Thousands of long-distance mobile mRNAs were identified from different grafting systems,based on high-throughput sequencing technology.Moreover,the long-distance delivery of RNAs was proved to involve multiple mechani...Thousands of long-distance mobile mRNAs were identified from different grafting systems,based on high-throughput sequencing technology.Moreover,the long-distance delivery of RNAs was proved to involve multiple mechanisms.Here,we analyzed the homology,motif,and tRNA-like structure(TLS)of long-distance mobile mRNAs identified by RNA-seq as well as the RNA-binding protein(RBP)in nine grafting combinations including Arabidopsis thaliana,Vitis vinifera,Cucumis sativus,Citrullus lanatus,Nicotiana benthamiana,Malus domestica,Pyrus spp.,Glycine max and Phaseolus vulgaris.Although several mRNAs were found to be shared in herbaceous,woody,and related species,the vast majority of long-distance mobile mRNAs were species-specific.Four non-specific movement-related motifs were identified,while the TLS was not necessary for mRNA long distance mobility.In addition,we found that RBPs were conserved among herbaceous and woody plants as well as related species.This paper reports a further in-depth analysis of the endogenous mechanisms by which the species-specific transportable m RNAs were selected by bioinformatics,in order to provide insights for future research on long-distance mobile mRNAs.展开更多
For a water supply system with long-distance diversion pipelines, in addition to the water hammer problems that occur beyond pumps, the safety of the water diversion pipeline in front of pumps also deserves attention....For a water supply system with long-distance diversion pipelines, in addition to the water hammer problems that occur beyond pumps, the safety of the water diversion pipeline in front of pumps also deserves attention. In this study, a water hammer protection scheme combined with an overflow surge tank and a regulating valve was developed. A mathematical model of the overflow surge tank was developed, and an analytical formula for the height of the overflow surge tank was derived. Furthermore, a practical water supply project was used to evaluate the feasibility of the combined protection scheme and analyze the sensitivity of valve regulation rules. The results showed that the combined protection scheme effectively reduced the height of the surge tank, lessened the difficulties related to construction, and reduced the necessary financial investment for the project. The two-stage closing rule articulated as fast first and then slow could minimize the overflow volume of the surge tank when the power failure occurred, while the two-stage opening rule articulated as slow first and then fast could be more conducive to the safety of the water supply system when the pump started up.展开更多

Invasion of the intertidal mudflats by Spartina alterniflora is an ideal example of stratified diffusion, involving both long-distance dispersal of seeds and local diffusion due to clonal growth. In conjunction with experimental data on range radius-versus-time curve, a traveling wave equation-based model is used to investigate the sensitivity of the spread rate of exotic S. alterniflora to parameters of long distance dispersal (c, maximum colonial establishment rate) and local colony diffusion (r, intrinsic growth rate) at two tidal marshes, the Eastern Chongming and the Jiuduansha Islands, at the Yangtze River estuary. Both Eastern Chong ming and Jiuduansha Islands are now national natural reserves in China, which were established in 2005. However, the mudflats and salt marshes in the two reserves are now heavily infested with introduced S. alterniflora, which may threaten the estuarine ecosystems and their biodiversity. S. alterniflora was first found in 1995 on Chongming. For rapid sediment accretion in mudflats in the estuary, S. alterniflora was also intentionally introduced to Jiuduansha in 1997 and Chongming in 2001, which has led to a rapid range expansion in the estuary. Our results show that range expansion of species with stratified diffusion is affected by both long-distance dispersal and local colony diffusion, and that there is a critical c*, below which the spread rate is more influenced by long-distance dispersal than by local diffusion. After applying this model to the invasion of S. alterniflora in the Yangtze River estuary, we derive that c = 1.7 × 10-3, c* = 0.126 and c = 4.8 × 10-3 km-2·yr-1, c* = 0.140 km-2·yr-1 at Chongming and Jiuduansha (Shanghai), respectively. Our results suggest that the range spread of S. alterniflora in the Yangtze River estuary is more influenced by long-distance dispersal than local colony diffusion, and that S. alterniflora generates about 1.7 × 10-3 to 4.8 × 10-3 colonies per square kilometers per year. <b><span>Background</span></b><span><span>: Often, long-distance truck drivers’ (LDTDs’) work predisposes them to sexually transmitted infections (STIs) whose outcomes are influen...