BACKGROUND Antinuclear antibodies(ANAs)are crucial in diagnosing autoimmune diseases,mainly systemic lupus erythematosus(SLE).This study aimed to compare the performance of chemiluminescence assay(CLIA)and line immuno...BACKGROUND Antinuclear antibodies(ANAs)are crucial in diagnosing autoimmune diseases,mainly systemic lupus erythematosus(SLE).This study aimed to compare the performance of chemiluminescence assay(CLIA)and line immunoassay(LIA)in detecting ANAs in patients with autoimmune diseases,evaluate their diagnostic accuracy for SLE,and develop a novel diagnostic model using CLIA-detected antibodies for SLE.Specimens from patients with autoimmune diseases and physical examination specimens were collected to parallel detect specific antibodies.Individual antibodies'diagnostic performance and a model combining multiple antibodies were assessed.The findings provide valuable insights into improving the diagnosis of SLE through innovative approaches.AIM To compare the performance of CLIA and LIA in detecting ANAs in patients with autoimmune diseases,assess their accuracy for SLE,and develop a novel diagnostic model using CLIA-detected antibodies for SLE.METHODS Specimens have been obtained from 270 patients with clinically diagnosed autoimmune disorders,as well as 130 physical examination specimens.After that,parallel detection of anti-double-stranded DNA(dsDNA)antibody,anti-histone(Histone)antibody,anti-nucleosome(Nuc)antibody,anti-Smith(Sm)antibody,anti-ribosomal P protein(Rib-P)antibody,anti-sicca syndrome A(Ro60)antibody,anti-sicca syndrome A(Ro52)antibody,anti-sicca syndrome(SSB)antibody,anticentromere protein B(Cenp-B)antibody,anti-DNA topoisomerase 1(Scl-70)antibody,anti-histidyl tRNA synthetase(Jo-1)antibody,and anti-mitochondrial M2(AMA-M2)antibody was performed using CLIA and LIA.The detection rates,compliance rates,and diagnostic performance for SLE were compared between the two methodologies,followed by developing a novel diagnostic model for SLE.RESULTS CLIA and LIA exhibited essentially comparable detection rates for anti-dsDNA antibody,anti-Histone antibody,anti-Nuc antibody,anti-Sm antibody,anti-Rib-P antibody,anti-Ro60 antibody,anti-Ro52 antibody,anti-SSB antibody,anti-Cenp-B antibody,anti-DNAScl-70 antibody,anti-Jo-1 antibody and anti-AMA-M2 antibody(P>0.05).The two methods displayed identical results for the detection of anti-dsDNA antibody,anti-Histone antibody,anti-Nuc antibody,anti-Sm antibody,anti-Ro60 antibody,anti-Ro52 antibody,anti-SSB antibody,anti-Cenp-B antibody,anti-Scl-70 antibody,and anti-AMA-M2 antibody(Kappa>0.7,P<0.05),but showed a moderate agreement for the detection of anti-Rib-P antibody and anti-Jo-1 antibody(Kappa=0.671 and 0.665;P<0.05).In addition,the diagnostic performance of these antibodies detected by both methods was similar for SLE.The diagnostic model's area under the curve values,sensitivity,and specificity,including an anti-dsDNA antibody and an anti-Ro60 antibody detected by CLIA,were 0.997,0.962,and 0.978,respectively.These values were higher than the diagnostic performance of individual antibodies.CONCLUSION CLIA and LIA demonstrated excellent overall consistency in detecting ANA profiles.A diagnostic model based on CLIA-detected antibodies can successfully contribute to developing a novel technique for detecting SLE.展开更多

On the basis of this, a novel method for the determination of trace amount of molybdenum has been established. The emission intensity was linear with molybdenum concentration in the range of 0.1-1000ng/mL; the detection limit was 0.02 ng/mL molybdenum, the relative standard deviation was less than 2% for the determination of 0.1 ng/mL molybdenum (n=11). Methods: In this crosssectional study, 276 blood samples were collec...Objective: To identify serodiagnosis and quantification of Toxoplasma gondii(T. gondii) infection among pregnant women in Salmas, northwest of Iran. Methods: In this crosssectional study, 276 blood samples were collected from pregnant women referred to the health care centers in Salmas city. The demographic variables were also recorded. Titers of antiToxoplasma IgM and IgG antibodies(Ab) were determined using the chemiluminescence immunoassay. Quantitative real-time PCR targeting the T. gondii repeated element gene was also performed on the blood sample. Results: Out of all, 19.92%(55/276) and 2.17%(6/276) patients were seropositive for anti-Toxoplasma IgG and IgM Ab, respectively. Moreover, the presence of T. gondii DNA was observed in 12.31%(34/276) blood samples. A significant relationship was observed between the IgG Ab seropositivity and contact with the cat as a risk factor(P=0.022). Conclusions: The seroprevalence rate of T. gondii infection in pregnant women is relatively low. Consequently, the seronegative pregnant women are at risk, and a considerable rate of positive blood samples for the presence of parasite's DNA should not be ignored. Besides, quantitative real-time PCR could be considered as an accurate method for diagnosis of acute toxoplasmosis especially when the precise results are of the most importance in pregnancy. Limiting contact with cats is also suggested for pregnant women.展开更多

