Methods: A high-performance liquid chromatography...Objective: To investigate the relationship between tissue distributions of modified Wuzi Yanzong prescription(加味五子衍宗方, MWP) in rats and meridian tropism theory. Methods: A high-performance liquid chromatography with Fourier transform-mass spectrometry(HPLC-FT) method was used to identify the metabolites of MWP in different tissues of rats after continued oral administration of MWP for 7 days. The relationship between MWP and meridian tropism theory was studied according to the tissue distributions of the metabolites of MWP in rats and the relevant literature. Results: Nineteen metabolites, mainly flavanoid compounds, were detected in the different rat tissues and classified to each herb in MWP. Further, it was able to establish that the tissue distributions of the metabolites of MWP were consistent with the descriptions of meridian tropism of MWP available in literature, this result might be useful in clarifying the mechanism of MWP on meridian tropism. In the long run, these data might provide scientific evidence of the meridian tropism theory to further promote the reasonable, effective utilization, and modernization of Chinese medicine. Conclusion: The tissue distributions of MWP in vivo were consistent with the descriptions of meridian tropism of MWP.展开更多

(RPL), a widely used medicinal plant, were evaluated. Results demonstrated that RPL extract possessed pronounced hepatoprotective effects against carbon tetrachloride (CCl4)-induced hepatic injury in mice, which was at least partially attributed to its strong antioxidant capacity. Treatment with RPL extract markedly attenuated the increases in serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels caused by CCl4 intoxication. It also significantly prevented the decrease in superoxide dismutase (SOD) activity and the increase in malondialdehyde (MDA) content of liver tissue. Meanwhile, histopathological changes of hepatic damage were also remarkably ameliorated. Phytochemical analysis based on high-performance liquid chromatography/tandem mass spectrometry (HPLC-MS/MS) revealed the presence of various phenolic compounds, including caffeic acid conjugates, ellagic acid glycosides, and flavonol glycosides, which might be responsible for the hepatoprotective and antioxidant activities of RPL.展开更多

A method was established to determine the activity of hexokinase by reverse-phase high-performance liquid chromatography(RP-HPLC).The activity of hexokinase was determined by detecting the amount of ADP in the reactio...A method was established to determine the activity of hexokinase by reverse-phase high-performance liquid chromatography(RP-HPLC).The activity of hexokinase was determined by detecting the amount of ADP in the reaction that converts glucose into glucose-6-phosphate.The separation degree of ATP and ADP was as good as 3.46(separation degree>1.5).In the range of 0.01-0.40 mmol/L,there was a good linear relationship between concentration of ADP and its peak areas(the correlation coefficient was 0.999).The relative standard deviation(RSD) of intraday was 1.43%-1.46%,that of interday was 2.12%-2.15%.At 30 °C(optimal temperature) and pH7.0(optimal pH),the recovery rate of hexokinase was 95.3%-97.6%.The results showed that the method had good precision and high recovery rate,and the enzyme activity was determined through only one-step reaction.To some extent,this method can reduce the cost.展开更多