<b><span>Background</span></b><span><span>: Often, long-distance truck drivers’ (LDTDs’) work predisposes them to sexually transmitted infections (STIs) whose outcomes are influenced by access and behavior of seeking sexual health care. </span><b></b></span><b><b><span>Methodology</span></b><span>:</span></b><span> In this study, we assessed the utilization of HIV/STI preventive services and associated factors among 296 LDTDs operating along the northern corridor highway using an interviewer-administered questionnaire for data collection at Mlolongo stopover in Machakos, Kenya. Responses for the investigated variables, including condom use, history of HIV testing, frequency of HIV testing, antiretroviral therapy (ART) use and follow-up for the HIV positive and STI treatment, were assigned a score of either 1 or 0 depending on the question’s dimension. Following summing up for each participant, we computed a weighted score ranging between 0 and 1 by dividing the summed responses by the number of eligible variables. We arbitrarily multiplied these scores by 8 to generate endpoint scores ranging from one to eight for each participant to help create a dichotomized outcome variable for utilization levels: limited utilization (1 to 4) and good utilization (5 to 8). Association between certain independent variables and the outcome variable (level of utilization of H.I.V./STIs preventive services) w</span><span>as</span><span style="font-family:;" "=""><span><span> analyzed using binomial logistic regression analysis in R statistical software. </span><b></b></span><b><b><span>Results</span></b><span>:</span></b><span> The mean age of the LDTDs was 38.4 years, ranging from 24 - 57 years. The majority (n = 287, 97%) of the LDTDs had been tested on HIV at least once since the beginning of their career. Only 4.9% of the LDTDs had been tested on HIV within the previous three months. Of the 175 LDTDs who reported a history of STI, most (n = 173, 98.9%) of them had sought treatment. Condom use rates were higher (97.1%) among the LDTDs who had sexual interactions with casual sexual partners compared to 47.2% among regular sexual partners. Analyses classed most of the respondents (n = 231, 78.0%) as having good utilization, while the rest (22%) had limited utilization. Cytokinins(CKs),primarily trans-zeatin(tZ)and isopentenyladenine(iP)types,play critical roles in plant growth,development,and various stress responses.Long-distance transport of tZ-type CKs meidated by Arabidopsis ATP...Cytokinins(CKs),primarily trans-zeatin(tZ)and isopentenyladenine(iP)types,play critical roles in plant growth,development,and various stress responses.Long-distance transport of tZ-type CKs meidated by Arabidopsis ATP-binding cassette transporter subfamily G14(AtABCG14)has been well studied;however,less is known about the biochemical properties of AtABCG14 and its transporter activity toward iP-type CKs.Here we reveal the biochemical properties of AtABCG14 and provide evidence that it is also required for long-distance transport of iP-type CKs.AtABCG14 formed homodimers in human(Homo sapiens)HEK293T,tobacco(Nicotiana tabacum),and Arabidopsis cells.Transporter activity assays of AtABCG14 in Arabidopsis,tobacco,and yeast(Saccharomyces cerevisiae)showed that AtABCG14 may directly transport multiple CKs,including iP-and tZ-type species.AtABCG14 expression was induced by iP in a tZ-type CKdeficient double mutant(cypDM)of CYP735A1 and CYP735A2.The atabcg14 cypDM triple mutant exhibited stronger CK-deficiency phenotypes than cypDM.Hormone profiling,reciprocal grafting,and 2H6-iP isotope tracer experiments showed that root-to-shoot and shoot-to-root long-distance transport of iP-type CKs were suppressed in atabcg14 cypDM and atabcg14.These results suggest that AtABCG14 participates in three steps of the circular long-distance transport of iP-type CKs:xylem loading in the root for shootward transport,phloem unloading in the shoot for shoot distribution,and phloemunloading in the root for root distribution.We found that AtABCG14 displays transporter activity toward multiple CK species and revealed its versatile roles in circular long-distance transport of iP-type CKs.These findings provide newinsights into the transport mechanisms of CKs and other plant hormones.展开更多
Plants program their meristem-associated developmental switches for timely adaptation to a changing environment.Potato(Solanum tuberosum L.)tubers differentiate from specialized belowground branches or stolons through...Plants program their meristem-associated developmental switches for timely adaptation to a changing environment.Potato(Solanum tuberosum L.)tubers differentiate from specialized belowground branches or stolons through radial expansion of their terminal ends.During this process,the stolon apex and closest axillary buds enter a dormancy state that leads to tuber eyes,which are reactivated the following spring and generate a clonally identical plant.The potato FLOWERING LOCUS T homolog SELF-PRUNING 6A(StSP6A)was previously identified as the major tuber-inducing signal that integrates day-length cues to control the storage switch.However,whether some other long-range signals also act as tuber organogenesis