Pall aerial tissue. Sscat1 (1.7 kb) contains a full open reading frame of 492 amino acids and Sscat2 (1.1 kb) is a partial clone. BLAST analysis indicates that the two clones share 71.9% identity in nucleotide sequence and 75% identity in deduced amino acid sequence within the last 287 amino acid residues of Sscat1. Southern blotting analysis showed that Sscat1 is multicopy in S. salsa genome, while Sscat2 is a single copy gene. Northern blotting analysis showed a rapid increase in the steady-level of both genes in roots after 48 It salt treatment, but only Sscat1 was induced in salinity treated leaves. Time-course analysis carried out in leaves confirmed that Sscat1 was induced by salt stress, in contrast to Sscat2. These implied that the expression of Sscat1 and Sscat2 genes are differentially regulated in S. salsa. The activity of total catalase is dramatically increased in response to salt stress.展开更多

We evaluated the effect of cadmium (Cd) on the defense responses of the Pacific oyster Crassostrea gigas to Listonella anguillarum challenge. The activities of several important defensive enzymes, including superoxide dismutase (SOD), glutathione peroxidase (GPx), acid phosphatase (ACP), Na+, K+-ATPase in gills and hepatopancreas, and phenoloxidase-like (POL) enzyme in hemolymph were assayed. In addition, the expression levels of several genes, including heat shock protein 90 (IrtSP9~)), metallothionein (MT), and bactericidal/permeability increasing (BPI) protein were quantified by fluorescent quantitative PCR. The enzyme activities of SOD, ACP, POL, and GPx in hepatopancreas, and the expression of HSP90 were down-regulated, whereas GPx activity in the gill, Na+, K+-ATPase activities in both tissues, and MT expression was increased in Cd- exposed oysters post L. anguillarum challenge. However, BPI expression was not significantly altered by co-stress of L. anguillarum infection and cadmium exposure. Our results suggest that cadmium exposure alters the oysters' immune responses and energy metabolism following vibrio infection.展开更多

Both extracts had high antioxidant properties and could inhibit FeE+ and sodium nitroprusside lipid peroxidation in the liver. Total phenol content of ethyl acetate fraction and aqueous extract were 4.50 ±2.26 and 1.21±3.12 mg/g, respectively. Both extracts had a maximal effect at the lowest concentrations tested (15 μg/mL for ethyl acetate and 150 μg/mL for the aqueous). Ethyl acetate fraction showed that the highest values of antioxidant activity is probably associated with its high phenolic content, Fe2+ chelating and DPPH scavenging activities. It is therefore concluded that ethyl acetate fraction and to a less extent the aqueous fractions are potent inhibitor of lipid peroxidation.展开更多

Histopathological examination of gill, liver and kidney of exposed fishes showed that gill was a target organ of butachlor. The butachlor seriously impaired the respiration of gills by a series of lesions such as edema, lifting and detachment of lamellar epithelium, breakdown of pillar cells, and blood congestion. The dysfunction of gill respiration caused suffocation to the exposed flounder with extremely high acute lethality. Antioxidant enzyme activity assay of the in vitro cultured flounder gill (FG) ceils exposed to butachlor indicated that butachlor markedly inhibited the antioxidant enzyme activities of Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX). Furthermore, along with the decline of antioxidant enzyme activities, ATP content in the exposed FG cells decreased, too. This infers that the oxidative stress induced by butachlor can inhibit the production of cellular ATP. Similar decrease of ATP content was also observed in the exposed flounder gill tissues. Taken together, as in FG cells, butachlor possibly induced a short supply of ATP in pillar cells by inhibiting the antioxidant enzyme activities and then affecting the contractibility of the pillar cells, which in turn resulted in the blood congestion and suffocation of exposed flounder.展开更多

Sperm motility and the activities of superoxide dismutase(SOD), catalase(CAT), glutathione peroxidase(GPx), glutathione reductase(Gr), and lipid peroxidation(measured via malondialdehyde(MDA) content) were determined after the milt was stored at 4°C for 12 h, cryopreserved without cryoprotectant in 12% propylene glycol(PG), cryopreserved in 12% PG+0.1 mol/L trehalose, or cryopreserved in 12% PG spermatozoa but centrifuged to decant the supernatant prior to cryopreservation(only sperm cells were cryopreserved). After chilled storage or cryopreservation, the SOD, CAT and GPx activities were reduced in sperm cells and increased in seminal plasma in almost all treatments; sperm motility parameters were also decreased. However, the addition of trehalose into the cryoprotectant could signifi cantly improve the postthaw sperm quality as revealed by the sperm average path velocity. This improvement might be attributed to the function of trehalose in scavenging reactive oxygen species. Chilled storage and cryopreservation had signifi cant eff ects on sperm motion characteristics, antioxidant enzyme activities, and lipid peroxidation in the Pacific cod.展开更多

