A green fluorescent protein (GFP) expressing plasmid pRP22-GFP was constructed for monitoring the colonization of two biocontrol agents, Brevibacillus brevis ZJY-1 and Bacillus subtilis ZJY-116, on the spikes of barley and their effect on suppression of FHB. Survival and colonization of the Brevibacillus brevis ZJY-1 and Bacillus subtilis ZJY-116 strains on spikes of barley were observed by tracking the bacterial transformants with GFP expression. Our field study revealed that plasmid pRP22-GFP was stably maintained in the bacterial strains without selective pressure. The retrieved GFP-tagged strains showed that the bacterial population fluctuation accorded with that of the rain events. Furthermore, both biocontrol strains gave significant protection against FHB on spikes of barley in fields. The greater suppression of barley FHB disease was resulted from the treat-ment of barley spikes with biocontrol agents before inoculation with F. graminearum.展开更多

Biological control is usually the first choice of control and prevention method for integrated pest management (IPM) strategies and has now been widely implemented by Indonesian oil palm plantations. Entomopathogeni...Biological control is usually the first choice of control and prevention method for integrated pest management (IPM) strategies and has now been widely implemented by Indonesian oil palm plantations. Entomopathogenic fungus, i.e., Metarhizium anisopliae, Cordyceps militaris and Beauveria bassiana have been demonstrated to control renowned pests of oil palm. Metarhizium has been used to control Oryctes larvae and the mortality has ranged from 91.67% to 100% in laboratory and 7.4% to 88.75% in the field. Metarhizium has been applied in combination with a termite baiting system (TBS) to control termites in the field for preventive and curative action as well. In many oil palm plantations in Indonesia, Cordyceps has been used to reduce the field moth population ofSetothosea asigna. Application of Cordyceps within the oil palm circle was able to infect S. asigna pupae up to 80%. Meanwhile, Beauveria in an effervescent formulation was demonstrated to have better efficacy on Darna trima larvae. A significant finding on the biological control of basal stem rot disease (Ganoderma) was the isolation of Trichoderma sp. and Gliocladium sp.. The efficacy was conducted with promising result and techniques on the application of Triehoderma have been developed, i.e., hole-in-hole system, surgery and a mounding method. However, as the roots developed, Trichoderma was no longer able to protect the palm from Ganoderma. In spite of that, the use of Trichoderma still prolonged the life ofoil palms by up to 2-3 years. Another fungi belonging to vesicular arbuscular mychorrhiza (VAM) has been developed to control Ganoderma. The efficacy in the nursery showed promising results and the Ganoderma incidence remained low compared to the untreated control. Large scale field trials are ongoing. Challenges on the implementation of biological control in oil palm plantations are because of application and availability of biopesticides/natural enemies. The threshold values for the existence and s...Based on the facts of releasing natural enemies and spraying pesticides at different time points, we propose a generalized predator-prey model with impulsive interventions. The threshold values for the existence and stability of pest eradication periodic solution are provided under the assumptions of releasing natural enemies either more or less frequent than spray. In order to address how the different pulse time points, control tactics affect the pest control (i.e. the threshold value), the Holling Type II Lotka-Volterra predator- prey system, as an example, with impulsive intervention at different time points axe investigated carefully. The numerical results show how the threshold values are affected by the factors including instantaneous killing rates of pesticides on pests and natural enemies, the release rate of natural enemies and release constant, timing of pesticide application and timing of release period. Furthermore, it is confirmed that the system has the coexistences of pests and natural enemies for a wide range of parameters and with quite different pest amplitudes.展开更多

For the model with fixed impulsive effects, the global stability of pest eradi- cation periodic solution and the dominance of dynamic behavior are investigated. This indicates that the model with fixed moments has the potential to protect the natural enemies from extinction, but under some conditions may also serve to extinction of the pest. The second model is constructed according to the practices of IPM, that is, when the pest population reaches the economic injury level, a combination of biological, cultural, and chemical tactics that reduce pests to tolerable levels is used. Numerical investigations imply that there are several different types of periodic solutions and their maximum amplitudes are always less than the given economic threshold. The results also show that the time series at which the IPM strategies are applied are quite complex, which means that the application and realization of IPM in practice are very difficult.展开更多
