Radon and its decay daughter products are the most widespread source of such irradiation. Miners (in both uranium and non-uranium mines) as well as laypeople in domestic life may be exposed to radon, making the problem of assessing the cytogenetic effects of exposure extremely crucial. One of the more promising test systems to assess the effect of radon is the cytokinesis-block micronucleus assay (CBMN) on peripheral blood lymphocytes, which has a number of advantages over other cytogenetic techniques. Recent progress and future prospects of this cytogenetic method are discussed here.展开更多

Results reveal that the cytokinesis-block micronuclei (CBMN) and conventional cultured micronuclei in peripheral blood lymphocytes, serum levels of lipid peroxide, superoxide dismutase, and the total antioxidation capacity by chemical colorimetry all increased significantly in aircrew members. There exists a linear relationship between the CBMN and the average annual effective doses of radiation received or the average annual flying hours. With both of them, a trend shows that the serum lipid peroxide levels increase as well. Either the lipid peroxide or CBMN can sensitively reflect the recent changes in flight load. These findings indicate that cosmic radiation impairs the stability of chromosomes and genome, and induces lipid oxidative damage in aircrews; Lymphocyte CBMN and serum lipid peroxide can be used as monitoring indicators in the cosmic radiation protection for aircrew members.展开更多

KM mice were exposed to test compounds with different doses of 0.1, 0.2 and 0.4mg/kg through intraperitoneal injection two times with an internal of 24 h, and then killed by cervical dislocation 6 h after the second injection. In SCGE assay, isolated mice peripheral blood lymphocytes were employed to determine DNA damaging degree after a 1 h treatment by test compounds and a following electrophoresis. Carbofuran and carbofuranphenol showed negative results in both test and had no obvious toxicity. 3-hydrocarbofuran and nitrosocarbofuran were positive.3-ketocarbofuran could not induce micronucleus formation but caused significant DNA migration in SCGE test. These tests revealed that 3-ketocarbofuran, 3-hydrocarbofuran and nitrosocarbofuran are potential mutagesis and further research is needed.展开更多