In sulfuric a...A new chemiluminescence (CL) method for the determination of tetracycline and oxytetracycline is developed, hased on the CL reaction of tetracycline and oxytetracycline with Ru(bipy)32+ and Ce(IV). In sulfuric acid medium, the CL emission is generated upon continuous oxidation of Ru(bipy)32+ by cerium (IV ). The emission intensity is greatly enhanced when tetracycline and oxytetracycline are introduced into the reaction system after acid degradation. Under the optimum conditions, the calibration curves are linear over the range of 8. 0 X 10-8-4. 0 X 10-6 mol/L for tetracycline and of 2. 0 X 10-74. 0 X 10-5 mol/L for oxytetracycline, with the detection limits are 4. 2 X 10-8 mol/L for tetracycline and 1. 5 X 10-7 mol/L for oxytetracycline, respectively. The proposed method was used for the determination of tetracycline and oxytetracycline in pharmaceutical formulations with good results.展开更多

It was applicable to the determination of penicillamine in the concentration range of 2×10-6~5×10-4 mol·L-1. The method was successfully applied to the determination of penicillamine in tablets. The light emission from the reaction of Cu(Ⅱ) oxi diz ed ascorbic acid and luminol was detected by a modified luminometer. The detecti on limit was 3.6×10 -6 mol/L for ascorbic acid at a S/N ratio of 3, and the linea r calibration range was 2×10 -4\| 2×10 -3 mol/L. The relative standard deviation for 5 replicate injections of 1×10 -3 mol/L ascorbic acid was calculated as 4.3 % . In the present investigation,measurement of leukocyte chemiluminescence (CL) wasundertaken using micro-whole blood assay in 76 patients with coronary heart diseases (CHDs).The findings showed that there was a signific...In the present investigation,measurement of leukocyte chemiluminescence (CL) wasundertaken using micro-whole blood assay in 76 patients with coronary heart diseases (CHDs).The findings showed that there was a significant increase in leukocyte CL in patients with un-stable angina pectoris (UAP) and acute myocardial infarction (AM1) (P【0.01) and the in-crease was greater in AMI than that in UAP (P【0,01).It was found that leukocyte CL inAMI was correlated with the extent of the infarct and increased with the size of the infarct.Leukocyte CL in AMI patients who had received fibrinolytic treatment was markedly greaterthan that before treatment (P【0.05),suggesting that leukocytes may play an important rolein the pathogenesis of CHDs and may be involved in myocardial ischemia-reperfusion induced in-jury.The findings of the present study demonstrate that measurement of leukocyte CL may beof benefits for prognosis and treatment of CHDs.展开更多

Spermatozoa were the ...The presence of high reactive oxygen species (ROS) levels in semen is a major factor involved in the decline of male fertility. In seminal plasma, ROS are mainly produced by activated leucocytes. Spermatozoa were the first cell type reported to show a potential susceptibility to oxidative damage. The aim of our study was to evaluate the impact of leucocytospermia on basal and FMLP (Formyl-Methionyl-Leucyl-Phenylalanine) induced oxidative status in semen of infertile men. We also analyzed the correlations of the spermatic parameters with amounts of ROS in semen. Our study included 50 semen samples of infertile men. Sperm analysis was performed using WHO standardized method. Seminal leucocytes were quantified using peroxidase technique. The measurement of ROS levels in semen was made by chemiluminescence assay. We measure respectively ROS amounts in neat semen and in washed sperm cells suspension from the same ejaculate. We also applied the test of provocation of leucocytes by FMLP on neat and washed samples to assess the spermatic oxidative status after leucocyte stimulation. Our results showed significant correlations between ROS levels in neat semen and many sperm parameters: motility, sperm concentration, leucocytes concentration and the rate of sperm cytoplasmic droplets. The studied samples were divided into 2 groups: (G1) composed of 36 samples without leucocytospermia and (G2) composed of 14 leucospermic samples. ROS levels were significantly lower in G1 than in G2 (p = 0.002). ROS production was significantly increased after application of FMLP in washed leucospermic samples (p = 0.001). The measurement of ROS in neat semen is a considerable contribution to explore the impairment of semen quality in infertile men. ROS levels in washed semen reflect the oxidative status generated by sperm preparation techniques used in assisted reproductive procedures. Levels of ROS are highly influenced by the presence of leucocytes and associated with decreased seminal parameters.展开更多

