目的本研究旨在探索miR-645能否通过靶向SING来影响结直肠癌转移,进一步探索其作用机制。方法采用细胞实验和动物模型来研究miR-645在结直肠癌中的功能。我们使用实时定量聚合酶链反应(qPCR)技术检测了miR-645在结直肠癌细胞系和组织样... 目的本研究旨在探索miR-645能否通过靶向SING来影响结直肠癌转移,进一步探索其作用机制。方法采用细胞实验和动物模型来研究miR-645在结直肠癌中的功能。我们使用实时定量聚合酶链反应(qPCR)技术检测了miR-645在结直肠癌细胞系和组织样本中的表达水平。结果发现miR-645在结直肠癌组织中高度表达,并与肿瘤的恶性特征相关。miR-645过表达显著促进了结直肠癌细胞的迁移和侵袭能力,而miR-645沉默则具有相反的效果。进一步的研究揭示了miR-645通过抑制STING蛋白的表达来促进结直肠癌的转移过程。结论本研究结果表明,miR-645通过靶向STING调控结直肠癌转移。miR-645的上调与结直肠癌细胞的迁移和侵袭能力增强相关,同时也与肿瘤的恶性程度有关。这一研究结果有助于深入了解结直肠癌转移的分子机制,并为miR-645作为潜在治疗靶点提供了依据。 展开更多

Colorectal cancer(CRC)remains one of the major causes of death worldwide,despite steady improvement in early detection and overall survival over the past decade.Current treatment paradigms,with chemotherapy and biologics,appear to have reached their maximum benefit.Immunotherapy,especially with checkpoint inhibitors,has shown considerable clinical benefit in various cancers,including mismatch-repair-deficient CRC.This has led to the planning and initiation of several clinical trials evaluating novel immunotherapy agents—as single agents,combinations and in conjunction with chemotherapy—in patients with CRC.This article reviews biological and preclinical data for checkpoint inhibitors and discusses various immunotherapy trials in CRC,as well as current efforts in CRC immunotherapy. 展开更多
