Each sample animal was injected with 40 nanogram of synthetic gonadotropin-releasing hormone analogue (GnRH-A) in distilled water every other day for up to 16 days; control animals received similar volumes of distilled wnter only. Both hormone-treated samples and control animals were examined under microscope every eight days and every twelve days. Then all the animals were killed on the 16th day. Gonads earlier development and a more advanced degree of gametogenesis are in the hormone-treated group than in the control group. Thus, GnRH-A can initiate and accelerate the gonadal development in gonadally regressed am-phioxus. GnRH-A in some form may be important in regulating the gonadal development in amphioxus as well as in vertebrates.展开更多

Methods: Prospective cohort study of 50 women undergoing in vitro ferti...Purpose: Can Cortisol Releasing Hormone (CRH) levels in follicular fluid predict outcomes following assisted reproductive treatment (ART) cycles? Methods: Prospective cohort study of 50 women undergoing in vitro fertilisation (IVF)/intra-cytoplasmic sperm injection (ICSI) cycles over a two month study period. All patients were treated on the long stimulation protocol;follicular fluid was aspirated and pooled for each patient. The samples were processed appropriately and assayed using CRH radioimmunoassay (RIA). Results: This study confirmed that CRH was present in follicular fluid. The average level detected was 173 ± 9 pg/mL (mean ± standard error of mean [SEM]). The data suggests a positive correlation of CRH follicular fluid levels greater than 145 pg/mL with successful ART outcomes. Conclusion: The data indicates a positive correlation between ART outcomes and the presence of follicular fluid CRH levels greater than 145 pg/mL. The results should be interpreted with caution due to the small sample size and pooling of follicular fluid per patient. Furthermore, the pooling of follicular fluid is not representative of CRH levels in an individual follicle, and thus, mature oocyte. This study serves as a reminder to what has previously been hypothesised.展开更多
In recent years, one of the most exciting advances in the researches of pituitary adenomas is the discovery that 30 %-40 % of human pituitary somatotrophinomas carry somatic mutations of the gene for the α subunit of...In recent years, one of the most exciting advances in the researches of pituitary adenomas is the discovery that 30 %-40 % of human pituitary somatotrophinomas carry somatic mutations of the gene for the α subunit of the stimulatory GTP binding protein, G s (G sα). These mutations, termed gsp oncogenes, may play an important role in the tumorigenesis of pituitary adenomas. Of 10 somatotrophinomas examined, 3 (30 %) were proved to be gsp positive, as determined by sequence analysis of DNA generated by the polymerase chain reaction (PCR). GHRH exerted a significant stimulatory effect on GH secretion in 2 of 3 gsp positive and 4 of 7 gsp negative tumors. Moreover, phorbol ester, 1, 2 tetradecanoylphorbol 13 acetate (TPA), enhanced stimulation of lated the GH secretion effect exerted by GHRH in gsp positive somatotrophinomas, whereas this effect was not observed in gsp negative tumors. This result suggests that the protein kinase C signal system as well as adenylyl cyclase cAMP protein kinase A intracellular signal transduction system plays a pivotal role in GH secretory control of GHRH, which may work together via a cross talk mechanism.展开更多
Objective It is well known that a dual trigger treatment can improve clinical outcomes of in vitro fertilization(IVF)in high or normal ovarian responders.However,it is not clear whether dual triggering also benefits p...Objective It is well known that a dual trigger treatment can improve clinical outcomes of in vitro fertilization(IVF)in high or normal ovarian responders.However,it is not clear whether dual triggering also benefits patients with diminished ovarian reserve(DOR).The aim of this study was to investigate whether a dual trigger treatment of gonadotropin-releasing hormone(GnRH)agonist combined with human chorionic gonadotropin(hCG)for final follicular maturation improves the cumulative live birth rate(CLBR)during the GnRH-antagonist cycle in patients with DOR.Methods This retrospective study included patients with DOR who received a GnRH-antagonist protocol during IVF and intracytoplasmic sperm injection(IVF-ICSI)cycles at Peking University People’s Hospital from January 1,2017 through December 31,2017.Oocyte maturation was triggered by GnRH combined with hCG(n=110)or hCG alone(n=71).Embryos were transferred on the third day after oocyte retrieval or during a subsequent freeze-thaw cycle.Patients were followed up for 3 years.Results The dual trigger treatment did not affect CLBR,which is an overall determinant of the success rate of assisted reproductive technology(ART).Women in the dual trigger group had significantly higher rates of fertilization than those in the hCG group(90.1%vs.83.9%,P=0.040).Conclusion Dual trigger with GnRH agonist and hCG did not improve CLBR in patients with DOR,but did slightly improve fertilization rate,oocyte count,and embryo quality.展开更多

