目的探讨白介素⁃17A(interleukin⁃17A,IL⁃17A)对大鼠支气管成纤维细胞(bronchial fibro⁃blasts,BF)增殖、自噬及自噬后胶原降解的影响。方法采用酶联合消化法+组织块贴壁法培养BF并鉴定,CCK⁃8法检测细胞增殖能力;实验分组:对照组、雷帕... 目的探讨白介素⁃17A(interleukin⁃17A,IL⁃17A)对大鼠支气管成纤维细胞(bronchial fibro⁃blasts,BF)增殖、自噬及自噬后胶原降解的影响。方法采用酶联合消化法+组织块贴壁法培养BF并鉴定,CCK⁃8法检测细胞增殖能力;实验分组:对照组、雷帕霉素组、雷帕霉素+IL⁃17A组、IL⁃17A组;透射电镜观察细胞自噬泡的形成情况;Western blot检测BF中自噬标记蛋白LC3、p62的表达;ELISA法检测各组中基质金属蛋白酶⁃1(matrix metalloproteinase⁃1,MMP⁃1)与基质金属蛋白酶抑制剂(matrix metalloproteinase inhibitor,TIMP⁃1)的表达。结果IL⁃17A浓度为20 ng/mL时BF细胞增殖能力最佳。IL⁃17A组中自噬泡数量明显减少,LC3⁃Ⅱ/LC3⁃I的比值及MMP⁃1的表达水平明显下调,p62及TIMP⁃1的表达水平明显上调(P<0.05)。雷帕霉素部分逆转了IL⁃17A的作用。结论(1)IL⁃17A可促进BF细胞增殖;(2)IL⁃17A通过抑制自噬,发挥抑制胶原降解的作用,促进气道重塑的发生发展。 展开更多

目的通过统计学方法分析白介素⁃17与骨质疏松症的相关性。方法检索中国知网、中国生物医学文献检索数据库、万方科技期刊全文数据库、维普中文科技期刊全文数据库、美国国立图书馆、Web of Science、EMBASE、Cochrane图书馆,查找白介素... 目的通过统计学方法分析白介素⁃17与骨质疏松症的相关性。方法检索中国知网、中国生物医学文献检索数据库、万方科技期刊全文数据库、维普中文科技期刊全文数据库、美国国立图书馆、Web of Science、EMBASE、Cochrane图书馆,查找白介素⁃17与骨质疏松症相关性的研究文献,检索时间段为建库至2019年6月1日。采用RevMan 5.3软件进行统计学分析。结果初步检索出732篇文献,最终纳入14篇,共涉及1150人。结果显示绝经后骨质疏松症患者血清白介素⁃17水平高于绝经后骨量减少患者及骨质正常者[标准化均数差=3.1,95%可信区间(1.46,4.73),P=0.0002;标准化均数差=0.71,95%可信区间(0.37,1.05),P<0.0001]。类风湿关节炎患者骨量水平与血清白介素⁃17水平相关性不显著[均数差=6.15,95%可信区间(-2.61,14.91),P=0.17]。结论血清白介素⁃17水平升高可能与绝经后骨质疏松症相关,仍需更多大样本量、高质量的研究探索白介素⁃17表达与骨质疏松发生的关系及机制。 展开更多

健脾润肺丸对肺结核患者β⁃CAR、VEGF、IFN⁃γ、IL⁃17水平的影响

目的研究健脾润肺丸对肺结核患者β⁃胡萝卜素(β⁃CAR)、血管内皮生长因子(VEGF)、干扰素⁃γ(IFN⁃γ)、白介素⁃17(IL⁃17)水平的影响。方法以2020年1月至2023年4月张家口市肺科医院收治的肺结核患者120例为研究对象,采用随机数字表法分为... 目的研究健脾润肺丸对肺结核患者β⁃胡萝卜素(β⁃CAR)、血管内皮生长因子(VEGF)、干扰素⁃γ(IFN⁃γ)、白介素⁃17(IL⁃17)水平的影响。方法以2020年1月至2023年4月张家口市肺科医院收治的肺结核患者120例为研究对象,采用随机数字表法分为进行常规治疗的常规组(60例)和联合健脾润肺丸治疗的联合组(60例),两组均治疗90 d。比较两组疗效、临床症状、体征改善情况及血清细胞因子水平(β⁃CAR、VEGF、IFN⁃γ、IL⁃17)、免疫功能(CD_(3)^(+)、CD_(4)^(+)、CD_(8)^(+)、CD_(4)^(+)/CD_(8)^(+))、肺功能[用力呼气量(FEV1)、用力肺活量(FVC)、FEV1/FVC]。结果治疗90 d后,联合组总有效率高于常规组,差异有统计学意义(P<0.05)。联合组咳嗽、咳痰、胸憋气短、咯血胸痛症消失时间、痰涂片抗酸杆菌连续3次阴转时间及肺部胸片病灶缩小时间均短于常规组,差异有统计学意义(P<0.05)。两组治疗90 d后血清β⁃CAR、VEGF、IL⁃17水平、外周血CD_(8)^(+)百分比低于治疗前,且联合组低于常规组,差异有统计学意义(P<0.05);血清IFN⁃γ水平、外周血CD_(3)^(+)、CD4^(+)百分比及CD_(4)^(+)/CD_(8)^(+)、FEV1、FVC、FEV1/FVC高于治疗前,且联合组高于常规组,差异有统计学意义(P<0.05)。结论健脾润肺丸可改善肺结核患者血清β⁃CAR、VEGF、IFN⁃γ、IL⁃17水平及免疫功能、肺功能,促进患者临床症状、体征改善,具有较好的治疗效果。 展开更多