Glucose oxidase(GOD) and horseradish peroxidase(HRP) were co-immobilized by microencapsulation in a sol-gel film derived from tetraethyl orthosilicate(TEOS). The calibration plots for glucose were established by the optical fiber glucose sensor fabricated by attaching the bienzyme silica gel onto the glass window of the fiber bundle. The linear range was 0 2-2 mmol/L and the detection limit was approximately 0 12 mmol/L. The relative standard deviation was 5.3% ( n =6). The proposed biosensor was applied to glucose assay in ofloxacin injection successfully.展开更多
A novel flow-injection chemiluminescence(FI-CL) method free of CL reagent was developed for the determination of captopril based on its enhancing effect on the CL derived from diperiodatoargentate(Ⅲ)-sulfuric acid sy...A novel flow-injection chemiluminescence(FI-CL) method free of CL reagent was developed for the determination of captopril based on its enhancing effect on the CL derived from diperiodatoargentate(Ⅲ)-sulfuric acid system. Compared with the conventional CL system, the CL system based on trivalent silver was characterized of good selectivity for the absence of CL reagent. The CL mechanism was discussed through CL spectra and UV–vis absorption spectra. The conditions of the FI-CL system were investigated and optimized.Under the optimal conditions, the relative CL intensity was linear with the captopril concentration in the range of 0.3–15.0 μg/m L. The detection limit for captopril was 0.05 μg/m L, and the relative standard deviation(n=11) was 2.0% for 5.0 μg/m L captopril. The proposed method was applied to the analysis of captopril in tablet and human urine with the recoveries of 83.1%–112.5%, and the relative standard deviations of 0.5%–4.4%. The results obtained by the proposed method agreed well with those obtained from HPLC method. The proposed method is fast, convenient, and cost-effective for the determination of captopril in medicine and biological samples.展开更多

文摘A chemiluminescence enzyme immunoassay based on magnetic microparticles (MmPs-CLEIA) was developed to evaluate serum a-fetoprotein (AFP) in parallel with traditional colorimetric enzyme-linked immunosorbent assay (ELISA).A systematic comparison between the MmPs-CLEIA and colorimetric ELISA concluded that the MPs-CLEIA exhibited fewer dosages of immunoreagents,less total assay time,and better linearity,recovery,precision,sensitivity and validity.AFP was detected in forty human serum samples by the proposed MPs-CLEIA and ELISA,and the results were compared with commercial electrochemiluminescence immunoassay (ECLIA) kit.The correlation coefficient between MPs-CLEIA and ELISA was obtained with R 2 0.6703;however,the correlation between MPs-CLEIA and ECLIA (R 2 0.9582) was obviously better than that between colorimetric ELISA and ECLIA (R 2 0.6866).

文摘Objective: To identify serodiagnosis and quantification of Toxoplasma gondii(T. gondii) infection among pregnant women in Salmas, northwest of Iran. Methods: In this crosssectional study, 276 blood samples were collected from pregnant women referred to the health care centers in Salmas city. The demographic variables were also recorded. Titers of antiToxoplasma IgM and IgG antibodies(Ab) were determined using the chemiluminescence immunoassay. Quantitative real-time PCR targeting the T. gondii repeated element gene was also performed on the blood sample. Results: Out of all, 19.92%(55/276) and 2.17%(6/276) patients were seropositive for anti-Toxoplasma IgG and IgM Ab, respectively. Moreover, the presence of T. gondii DNA was observed in 12.31%(34/276) blood samples. A significant relationship was observed between the IgG Ab seropositivity and contact with the cat as a risk factor(P=0.022). Conclusions: The seroprevalence rate of T. gondii infection in pregnant women is relatively low. Consequently, the seronegative pregnant women are at risk, and a considerable rate of positive blood samples for the presence of parasite's DNA should not be ignored. Besides, quantitative real-time PCR could be considered as an accurate method for diagnosis of acute toxoplasmosis especially when the precise results are of the most importance in pregnancy. Limiting contact with cats is also suggested for pregnant women.

