In this stud, we investigated the anti-tumor activity of CIK cells in vitro, and conducted a preliminary investig...OBJECTIVE Cytokine-induced killer (CIK) cells are T-cells that display effective anti-tumor activity. In this stud, we investigated the anti-tumor activity of CIK cells in vitro, and conducted a preliminary investigation using autologous CIK cells to treat glioma patients through local administration. METHODS The CIK cells were derived from peripheral blood monocytes (PBMCs) of the glioma patients. The anti-tumor activity of the CIK cells against human T98-G glioma cell was tested in vitro. In addition, the autologous CIK cells were locally administrated into the tumor cavity in the malignant glioma patients through an Ommaya reservoir which was pre-inserted during tumor resection. The 4 x 108 CIK cells in a 5 ml suspension were injected once a week 2 times per cycle. Five hundreds KU of IL-2 was injected every other day. RESULTS (i) With incubation, the CIK cells showed dual staining of CD3^+CD56^+ with a positive rate of 3.45% on day 10 and 55.2% on day 30. In vitro anti-tumor activity (against T98-G cells) of the CIK cells reached the highest level after 18 days of incubation with different effector/target (E:T) ratios. (ii) Six patients received autologous CIK cell treatment (10 cycles). Two patients showed no recurrence and are still alive (24 and 10 months), while 4 cases had a recurrence 3 of which have died. The mean survival time from the first CIK cell treatment to the end of follow-up was 12.5 months. The main side-effects of the local CIK cell treatment was brain edema, which was controlled by mannitol in most of the cases. However for one patient injection of CIK cells and IL-2 had to be discontinued. CONCLUSION In vitro CIK cells are effective anti-glioma T-cells. Local therapy with CIK cells has potential anti-glioma efficacy and tolerable side-effects.展开更多

Methods: The clinical and pathologic features in six cases of PMA were analyzed. Immunohisto- chemical staining for glial fibrillary acidic protein (GFAP), synaptophysin (Syn), Chromogranin A (CgA), cytokeratin (AEI/AE3), epithelial membrane antigen (EMA) and Ki67 was performed on paraffin-embedded sections. Results: Among the six cases, five occurred in female patients, one was male, the age at diagnosis ranged from 2 to 15 years. Four cases were located in the hypothalamic area and optic pathway, one case in the third ventricle, and one case in left parietal lobe. On imaging, PMAs often appears as well-circumscribed mass. Microscopically, the tumor was composed of monomorphous bipolar (piloid) cells setting in a prominent myxoid background with an angiocentric radiating growth pattern in some areas. PMA lacked biphasic pattern, Rosenthal fibers and eosinophilic granular bodies which were usually typical in a classic pilocytic astrocytoma (PA). Immunohistochemcal study showed that the tumor cells were diffusely positive for GFAP. Syn positive staining was observed in one case. The Ki67 labeling index measured less than 5%. Conclusion: PMA is a distinct aggressive variant of pilocytic astrocytoma with special histological and immunohistochemical features. It is typically a rare tumor of early childhood. Im- munohistochemical staining for GFAP and Syn is helpful in differential diagnosis.展开更多

Therefore, DC-based immunotherapy has been one of the hotspots in cancer immunotherap...As professional antigen presenting cells, dendritic cells(DCs) greatly determine the quality of the innate and adaptive immunities. Therefore, DC-based immunotherapy has been one of the hotspots in cancer immunotherapy in recent years. Although this unique therapeutic strategy has been approved by U.S. Food and Drug Administration for prostate cancer treatment, the efficacy of DC-based immunotherapy remains to be further improved. Moreover, it is still not completely clear about the immunological basis of DCs, which is another hurdle for the progress of DC-based immunotherapy. Due to their unique physicochemical properties, nanomaterials have shown potentials in addressing these above mentioned problems and have provided important guidelines for optimizing DC-based immunotherapy. However, it is still at the starting stage for this emerging field and there are many critical questions in the rational design of this therapeutic strategy to be answered. Therefore, it is greatly necessary to review and analyze recent progresses in this field. In this review, we mainly focus on the development of various types nanoparticles for DC-based immunotherapy. Psoriasis is one type of autoimmune chronic skin diseases that is differentiated by T-Cells mediated hyper-proliferation of epidermal Keratinocytes. Dendritic Cells and CD8+ T-Cells have a significant role for the occurrence of this disease. In this paper, the authors have developed a mathematical model of Psoriasis involving CD4+ T-Cells, Dendritic Ceils, CD8+ T-Cells and Keratinocyte cell populations using the fractional differential equations with the effect of Cytokine release to observe the impact of memory on the cell-biological system. Using fractional calculus, the authors try to explore the suppressed memory, associated with the cell-biological system and to locate the position of Keratinocyte cell population as fractional derivative possess non-local property. Thus, the dynamics of Psoriasis can be predicted in a better way using fractional differential equations rather than its corresponding integer order model. Finally, the authors introduce drug into the system to obstruct the interaction between CD4+ T-Cells and Keratinocytes to restrict the disease Psoriasis. The authors derive the Euler-Lagrange conditions for the optimality made through Matlab by developing iterative of the drug induced system. Numerical simulations are schemes.展开更多
