一株易转化、高生物量地衣芽孢杆菌的筛选与鉴定

【目的】地衣芽孢杆菌(Bacillus licheniformis)是发酵工业中表达异源蛋白的重要底盘细胞,筛选易转化和高生物量的B.licheniformis菌株,为有效提升宿主的改造效率和蛋白表达水平提供菌种资源。【方法】经过富集、gyrB扩增与16S rDNA序... 【目的】地衣芽孢杆菌(Bacillus licheniformis)是发酵工业中表达异源蛋白的重要底盘细胞,筛选易转化和高生物量的B.licheniformis菌株,为有效提升宿主的改造效率和蛋白表达水平提供菌种资源。【方法】经过富集、gyrB扩增与16S rDNA序列分析从土壤中筛选与地衣芽孢杆菌同源性较高的菌株,将所筛菌株的电转效率和生物量与工业常用底盘细胞B.licheniformis 2709进行比较分析。对筛选出的易电转和高生物量菌株进行形态学与生理生化鉴定,并进行抗生素敏感性测定和高温碱性环境生长情况测定。【结果】筛选出10株与地衣芽孢杆菌同源性较高的菌株,其中菌株1-33电转化效率为6700 CFU/μg DNA,是B.licheniformis 2709的13.7倍。在豆粕玉米粉半固体培养基和SR液体培养基中培养,菌株1-33最大生物量达到5.2×10^(10) CFU/mL和6.8 g/L,分别为B.licheniformis 2709的1.71倍和1.3倍。进一步经过形态学、生理生化特性实验将菌株1-33最终鉴定为B.licheniformis,并表现出多种常用抗生素的敏感性和良好的耐碱与耐高温的能力。【结论】筛选出一株易电转和高生物量的B.licheniformis 1-33,为开发成高效表达外源蛋白的宿主菌奠定基础。 展开更多

Glut-4 is translocated to both caveolae and non-caveolar lipid rafts, but is partially internalized through caveolae in insulin-stimulated adipocytes 被引量:6

Caveolae and non-caveolar lipid rafts are two types of membrane lipid microdomains that play important roles in insulin-stimulated glucose uptake in adipocytes. In order to ascertain their specific functions in this process, caveolae were ablated by caveolin-1 RNA interference. In Cav-1 RNAi adipocytes, neither insulin-stimulated glucose uptake nor Glut-4 (glucose transporter 4) translocation to membrane lipid microdomains was affected by the ablation of caveolae. With a modified sucrose density gradient, caveolae and non-caveolar lipid rafts could be separated. In the wild-type 3T3- L l adipocytes, Glut-4 was found to be translocated into both caveolae and non-caveolar lipid rafts. However, in Cav1 RNAi adipocytes, Glut-4 was localized predominantly in non-caveolar lipid rafts. After the removal of insulin, caveolaelocalized Glut-4 was internalized faster than non-caveolar lipid raft-associated Glut-4. The internalization of Glut-4 from plasma membrane was significantly decreased in Cav-1 RNAi adipocytes. These results suggest that insulin-stimulated Glut-4 translocation and glucose uptake are caveolae-independent events. Caveolae play a role in the internalization of Glut-4 from plasma membrane after the removal of insulin. 展开更多

Based on an empirical study of macroeconomic data for 284 prefecture-level cities, this paper argues that the dual economy transition in China involves the coexistence of urbanization, industrialization and open economy. However, China has not yet arrived at the Lewisian Turning Point, because wage gaps between different industries and regions still exist and surplus labor is still being attracted to the labor-intensive manufacturing and tertiary industries in the eastern provinces. External economic shock and the mismatch between industrialization and urbanization are the primary causes of the coexistence of labor surplus and labor shortage. 展开更多

Among these organocatalysts, Lewis base-CO_2 adducts(LB-CO_2) were found to be more efficient.The used Lewis base has great effect on the catalytic activity of its CO_2 adduct. This review reports the recent progress in LB-CO_2 adducts catalyzed the cyclization of CO_2 with epoxides or aziridines to afford cyclic carbonates or oxazolidinones,the carboxylation of CO_2 with propargylic alcohols to α-alkylidene cyclic carbonates, and the reduction of CO_2 to methanol,formamides and methylamines, with the focus on the catalytic mechanism. 展开更多
