However, the precise neuroprotective mechanism of Shuxuetong injection remains poorly understood. Here, cerebral microvascular endothelial cells(bEnd.3) were incubated in glucose-free Dulbecco's modified Eagle's medium containing 95% N_2/5% CO_2 for 6 hours, followed by high-glucose medium containing 95% O_2 and 5% CO_2 for 18 hours to establish an oxygen-glucose deprivation/reperfusion model. This in vitro cell model was administered Shuxuetong injection at 1/32, 1/64, and 1/128 concentrations(diluted 32-, 64-, and 128-times). Cell Counting Kit-8 assay was used to evaluate cell viability. A fluorescence method was used to measure lactate dehydrogenase, and a fluorescence microplate reader used to detect intracellular reactive oxygen species. A fluorescent probe was also used to measure mitochondrial superoxide production. A cell resistance meter was used to measure transepithelial resistance and examine integrity of monolayer cells. The fluorescein isothiocyanate-dextran test was performed to examine blood-brain barrier permeability. Real-time reverse transcription polymerase chain reaction was performed to analyze mRNA expression levels of tumor necrosis factor alpha, interleukin-1β, interleukin-6, and inducible nitric oxide synthase. Western blot assay was performed to analyze expression of caspase-3, intercellular adhesion molecule 1, vascular cell adhesion molecule 1, occludin, vascular endothelial growth factor, cleaved caspase-3, B-cell lymphoma 2, phosphorylated extracellular signal-regulated protein kinase, extracellular signal-regulated protein kinase, nuclear factor-κB p65, I kappa B alpha, phosphorylated I kappa B alpha, I kappa B kinase, phosphorylated I kappa B kinase, claudin-5, and zonula occludens-1. Our results show that Shuxuetong injection increases bEnd.3 cell viability and B-cell lymphoma 2 expression, reduces cleaved caspase-3 expression, inhibits production of reactive oxygen species and mitochondrial superoxide, suppresses expression of tumor necrosis factor alpha, interleukin-1β, interleukin-6, inducible nitric oxide synthase mRNA, intercellular adhesion molecule-1, and vascular cell adhesion molecule-1, markedly increases transepithelial resistance, decreases blood-brain barrier permeability, upregulates claudin-5, occludin, and zonula occludens-1 expression, reduces nuclear factor-κB p65 and vascular endothelial growth factor expression, and reduces I kappa B alpha, extracellular signal-regulated protein kinase 1/2, and I kappa B kinase phosphorylation levels. Overall, these findings suggest that Shuxuetong injection has protective effects on brain microvascular endothelial cells after oxygen-glucose deprivation/reperfusion. Moreover, its protective effect is associated with reduction of mitochondrial superoxide production, inhibition of the inflammatory response, and inhibition of vascular endothelial growth factor, extracellular signal-regulated protein kinase 1/2, and the nuclear factor-κB p65 signaling pathway.展开更多

The MAT-CI is expected to acquire high quality image and embraces a wide application. Its principle is to put the conductive sample in the Static Magnetic Field(SMF) and inject a time-varying current, during which the SMF and the current interact and generate the Lorentz Force that inspire ultrasonic signal received by the ultrasonic transducers positioned around the sample. And then according to related reconstruction algorithm and ultrasonic signal, electrical conductivity image is obtained. In this paper, a forward problem mathematical model of the MAT-CI has been set up to deduce the theoretical equation of the electromagnetic field and solve the sound source distribution by Green’s function. Secondly, a sound field restoration by Wiener filtering and reconstruction of current density by time-rotating method have deduced the Laplace’s equation that caters to the current density to further acquire the electrical conductivity distribution image of the sample through iteration method. In the end, double-loop coils experiments have been conducted to verify its feasibility.展开更多

