlusion, and were housed and fed under standard conditions. On postoperative days 7 and 14, a beam-walking test was used to assess coordination, balance, and spatial learning. On postoperative days 16–20, a Morris water maze test was used to assess spatial learning and memory. On postoperative day 15, the expression levels of FDNC5 and BDNF proteins in the ipsilateral cerebral cortex were analyzed by western blot assay. The results showed that compared with the standard housing condition group, the motor balance and coordination functions(based on beam-walking test scores 7 and 14 days after operation), spatial learning abilities(based on the spatial learning scores from the Morris water maze test 16–19 days after operation), and memory abilities(based on the memory scores of the Morris water maze test 20 days after operation) of the enriched environment group improved significantly. In addition, the expression levels of FDNC5 and BDNF proteins in the ipsilateral cerebral cortex increased in the enriched environment group compared with those in the standard housing condition group. Furthermore, the Pearson correlation coefficient showed that neurobehavioral functions were positively associated with the expression levels of FDNC5 and BDNF(r = 0.587 and r = 0.840, respectively). These findings suggest that an enriched environment upregulates FDNC5 protein expression in the ipsilateral cerebral cortex after cerebral ischemia, which then activates BDNF protein expression, improving neurological function. BDNF protein expression was positively correlated with improved neurological function. 本文用近交克隆系(Close cloned)大鼠研究了内源性Laminin和Fibronectin对周围神经的再生轴突及非神经元的雪旺氏细胞、成纤维细胞等的作用和影响。将从供体鼠获得的坐骨神中段经冷冻、加热处理后再用Laminin或Fibronectin抗血清处理;... 本文用近交克隆系(Close cloned)大鼠研究了内源性Laminin和Fibronectin对周围神经的再生轴突及非神经元的雪旺氏细胞、成纤维细胞等的作用和影响。将从供体鼠获得的坐骨神中段经冷冻、加热处理后再用Laminin或Fibronectin抗血清处理;对照组则用正常兔血清处理。将处理后的神经段(10mm)分别植入三组受体鼠体内,术后不同时期取材,电镜观察。术后15天,抗Laminin组和抗Fibronectin组的轴突总数,只有对照组的50%左右。对照组和抗Fibronectin组约90%的轴突走在基底膜管内,而抗Laminin组,再生轴突似不能识别基底膜而生长在基底膜管外。轴突生长总是先于雪旺氏细胞的迁移,而后雪旺氏细胞才生长粘附并包绕轴突。成纤维细胞能够识别伴随有雪旺氏细胞的轴突,并形成神经周膜包绕这些轴突,但它们不能识别空陷的基底膜管,只有当组织中缺乏Fibronectin时,增生的成纤维细胞方在基底膜管外形成神经周膜。在缺乏Laminin的神经段内,巨噬细胞不仅大量增生,还有包随走在基底膜管外单个轴突的趋向。这些结果提示在神经再生的早期,内源性Laminin和Fibronectin不但调节再生神经纤维的生长,对在神经损伤和再生中起重要作用的巨噬细胞和成纤维细胞也有积极的影响。 展开更多

Objective: To observe the expression and significance of fibronectin and metalloproteinase-3(MMP-3) in patients with ankylosing spondylitis(AS). Methods: A total of 30 AS patients in our hospital and 30 healthy volunteers were selected in our study. Fibronectin and MMP-3 were measured and compared between these two groups. The AS group received sulfasalazine2 g daily for 3 months. Bath ankylosing spondylitis disease activity index, bath ankylosing spondylitis functional index, bath ankylosing spondylitis metrology index, erythrocyte sedimentation rate and C-reactive protein were compared before and after treatment.Pearson's linear-correlation analysis was used to determine relationships between parameters.Results: Totally 28 patients in the AS group completed the study. Fibronectin and MMP-3 in peripheral blood of AS patients were evidently higher than that in the normal control group(P<0.05). After treated by sulfasalazine, the level of expressing Fibronectin and MMP-3 significantly decreased compared with baseline values(P<0.05). Pearson's linear-correlation analysis showed that serum fibronectin and MMP-3 level had a positive correlation with bath ankylosing spondylitis disease activity index global assessment, spine pain, night pain, general pain, erythrocyte sedimentation rate and C-reactive protein(P<0.05). Conclusions: The expression of fibronectin and MMP-3 in AS patients were significantly higher than that in the normal control group, and they all decreased significantly after treatment. It indicated that both fibronectin and MMP-3 were correlated closely with the onset of AS. 展开更多

The adsorption behavior of heparin and fibronectin was studied by quartz crystal microbalance with dissipation(QCM-D), and the interaction between heparin and fibronectin was evaluated using immunochemistry and isothermal titration calorimetry(ITC) measurement. The results showed that there was competitive adsorption between heparin and fibronectin, and the preadsorption of fibronectin could prevent subsequent heparin adsorption to some extent, and the adsorbed Hep/Fn complex on the surface was in a rigid form. The bioactivity of heparin and fibronectin could be affected by the bulk concentration of each, and both heparin and fibronectin in Hep/Fn complex formed under p H 4 condition displayed larger bioactivity than that formed under p H 7 condition. Moreover, the fibronectin showed more exposed cell-binding sites at the p H value lower than physiological condition. The results of ITC further suggested that the interaction between heparin and fibronectin under p H 4 was stronger than under p H 7, and the complex was also more stable. The study brings forth the detailed interaction between heparin and fibronectin, which will be helpful for better understanding the interaction mechanism of the two biomolecules. The results may be potentially useful for the development of new generation of cardiovascular biomaterials. 展开更多

The cDNAs in plasmids code for two polypeptides, CH62 (Pro1239-Ser1515 of FN linked with Ala1690 -Val2049 through Met) and CH63 (CH62 without Ile1850-Glu1978). The expression level of CH62 in E. coli was very low, but that of CH63 was very high.The results suggests that Asp1961-Glu1978 in FN is a key sequence influencing the expression of triple-domain polypeptide in E. Coli. After being dissolved and renatured, CH63 can be purified by heparin-agarose affinity chromatography.Both of the cell-binding domains in the recombinant polypeptide were functional.The production of CH63 provides a fundamental basis for further study of recombinant products with better anti-metastasis function. 展开更多

We prepared an anti-metastatic polypeptide, recombinant fibronectinpolypeptide CH50, and finished the preliminary identification of its functions. In this paper, we studied the effect of this polypeptide on the function of macrophages. CH50 can significantly augment the production of nitric oxide(NO) by macrophages in a dose-dependent manner. The continuous presence of CH50 had a much stronger effect. In the presence of CH50, the cytotoxicity of macrophages to melanoma B16/F1 cells was significantly enhanced, and a stronger effect was obtained if CH50 was present continuously. CH50 polypeptide and IFN-r have a synergistic effect on the production of NO by macrophages and the cytotoxicity of macrophages on tumor cells. In the in vivo experiments, CH50 can inhibit the growth of tumor cells, and have a better effect in the presence of IFN-r. Our results suggest that recombinant fibronectin polypeptide CH50 has two functions: one is to inhibit the metastasis of tumor cells, and the other one is to augment the function of macrophages. And this polypeptide will be potentially useful in tumor therapy. 展开更多
