The results suggest...Detailed circular dichroism(CD) and Fourier transform infrared(FTIR) studies have been carried out to monitor thermal unfolding of horseradish peroxidase isoenzyme C(HRP) inhibited by CN -(HRP CN). The results suggest that HRP CN is quite different from native HRP with different spin states of Fe of heme and different coordinated states. Cyanide becomes the sixth ligand of Fe(Ⅲ) of heme and the hydrogen binding network is destroyed partly at the same time, which cause the drastic decrease of thermal stability of HRP. The FTIR and Soret CD spectra analysis demonstrate that during the heating process there is an intermediate state(I) which has both partly destroyed secondary and tertiary structures of native HRP, then it is the appearance of protein aggregation state(A) after fully unfolding. 目的:为不同类型中药小分子体外筛选及设计合成改造提供参考。方法:以"圆二色光谱""中药小分子与DNA""中药小分子与蛋白质""Circular dichroism""Interaction of traditional Chinese med...目的:为不同类型中药小分子体外筛选及设计合成改造提供参考。方法:以"圆二色光谱""中药小分子与DNA""中药小分子与蛋白质""Circular dichroism""Interaction of traditional Chinese medicine and DNA""Interaction of traditional Chinese medicine and protein"等为关键词,组合查询2005年1月-2016年4月在PubMed、Web of Science、Wiley Online Library、Springer Link、中国知网、万方、维普等数据库中的相关文献,对圆二色光谱(CD)在中药小分子与DNA、中药小分子与蛋白质相互作用中的应用进行综述。结果与结论:共检索到相关文献349篇,其中有效文献54篇。生物碱、黄酮类、醌类、苯丙素类、植物色素等类型的中药小分子,与DNA相互作用以沟槽结合和嵌插结合2种模式为主;苯丙素类、黄酮类、萜类、生物碱、醌类等类型的中药小分子,与蛋白质结合后,通常使其α-螺旋、β-转角和无规则卷曲等二级结构发生改变。后续进行诱导圆二色谱的探索性研究,可得到更多两者相互作用的结合信息,如中药小分子与DNA的结合序列及与蛋白质的结合位点;还可将CD与紫外-可见吸收光谱法、荧光光谱、分子模拟等技术联用,以实现多种技术互为验证、互为补充。展开更多
