Nano β-TCP particles were added into Mg-Zn-Zr alloy to improve its microstructure and the properties. As-extruded Mg-3Zn-0.8Zr alloy and Mg-3Zn-0.8Zr/xβ-TCP (x=0.5%, 1.0% and 1.5%) composites were respectively fabricated. The grains of Mg-Zn-Zr/β-TCP composites were significantly refined. The results of the tensile tests indicate that the ultimate tensile strength and the elongation of composites were improved with the addition of β-TCP. The electrochemical test result in simulation body fluid shows that the corrosion resistance of the composites was strongly enhanced comparing with that of the alloy. The corrosion potential of Mg-3Zn0.8-Zr/1.0β-TCP composite is 1.547 V and its corrosion current density is 1.20×10 6 A/cm 2 .展开更多
With an elemental composition similar to bone mineral,and the ability to release phosphorus and calcium that benefit bone regeneration,Calcium Phosphate Glass(CPG)serves as a promising component of bone tissue enginee...With an elemental composition similar to bone mineral,and the ability to release phosphorus and calcium that benefit bone regeneration,Calcium Phosphate Glass(CPG)serves as a promising component of bone tissue engineering scaffolds.However,the degradation of CPG composites typically results in increased acidity,and its impact on bone-forming activity is less studied.In this work,we prepared 3D-printed composite scaffolds comprising CPG,Poly-ε-caprolactone(PCL),and various Magnesium Oxide(MgO)contents.Increasing the MgO content effectively suppressed the degradation of CPG,maintaining a physiological pH of the degradation media.While the degradation of CPG/PCL scaffolds resulted in upregulated apoptosis of Rat Bone Marrow-derived Stem Cells(rBMSC),scaffolds containing MgO were free from these negative impacts,and an optimal MgO content of 1 wt%led to the most pronounced osteogenic differentiation of rBMSCs.This work demonstrated that the rapid degradation of CPG impaired the renewability of stem cells through the increased acidity of the surrounding media,and MgO effectively modulated the degradation rate of CPG,thus preventing the negative effects of rapid degradation and supporting the proliferation and osteogenic differentiation of the stem cells.展开更多

The nBCP with composition of 63/37 (...In this paper, preparation of nano-biphasic calcium phosphate (nBCP), mechanical behavior and load-bearing of poly (lactide-co-glycolide) (PLGA) and PLGA/nBCP are presented. The nBCP with composition of 63/37 (w/w) HA/-TCP (hydroxyapatite/fl-tricalcium phosphate) was produced by heating of bovine bone at 700℃. Composite scaffolds were made by using PLGA matrix and 10-50 wt% nBCP powders as reinforcement material. All scaffolds were prepared by thermally induced solid-liquid phase separation (TIPS) at -60~C under 4 Pa (0.04 mbar) vacuum. The results of elastic modulus testing were adjusted with Ishai-Cohen and Narkis models for rigid polymeric matrix and compared to each other. PLGA/nBCP scaffolds with 30 wt% nBCP showed the highest value of yield strength among the scaffolds. In addition, it was found that by increasing the nBCP in scaffolds to 50 wt%, the modulus of elasticity was highly enhanced. However, the optimum value of yield strength was obtained at 30 wt% nBCP, and the agglomeration of reinforcing particles at higher percentages caused a reduction in yield strength. It is clear that the elastic modulus of matrix has the significant role in elastic modulus of scaffolds, as also the size of the filler particles in the matrix.展开更多

Unfortunately repair of articular cartilage defects do not always provide satisfactory outcomes. Methods Chondrocyte and osteoblast composites were co-cultured using a bioreactor. The cartilage defects were treated with cell-β-tricalcium phosphate (β-TCP) composites implanted into osteochondral defects in dogs, in vivo, using mosaicplasty, by placing chondrocyte-β-TCP scaffold composites on top of the defect and osteoblast-β-TCP scaffold composites below the defect.Results Electron microscopy revealed that the induced chondrocytes and osteoblast showed fine adhesive progression and proliferation in the β-TCP scaffold. The repaired tissues in the experimental group maintained their thickness to the full depth of the original defects, as compared with the negative control group (q=12.3370, P 〈0.01; q=31.5393, P 〈0.01). Conclusions Perfusion culture provided sustained nutrient supply and gas exchange into the center of the large scaffold. This perfusion bioreactor enables the chondrocytes and osteoblasts to survive and proliferate in a three-dimensional scaffold.展开更多
In bone tissue engineering,scaffolds with excellent mechanical and bioactive properties play prominent roles in space maintaining and bone regeneration,attracting increasingly interests in clinical practice.In this st...In bone tissue engineering,scaffolds with excellent mechanical and bioactive properties play prominent roles in space maintaining and bone regeneration,attracting increasingly interests in clinical practice.In this study,strontium-incorporatedβ-tricalcium phosphate(β-TCP),named Sr-TCP,bioceramic triply periodic minimal surface(TPMS)structured scaffolds were successfully fabricated by digital light processing(DLP)-based 3D printing technique,achieving high porosity,enhanced strength,and excellent bioactivity.The Sr-TCP scaffolds were first characterized by element distribution,macrostructure and microstructure,and mechanical properties.Notably,the compressive strength of the scaffolds reached 1.44 MPa with porosity of 80%,bringing a great mechanical breakthrough to porous scaffolds.Furthermore,the Sr-TCP scaffolds also facilitated osteogenic differentiation of mouse osteoblastic cell line(MC3T3-E1)cells in both gene and protein aspects,verified by alkaline phosphatase(ALP)activity and polymerase chain reaction(PCR)assays.Overall,the 3D-printed Sr-TCP bioceramic TPMS structured scaffolds obtained high porosity,boosted strength,and superior bioactivity at the same time,serving as a promising approach for bone regeneration.展开更多