We construct adversarial prompts using strategies like role emulation and manipulative assistance to simulate real threats. We introduce a comprehensive, multi-tiered defense framework named GUARDIAN (Guardrails for Upholding Ethics in Language Models) comprising a system prompt filter, pre-processing filter leveraging a toxic classifier and ethical prompt generator, and pre-display filter using the model itself for output screening. Extensive testing on Meta’s Llama-2 model demonstrates the capability to block 100% of attack prompts. The approach also auto-suggests safer prompt alternatives, thereby bolstering language model security. Quantitatively evaluated defense layers and an ethical substitution mechanism represent key innovations to counter sophisticated attacks. The integrated methodology not only fortifies smaller LLMs against emerging cyber threats but also guides the broader application of LLMs in a secure and ethical manner.展开更多

Therefore, mesenchymal stem cells have been shown to exhibit prospects in the treatment of amyotrophic lateral sclerosis. However, the safety of their clinical application needs to be validated. To investigate the safety of intrathecal injection of Wharton's jelly-derived mesenchymal stem cells in amyotrophic lateral sclerosis therapy, 43 patients(16 females and 27 males, mean age of 57.3 years) received an average dose of 0.42 × 106 cells/kg through intrathecal administration at the cervical, thoracic or lumbar region depending on the clinical symptoms. There was a 2 month interval between two injections. The adverse events occurring during a 6-month treatment period were evaluated. No adverse events occurred. Headache occurred in one case only after first injection of stem cells. This suggests that intrathecal injection of Wharton's Jelly-derived mesenchymal stem cells is well tolerated in patients with amyotrophic lateral sclerosis. We present a unique treatment method and magnetic resonance (MR) images of this condition. The patient was a 43-year-old male with a chief complaint of abnormal occlusion. Regarding the present status, there was no occlusion on the right side. No temporomandibular joint pain, myalgia, or clicking were observed while mouth opening. On MR images, the posterior disc displacement without reduction on the right was observed and it was assumed that occlusal abnormality was due to this condition. We treated him with the following methods. After injection into the superior articular cavity with 2% lidocaine, a sodium hyaluronate preparation was injected followed by intermaxillary traction applied using rubber. The recovery of occlusion was confirmed in follow-up at 5 days after treatment. In the 13th days, the intermaxillary traction was removed. On MR images, the right disc condition was changed to anterior disc displacement with reduction. We consider our treatment methods are effective for this clinical condition.展开更多

Methods:Sixty Balb/c nude mice were randomly divided into groups A, B and C with 20 mice in each group. Group A mice were used as blank control, group B mice as model group and group C mice as treatment group. The PASI score of psoriasis, skin thickness, inflammatory factors, serum levels of Caspase-14, SOCS1 and STAT3 in three groups of mice were analyzed after 2 weeks of treatment. Result: After treatment, the P ASI score of group B was significantly higher than that of group C, with statistical significance (P < 0.05);there was statistical significance in the measurements of lesion skin of three groups of mice after treatment (P <0.05). Compared with the blank control group, the thickness of lesion skin in group B and C was significantly higher, and the thickness of lesion skin in treatment group was significantly lower than that in control group (P < 0.05). Compared with the blank control group, the inflammatory factors IL-17, IL-22 and IL-23 in the B and C groups were significantly increased, and the inflammatory factors IL-17, IL-22 and IL-23 in the treatment group were significantly lower than those in the model group. The levels of serum C aspase-14, SOCS1 and STAT3 in three groups of mice were significantly different after treatment (P < 0.05). Compared with the blank control group, the levels of serum C aspase-14 and SOCS1 in B and C groups were significantly lower and the levels of STAT3 were significantly higher, and the levels of inflammatory factors aspase-14 and SO in treatment group were significantly higher than those in control group. The level of CS1 was significantly lower than that of model group, and the level of STAT3 was significantly higher than that of model group (P <0.05). Conclusion: Astragalus membranaceus injection can effectively improve the degree of psoriasis in Balb/c nude mice. Its possible mechanism is that it can decrease the expression of Caspase-14 and SOCS1, reduce the degree of keratosis in the lesion site of mice, improve the local surface hyperplasia, increase the level of STAT3 and enhance the level of local cell proliferation, which is of positive significance for the rehabilitation of psoriasis.展开更多