The experiments were performed on mouse bone marrow cells. THH showed similar gentoxic effects to colchicine (COL) in CM, MN and CA analyses; positive CM effects were observed accompanied with increases of mitotic index and frequencies of C-mltotic cells as well as decreased frequencies of anaphase in all of the THH-treated groups. The compound showed a positive MN response in bone marrow polychromatic erythrocytes but was negative in CA analyses. The preliminary results suggested that THH is an aneuploldy inducer in mouse bone marrow cells under present experiment conditions.展开更多
Objective:To identify the available phytochemicals and carotenoids in the selected green algae and evaluate the potential genotoxic/antigenotoxic effect using lymphocytes.Methods:Organic,solvent extracts of Chlorococc...Objective:To identify the available phytochemicals and carotenoids in the selected green algae and evaluate the potential genotoxic/antigenotoxic effect using lymphocytes.Methods:Organic,solvent extracts of Chlorococcum humicola(C.humicola)were used for the phytochemical analysis.The available carotenoids were assessed by HPLC,and LC-MS analysis.The genotoxicity was induced by the benzo(a)pyrene in the lymphocyte culture,the genotoxic and antigenotoxic effects of algal carotenoids with and without genotoxic inducer were evaluated by chromosomal aberration(CA),sister chromatid exchange(SCE)and micronucleus assay(MN).Results:The results of the analysis showed that the algae were rich in carotenoids and fatty acids.In the total carotenoids lutein,β-carotene andα-carotene were found to be present in higher concentration.The frequency of CA and SCE increased by benzo(a)pyrene were significantly decreased by the carotenoids(P<0.05 for CA,P<0.001 for SCE).The MN frequencies of the cells were significantly decreased by the treatment with carotenoids when compared with the positive controls(P<0.05).Conclusions:The findings of the present study demonstrate that,the green algae C.humicola is a rich source of bioactive compounds especially carotenoids which effectively fight against environmental genotoxic agents,the carotenoids itself is not a genotoxic substance and should be further considered for its beneficial effects.展开更多
The radiation is considered as a double edged sword, as its beneficial and detrimental effects have been demonstrated. The potential benefits are being exploited to its maximum by adopting safe handling of radionuclid...The radiation is considered as a double edged sword, as its beneficial and detrimental effects have been demonstrated. The potential benefits are being exploited to its maximum by adopting safe handling of radionuclide stipulated by the regulatory agencies. While the occupational workers are monitored by personnel monitoring devices, for general publics, it is not a regular practice. However, it can be achieved by using biomarkers with a potential for the radiation triage and medical management. An ideal biomarker to adopt in those situations should be rapid, specific, sensitive, reproducible, and able to categorize the nature of exposure and could provide a reliable dose estimation irrespective of the time of the exposures. Since cytogenetic markers shown to have many advantages relatively than other markers, the origins of various chromosomal abnormalities induced by ionizing radiations along with dose-response curves generated in the laboratory are presented. Current status of the gold standard dicentric chromosome assay, micronucleus assay, translocation measurement by fluorescence insitu hybridization and an emerging protein marker the g-H2 AX assay are discussed with our laboratory data. With the wide choice of methods, an appropriate assay can be employed based on the net.展开更多
[Objectives]This study was conducted to analyze the genetic toxicity of Solidago decurrens,with a view to scientifically and safely using S.decurrens.[Methods]Using commercially available S.decurrens as an experimenta...[Objectives]This study was conducted to analyze the genetic toxicity of Solidago decurrens,with a view to scientifically and safely using S.decurrens.[Methods]Using commercially available S.decurrens as an experimental material,according to Vicia faba root tip micronucleus technique,V.faba root tips were treated with different doses of aqueous extract from S.decurrens for different times,and the mitotic index,frequency of micronucleus and chromosomal aberration of root tips in V.faba were detected,so as to reveal the allelopathic potential and the potential ecological risks of S.decurrens at the cellular level.[Results]The decrease in the mitotic index caused by the aqueous extract of S.decurrens was positively related to the concentration of the aqueous extract and the treatment time.The aqueous extract of S.decurrens increased the frequency of micronucleus in root tip cells of V.faba,and the increase was positively related to the concentration of the aqueous extract and the treatment time.The aqueous extract of S.decurrens caused abnormal phenomena such as chromosome bridges,chromosome fragmentation,and chromosome lag in V.faba root tip cells,and the effect of the aqueous extract on V.faba root tips had significant effects dependent on time and concentration.Above results indicate that the aqueous extract of S.decurrens inhibited and damaged the mitosis of V.faba root tip cells to some degrees,had certain genetic toxicity and had the ability to rapidly spread.[Conclusions]This study provides a theoretical basis for scientifically and rationally using S.decurrens and preventing the spread of S.decurrens.展开更多
AIM:To evaluate the occurrence of micronucleus(MN),nucleoplasmic bridges(NPBs)and nuclear buds(NBUDs)in the mitogen-stimulated lymphocytes of patients with non-alcoholic steatohepatitis(NASH).METHODS:The study was per...AIM:To evaluate the occurrence of micronucleus(MN),nucleoplasmic bridges(NPBs)and nuclear buds(NBUDs)in the mitogen-stimulated lymphocytes of patients with non-alcoholic steatohepatitis(NASH).METHODS:The study was performed in 25(9 females,16 males)patients newly diagnosed with NASH,and 25healthy subjects of similar ages and genders were used as a control group.None of the controls was known to be receiving any drugs for medical or other reasons or using alcohol.Hepatosteatosis was further excluded by abdominal ultrasound imaging in the control group.The numbers of MN,NPBs and NBUDs scored in binucleated(BN)cells were obtained from the mitogen-stimulated lymphocytes of patients and control subjects.Statistical comparisons of the numbers of BN cells with MN,NPBs and NBUDs and ages between the patients with NASH and control subjects were performed.RESULTS:The mean ages of the patients and the control group were 41.92±13.33 and 41.80±13.09 years(P>0.05),respectively.The values of the mean body mass index(BMI),HOMA-IR,hemoglobin,creatinin,aspartate aminotransferase,alanine aminotransferase,triglyceride,high density lipoprotein,and low density lipoprotein were 31.19±4.62 kg/m2vs 25.07±4.14 kg/m2,6.71±4.68 vs 1.40±0.53,14.73±1.49 g/dL vs 14.64±1.30 g/dL,0.74±0.15 mg/dL vs 0.80±0.13 mg/dL,56.08±29.11 U/L vs 16.88±3.33 U/L,92.2±41.43U/L vs 15.88±5.88 U/L,219.21±141.68 mg/dL vs102.56±57.98 mg/dL,16.37±9.65 mg/dL vs 48.72±15.31 mg/dL,and 136.75±30.14 mg/dL vs 114.63±34.13 mg/dL in the patients and control groups,respectively.The total numbers and frequencies of BN cells with MN,NPBs and NBUDs,which were scored using the CBMN cytome assay on PHA-stimulated lymphocytes,were evaluated in the patients with NASH and control group.We found significantly higher numbers of MN,NPBs and NBUDs in the BN cells of patients with NASH than in those of the control subjects(21.60±9.32vs 6.88±3.91;29.28±13.31 vs 7.84±3.96;15.60±5.55 vs 4.20±1.63,respectively,P<0.0001).CONCLUSION:The increased numbers of MN,NPBs and NBUDs observed in the lymphocytes obtained from patients with NASH may reflect genomic instability.展开更多