Results: TRH, administered intravenously after head injury. promoted the recovery of consciousness andGCS score, alleviated the traumatic brain edema, controlled and lowered the intracranial pressure. decreased thelevel of lipid superoxides, decreased the mortality rate. and improved the life quality of the survivals. This study was conducted...The aim of this investigation is preparation of Mitomycin-C encapsulated with chitosan nanoparticles synthesis using ionic gelation technique for intravesical controlled drug delivery systems. This study was conducted in vitro. Cumulative amount of drug released from the nanoparticles was calculated. Mitomycin-C release studies were examined for different pH values. During the drug loading and release studies, initial amount of drug was changed (i.e., 0.5, 1.25 and 2.5 mg) to get different release profiles and the release studies were repeated (n = 6). The loading efficiencies of Mitomycin-C with three different initial concentrations 0.5mg/ml, 1.25 mg/ml and 2.5 mg/ml into chitosan nanoparticles were 54.5%, 47.1% and 36.4%, respectively. For different pH values, the cumulative releases of Mitomycin-C from chitosan nanoparticles were 47% and 53% for pH 6.0 and 7.4, respectively (p < 0.01). For different drug doses, the cumulative releases of Mitomycin-C (MMC) from Chitosan nanoparticles were 44%, 53% and 65% for 0.5 mg/mL, 1.25 mg/mL and 2.5 mg/mL respectively (p < 0.01). The anticancer activity of Mitomycin-C loaded chitosan nanoparticles was measured in T24 bladder cancer cell line in vitro, and the results revealed that the 2.5 MMC coated Chitosan nanoparticles had better tumor cells decline activity. From this investigation, we conclude that the drug encapsulated synthesized chitosan nanoparticles possess a high ability to be used as pH and dose responsive drug delivery system. This systematic investigation demonstrates a promising future for the intravesical installation in treatment of the superficial bladder cancer.展开更多

The volume ratio of water phase to oily phase is one of the critical parameters in forming core-sheath fibers. In this study, water phase was presented by hydrophilic tetracycline hydrochloride and oily phase by hydrophobic poly (E-caprolactone) (PCL). The effects of volume ratios of water phase to oily phase on fiber morphology and in vitro drug release were investigated. Scanning electron microscopy ( SEM ), transmission electron microscopy ( TEM), and eonfoeal laser scanning microscopy(CLSM) were used to observe the morphology, core.sheath structure of the fibers and drug loading in the fibers, respectively. Samples of three different volume ratios of water phase to oily phase, 1: 25, 1:15, and 1:10, were prepared with the same concentration of drug solution. Experiment results showed that, with an increase in the volume ratios of water phase to oily phase, the fiber diameter increased and diameter distribution scattered. The drug entrapment efficiency of the fibers reduces with the increase in volume ratios, L e. , from 73.48 % in the ratio of 1 : 25, 62.23 % in 1 : 15, down to 45.63 % in 1:10. In vitro release tests showed that a higher volume ratio of water phase to oily phase would lead to a lower release rate resulted from thicker fiber sheath.展开更多

We synthesized MINPs via precipitation polymerization with vinblastine(VBL) as a model drug, and investigated the drug loading,releasing property in vitro and bio-distribution in vivo. The obtained MINPs, from 300 to 450 nm,had smooth surface and favorable dispersibility. The entrapment efficacy and drug loading capacity of VBL loaded MINPs(MINPs-VBL) were 83.25% and 8.72% respectively. In PBS(pH 7.4),MINPs-VBL showed sustained release behavior. The cumulative release percentage reached about 70% during 216 h and no burst release was observed. The releasing behavior of MINPsVBL in vitro conformed to the first-order kinetics model. MINPs-VBL and commercially available vinblastine sulfate injection(VBL injection) were injected via tail vein of SD rats respectively to investigate the bio-distribution. MINPs-VBL group showed higher concentration of VBL in tissues and serum than VBL injection group after 60 min, and the drug level in liver was the highest. MINPs-VBL exhibited liver targeting trend to some extent, which was based on the evaluation of drug targeting index(DTI) and drug selecting index(DSI).展开更多