文摘Shuxuetong injection composed of leech(Hirudo nipponica Whitman) and earthworm(Pheretima aspergillum) has been used for the clinical treatment of acute stroke for many years in China. However, the precise neuroprotective mechanism of Shuxuetong injection remains poorly understood. Here, cerebral microvascular endothelial cells(bEnd.3) were incubated in glucose-free Dulbecco's modified Eagle's medium containing 95% N_2/5% CO_2 for 6 hours, followed by high-glucose medium containing 95% O_2 and 5% CO_2 for 18 hours to establish an oxygen-glucose deprivation/reperfusion model. This in vitro cell model was administered Shuxuetong injection at 1/32, 1/64, and 1/128 concentrations(diluted 32-, 64-, and 128-times). Cell Counting Kit-8 assay was used to evaluate cell viability. A fluorescence method was used to measure lactate dehydrogenase, and a fluorescence microplate reader used to detect intracellular reactive oxygen species. A fluorescent probe was also used to measure mitochondrial superoxide production. A cell resistance meter was used to measure transepithelial resistance and examine integrity of monolayer cells. The fluorescein isothiocyanate-dextran test was performed to examine blood-brain barrier permeability. Real-time reverse transcription polymerase chain reaction was performed to analyze mRNA expression levels of tumor necrosis factor alpha, interleukin-1β, interleukin-6, and inducible nitric oxide synthase. Western blot assay was performed to analyze expression of caspase-3, intercellular adhesion molecule 1, vascular cell adhesion molecule 1, occludin, vascular endothelial growth factor, cleaved caspase-3, B-cell lymphoma 2, phosphorylated extracellular signal-regulated protein kinase, extracellular signal-regulated protein kinase, nuclear factor-κB p65, I kappa B alpha, phosphorylated I kappa B alpha, I kappa B kinase, phosphorylated I kappa B kinase, claudin-5, and zonula occludens-1. Our results show that Shuxuetong injection increases bEnd.3 cell viability and B-cell lymphoma 2 expression, reduces cleaved caspase-3 expression, inhibits production of reactive oxygen species and mitochondrial superoxide, suppresses expression of tumor necrosis factor alpha, interleukin-1β, interleukin-6, inducible nitric oxide synthase mRNA, intercellular adhesion molecule-1, and vascular cell adhesion molecule-1, markedly increases transepithelial resistance, decreases blood-brain barrier permeability, upregulates claudin-5, occludin, and zonula occludens-1 expression, reduces nuclear factor-κB p65 and vascular endothelial growth factor expression, and reduces I kappa B alpha, extracellular signal-regulated protein kinase 1/2, and I kappa B kinase phosphorylation levels. Overall, these findings suggest that Shuxuetong injection has protective effects on brain microvascular endothelial cells after oxygen-glucose deprivation/reperfusion. Moreover, its protective effect is associated with reduction of mitochondrial superoxide production, inhibition of the inflammatory response, and inhibition of vascular endothelial growth factor, extracellular signal-regulated protein kinase 1/2, and the nuclear factor-κB p65 signaling pathway.

文摘The paper proposes a biomass cross-upgrading process that combines hydrothermal carbonization and pyrolysis to produce high-quality blast furnace injection fuel. The results showed that after upgrading, the volatile content of biochar ranged from 16.19%to 45.35%,and the alkali metal content, ash content, and specific surface area were significantly reduced. The optimal route for biochar pro-duction is hydrothermal carbonization-pyrolysis(P-HC),resulting in biochar with a higher calorific value,C=C structure, and increased graphitization degree. The apparent activation energy(E)of the sample ranges from 199.1 to 324.8 kJ/mol,with P-HC having an E of 277.8 kJ/mol,lower than that of raw biomass, primary biochar, and anthracite. This makes P-HC more suitable for blast furnace injection fuel.Additionally, the paper proposes a path for P-HC injection in blast furnaces and calculates potential environmental benefits.P-HC of-fers the highest potential for carbon emission reduction, capable of reducing emissions by 96.04 kg/t when replacing 40wt%coal injec-tion.
文摘The Magneto-acoustic Tomography with Current Injection (MAT-CI) is a new biological electrical impedance imaging technique that combines Electrical Impedance Tomography (EIT) with Ultrasonic Imaging (UI), which possesses the non-invasive and high-contrast of the EIT and the high-resolution of the UI. In the end, double-loop coils experiments have been conducted to verify its feasibility.
文摘This paper introduces a novel multi-tiered defense architecture to protect language models from adversarial prompt attacks. The integrated methodology not only fortifies smaller LLMs against emerging cyber threats but also guides the broader application of LLMs in a secure and ethical manner.