文摘Based on the fact that some metal ions can catalyze the chemiluminescence(CL)reaction of luminol with K<sub>3</sub>Fe(CN)<sub>6</sub>,a novel capillary electrophoresis CL method was developed for the determination of Co(Ⅱ)and Cu(Ⅱ).The separation was carried out with a 10 mmol/L sodium acetate solution containing 0.8 mmol/L luminol and 2.0 mmol/Lα-HIBA(adjusted to pH 4.8 by HAc solution).The post-capillary reagent was 2.0 mmol/L K<sub>3</sub>Fe(CN)<sub>6</sub> which was adjusted to pH 13.0 by NaOH solution.Under the optimum conditions,the detection limits(S/N=3)for Co(Ⅱ)and Cu(Ⅱ)were 7.5×10<sup>-11</sup>mol/L and 7.5×10<sup>-9</sup>mol/L,with the linear range of 7.5×10<sup>-9</sup>mol/L to 1.0×10<sup>-6</sup>mol/L and 7.5×10<sup>-8</sup>mol/L to 5.0×10<sup>-5</sup>mol/L, respectively.

文摘A new chemiluminescence (CL) method for the determination of tetracycline and oxytetracycline is developed, hased on the CL reaction of tetracycline and oxytetracycline with Ru(bipy)32+ and Ce(IV). In sulfuric acid medium, the CL emission is generated upon continuous oxidation of Ru(bipy)32+ by cerium (IV ). The emission intensity is greatly enhanced when tetracycline and oxytetracycline are introduced into the reaction system after acid degradation. Under the optimum conditions, the calibration curves are linear over the range of 8. 0 X 10-8-4. 0 X 10-6 mol/L for tetracycline and of 2. 0 X 10-74. 0 X 10-5 mol/L for oxytetracycline, with the detection limits are 4. 2 X 10-8 mol/L for tetracycline and 1. 5 X 10-7 mol/L for oxytetracycline, respectively. The proposed method was used for the determination of tetracycline and oxytetracycline in pharmaceutical formulations with good results.

文摘A new flow-injection procedure has been developed for the determination of penicillamine based on the enhancement of the chemiluminescence(CL) reaction of the penicillamine-cerium(Ⅳ)-hydrocortisone system. The method was simple, selective and sensitive with a detection limit of 6.2×10-5 mol·L-1(S/N=3). It was applicable to the determination of penicillamine in the concentration range of 2×10-6~5×10-4 mol·L-1. The method was successfully applied to the determination of penicillamine in tablets. The relative standard deviation (RSD) was 1.42% (n=7) and the recovery was 97%~103% (n=4).
文摘The chemiluminescence reactions between lucigenin and reductants such as M o(III),V(II ), U(III), W(III), C r(II ), T i(III) and Fe(II ), which were produced on-line by passing M o(VI),V(V ),U(VI). W(VI), C r(VI,III),T i(IV) and Fe(III) through a micro Jones column, are studied in detail. Results show that the reactions can be used directly for the determination of these substances. The mechanism of the reactions is also investigated.