Three antioxidant enzymes (superoxide dismutase, SOD; catalase, CAT; glutathione peroxidase, GPx), two immune defense enzymes (acid phosphatase, ACP; alkaline phosphatase, ALP), and one lipid peroxidation product (malondialdehyde, MDA) were measured in the gills and the hepatopancreas of the clam exposed to 0, 25, 75, and 125 μg/L cadmium for 0, 1, 3, 5, and 7 d. The results show that the concentrations of antioxidant enzymes in the organs soared to a peak value on the first day and then decreased afterwards in most cases. CAT and GPx activities in the hepatopancreas were higher than in the gills, but the SOD activity was lower in the hepatopancreas. ACP activity was unchanged until Day 3 in the hepatopancreas and until Day 5 in gills, when it began to increase. ALP activity showed no significant relationship with Cd treatment. MDA concentrations increased in the two tissues after Cd exposure, peaked on Day 3 in gills, and on Day 5 in hepatopancreas. These observations show that changes in the activities of antioxidant enzymes and ACP reflect the time course of oxidative stress in the clam caused by Cd, and could be used as potential biomarkers for ecotoxicological bioassays of heavy metals.展开更多

In this study, we cloned an 866 bp GST(phi) gene in Lemna minor and investigated its characteristics, expression and enzymatic activities under 75 lamol/L cadmium concentrations in comparison with other ROS scavengers. GST(phi) gene expression patterns were similar to those of other scavengers of ROS. This suggests that GST(phi) might be involved in responding to heavy metal (cadmium) stress and that its expression level could be used as a bio-indicator in monitoring cadmium pollution.展开更多

The germination percentage and the soluble protein content of the embryonic tissues were found to decrease with increasing of Cd concentration. There was elevation in superoxide dismutase (SOD) and decline in catalase (CAT) and peroxidase (POX) activities. The increasing of lipid peroxidation levels indicated the prevalence of oxidative stress in the tissues which was probably due to the alteration of antioxidative enzymes activities. The adding of ascorbate, along with CdCl2, has resulted in restoration of the Cd induced decline in CAT activity. Weakening in H2O2 detoxification system seems to be the principal reason behind Cd induced oxidative stress in germinating seeds. Thus, imposition of oxidative stress might be the consequence of cadmium stress and this finding may help in elucidating the mechanisms underlying cadmium mediated toxicity in germinating seeds.展开更多

The hydroquinone would be degraded completely after treated by Catalytic wet peroxide oxidation method with Co(II)/active carbon catalyst. After activate treatment, the active carbon was immerged in cobaltous nitrate solution, then put into a drying oven, Co(II) could be loaded on the micro-surface of carbon. Taking the static bed as the equipment, the absorption of active carbon and catalysis of Co(II) was used to reduce activation energy of hydroquinone. Thus hydroquinone could be drastically degraded and the effluent can be drained under the standard. Referring to Fenton reaction mechanism, experiment had been done to study the heterogeneous catalyzed oxidation mechanism of Co(II). The degradation rate of hydroquinone effluent could be achieved to 92% when treated in four columns at H2O2 concentration 10%, reaction temperature 40℃ , pH 5 and reaction time 2.5h.展开更多

The results showed that the linear effects of temperature were significant(P <0.01), the quadratic effects of temperature were significant( P <0.05), the linear effects of copper ion concentration were not significant(P >0.05), and the quadratic effects of copper ion concentration were significant(P <0.05). Additionally, the synergistic effects of temperature and copper ion concentration were not significant(P >0.05), and the effect of temperature was greater than that of copper ion concentration. A model equation of CAT enzyme activity in the digestive gland of C. ariakensis toward the two factors of interest was established, with R 2, Adj. R 2 and Pred. R 2 values as high as 0.943 7, 0.887 3 and 0.838 5, respectively. These findings suggested that the goodness of fit to experimental data and predictive capability of the model were satisfactory, and could be practically applied for prediction under the conditions of the study. Overall, the results suggest that the simultaneous variation of temperature and copper ion concentration alters the activity of the antioxidant enzyme CAT by modulating active oxygen species metabolism, which may be utilized as a biomarker to detect the effects of copper pollution.展开更多