文摘Future 6G communications are envisioned to enable a large catalogue of pioneering applications. These will range from networked Cyber-Physical Systems to edge computing devices, establishing real-time feedback control loops critical for managing Industry 5.0 deployments, digital agriculture systems, and essential infrastructures. The provision of extensive machine-type communications through 6G will render many of these innovative systems autonomous and unsupervised. While full automation will enhance industrial efficiency significantly,it concurrently introduces new cyber risks and vulnerabilities. In particular, unattended systems are highly susceptible to trust issues:malicious nodes and false information can be easily introduced into control loops.Additionally,Denialof-Service attacks can be executed by inundating the network with valueless noise.Current anomaly detection schemes require the entire transformation of the control software to integrate new steps and can only mitigate anomalies that conform to predefined mathematical models.Solutions based on an exhaustive data collection to detect anomalies are precise but extremely slow.Standard models, with their limited understanding of mobile networks, can achieve precision rates no higher than 75%.Therefore,more general and transversal protection mechanisms are needed to detect malicious behaviors transparently. This paper introduces a probabilistic trust model and control algorithm designed to address this gap.The model determines the probability of any node to be trustworthy.Communication channels are pruned for those nodes whose probability is below a given threshold. The trust control algorithmcomprises three primary phases, which feed themodel with three different probabilities, which are weighted and combined.Initially, anomalous nodes are identified using Gaussian mixture models and clustering technologies.Next, traffic patterns are studied using digital Bessel functions and the functional scalar product. Finally, the information coherence and content are analyzed. The noise content and abnormal information sequences are detected using a Volterra filter and a bank of Finite Impulse Response filters. An experimental validation based on simulation tools and environments was carried out. Results show the proposed solution can successfully detect up to 92%of malicious data injection attacks.

文摘Animal experiments have confirmed that mesenchymal stem cells can inhibit motor neuron apoptosis and inflammatory factor expression and increase neurotrophic factor expression. 文摘BACKGROUND Intramuscular corticosteroid injection may cause adverse effects such as dermal and/or subcutaneous atrophy, alopecia, hypopigmentation, and hyperpigmentation.Although cutaneous atrophy can spontaneously resolve, several treatment options have been suggested for this condition.CASE SUMMARY In this paper,we report a case of corticosteroid injection induced lipoatrophy treated with autologous whole blood(AWB)injection,as the condition had been unresponsive to fractional laser therapy.A 29-year-old female patient visited the dermatology clinic complaining of skin depression on her right buttock area, which had appeared six months earlier. There had been only subtle improvement at the margins after fractional CO2 laser treatment;therefore, after obtaining informed consent from the patient,AWB treatment was initiated. One month after the first AWB injection, the size and depth of the lesion had noticeably improved, and a slight improvement was also observed in discoloration.CONCLUSION Close observation is the initial treatment of choice for steroid induced skin atrophy;however, for patients in need of immediate cosmetic improvement,AWB injection may be a safe and cost-effective alternative.