文摘The chemiluminescence (CL) of luminol-Cu(Ⅱ) was applied to HPLC determination o f ascorbic acid, which was separated by a C 18 reverse-phase column with a mobile phase of 0.25 mol/L HAc. The eluted ascorbic acid was mixed with 0.3 mmol/L l um inol and 0.05 mol/L CuSO 4. The light emission from the reaction of Cu(Ⅱ) oxi diz ed ascorbic acid and luminol was detected by a modified luminometer. The detecti on limit was 3.6×10 -6 mol/L for ascorbic acid at a S/N ratio of 3, and the linea r calibration range was 2×10 -4\| 2×10 -3 mol/L. The relative standard deviation for 5 replicate injections of 1×10 -3 mol/L ascorbic acid was calculated as 4.3 % . The method was successfully applied to determination of ascorbic acid in juice beverage.
文摘In the present investigation,measurement of leukocyte chemiluminescence (CL) wasundertaken using micro-whole blood assay in 76 patients with coronary heart diseases (CHDs).The findings showed that there was a significant increase in leukocyte CL in patients with un-stable angina pectoris (UAP) and acute myocardial infarction (AM1) (P【0.01) and the in-crease was greater in AMI than that in UAP (P【0,01).It was found that leukocyte CL inAMI was correlated with the extent of the infarct and increased with the size of the infarct.Leukocyte CL in AMI patients who had received fibrinolytic treatment was markedly greaterthan that before treatment (P【0.05),suggesting that leukocytes may play an important rolein the pathogenesis of CHDs and may be involved in myocardial ischemia-reperfusion induced in-jury.The findings of the present study demonstrate that measurement of leukocyte CL may beof benefits for prognosis and treatment of CHDs.
文摘The presence of high reactive oxygen species (ROS) levels in semen is a major factor involved in the decline of male fertility. In seminal plasma, ROS are mainly produced by activated leucocytes. Spermatozoa were the first cell type reported to show a potential susceptibility to oxidative damage. The aim of our study was to evaluate the impact of leucocytospermia on basal and FMLP (Formyl-Methionyl-Leucyl-Phenylalanine) induced oxidative status in semen of infertile men. We also analyzed the correlations of the spermatic parameters with amounts of ROS in semen. Our study included 50 semen samples of infertile men. Sperm analysis was performed using WHO standardized method. Seminal leucocytes were quantified using peroxidase technique. The measurement of ROS levels in semen was made by chemiluminescence assay. We measure respectively ROS amounts in neat semen and in washed sperm cells suspension from the same ejaculate. We also applied the test of provocation of leucocytes by FMLP on neat and washed samples to assess the spermatic oxidative status after leucocyte stimulation. Our results showed significant correlations between ROS levels in neat semen and many sperm parameters: motility, sperm concentration, leucocytes concentration and the rate of sperm cytoplasmic droplets. The studied samples were divided into 2 groups: (G1) composed of 36 samples without leucocytospermia and (G2) composed of 14 leucospermic samples. ROS levels were significantly lower in G1 than in G2 (p = 0.002). ROS production was significantly increased after application of FMLP in washed leucospermic samples (p = 0.001). The measurement of ROS in neat semen is a considerable contribution to explore the impairment of semen quality in infertile men. ROS levels in washed semen reflect the oxidative status generated by sperm preparation techniques used in assisted reproductive procedures. Levels of ROS are highly influenced by the presence of leucocytes and associated with decreased seminal parameters.
文摘Combined with flow-injection (Fl) technology, a simple chemiluminescence(CL)method was developed for hydrazine determination in this paper. It was found that hydrazine could greatly decrease the strong CL signal produced by the reaction between luminol and hexacyanoferrate (Ⅲ) in alkaline medium. The decreased CL intensity was linear with hydrazine concentration in the range of 5.0× 10-9 gmL-1 to 4.0× 10-5 gmL1, and the limit of detection was 2.0× 10-9 g mL-1 (3σ) with a relative standard deviation of 2.4~4.1% (n=5).

文摘The chemiluminescence reaction between luminol and reductant was investigated by a flow-injection system. The results show that this reaction can be used for the determination of the reductant which is active to chemiluminescence in alkaline lumminol, and of the substance which can, in appropriate manner, be converted into the active reductant. The mechanism of the chemiluminescence reaction may be that the reductant reduces dissolved oxygen to O2 , then O2 reacts with alkaline luminol to generate chemiluminescence.
文摘An optical fiber bienzyme sensor based on the luminol chemiluminescent reaction was developed and demonstrated to be sensitive to glucose. Glucose oxidase(GOD) and horseradish peroxidase(HRP) were co-immobilized by microencapsulation in a sol-gel film derived from tetraethyl orthosilicate(TEOS). The calibration plots for glucose were established by the optical fiber glucose sensor fabricated by attaching the bienzyme silica gel onto the glass window of the fiber bundle. The linear range was 0 2-2 mmol/L and the detection limit was approximately 0 12 mmol/L. The relative standard deviation was 5.3% ( n =6). The proposed biosensor was applied to glucose assay in ofloxacin injection successfully.

文摘A novel flow-injection chemiluminescence(FI-CL) method free of CL reagent was developed for the determination of captopril based on its enhancing effect on the CL derived from diperiodatoargentate(Ⅲ)-sulfuric acid system. Compared with the conventional CL system, the CL system based on trivalent silver was characterized of good selectivity for the absence of CL reagent. The CL mechanism was discussed through CL spectra and UV–vis absorption spectra. The conditions of the FI-CL system were investigated and optimized.Under the optimal conditions, the relative CL intensity was linear with the captopril concentration in the range of 0.3–15.0 μg/m L. The detection limit for captopril was 0.05 μg/m L, and the relative standard deviation(n=11) was 2.0% for 5.0 μg/m L captopril. The proposed method was applied to the analysis of captopril in tablet and human urine with the recoveries of 83.1%–112.5%, and the relative standard deviations of 0.5%–4.4%. The results obtained by the proposed method agreed well with those obtained from HPLC method. The proposed method is fast, convenient, and cost-effective for the determination of captopril in medicine and biological samples.