stimuli remains unknown.Here,we show that the florigen SELF PRUNING 3D(StSP3D)and FLOWERING LOCUS T-like 1(StFTL1)genes are activated by short days,analogously to StSP6A.Overexpression of StSP3D or StFTL1 promotes tuber formation under non-inductive long days,and the tuber-inducing activity of these proteins is graft transmissible.Using the non-tuber-bearing wild species Solanum etuberosum,a natural SP6A null mutant,we show that leaf-expressed SP6A is dispensable for StSP3D long-range activity.StSP3D and StFTL1 mediate secondary activation of StSP6A in stolon tips,leading to amplification of this tuberigen signal.StSP3D and StFTL1 were observed to bind the same protein partners as StSP6A,suggesting that they can also form transcriptionally active complexes.Together,our findings show that additional mobile tuber-inducing signals are regulated by the photoperiodic pathway.展开更多
Mountain road tunnels are prone to water leakage and lining corrosion under the complex geological conditions and corrosive envi-ronments,which will reduce the strength of the lining structure until it loses its load-...Mountain road tunnels are prone to water leakage and lining corrosion under the complex geological conditions and corrosive envi-ronments,which will reduce the strength of the lining structure until it loses its load-bearing capacity;eventually,the definitive lining will need to be replaced.In this paper,a highway tunnel in a mountainous area in Southwest China is taken as an example.Field investi-gation found that the tunnel was seriously corroded by sulfate,the strength of the definitive lining decreased,and large-scale cracks and spalling appeared on the surface,so the operator decided to replace the definitive lining by the method of interval replacement.Based on the data obtained from drilling and coring,a numerical model of long-distance replacement of the definitive lining of the damaged tunnel is established.First,the back analysis of the calculation parameters is carried out,and the modified calculation results are com-pared with the field monitoring results for verification.Then,the deformation trend of the tunnel and the development of the plastic zone during the process of long-distance replacement of the definitive lining are studied.Finally,the construction scheme is optimized.Numer-ical analysis results show that the replacement of the definitive lining of the tunnel mainly leads to the settlement of the arch crown and the uplift of the inverted arch.The deformation of the tunnel shows two rapid growth stages and two stable stages during the replace-ment process;after replacement,the deformation of the arch crown and the inverted arch is divided into two buffer zones and one stable zone.In the progress of the replacement of the definitive lining,the plastic zone does not change.Regarding the reinforcement measures,with the increase in the grouting range,the grouting efficiency decreases,and the effect of the temporary steel arch on controlling the overall deformation is not obvious.The length of the replacement of the single section should be determined according to the geological conditions of the replacement section and the monitoring data during construction.The research results can provide a reference for sim-ilar projects for the replacement of the definitive lining.展开更多

文摘Cytokinins(CKs),primarily trans-zeatin(tZ)and isopentenyladenine(iP)types,play critical roles in plant growth,development,and various stress responses.Long-distance transport of tZ-type CKs meidated by Arabidopsis ATP-binding cassette transporter subfamily G14(AtABCG14)has been well studied;however,less is known about the biochemical properties of AtABCG14 and its transporter activity toward iP-type CKs.Here we reveal the biochemical properties of AtABCG14 and provide evidence that it is also required for long-distance transport of iP-type CKs.AtABCG14 formed homodimers in human(Homo sapiens)HEK293T,tobacco(Nicotiana tabacum),and Arabidopsis cells.Transporter activity assays of AtABCG14 in Arabidopsis,tobacco,and yeast(Saccharomyces cerevisiae)showed that AtABCG14 may directly transport multiple CKs,including iP-and tZ-type species.AtABCG14 expression was induced by iP in a tZ-type CKdeficient double mutant(cypDM)of CYP735A1 and CYP735A2.The atabcg14 cypDM triple mutant exhibited stronger CK-deficiency phenotypes than cypDM.Hormone profiling,reciprocal grafting,and 2H6-iP isotope tracer experiments showed that root-to-shoot and shoot-to-root long-distance transport of iP-type CKs were suppressed in atabcg14 cypDM and atabcg14.These results suggest that AtABCG14 participates in three steps of the circular long-distance transport of iP-type CKs:xylem loading in the root for shootward transport,phloem unloading in the shoot for shoot distribution,and phloemunloading in the root for root distribution.We found that AtABCG14 displays transporter activity toward multiple CK species and revealed its versatile roles in circular long-distance transport of iP-type CKs.These findings provide newinsights into the transport mechanisms of CKs and other plant hormones.