文摘Based on the independently developed true triaxial multi-physical field large-scale physical simulation system of in-situ injection and production,we conducted physical simulation of long-term multi-well injection and production in the hot dry rocks of the Gonghe Basin,Qinghai Province,NW China.Through multi-well connectivity experiments, the spatial distribution characteristics of the natural fracture system in the rock samples and the connectivity between fracture and wellbore were clarified. The injection and production wells were selected to conduct the experiments, namely one injection well and two production wells, one injection well and one production well. The variation of several physical parameters in the production well was analyzed, such as flow rate, temperature, heat recovery rate and fluid recovery. The results show that under the combination of thermal shock and injection pressure, the fracture conductivity was enhanced, and the production temperature showed a downward trend. The larger the flow rate, the faster the decrease. When the local closed area of the fracture was gradually activated, new heat transfer areas were generated, resulting in a lower rate of increase or decrease in the mining temperature. The heat recovery rate was mainly controlled by the extraction flow rate and the temperature difference between injection and production fluid. As the conductivity of the leak-off channel increased, the fluid recovery of the production well rapidly decreased. The influence mechanisms of dominant channels and fluid leak-off on thermal recovery performance are different. The former limits the heat exchange area, while the latter affects the flow rate of the produced fluid. Both of them are important factors affecting the long-term and efficient development of hot dry rock.
文摘The design of three-dimensional printing based conformal cooling channels(CCCs)in injection molding holds great significance.Compared to CCCs,conformal cooling(CC)cavity solutions show promise in delivering enhanced cooling performance for plastic products, although they have been underexplored. In this research,CC cavity is designed within the mold geometry, reinforced by body-centered cubic(BCC)lattice structures to enhance mechanical strength.Three distinct BCC lattice variations have been integrated into the CC cavity:the BCC structure,BCC with cubes, and BCC with pillars. The thermal performances of the BCC lattice-added CC cavity are assessed numerically after experimental validation. To provide feasible solutions from viewpoints of thermal performances, various BCC lattice structure thicknesses are analyzed in the range of 0.8–1.2mm.Thermal simulation outcomes reveal that thicker lattice structures enhance mechanical strength but simultaneously lead to an increase in cooling time.Upon examining all the proposed CC cavity solutions supported by BCC,the cooling times range from 2.2 to 4 s,resulting in a reduction of 38.6%to 66.1%when compared to conventional straightdrilled channels. In contrast to CCCs,CC cavities have the potential to decrease the maximum temperature nonuniformity from 8.5 to 6 K.Nevertheless, the presence of lattice structures in CC cavity solutions results in an elevated pressure drop, reaching 2.8MPa,whereas the results for CCCs remain below2.1MPa.

文摘Posterior disc displacement of the temporomandibular joint is rare. We consider our treatment methods are effective for this clinical condition.

文摘Objective: To investigate the effect of Astragalus Injection on the Skin Lesion Degree and Caspase-14, SOCS1 and STAT3 Levels of Psoriasis Model in Balb/c Nude Mice. Its possible mechanism is that it can decrease the expression of Caspase-14 and SOCS1, reduce the degree of keratosis in the lesion site of mice, improve the local surface hyperplasia, increase the level of STAT3 and enhance the level of local cell proliferation, which is of positive significance for the rehabilitation of psoriasis.

文摘Low thermal evolution degree of gas and connected fracture network are key features in the continental shale gas reservoirs. When the pressure decreases lower than the dew point pressure, gas condensate diminishes the mobility of subsurface fluid forms in the near well region, which further reduces the productivity and final recovery factor in addition to the low connected fracture network. This work is motivated to investigate the application of gas injection, proven to be a feasible technique in conventional/marine-shale gas condensate reservoirs,in continental shale gas condensate reservoirs. We apply a compositional model to investigate the mechanism of gas injection treatment taking a continental shale gas condensate reservoir in the Fuxing area,Sichuan Basin,China as an example. The results demonstrate that for the removal of gas condensate during CO_(2)injection,re-vaporization plays the main mechanism through two processes including pressurization and development of miscibility, while the former is overwhelming.Considering the variation trend of the total volume of gas condensate,we divide the whole injection process into four and six stages in the single fracture model and fracture network model, respectively, and reason the trend with respect to phase behavior and flow pattern.A sensitive study on the injected gas type indicates that the sorting of the gas injection performance to remove gas condensate produced gas followed by CH4,and followed by CO_(2).Finally,we investigate the performance of cyclic produced-gas injection and optimize the injection scheme. This work provides technical and theoretical support for the development of continental shale gas reservoirs.
文摘Optimal spacing for vertical wells can be effectively predicted with several published methods, but methods suitable for assessing the proper horizontal well spacing are rare. This work proposes a method for calculating the optimal horizontal well spacing for an ultra-low permeability reservoir e the Yongjin reservoir in the Juggar Basin, northwestern China. The result shows that a spacing of 640m is the most economical for the development of the reservoir. To better develop the reservoir, simulation approaches are used and a new model is built based on the calculated well spacing.Since the reservoir has an ultralow permeability, gas injection is regarded as the preferred enhanced oil recovery(EOR)method.Injection of different gases including carbon dioxide, methane, nitrogen and mixed gas are modelled. The results show that carbon dioxide injection is the most efficient and economical for the development of the reservoir. However,if the reservoir produces enough methane, reinjecting methane is even better than injecting carbon dioxide.