Then various concentrations(2, 4, 8, 16, 32, 64, 128 and256 mmol/L) of the peptide were incubated with mast cells and lactate dehydrogenase assay was performed.Results: No significant effect was observed in lactate dehydrogenase absorbance under128 mmol/L concentration. This implied that the peptide did not cause cell death in mast cells and consequently, histamine release did not happen. Moreover, the results showed the IC50 of mast cells degranulation at 126 mmol/L, which was approximately high implying that this peptide had high selectivity for normal cells and did not cause histamine release from these cells.Conclusions: This would be a great aim in new drug development, in which an agent acts potentially on its target tissue without activating the immune system.展开更多
The yield of tar and syngas has been investigated by catalytic pyrolysis of Pingzhuang lignite(PZL)over Ca(OH)2 catalyst in temperature range of 600℃-1000℃in a tube furnace.The results show that the yield of volatil...The yield of tar and syngas has been investigated by catalytic pyrolysis of Pingzhuang lignite(PZL)over Ca(OH)2 catalyst in temperature range of 600℃-1000℃in a tube furnace.The results show that the yield of volatile pyrolysis increases and char decreases with rising temperature for both raw and catalyzed Pingzhuang lignite.The hydrogen fraction(H2)increased from 20%to 40%for the PZL sample;but,for the PZL-Ca(OH)2 sample,H2 fraction fluctuated randomly between 35%to 42%,with the maximum H2 fraction found at 1000℃.The Gaschromatography mass-spectroscopic(GC-MS)analysis revealed that the maximum tar yield at 800℃and 700℃was obtained for PZL and PZL-Ca(OH)2,respectively.The surface morphology of PZL and PZL-Ca(OH)2 chars underwent different transformation in the presence of catalyst as illustrated by SEM/EDX,FTIR,and BET analysis.Furthermore,char sample was investigated for the carbon conversion and reactivity index using TGA analysis under N2 and CO atmosphere.展开更多

Biomimetic delivery of growth factors based on stem cell recruitment and osteogenic differentiation, as the key steps in natural alveolar bone regenerative process, has been an area of intense research in recent years. A mesoporous self-healing hydrogel(DFH) with basic fibroblast growth factor(bFGF) entrapment and transforming growth factor β3(TGFβ3)-loaded chitosan microspheres(CMs) was developed. The formulation was optimized by multiple tests of self-healing, in-bottle inversion, SEM, rheological, swelling rate and in vitro degradation. In vitro tubule formation assays, cell migration assays, and osteogenic differentiation assays confirmed the ability of DFH to promote blood vessels, recruit stem cells, and promote osteogenic differentiation. The optimum DFH formula is 0.05 ml 4ArmPEG-DF(20%) added to 1 ml CsGlu(2%) containing bFGF(80 ng) and TGFβ3-microspheres(5 mg). The results of in vitro release studied by Elisa kit, indicated an 95% release of b FGF in7 d and long-term sustained release of TGFβ3. For alveolar defects rat models, the expression levels of CD29 and CD45, the bone volume fraction, trabecular number, and trabecular thickness of new bone monitored by Micro-CT in DFH treatment groups were significantly higher than others(*P < 0.05, vs Model). HE and Masson staining show the same results.In conclusion, DFH is a design of bionic alveolar remodelling microenvironment, that is in early time microvessels formed by b FGF provide nutritious to recruited endogenous stem cells, then TGFβ3 slowly released speed up the process of new bones formation to common facilitate rat alveolar defect repair. The DFH with higher regenerative efficiency dovetails nicely with great demand due to the requirement of complicated biological processes.展开更多