文摘Plants program their meristem-associated developmental switches for timely adaptation to a changing environment.Potato(Solanum tuberosum L.)tubers differentiate from specialized belowground branches or stolons through radial expansion of their terminal ends.During this process,the stolon apex and closest axillary buds enter a dormancy state that leads to tuber eyes,which are reactivated the following spring and generate a clonally identical plant.The potato FLOWERING LOCUS T homolog SELF-PRUNING 6A(StSP6A)was previously identified as the major tuber-inducing signal that integrates day-length cues to control the storage switch.However,whether some other long-range signals also act as tuber organogenesis stimuli remains unknown.Here,we show that the florigen SELF PRUNING 3D(StSP3D)and FLOWERING LOCUS T-like 1(StFTL1)genes are activated by short days,analogously to StSP6A.Overexpression of StSP3D or StFTL1 promotes tuber formation under non-inductive long days,and the tuber-inducing activity of these proteins is graft transmissible.Using the non-tuber-bearing wild species Solanum etuberosum,a natural SP6A null mutant,we show that leaf-expressed SP6A is dispensable for StSP3D long-range activity.StSP3D and StFTL1 mediate secondary activation of StSP6A in stolon tips,leading to amplification of this tuberigen signal.StSP3D and StFTL1 were observed to bind the same protein partners as StSP6A,suggesting that they can also form transcriptionally active complexes.Together,our findings show that additional mobile tuber-inducing signals are regulated by the photoperiodic pathway.

文摘Mountain road tunnels are prone to water leakage and lining corrosion under the complex geological conditions and corrosive envi-ronments,which will reduce the strength of the lining structure until it loses its load-bearing capacity;eventually,the definitive lining will need to be replaced.In this paper,a highway tunnel in a mountainous area in Southwest China is taken as an example.Field investi-gation found that the tunnel was seriously corroded by sulfate,the strength of the definitive lining decreased,and large-scale cracks and spalling appeared on the surface,so the operator decided to replace the definitive lining by the method of interval replacement.Based on the data obtained from drilling and coring,a numerical model of long-distance replacement of the definitive lining of the damaged tunnel is established.First,the back analysis of the calculation parameters is carried out,and the modified calculation results are com-pared with the field monitoring results for verification.Then,the deformation trend of the tunnel and the development of the plastic zone during the process of long-distance replacement of the definitive lining are studied.Finally,the construction scheme is optimized.Numer-ical analysis results show that the replacement of the definitive lining of the tunnel mainly leads to the settlement of the arch crown and the uplift of the inverted arch.The deformation of the tunnel shows two rapid growth stages and two stable stages during the replace-ment process;after replacement,the deformation of the arch crown and the inverted arch is divided into two buffer zones and one stable zone.In the progress of the replacement of the definitive lining,the plastic zone does not change.Regarding the reinforcement measures,with the increase in the grouting range,the grouting efficiency decreases,and the effect of the temporary steel arch on controlling the overall deformation is not obvious.The length of the replacement of the single section should be determined according to the geological conditions of the replacement section and the monitoring data during construction.The research results can provide a reference for sim-ilar projects for the replacement of the definitive lining.