文摘Multi-gradient drilling is a new offshore drilling method. The accurate calculation of the related wellbore temperature is of great significance for the prediction of the gas hydrate formation area and the precise control of the wellbore pressure. In this study,a new heat transfer model is proposed by which the variable mass flow is properly taken into account.Using this model, the effects of the main factors influencing the wellbore temperature are analyzed. The results indicate that at the position where the separation injection device is installed, the temperature increase of the fluid in the drill pipe is mitigated due to the inflow/outflow of hollow spheres, and the temperature drop of the fluid in the annulus also decreases. In addition,a lower separation efficiency of the device,a shallower installation depth and a smaller circulating displacement tend to increase the temperature near the bottom of the annulus, thereby helping to reduce the hydrate generation area and playing a positive role in the prevention and control of hydrates in deepwater drilling.
文摘BACKGROUND Hemolymphangiomas are rare malformations composed of both lymphatic and vascular vessels and are located in the pancreas, spleen, mediastinum, etc.Small intestinal hemolymphangioma is extremely rare and often presents as obscure gastrointestinal bleeding. It is rarely diagnosed correctly before the operation.Endoscopic injection sclerotherapy is usually used as a management of bleeding in esophageal varices and was occasionally reported as a treatment of vascular malformation. The treatment of small intestinal hemolymphangioma with enteroscopic injection sclerotherapy has not been reported.CASE SUMMARY A 42-year-old male complained of recurrent episodes of melena and dizziness, fatigue and reduced exercise capacity for more than 2 mo.Gastroduodenoscopy and blood test revealed a gastric ulcer and anemia.Treatment with oral protonpump inhibitors and iron did not improve symptoms. We then performed a capsule endoscopy and anterograde balloon-assisted enteroscopy and revealed a hemolymphangioma.Considering it is a benign tumor without malignant potential,we performed enteroscopic injection sclerotherapy. He was discharged 4 days later. At follow-up 3 mo later, the melena disappeared.Balloon-assisted enteroscopy revealed an atrophied tumor atrophied and no bleeding.Argon plasma coagulation was applied to the surface of the hemolymphangioma to accelerated healing. When he returned for follow-up 1 year later, anemia was resolved and the tumor had been cured.CONCLUSION Balloon-assisted enteroscopy and capsule endoscopy are effective methods for diagnosis of hemolymphangioma.Enteroscopic injection sclerotherapy is an effective treatment.
文摘Experimental methods, including mercury pressure, nuclear magnetic resonance(NMR)and core(wateroil)displacement, are used to examine the effects of high-multiple water injection(i.e.water injection with high injected pore volume)on rock properties, pore structure and oil displacement efficiency of an oilfield in the western South China Sea.The results show an increase in the permeability of rocks along with particle migration,an increase in the pore volume and the average pore throat radius, and enhanced heterogeneity after high-multiple water injection.Compared with normal water injection methods,a high-multiple water injection is more effective in improving the oil displacement efficiency. The degree of recovery increases faster in the early stage due to the expansion of the swept area, and the transition from oil-wet to water-wet.The degree of recovery increases less in the late stage due to various factors, including the enhancement of heterogeneity in the rocks.Considering both the economic aspect and the production limit of water flooding,it is recommended to adopt other technologies to further enhance oil recovery after 300 PV water injection.