Peroxidase enzyme is one of the most important enzymes in plant tissue which can bind to hydrogen peroxide and produce an activated complex that can react with a wide range of donor molecules. Therefore inactivation of the enzyme may increase the shelf life of raw and un-blanched frozen vegetables. In order to inactivate of the enzyme several physical and chemical treatments among them heating (blanching), lowering pH or aw or adding chemical additives can be used, however each of the above mentioned methods has a kind of shortcoming. Our results indicated that crude extract of red cabbage and cabbage has the highest and potato and lettuce has the lowest peroxidase activity respectively (P〈0.05). Although peroxidase resistance to essential oils varied among different vegetables but clove in all concentration had the best effects in decrease ofperoxidase activity (P〈0.05) whereas cumin and fennel were partially able to reduce the enzyme activity. (P〈0.05).展开更多

The pre-reduced catalysts show significantly higher activities than the pre-oxidized ones. Model studies were performed to better understand the oxidation state, reactivities and stabilities of partially oxidized Pd surfaces under CO oxidation reaction condi tions using an in situ infrared reflection absorption spectrometer (IRAS). Three O/Pd(100) model surfaces, chemisorbed oxygen covered surface, surface oxide and bulk-like surface oxide, were prepared and characterized by low-energy electron diffraction (LEED) and Auger electron spectroscopy (AES). The present work demonstrates that the oxidized palladium surface is less active for CO oxidation than the metallic surface, and is unstable under the reaction conditions with sufficient CO.展开更多

文摘We prepared the Fe3O4/g‐C3N4nanoparticles(NPs)through a simple electrostatic self‐assembly method with a3:97weight ratio to investigate their Fenton, photo‐Fenton and oxidative functionalities besides photocatalytic functionality. We observed an improvement of the Fenton and photo‐Fenton activities of the Fe3O4/g‐C3N4nanocomposites.This improvement was attributed to efficient charge transfer between Fe3O4and g‐C3N4at the heterojunctions, inhibition of electron‐hole recombination,a high surface area, and stabilization of Fe3O4against leaching by the hydrophobic g‐C3N4.The obtained NPs showed a higher degradation potential for rhodamine B(RhB)dye than those of Fe3O4and g‐C3N4.As compared to photocatalysis, the efficiency of RhB degradation in the Fenton and photo‐Fenton reactions was increased by20%and90%,respectively.Additionally,the horseradish peroxidase(HRP)activity of the prepared nanomaterials was studied with3,3,5,5‐tetramethylbenzidinedihydrochloride(TMB)as a substrate.Dopamine oxidation was also examined. Results indicate that Fe3O4/g‐C3N4nanocomposites offers more efficient degradation of RhB dye in a photo‐Fenton system compared with regular photocatalytic degradation, which requires a long time. Our study also confirmed that Fe3O4/g‐C3N4nanocomposites can be used as a potential material for mimicking HRP owing to its high affinity for TMB.These findings suggest good potential for applications in biosensing and as a catalyst in oxidation reactions.

文摘To assess the toxicity of heavy metal pollution to marine intertidal shellfish, enzymatic responses and lipid peroxidation were investigated in the clam Mactra vereformis exposed to cadmium under laboratory conditions. 文摘Germinating wheat (Triticum aestivum L.) seeds were exposed to CdCI2 (50, 100 and 200 μM) for 48 h and some aspects of oxidative metabolism was assessed in the embryonic tissues. Thus, imposition of oxidative stress might be the consequence of cadmium stress and this finding may help in elucidating the mechanisms underlying cadmium mediated toxicity in germinating seeds.

文摘Catalysts based on Co(II) supported on active carbon were prepared and loaded in static bed. The degradation rate of hydroquinone effluent could be achieved to 92% when treated in four columns at H2O2 concentration 10%, reaction temperature 40℃ , pH 5 and reaction time 2.5h.

文摘A central composite experimental design and response surface method were used to investigate the combined effects of water temperature(18–34℃) and copper ion concentration(0.1–1.5 mg/L) on the catalase(CAT) activity in the digestive gland of C rassostrea ariakensis. Overall, the results suggest that the simultaneous variation of temperature and copper ion concentration alters the activity of the antioxidant enzyme CAT by modulating active oxygen species metabolism, which may be utilized as a biomarker to detect the effects of copper pollution.
文摘The aim of this work was to study the effect of essential oils as natural antioxidant for inhibition of peroxidase enzyme activity. (P〈0.05).

文摘CO oxidation was investigated on various powder oxide supported Pd catalysts by temperature-programined reaction. The present work demonstrates that the oxidized palladium surface is less active for CO oxidation than the metallic surface, and is unstable under the reaction conditions with sufficient CO.
