[ Method ] In the process of screening and identification of Bar-transgenic rice, a biomass mutant was found in 10 lines of T1 progenies. The mutant was investigated for genetic analysis and agronomic traits by herbicide spraying and PCR amplification. [ Result] The segregation ratio is consistent with mendelian law(3:1). The mutant assumed not only higher plant height, wider straw and earlier florescence, but also more tillers, bigger spikes and resultantly higher biomass. PCR detections indicated that no co-segregation was observed between mutant traits and target gene(Bar) in the T-DNA inserted, proving that the mutant is not caused by the insertion of T-DNA containing target gene (Bar). [ Conclusion] Our study may avail to understand the cloning of mutant gene and the mechanism of the mutant gene on biomass.展开更多
According to the bio-characteristics of the lower and upper cavity surfaces of dental restoration, a stitching approach is proposed based on a virtual zipper working mechanism and a minimization of the surface total c...According to the bio-characteristics of the lower and upper cavity surfaces of dental restoration, a stitching approach is proposed based on a virtual zipper working mechanism and a minimization of the surface total curvature energy, which is used to resolve the stitching problems existing during computer-aided design for dental restorations. First, the two boundaries corresponding to the lower and upper surfaces are triangulated based on the zipper working mechanism to generate the initial stitching surface patch, of which the edges are distributed uniformly between the boundaries. Secondly, the initial stitching surface patch is subdivided and deformed to reconstruct an optimized surface patch according to the bio-characteristics of the teeth. The optimized surface patch is minimally distinguishable from the surrounding mesh in smoothness and density, and it can stitch the upper and lower cavity surfaces naturally. The experimental results show that the dental restorations obtained by the proposed method can satisfy both the shape aesthetic and the fitting accuracy, and meet the requirements of clinical oral medicine.展开更多
Heterosis plays an important role in the development of new crop varieties with high-yielding, good-quality and biotic/abiotic stresses while male sterile line de- velopment is the key step to determine the success of...Heterosis plays an important role in the development of new crop varieties with high-yielding, good-quality and biotic/abiotic stresses while male sterile line de- velopment is the key step to determine the success of heterosis utilization. A male sterile mutant, M207A was created in proso millet (Panicum mi/iaceurn, 2n=4x=36) for the first time using 60Co-y ray mutagenesis. Fertility identification and genetic analysis were carried out to characterize the mutant for its possible use for hetero- sis utilization in proso millet. First the sterility was investigated using both field sur- vey and indoor pollen microscopy identification. Then Pollinated by normal fertile proso millet cultivars, F1 and F2 populations from the mutant were obtained. Mean- while primary genetic analysis was also conducted using above populations in dif- ferent experimental sites, seasons and years. The results showed that the male sterile plant exhibited closed glumes, browning and dry anthers with few normal pollens. The sterility was stable and sterility rate was above 95% on average. The segregation ratio of fertile to sterile plants was 35:1 in the fertile selfing F2 popula- tion indicating that the mutant was a genic male sterility belonging to a pollen-less type controlled by a single recessive gene. The creation of the mutant, M207A can play a key role for heterosis utilization in proso millet.展开更多

In both DC and AC electric fields, the ERF material showed a distinct ER effect. Especially, in the alternating electric field, the shear stress of this material versus AC voltage has a better quadratic relation than that of the other materials. The experimental data showed that organic semiconductor polymers with big π bond conjugate system are a new type of electrosensitive particle materials which are worth well developing.展开更多

Forty-two putative low-k(+)-tolerant (lkt) mutants were selected from 150 000 tested M-2 seedlings, and two of these mutants maintained their low-K+-tolerant phenotype in their M-3 generations, respectively. The results showed that the absorption spectra of phycoerythrins ( PE) from different material changed significantly, while those of phycocyanins (PC) and allophycocyanins (APC) were basically similar. In order to disclose the essence of die difference, partial sequences of die subunit genes of PE of Qingdao strain of G. lemaneiformis (qd) and its pigmental mutants were determined. The amino acid sequences were deduced and used to explain spectral shifts of PE from the pigmental mutants. The amino acid sequences of PE resembled each other, and several residues changed among qd and its pigmental mutants. Residue substitutions were found in a region consisting of amino acids which determined are secondary structure and subunits interactions, thus might influence the confirmation and interaction of subunits, and further caused spectral deviation.展开更多
To improve the understanding of coupling effect between dynamic recrystallization(DRX)behaviors and flow behaviors of as-cast AlCu4 SiMg, a finite element(FE) simulation equipped with the models of DRX evolution was i...To improve the understanding of coupling effect between dynamic recrystallization(DRX)behaviors and flow behaviors of as-cast AlCu4 SiMg, a finite element(FE) simulation equipped with the models of DRX evolution was implemented. A series of isothermal compression tests were performed primarily on a Gleeble-3500 thermo-mechanical simulator in a temperature range of 648-748 K and a strain rate range of 0.01-10 s-1.According to the measured true stress-strain data,the strain hardening rate curves(dσ/dε versus σ) were plotted to identify the critical strains for DRX initiation(εc). By further derivation of the related material constants, the DRX volume fraction equation and the strain for 50% DRX(ε0.5) equation were solved. Accordingly, the aforementioned DRX equations were implanted into the FE model to conduct a series of simulations for the isothermal compression tests. The results show that during the evolution of DRX volume fraction at a fixed strain rate, the strain required for the same amount of DRX volume fraction increases with decreasing temperature. In contrast, at a fixed temperature, it increases with increasing strain rate. Ultimately, the DRX kinetics model of AlCu4 SiMg alloy and the consequence of the FE analysis were validated by microstructure observations.展开更多