The 50%-affected dose ofγand UV radiation for the GA rate was 10 Gy and 159 microwatt/cm for 0.5 seconds, respectively.After X-ray exposure,there was no apparent effect on RAW264.7 cells,even with a fortyfold human diagnostic dose.Two exposures toγradiation at 20 Gy resulted in a significantly higher MP rate than 20 Gy single exposure or control(P【0.05).The Thai medicinal plants(Kamin-chun capsules,Curcuma longa Linn;Hed lingeu,Ganoderma lucidum;Ya Pakking capsule,Murdannia loriformis) could not prevent cell damage,but epigallocatechin gallate and L-cysteine could provide protection from 2 Gyγ-ray exposure. Conclusion:γradiation caused chromosomal damage during cell division and UV caused cell death, while X-ray radiation was safe.The radioprotective effects of Thai medicinal plants,Kamin-chun,Hed lingeu, and Ya Pakking,could not prevent cell damage in this study.展开更多

文摘Objective:To identify the available phytochemicals and carotenoids in the selected green algae and evaluate the potential genotoxic/antigenotoxic effect using lymphocytes.Methods:Organic,solvent extracts of Chlorococcum humicola(C.humicola)were used for the phytochemical analysis.The available carotenoids were assessed by HPLC,and LC-MS analysis.The genotoxicity was induced by the benzo(a)pyrene in the lymphocyte culture,the genotoxic and antigenotoxic effects of algal carotenoids with and without genotoxic inducer were evaluated by chromosomal aberration(CA),sister chromatid exchange(SCE)and micronucleus assay(MN).Results:The results of the analysis showed that the algae were rich in carotenoids and fatty acids.In the total carotenoids lutein,β-carotene andα-carotene were found to be present in higher concentration.The frequency of CA and SCE increased by benzo(a)pyrene were significantly decreased by the carotenoids(P<0.05 for CA,P<0.001 for SCE).The MN frequencies of the cells were significantly decreased by the treatment with carotenoids when compared with the positive controls(P<0.05).Conclusions:The findings of the present study demonstrate that,the green algae C.humicola is a rich source of bioactive compounds especially carotenoids which effectively fight against environmental genotoxic agents,the carotenoids itself is not a genotoxic substance and should be further considered for its beneficial effects.

文摘The radiation is considered as a double edged sword, as its beneficial and detrimental effects have been demonstrated. The potential benefits are being exploited to its maximum by adopting safe handling of radionuclide stipulated by the regulatory agencies. While the occupational workers are monitored by personnel monitoring devices, for general publics, it is not a regular practice. However, it can be achieved by using biomarkers with a potential for the radiation triage and medical management. An ideal biomarker to adopt in those situations should be rapid, specific, sensitive, reproducible, and able to categorize the nature of exposure and could provide a reliable dose estimation irrespective of the time of the exposures. Since cytogenetic markers shown to have many advantages relatively than other markers, the origins of various chromosomal abnormalities induced by ionizing radiations along with dose-response curves generated in the laboratory are presented. Current status of the gold standard dicentric chromosome assay, micronucleus assay, translocation measurement by fluorescence insitu hybridization and an emerging protein marker the g-H2 AX assay are discussed with our laboratory data. With the wide choice of methods, an appropriate assay can be employed based on the net.