Mucosal biopsy samples we... AIM: To determine the role of interleukin (IL)-17 in gastric ulcerogenesis. METHODS: Thirty-six gastric ulcer (GU) patients and 29 non-ulcer (NU) patients were enrolled in this study. Mucosal biopsy samples were obtained from the gastric antrum and GU site during endoscopy. Samples were used in in situ stimulation for 48 h in the presence of 10 ug/mL phytohemagglutinin-P (PHA), histological examination, and Helicobacter pylori(Hpylon) culture. IL-17 and IL-8 protein levels in culture supematants were assayed by ELISA. IL- 17 mRNA expression was analyzed by reverse transcriptasepolymerase chain reaction (RT-PCR). Hpylori cagA and vacA status was assessed by reverse hybridization using a line probe assay (UPA). IL-8 levels in culture supematants were assayed after AGS cells were co-cultured with Hpylori strain 26 695 or recombinant human (rh) IL-17. RESULTS: All 36 GU patients and 15 of 29 NU patients were found to be Hpy/or/-positive, while 14 NU patients were Hpylori-nogative. All 51 H pylori strains from both GU and NU patients were cagA- and vacAsl/ml-positive. Antral mucosal tissues from H pylori-positive patients contained significantly (H pylori-positive NU patients: median 467 pg/mg/protein, range 53-2 499; Hpylori negative NU patients: median 104 pg/mg/protein, range 16-312, P〈0.0005) higher levels of IL-17 than those from uninfected patients. IL-17 levels at the ulcer site were significantly (ulcer site: median 1 356 pcj/mg/protein, range 121-1 3730; antrum: median 761 pg/mg/protein, range 24-7 620, P〈0.005) higher than those at distant sites in the antrum. Biopsies from H pylori-positive GU and NU patients showed IL-17 mRNA expression in all samples whereas those from the antrum of the Hpylori-negativecontrols showed no detectable expression. A significant correlation was seen between IL-17 and IL-8 levels at each biopsy site (ulcer: r = 0.62,P〈0.0001; antrum: r = 0.61, P〈0.0001) in GU patients. RhIL-17 and Hpylori strain 26 695 each stimulated IL-8 production from AGS cells. CONCLUSION: IL-17 may play an important role in the inflammatory response to Hpyloricolonization, and may ultimately influence the outcome of H pylori-associated diseases that arise within the context of gastritis. 展开更多

AIM: To assess vitamin D (Vit D) abnormalities in hepatitis C infected patients and their relationship with interleukin (IL)-17, IL-23 and N-terminal propeptide of type Ⅲ pro-collagen (PⅢNP) as immune response mediators. METHODS: The study was conducted on 50 Egyptian patients (36 male, 14 female) with hepatitis C virus (HCV) infection, who visited the Hepatology Outpatient Clinic in the Endemic Disease Hospital at Cairo University. Patients were compared with 25 ageand sexmatched healthy individuals. Inclusion criteria were based on a history of liver disease with HCV genotype 4 (HCV-4) infection (as new patients or under followup). Based on ultrasonography, patients were classified into four subgroups; 14 with bright hepatomegaly; 11 with perihepatic fibrosis; 11 with hepatic cirrhosis; and 14 with cirrhosis and hepatocellular carcinoma (HCC).Total Vit D (i.e., 25-OH-Vit D) and active Vit D [i.e., 1,25-(OH) 2 -Vit D] assays were carried out using commercial kits. IL-17, IL-23 and PⅢNP levels were assayed using enzyme linked immunosorbent assay kits, while HCV virus was measured by quantitative and qualitative polymerase chain reaction. RESULTS: Levels of Vit D and its active form were significantly lower in advanced liver disease (hepatic cirrhosis and/or carcinoma) patients, compared to those with bright hepatomegaly and perihepatic fibrosis. IL-17, IL-23 and PⅢNP levels were markedly increased in HCV patients and correlated with the progression of hepatic damage. The decrease in Vit D and active Vit D was concomitant with an increase in viral load, as well as levels of IL-17, IL-23 and PⅢNP among all subgroups of HCV-infected patients, compared to normal healthy controls. A significant negative correlation was evident between active Vit D and each of IL-17, IL-23 and PⅢNP (r = -0.679, -0.801 and -0.920 at P < 0.001, respectively). HCV-infected men and women showed no differences with respect to Vit D levels. The viral load was negatively correlated with Vit D and active Vit D (r = -0.084 and -0.846 at P < 0.001, respectively), and positively correlated with IL-17, IL-23 and PⅢNP (r = 0.951, 0.922 and 0.94 at P < 0.001, respectively). Whether the deficiency in Vit D was related to HCVinduced chronic liver disease or was a predisposing factor for a higher viral load remains to be elucidated. CONCLUSION: The negative correlations between Vit D and IL-17, IL-23 and PⅢNP highlight their involvement in the immune response in patients with HCV-4related liver diseases in Egypt. 展开更多