Results: TRH, administered intravenously after head injury. promoted the recovery of consciousness andGCS score, alleviated the traumatic brain edema, controlled and lowered the intracranial pressure. decreased thelevel of lipid superoxides, decreased the mortality rate. and improved the life quality of the survivals. Nocomplications or adverse and toxic effects were noted during the course of TRH treatment. Conclusion: TRH hasbeneficial effects on patients with severe head injury.
文摘In 1453,Xu Youzhen,a scholar-bureaucrat and hydrologist in Ming China,was dispatched to Shandong to find a way to harness the Yellow River.He proposed the opening up of multiple channels to diverge the flood waters.An experiment was conducted to compare the efficiency of releasing water using one large opening with the use of a number of small openings.This experiment proved that opening up multiple channels outperformed the construction of only one,thereby convincing the emperor and officials of the efficacy of this method.Xu’s method for water control proved to be successful in averting a flood that occurred in 1456.
文摘The aim of this investigation is preparation of Mitomycin-C encapsulated with chitosan nanoparticles synthesis using ionic gelation technique for intravesical controlled drug delivery systems. This study was conducted in vitro. Cumulative amount of drug released from the nanoparticles was calculated. Mitomycin-C release studies were examined for different pH values. During the drug loading and release studies, initial amount of drug was changed (i.e., 0.5, 1.25 and 2.5 mg) to get different release profiles and the release studies were repeated (n = 6). The loading efficiencies of Mitomycin-C with three different initial concentrations 0.5mg/ml, 1.25 mg/ml and 2.5 mg/ml into chitosan nanoparticles were 54.5%, 47.1% and 36.4%, respectively. For different pH values, the cumulative releases of Mitomycin-C from chitosan nanoparticles were 47% and 53% for pH 6.0 and 7.4, respectively (p < 0.01). For different drug doses, the cumulative releases of Mitomycin-C (MMC) from Chitosan nanoparticles were 44%, 53% and 65% for 0.5 mg/mL, 1.25 mg/mL and 2.5 mg/mL respectively (p < 0.01). The anticancer activity of Mitomycin-C loaded chitosan nanoparticles was measured in T24 bladder cancer cell line in vitro, and the results revealed that the 2.5 MMC coated Chitosan nanoparticles had better tumor cells decline activity. From this investigation, we conclude that the drug encapsulated synthesized chitosan nanoparticles possess a high ability to be used as pH and dose responsive drug delivery system. This systematic investigation demonstrates a promising future for the intravesical installation in treatment of the superficial bladder cancer.

文摘The sub-regions are divided for the seismicity of the Chinese mainland based on the hypothesis of the active crustal blocks and the division of the active boundaries. On this result, the seismicity of each active crustal blocks are studied by calculating the accumulated and released strain of the earthquakes based on strain accumulating and releasing model, and the different seismicity stages of the sub-regions are discussed basically. Finally we have discussed the premise of the model application and the potential problems of the model results.

文摘Emulsion electrospinning as a novel process in spinning core-sheath fibers shows a promising potential in drug release control. The volume ratio of water phase to oily phase is one of the critical parameters in forming core-sheath fibers. In this study, water phase was presented by hydrophilic tetracycline hydrochloride and oily phase by hydrophobic poly (E-caprolactone) (PCL). The effects of volume ratios of water phase to oily phase on fiber morphology and in vitro drug release were investigated. Scanning electron microscopy ( SEM ), transmission electron microscopy ( TEM), and eonfoeal laser scanning microscopy(CLSM) were used to observe the morphology, core.sheath structure of the fibers and drug loading in the fibers, respectively. Samples of three different volume ratios of water phase to oily phase, 1: 25, 1:15, and 1:10, were prepared with the same concentration of drug solution. Experiment results showed that, with an increase in the volume ratios of water phase to oily phase, the fiber diameter increased and diameter distribution scattered. The drug entrapment efficiency of the fibers reduces with the increase in volume ratios, L e. , from 73.48 % in the ratio of 1 : 25, 62.23 % in 1 : 15, down to 45.63 % in 1:10. In vitro release tests showed that a higher volume ratio of water phase to oily phase would lead to a lower release rate resulted from thicker fiber sheath.