文摘[Objectives]This study was conducted to analyze the genetic toxicity of Solidago decurrens,with a view to scientifically and safely using S.decurrens.[Methods]Using commercially available S.decurrens as an experimental material,according to Vicia faba root tip micronucleus technique,V.faba root tips were treated with different doses of aqueous extract from S.decurrens for different times,and the mitotic index,frequency of micronucleus and chromosomal aberration of root tips in V.faba were detected,so as to reveal the allelopathic potential and the potential ecological risks of S.decurrens at the cellular level.[Results]The decrease in the mitotic index caused by the aqueous extract of S.decurrens was positively related to the concentration of the aqueous extract and the treatment time.The aqueous extract of S.decurrens increased the frequency of micronucleus in root tip cells of V.faba,and the increase was positively related to the concentration of the aqueous extract and the treatment time.The aqueous extract of S.decurrens caused abnormal phenomena such as chromosome bridges,chromosome fragmentation,and chromosome lag in V.faba root tip cells,and the effect of the aqueous extract on V.faba root tips had significant effects dependent on time and concentration.Above results indicate that the aqueous extract of S.decurrens inhibited and damaged the mitosis of V.faba root tip cells to some degrees,had certain genetic toxicity and had the ability to rapidly spread.[Conclusions]This study provides a theoretical basis for scientifically and rationally using S.decurrens and preventing the spread of S.decurrens.
文摘AIM:To evaluate the occurrence of micronucleus(MN),nucleoplasmic bridges(NPBs)and nuclear buds(NBUDs)in the mitogen-stimulated lymphocytes of patients with non-alcoholic steatohepatitis(NASH).METHODS:The study was performed in 25(9 females,16 males)patients newly diagnosed with NASH,and 25healthy subjects of similar ages and genders were used as a control group.None of the controls was known to be receiving any drugs for medical or other reasons or using alcohol.Hepatosteatosis was further excluded by abdominal ultrasound imaging in the control group.The numbers of MN,NPBs and NBUDs scored in binucleated(BN)cells were obtained from the mitogen-stimulated lymphocytes of patients and control subjects.Statistical comparisons of the numbers of BN cells with MN,NPBs and NBUDs and ages between the patients with NASH and control subjects were performed.RESULTS:The mean ages of the patients and the control group were 41.92±13.33 and 41.80±13.09 years(P>0.05),respectively.The values of the mean body mass index(BMI),HOMA-IR,hemoglobin,creatinin,aspartate aminotransferase,alanine aminotransferase,triglyceride,high density lipoprotein,and low density lipoprotein were 31.19±4.62 kg/m2vs 25.07±4.14 kg/m2,6.71±4.68 vs 1.40±0.53,14.73±1.49 g/dL vs 14.64±1.30 g/dL,0.74±0.15 mg/dL vs 0.80±0.13 mg/dL,56.08±29.11 U/L vs 16.88±3.33 U/L,92.2±41.43U/L vs 15.88±5.88 U/L,219.21±141.68 mg/dL vs102.56±57.98 mg/dL,16.37±9.65 mg/dL vs 48.72±15.31 mg/dL,and 136.75±30.14 mg/dL vs 114.63±34.13 mg/dL in the patients and control groups,respectively.The total numbers and frequencies of BN cells with MN,NPBs and NBUDs,which were scored using the CBMN cytome assay on PHA-stimulated lymphocytes,were evaluated in the patients with NASH and control group.We found significantly higher numbers of MN,NPBs and NBUDs in the BN cells of patients with NASH than in those of the control subjects(21.60±9.32vs 6.88±3.91;29.28±13.31 vs 7.84±3.96;15.60±5.55 vs 4.20±1.63,respectively,P<0.0001).CONCLUSION:The increased numbers of MN,NPBs and NBUDs observed in the lymphocytes obtained from patients with NASH may reflect genomic instability.

文摘Objective:To investigate the effects of radiation on growth-arrested(GA) and micronucleus-production(MP) rates,and the radioprotective properties of Thai medicinal plants in mouse macrophage cell line RAW264.7 in vitro.Methods:Mouse macrophage cell line(RAW264.7) was cultured in vitro.Various radiation exposures, growth-arrested rate assay,micronucleus production assay,and radioprotection by Thai medicinal plants were performed.Results:The results showed that GA and MP rates forγ-rays and UV were dose-dependent. The 50%-affected dose ofγand UV radiation for the GA rate was 10 Gy and 159 microwatt/cm for 0.5 seconds, respectively.After X-ray exposure,there was no apparent effect on RAW264.7 cells,even with a fortyfold human diagnostic dose.Two exposures toγradiation at 20 Gy resulted in a significantly higher MP rate than 20 Gy single exposure or control(P【0.05).The Thai medicinal plants(Kamin-chun capsules,Curcuma longa Linn;Hed lingeu,Ganoderma lucidum;Ya Pakking capsule,Murdannia loriformis) could not prevent cell damage,but epigallocatechin gallate and L-cysteine could provide protection from 2 Gyγ-ray exposure. Conclusion:γradiation caused chromosomal damage during cell division and UV caused cell death, while X-ray radiation was safe.The radioprotective effects of Thai medicinal plants,Kamin-chun,Hed lingeu, and Ya Pakking,could not prevent cell damage in this study.
