Cloning and Functional Analysis of the Porcine Growth Hormone Gene Promoter 被引量:1

[Objective] This study aimed to clone the porcine growth hormone gene promoter and determine the core promoter sequences and the cis-acting elements. [Method] Sequence of the 5'flanking region of porcine growth hormone gene was searched out and downloaded from the NCBI website. According to the targeted se- quence, primers were designed and synthesized for the PCR amplification. The 1 882 bp (-1 821 bp-+61 bp) fragment was amplified by PCR. Nine promoter frag- ments with different lengths were obtained by genome-walking deletion method and then cloned into luciferase reporter vectors. Relative transcriptional activities of these 5' terminal-deleted plasmids in pituitary and non-pituitary cells were determined by transient transfection of the rat pituitary adenoma cell (GH3), porcine lilac endotheli- um cell (PIEC) and porcrne Kidney-15 (PK15) with the constructed dual-luciferase vectors. [Result] Result of DNA sequencing showed that the 1 882 bp fragment of GH 5' promoter was successfully cloned. Nine luciferase reporter gene plasmids were constructed. DuaI-Luciferase reporter assay indicated that the promoter inserted into reporter gene vector had very strong cell specificity. [Conclusion] Porcine growth hormone gene specifically expresses in pituitary cells. The minimal promoter of the porcine growth hormone gene is mapped at the region -110 bp-+61 bp. Promoter regions 218 bp--110 bp and -429 bp--218 bp contain positive regulatory elements. 展开更多

众多广视角技术中,高级超维场转换技术(Advanced Super Dimension Switch,ADS)具有高分辨率、高透过率、低功耗、宽视角、高开口率、低色差、无挤压水波纹(Push Mura)等优点。与垂直取向(Vertical Aligned,VA)模式相比,ADS模式存在对比... 众多广视角技术中,高级超维场转换技术(Advanced Super Dimension Switch,ADS)具有高分辨率、高透过率、低功耗、宽视角、高开口率、低色差、无挤压水波纹(Push Mura)等优点。与垂直取向(Vertical Aligned,VA)模式相比,ADS模式存在对比度(Contrast Ratio,CR)偏低的问题。本文主要从设计及材料两个方面对ADS显示模式超高清(Ultra High Definition,UHD)产品对比度提升进行研究。与传统的产品相比较,在设计方面采用了高透像素设计,在提升L255亮度的同时,保证L0亮度不变;同时开发了一种低散射液晶,降低L0亮度,明显改善对比度。将高透像素设计搭载55UHD和49UHD ADS产品进行了实验验证,将低散射液晶搭载49UHD ADS产品进行了实验验证。结果表明,采用高透像素设计的面板与传统设计面板相比,对比度提升约8.3%;采用高对比度液晶的面板的与传统设计面板相比,对比度约提升22%。二者共同作用,UHD产品对比度提升约30%。 展开更多

Methotis Diels-Alder reaction between furan and maleic anhydride afforded dehydronorcantharidin, and subsequent hydrogenation, phosphorylation, and basification gave compounds 1 and 2, separately. Resuits The structures of compounds were confirmed by IR, NMR and FAB-MS. The aqueous solubility of 1 and 2 were improved, compared with the parent compounds, and their activities were more potent than norcantharidin 4 against MGCS03 cell lines. Conclusion The phosphorylation of norcantharidin analogues is an effective way to increase the activity and solubility. 展开更多

Effect of Cultivation Conditionss on Quality Parameters of High Grade Hybrid Rape of Brassica napus L.