文摘Molecular imprinted nanoparticles(MINPs) can memorize the shape and functional group positions complementary to template, which account for the large drug loading capacity and slow drug release behavior as drug carriers. We synthesized MINPs via precipitation polymerization with vinblastine(VBL) as a model drug, and investigated the drug loading,releasing property in vitro and bio-distribution in vivo. The obtained MINPs, from 300 to 450 nm,had smooth surface and favorable dispersibility. The entrapment efficacy and drug loading capacity of VBL loaded MINPs(MINPs-VBL) were 83.25% and 8.72% respectively. In PBS(pH 7.4),MINPs-VBL showed sustained release behavior. The cumulative release percentage reached about 70% during 216 h and no burst release was observed. The releasing behavior of MINPsVBL in vitro conformed to the first-order kinetics model. MINPs-VBL and commercially available vinblastine sulfate injection(VBL injection) were injected via tail vein of SD rats respectively to investigate the bio-distribution. MINPs-VBL group showed higher concentration of VBL in tissues and serum than VBL injection group after 60 min, and the drug level in liver was the highest. MINPs-VBL exhibited liver targeting trend to some extent, which was based on the evaluation of drug targeting index(DTI) and drug selecting index(DSI).

文摘Objective: To investigate the antihistamine-releasing effect of a peptide isolated from wasp venom of Vespa orientalis.Methods: This peptide was separated from crude venom by chromatography methods and mass spectrometry. Then various concentrations(2, 4, 8, 16, 32, 64, 128 and256 mmol/L) of the peptide were incubated with mast cells and lactate dehydrogenase assay was performed.Results: No significant effect was observed in lactate dehydrogenase absorbance under128 mmol/L concentration. This implied that the peptide did not cause cell death in mast cells and consequently, histamine release did not happen. Moreover, the results showed the IC50 of mast cells degranulation at 126 mmol/L, which was approximately high implying that this peptide had high selectivity for normal cells and did not cause histamine release from these cells.Conclusions: This would be a great aim in new drug development, in which an agent acts potentially on its target tissue without activating the immune system.

文摘The yield of tar and syngas has been investigated by catalytic pyrolysis of Pingzhuang lignite(PZL)over Ca(OH)2 catalyst in temperature range of 600℃-1000℃in a tube furnace.The results show that the yield of volatile pyrolysis increases and char decreases with rising temperature for both raw and catalyzed Pingzhuang lignite.The hydrogen fraction(H2)increased from 20%to 40%for the PZL sample;but,for the PZL-Ca(OH)2 sample,H2 fraction fluctuated randomly between 35%to 42%,with the maximum H2 fraction found at 1000℃.The Gaschromatography mass-spectroscopic(GC-MS)analysis revealed that the maximum tar yield at 800℃and 700℃was obtained for PZL and PZL-Ca(OH)2,respectively.The surface morphology of PZL and PZL-Ca(OH)2 chars underwent different transformation in the presence of catalyst as illustrated by SEM/EDX,FTIR,and BET analysis.Furthermore,char sample was investigated for the carbon conversion and reactivity index using TGA analysis under N2 and CO atmosphere.

文摘Resorption and loss of alveolar bone leads to oral dysfunction and loss of natural or implant teeth. Biomimetic delivery of growth factors based on stem cell recruitment and osteogenic differentiation, as the key steps in natural alveolar bone regenerative process, has been an area of intense research in recent years. A mesoporous self-healing hydrogel(DFH) with basic fibroblast growth factor(bFGF) entrapment and transforming growth factor β3(TGFβ3)-loaded chitosan microspheres(CMs) was developed. The formulation was optimized by multiple tests of self-healing, in-bottle inversion, SEM, rheological, swelling rate and in vitro degradation. In vitro tubule formation assays, cell migration assays, and osteogenic differentiation assays confirmed the ability of DFH to promote blood vessels, recruit stem cells, and promote osteogenic differentiation. The optimum DFH formula is 0.05 ml 4ArmPEG-DF(20%) added to 1 ml CsGlu(2%) containing bFGF(80 ng) and TGFβ3-microspheres(5 mg). The results of in vitro release studied by Elisa kit, indicated an 95% release of b FGF in7 d and long-term sustained release of TGFβ3. For alveolar defects rat models, the expression levels of CD29 and CD45, the bone volume fraction, trabecular number, and trabecular thickness of new bone monitored by Micro-CT in DFH treatment groups were significantly higher than others(*P < 0.05, vs Model). HE and Masson staining show the same results.In conclusion, DFH is a design of bionic alveolar remodelling microenvironment, that is in early time microvessels formed by b FGF provide nutritious to recruited endogenous stem cells, then TGFβ3 slowly released speed up the process of new bones formation to common facilitate rat alveolar defect repair. The DFH with higher regenerative efficiency dovetails nicely with great demand due to the requirement of complicated biological processes.
