The...Since the 1960s, we have used acupuncture to induce ovulation for patients in the hospital in rural areas according to TCM theory that channels of the kidney, spleen, liver, Ren and Du are related to reproduction. The success rate was around 30-50%. Since then, we tired to study factors related to the effect. In the 1980s, the result was found to be closely related to endogenous estrogen level (P<0. 05), sympathetic reaction (P<O. 05), gonadotropic pulsatile secretion (P<0. 05) and central oploid activity (P<0. 05). Perlpheral estrogen levels may predict central opioid levels. Push-pull perfusion, in situ hybridization, immunohistochemistry, autoradiography and image procession were used to investigate the relationship between estradiol (E2) and hypothalamic ↓ -endorphinergic ( ↓ -EP) activity during the process of gonadotropin-releasing hormone (GnRH)-LH surge in rats and rabbits. Before ovulation, serum E2 surge (P <0. 01) was accompanied by a down-regulation on its receptor ER and proopiomelanocortin (POMC) mRNA level (P <0. 01) in the arcuate nucleus in rats. Pulsatile secretion of ↓-EP and fluctuation of μ-receptor density in MBH and MPO in rabbits (P<0. 01 ) appeared after cupric acid was injected transvenoulsy while at nadir leves GnRH-LH surge followed and ovulation resulted. Central GnRH levels and μ-receptor densities in MBH and MPO did not show change when Ez was injected alone but fluctuated and ovulation followed after acupuncture. Puberty is the time for maturity of GnRH pulsatile secretion that is related to central opioid activity. Serum Ez level in patients with pubertal dysfunctional uterine bleeding were higher than in patients with pubertal amenorrhea. Ovulation rate induced by acupuncture was 86.7% in the former group and 60% in the latter group (P <0. 05% ). It suggests that Ez level is important for the synthesis of central ↓-endorphin and that only with this can acupuncture induce hypothalamic pulsatile release of ↓ -endorphin to cause GnRH-LH surge and ovulation.展开更多
