In contrast, the Health at Every Size (HAES) paradigm fosters improved health behaviors for people of all sizes by emphasizing natural diversity of body type and attention to social, emotional, and spiritual factors in addition to physical ones. This study examined introduction of the HAES paradigm to student teachers using different communication models while moving across the learning domains of Bloom’s Taxonomy. The qualitative research tested a face-to-face verbal communication model and a Computer-Mediated Communication (CMC) module and its blog. It was also based upon reflection diaries recorded by participants using both models. Participants included Druze, Jewish, and Bedouin students at a teacher training program in northern Israel. The student teachers succeeded in developing the HAES concepts further while demonstrating the higher domain levels across Bloom’s Taxonomy. These higher levels emerged when engaging with theoretical concepts and practical dilemmas relating to the HAES concepts of health promotion, body image, well-being, and self-worth. Challenging the appreciation of HAES using different communication models can be translated into diverse contexts of Health Education Practices.展开更多

The basic educational model proposed here is my seven-step process model of rehabilitation practice. Knowledge consists of four aspects: 1) clinical, 2) psychological, 3) environmental, and 4) disability;skills consist of two steps: 5) identifying intervention points and 6) setting feasible goals;and attitudes 7) of communicating and sharing policies and paths with patients, families, and other professionals. This constitutes the process of rehabilitation practice, and a framework that integrates the three elements is developed here. This study focuses on integrating knowledge, skills, and attitudes into what Bloom described as “the integration of instruction and assessment” so that learners and instructors can reconcile them. Therefore, a typology that explains each other for advancing and deepening individual skills is adopted. In Bloom’s original taxonomy of educational goals, the cognitive domain has five layers in the pyramid of knowledge;the psychomotor domain of Simpson’s has seven layers, and Bloom’s affective domain is represented by five in another pyramid. In addition, the above seven layers of the process model and the seven layers of the skill level of the Dreyfus model were brought together. The integration of the above five typologies becomes a useful educational evaluation model when the relationships are clarified.展开更多

Recently, algicidal bacteria have attracted wide attention as possible agents for inhibiting algal water blooms. In this study, one strain of algicidal bacterium B5 was isolated from activated sludge. On the basis of analysis of its physiological characteristics and 16S rDNA gene sequence, it was identified as Bacillusfusiformis. Its algaelysing characteristics on Microcystis aeruginosa, Chlorella and Scenedesmus were tested. The results showed that: (1) the algicidal bacterium B5 is a Gram-negative bacterium. The 16S rDNA nucleotide sequence homology of strain B5 with 2 strains of B. fusiformis reached 99.86%, so B5 was identified as B. fusiformis; (2) the algal-lysing effects of the algicidal bacterium B5 on M. aeruginosa, Chlorella and Scenedesrnus were pronounced. The initial bacterial and algal cell densities strongly influence the removal rates of chlorophyll-a. The greater the initial bacterial cell density, the faster the degradation of chlorophyll-a. The greater the initial algal cell density, the slower the degradation of chlorophyll-a. When the bacterial cell density was 3.6 × 10^7 cells/ml, nearly 90% of chlorophyll-a was removed. When the chlorophyll-a concentration was less than 550 μg/L, about 70% was removed; (3) the strain B5 lysed algae by secreting metabolites and these metabolites could bear heat treatment.展开更多

To solve this problem, we developed a real-time PCR assay (SYBR Green I) to identify planulae in both cultured and natural seawater samples. Species-specific primers targeting Aurelia sp.1 mitochondrial 16S rDNA (mr 16S rDNA) regions were designed. Using a calibration curve constructed with plasmids containing the Aurelia sp. 1 mt 16S rDNA fragment and a standard curve for planulae, the absolute number of mt 16S rDNA copies per planula was determined and from that the total number ofplanulae per sample was estimated. For the field samples, a 100-fold dilution of the sample DNA combined with a final concentration of 0.2 μg/μL BSA in the PCR reaction mixture was used to remove real- time PCR inhibitors. Samples collected in Jiaozhou Bay from July to September 2012 were subsequently analyzed using this assay. Peak Aurelia sp.1 planula abundance occurred in July 2012 at stations near Hongdao Island and Qingdao offshore; abundances were very low in August and September. The real-time PCR assay (SYBR Green I) developed here negates the need for traditional microscopic identification, which is laborious and time-consuming, and can detect and quantify jellyfish planulae in field plankton samples rapidly and specifically.展开更多

文摘Over the past decades, jellyfish occurred increasingly and abundantly in coastal areas worldwide.Usually, biomass of jellyfish, especially when they bloom, can be determined by visual counting. However, tiny individuals of jellyfish(e.g.,planulae,polyps,and ephyrae)are difficult to detect in the field. In this study,species-specific quantitative real-time PCR assays(qPCR)(SYBR Green I)targeting the mitochondrial 16S rDNA(mt-16S rDNA)of jellyfish were developed and were used to estimate the distribution and seasonal fluctuations of four jellyfish species(Nemopilema nomurai,Cyanea nozakii,Rhopilema esculentum, and Aurelia coerulea)in Jiaozhou Bay(JZB),China in 2013.The mt-16S rDNA of A.coerulea and N.nomurai was detected in most of the survey months and it peaked in July(1.03×10^(4) copies/L)and September(1.08×10^(6) copies/L),respectively.The mt-16S rDNA of C.nozakii occurred from August to October only with monthly mean values of 7.18-46.17 copies/L and was mainly located from the middle part to the outer part of the bay.The mt-16S rDNA of R.esculentum was the least abundant among the four species and was detected in only one sample(B2 station in March),with a value of 88.49 copies/L.The Spearman correlation test revealed that phytoplankton biomass was significantly and positively correlated with the mt-16S rDNA abundance of A.coerulea(R=0.37,P<0.01)and negatively with the mt-16S rDNA of N.nomurai(R=-0.36,P<0.01).The qPCR assay will enable the identification and quantification of jellyfish species in their whole life history and can be used as an approach in combination of the traditional jellyfish survey.

文摘The complicated life cycle ofAurelia spp., comprising benthic asexually-reproducing polyps and sexually-reproducing medusae, makes it hard for researchers to identify and track them, especially for early stage individuals, such as planulae. The real-time PCR assay (SYBR Green I) developed here negates the need for traditional microscopic identification, which is laborious and time-consuming, and can detect and quantify jellyfish planulae in field plankton samples rapidly and specifically.