[Objective] This study aimed to investigate the combined effects of sowing date, plant density, nitrogenous fertilizer, phosphorus fertilizer and potassium fertilizer on quality parameters of high grade hybrid rapeseed. [Method] By using Youyan 599 and Sanbei 98 as experimental materials, quadratic orthogonal rotation combination with five factors was designed for this study. [Result] Erucic acid content was nega- tively correlated with sowing date and nitrogen fertilizer, positively correlated with plant density. Glucosinolate content was negatively correlated with sowing date, positively correlated with potash fertilizer amount. Oil content was negatively correlated with sowing date and nitrogen fertilizer amount, positively correlated with plant density and potash fertilizer amount. Protein content was positively correlated with sowing date and nitrogen fertilizer amount, negatively correlated with plant density and potash fer- tilizer amount. Oleic acid content was negatively correlated with phosphate fertilizer amount, positively correlated with nitrogen fertilizer amount. Palmitic acid was nega- tively correlated with potash fertilizer amount, positively correlated with sowing date, plant density, nitrogen fertilizer amount and phosphate fertilizer amount in Youyan 599, but negatively correlated with sowing date, plant density, nitrogen fertilizer amount and phosphate fertilizer amount in Sanbei 98. Stearic acid content was posi- tively correlated with sowing date, plant density, nitrogen fertilizer amount and phos- phate fertilizer amount. Linoleic acid was positively correlated with density and nitroge- nous fertilizer, but negatively correlated with potash fertilizer amount. Linolenic acid was positively correlated with nitrogen fertilizer amount and potash fertilizer amount. Eicosenoic acid was negatively correlated with sowing date. There were positive cor- relations and negative correlations among cultivation conditions and quality parame- ters, and the positive correlations and negative correlations could counteract each other. Expression of quality parameters for each treatment was the combined effect. Among the 50 pairs of simple correlations of five cultivation factors and ten quality parameters, 26 pairs were consistent between two varieties, 24 pairs of simple corre- lation were not. Sowing date, plant density, nitrogen fertilizer amount and phosphate fertilizer amount showed significant influence on erucic acid of two varieties, but small effect on palmitic acid. [Conclusion] The combined effect of the five cultivation factors could not fundamentally improve the quality traits of high grade hybrid rapeseed. 展开更多

[Method] The test-tube seedlings were produced from seeds of Atractylodis Lances. At the same time, the effects of 6-BA, NAA and KT on the proliferation of aseptic seedling was studied through the orthogonal test. Effect of NAA concentration on the rooting of aseptic seedling was studied through nine types of light substrate groups, which consist of peat soil, fine sand and cotton seed hull, and their effects on Atractylodes lances growth were studied on with orthogonal design. [Result] The culture medium suitable for the proliferation of the Atractylodes lances tube seedling was MS+6-BA 1.0 mg/L+NAA 0.4 mg/L+KT 0,4 mg/L. And the medium suitable for rooting was 1/2MS+NAA 0.5 mg/L. The optimal ratio of soil, riversand and cotton seed hull was 3:3:2. [Conclusion] The research selected optimum culture medium, rooting medium and ratio of substrate, which provided technical guidance for laboratorial preservation of germplasm resource of Atractylodes lances and industrialization of its seedling production. 展开更多

A set of formulations about inverse kinematics, Jacobin matrix, and forward kinematics for the high precision 6-HTRT parallel robots is presented. The analysis of errors existing in the manipulator is discussed and a novel approach for error correction is advanced. By DSP technique, inverse kinematics is solved in real time conditions with high precision and the hardware control system is given. Cavity resonance noise of passenger car tires is generated by interacting excitation between a tire structure and the fill gas (air), and generally lies in a frequency range of 200?250 Hz. As such, this noise is strongly perceived and may be a serious source of driver annoyance. Thus, many studies regarding the cavity noise mechanism and its reduction have already been conducted. In this work, a vibro-acoustic coupled analysis was conducted between a tire structure and air cavity. Using this analysis, we can more accurately simulate the tire noise performance in the region of the cavity resonance frequency. An analysis of the effects of variation of tire contour design factors was conducted, using design-of-experiments methods. Finally, a multi-objective optimization was performed using in-house codes to reduce the cavity noise level while minimizing the loss of other performances, such as diminished ride comfort and handling caused by the variations of contour. As a result of this optimization, an optimized contour shape was derived, which satisfied the multi-objective performances. 展开更多
