在大鼠基因组数据库中搜索得到两个泌乳刺激素基因家族的新成员。进一步分析显示该基因家族起源于啮齿目和其他哺乳动物分歧之后,而且大部分基因座位的重排在大、小鼠分歧之前已经完成。但PL-I和PL-II基因簇却是例外,它们在基因树上以... 在大鼠基因组数据库中搜索得到两个泌乳刺激素基因家族的新成员。进一步分析显示该基因家族起源于啮齿目和其他哺乳动物分歧之后,而且大部分基因座位的重排在大、小鼠分歧之前已经完成。但PL-I和PL-II基因簇却是例外,它们在基因树上以物种特异的方式聚类。结合基因转换的检验、染色体上相对位置比较和基因重复时间估计的结果,认为啮齿目PL-I和PL-II基因是物种特异的,它们由一系列在大、小鼠分歧之后发生的基因重复事件形成。结果还揭示了在啮齿目泌乳刺激素基因家族进化过程中持续不断的发生了基因重复和基因分化事件。 展开更多

The Change of Serum Prolactin Level after Multiple Injection of Norethisterone Enanthate and Estradiol Valerate

Sixteen healthy fertile women were recruited in a longitudinal study,and monthly given an injectable containing Norethisterone Enanthate (NET-EN) 50mg and Estradiol Valerate (EV) 5mg on the 5th day of menstrual cycle for 12 months.Blood samples were collected from each subject prior to and on day 1,7,21 after the 1st,6th and 12th injection.Serum Prolactin (PRL) level was measured by RIA.The PRL level of each cycle in almost every individual showed an obvious increase after injection of NET-EN/EV,followed by a gradual decrease from the middle of cycle length,then returned to its basal level.The profile of PRL level for each treated cycle could be well described by polynomial equation.The results indicate that there is no obvious effects and constant increase tendency on PRL level after long-term (1 year) injections of NET-EN 50mg/EV 5mg in healthy women. 展开更多

[Objective]The aim of this study was to explore the technical system of induced expression in vitro of goat mammary gland epithelial cell,and evaluate expression efficiency of mammary gland specific vector and foreign protein at the cell level.[Method]Goat mammary gland epithelial cell transfected by human lactoferrin gene was inducted by culturing in DMEM/F12 medium supplemented with 5 mg/L insulin,5 mg/L prolactin and 1 mg/L hydrocortisone.Supernatant was collected per 6 hours and concentrated.Expression situation of foreign protein were detected by SDS-PAGE and Western blotting.[Result]There was target protein expression in the induced culture medium,which molecular weight was about 42 kD.[Conclusion]The method used in this study can induce goat mammary gland epithelial cell to express foreign gene,it lays a foundation for researching heterologous expression of foreign gene and producing mammary gland bioreactor. 展开更多

Episodic evolution of prolactingene in primates

In the present study, we obtained exon 2―5 of prolactin (PRL) gene from four primate species by PCR and sequencing. Adding other genes available in GenBank, we calculate amino acid substitution rates for prolactin gene in primate. Comparison of nonsynonymous substitution rate to synonymous substitution rate ratios shows no evidence of positive selection for any lineage of primate prolactin gene. According to this and the facts that (i) no sites under positive selection are inferred by using maximum-likelihood method; (ii) among 32 amino acid replacement that occurred along the rapid evolutionary phase, only two are included in the 40 functionally important residues, indicating that amino acid replacement tends to occur in those functionally unimportant residues; (iii) partial of prolactin function is replaced by pla-cental lactogen in primate at the rapid evolutionary phase of prolactin gene, we thus deem that it is relaxation of purifying selection to some extent rather than positive selection that enforces the rapid evolution of primate prolactin gene. 展开更多