The plastic behavior of the extruded alloys with compositions of Mg99.2Zn0.2Y0.6 and Mg89Zn4Y7 (molar fraction, %), which were almost the same compositions of Mg matrix phase and LPSO phase in Mg97Zn1Y2 Mg/LPSO two-phase alloy, respectively, were also prepared. By comparing their mechanical properties, the strengthening mechanisms operating in the Mg97Zn1 Y2 extruded alloy were discussed. Existence of the LPSO-phase strongly enhanced the refinement of Mg matrix grain size during extrusion, which led to a large increment of the strength of alloy. In addition, the LPSO-phases, which were aligned along the extrusion direction in Mg97Zn1Y2 extruded alloy, acted as hardening phases, just like reinforced fibers.展开更多

METHODS: Microsatellite markers were ...AIM: To establish the role of human T Cell Factor-4 (hTCF-4) gene exons 3-9 mutation status in association with sporadic rectal cancer with microsatellite instability (MSI). METHODS: Microsatellite markers were genotyped in 93 sporadic rectal cancer patients. Eleven cases were found to be high-frequency MSI (MSI-H). Sequence analysis of the coding region of the exons 3-9 of hTCF-4 gene was carried out for the 11 MSI-H cases and 10 controls (5 microsatellite stability (MSS) cases and 5 cases with normal mucosa). The sequencing and MSI identification were used. RESULTS: Several novel mutations and variants were revealed. In exon 4, one is a 4-position continuous alteration which caused amino acid change from Q131T and S132I (391insA, 392 G 〉 A, 393 A 〉 G and 395delC) and another nucleotide deletion (395delC) is present in MSI-H cases (5/10 and 4/10, respectively) but completely absent in the controls.CONCLUSION: Novel mutations in exon 4 of hTCF-4 gene were revealed in this study, which might be of importance in the pathogenesis of sporadic rectal cancer patients with MSI-H.展开更多

In this study, two strains of Nannochloropsis oceanica, the wild and the Zeocin-tolerant strains, were re-sequenced to verify such function of Zeocin, The results showed that Zeocin can mutate the N. oceanica genome and cause the structural variation. Zeocin either swept away or selected the alleles of genes functioning in ubiquitin-mediated proteolysis, alpha-linolenic acid metabolism, ascorbate and aldarate metabolism, ribosome biogenesis, and circadian rhythm, indicating that N. oceanica may have adjusted its metabolic performances for protein, carbohydrate, and lipid, and changed its ribosome biosynthesis and living rhythm to survive in Zeocin containing medium. In addition, Zeocin caused mutation may have influenced the expression of a set of tanscription factors. It was concluded that Zeocin effectively caused the structural variation of the genome of N. oceanica, and forced the microalgae to select out the alleles of a set of genes around these variations in order to adapt to Zeocin containing medium. Further studies on the genetic basis of the phenotypic adaptation of this haploid and asexual microalga and the application of Zeocin to its genetic improvement are very important.展开更多

The short term strain distributions of ship hull structural components are id...A method of reliability analysis based on a direct strength calculation employing the von Mises stress failure criterion is presented here. The short term strain distributions of ship hull structural components are identified through the statistical analysis of the wave-induced strain history and the long term distributions by the weighted summation of the short term strain distributions. The wave-induced long term strain distribution is combined with the still water strain. The extreme strain distribution of the response strain is obtained by statistical analysis of the combined strains. The limit state function of the reliability analysis is based on the von Mises stress failure criterion, including the related uncertainties due to the quality of the material and model uncertainty. The reliability index is calculated using FORM and sensitivity analysis of each variable that has effects on the reliability is also discussed.展开更多
