Methods The chromatographic conditions were SuperdexTM peptide column, 0.01 mol*L-1 phosphate buffer solution as mobile phase, and flow rate of 1 mL·min-1. Five hundred microliters of lincomycin hydrochloride solution (3 g of lincomycin hydrochloride dissolved in 10 mL of mobile phase) was injected into the chromatograph and the eluted solution was collected between 6 min and 14.5 min (protein eluted from column within this period), and the residual content of total protein in the eluted solution was assayed using Bradford assay method. Results The average recovery was more than 90% for bovine serum albumin, the calibration equation for the range of 0-12 μg·mL-1 of protein was y=-0.002 4x2+0.064 2x+0.002 9, r2=0.999 9, RSD=0.1%-0.9%, and the LOD and LOQ were 3 and 10 ng·mL-1 of protein, respectively. Conclusion The novel method for determining the residual protein in ferment antibio-tics is simple, rapid, and precise.展开更多

A predominant approach to CF is rating prediction recommender algorithm, aiming ...Collaborative Filtering(CF) is a leading approach to build recommender systems which has gained considerable development and popularity. A predominant approach to CF is rating prediction recommender algorithm, aiming to predict a user's rating for those items which were not rated yet by the user. However, with the increasing number of items and users, thedata is sparse.It is difficult to detectlatent closely relation among the items or users for predicting the user behaviors. In this paper,we enhance the rating prediction approach leading to substantial improvement of prediction accuracy by categorizing according to the genres of movies. Then the probabilities that users are interested in the genres are computed to integrate the prediction of each genre cluster. A novel probabilistic approach based on the sentiment analysis of the user reviews is also proposed to give intuitional explanations of why an item is recommended.To test the novel recommendation approach, a new corpus of user reviews on movies obtained from the Internet Movies Database(IMDB) has been generated. Experimental results show that the proposed framework is effective and achieves a better prediction performance.展开更多

Filtration experiments were performed at 60 ℃, 500 rpm and dif...The application of single nanofiltration (NF) and sequential filtration of microfiltration (MF) and NF for the concentration of vinasse were studied. Filtration experiments were performed at 60 ℃, 500 rpm and different transmembrane pressures using four commercial membranes: MF, PVDF 0.2 μm; NF, PES 300, 500 and 1000 Da. The process effectiveness was evaluated through the permeate flux and the total solids (TS) concentration in the retentate. Because the retention of TS and permeate fluxes were very similar for MF, PVDF-0.2 membrane at 0.10 MPa was selected because of its lower power consumption. The best conditions tested for NF were the following: PES-1000 at 3.50 MPa with a flux of 183.2 kg/m^2 h at a volume reduction factor (VRF) of 2, under which 68% of solids were retained. For the MF + NF, a PES-1000 at 3.50 MPa was selected because a higher permeate flux was achieved at a VRF of 4, and PES-500 at 3.50 MPa because a higher retention of TS was achieved compared to the other experiments. A single NF resulted in a retentate concentration that was almost twice as high as the hybrid MF and NF process, allowing reduced costs in its transportation from distilleries to distant areas.展开更多

By feeding whole...Membrane microfiltration fermentation (MMF) with cell recycling was successfully applied to the production of glucose oxidase (GOD). A plate microfiltration module was found suitable for such purpose. By feeding whole medium in MMF, the productivity of GOD was much higher than that by feeding glucose alone. With increasing dilution rate the enzyme productivity increased and average enzyme activity decreased. The enzyme productivity of MMF under D = 0.12h-1 and 0.20h-1 were 3871 and 3945U·h-1 respectively, which was about 3 times as that of batch fermentation (BF) and the average enzyme activity was still as high as STU·mL-1 under D = 0.12h-1. The relative efficiency of MMF applied to low yield strain was higher than that applied to high yield strain.展开更多