AIM:To investigate the impact of different anesthetic techniques on T-helper(Th) cell subsets in hepatocellular carcinoma(HCC) patients undergoing hepatectomy.METHODS:Sixty-one HCC patients who received hepatectomies were randomized into an epidural combined general anesthesia(G + E;n = 31) or a general anesthesia(G;n = 30) group.Blood samples were obtained the morning before the operation(d0),and on the second(d2) and seventh(d7) day after the operation.Th cell contents were evaluated using flow cytometry,realtime reverse transcription-polymerase chain reaction and enzyme-linked immunosorbent assay.RESULTS:In all 61 patients,Th1 and Th2 cell frequencies,and interferon-(IFN-) mRNA expression markedly increased on d2,compared to d0.They recovered slightly on d7,and the Th1/Th2 ratio increased markedly on d7,compared with d2.In contrast,Th17,regulatory T cell(Treg),and interleukin-17(IL-17) levels and FOXP3 mRNA expression showed no significant change on d2,and then markedly decreased on d7.Similarly,plasma IFN-concentration on d2 was much higher than that on d0,and then partly recovered on d7.As compared with the G group,in the G + E group,Th1 cell frequencies and the Th1/Th2 ratio were slightly higher on d2 and significantly higher on d7,while Th2,Th17,and Treg cell frequencies were slightly lower on d2,and significantly lower on d7.Consistently,on d7,IFN-mRNA and protein levels and the IFN-/IL-4 ratio in the G + E group were higher than those in the G group.In contrast,the IL-17 mRNA level,and IL-17 and transforming growth factor-1 concentrations in the G + E group were lower than those in the G group.CONCLUSION:G + E is superior to G in shifting the Th1/Th2 balance towards Th1,while decreasing Th17 and Treg,potentially benefiting HCC patients by promoting anti-tumor Th polarization. 展开更多

Objective: To observe the regulatory effects of herbal cake-partitioned moxibustion on Crohn's disease (CD) rat's colon inflammatory cytokine interleukin-17 (IL-17), IL-23 and their mRNAs, and to investigate the action mechanism of moxibustion in treating CD. Methods: Forty SPF grade Sprague-Dauley (SD) male rats were randomly divided into a normal group, a model group, an herbal cake-partitioned moxibustion group and a Western medicine group, with 20 rats in each group. Except the normal group, rats in the other three groups were used to make CD model by giving an enema in colon with Trinitro-benzene-sulfonic acid (TNBS). When the models were successful made, rats in the model group had no therapeutic intervention; rats in the herbal cake-partitioned moxibustion group were subjected to herbal cake-partitioned moxibustion treatment at bilateral Tianshu (ST 25) plus O.ihai (CV 6); and rats in the Western medicine group were given oral Mesalazine. After treatment, the histopathological changes and inflammatory cytokines IL-17, IL-23 and their mRNAs expressions were observed in descending colon by hematoxylin-eosin (HE) staining, immunohistochemistry (IHC) and real-time polymerase chain reaction (RT-PCR) methods. Results: Colon tissues of TNBS enema rat models showed cracks-like ulcers accompanied by mucous layer inflammation, granulomas, and inflammatory cytokines IL-27, IL-23 and their mRNAs expressions were all higher than those in the normal group (P〈O.01); after intervention, colon tissue cracks-like ulcers and inflammation degree reduced, inflammatory cytokines IL-17, IL-23 and their mRNAs expressions were all significantly decreased (P〈0.01) in the herbal cake-partitioned moxibustion group and the Western medicine group. Conclusion: Herbal cake-partitioned moxibustion may improve colon tissue ulcers and relieve intestinal inflammation by down-regulation of IL-17, IL-23 and their mRNAs expressions in CD model rats' colonic mucosa. 展开更多
